Cheonghunhwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CTG was CHT adding Aurantii FructusㆍGastrodae Rhizoma. CTG was significantly increased regional cerebral blood f1ow(rCBF) in a dose-dependent, and CTG was decreased mean arterial blood pressure(MABP) compared with normal MABP(100%) in a low dosage, but was increased MABP in a dose-dependent, was aliked with normal MABP in a high-dosage. Therefore, purpose of this Study was to investigate experimental mechanism of CHTAG on the cerebral hemodynamics(rCBF, MABP) in rats. The changes of rCBF and MABP were determinated by Laser-Doppler Flowmetry(LDF). The results were as follows ; Pretreatment with indomethacin(3㎎/㎏, i.v.) was significantly inhibited CTG induced increase of rCBF and pretreated with propranolol(3㎎/㎏, i.v.) was inhibited CTG induced increase of rCBF, but pretreatment with methylene blue(10㎍/㎏, i.v) was increased CTG induced increase of rCBF. Pretreatment with indomethacin was decreased CTG induced increase of MABP, but pretreatment with propranolol and methylene blue were increased CTG induced increase of MABP. This results suggest that the mechanism of CTG is mediated by cyclooxygenase.

Jaeumgenby-tang(JGT) have been used in oriental medicine for many centries as a a therapeutic agent of vertigo caused by deficiency of qi and blood. The effects of JGT on the regional cerebral blood flow(rCBF), mean arterial blood pressure(MABP) and cardiac muscle contractile force(CMF) is not known. The purpose of this Study was to investigate effects of JGT on the rCBF, MABP, CMF and mechanism of JGT induced changed rCBF, MABP, CMF. The changes of rCBF, MABP and CMF were determinated by Laser-Doppler Flowmetry(LDF). The results were as follows; JGT extract was increased rCBF, MABP and CMF in a dose-dependent, specially JGT extract was significantly increased rCBF and MABP. Pretreatment with propranolol was significantly inhibited JGT induced increase of rCBF but pretreatment with indomethacin and methylene blue were accelerated JGT induced increase of rCBF. Pretreatment with propranolol and indomethacin were inhibited JGT induced increase of MABP, but pretreatment with methylene blue was accelerated JGT induced increase of MABP. Pretreatment with propranolol was significantly inhibited JGT induced increase of CMF but pretreatment with indomethacin and methylene blue were accelerated JGT induced increase of CMF. This results suggest that JGT increased rCBF by increasing MABP and CMF and the action of JGT is mediated by adrenergic β-receptor.

Objectives: This study was designed to investigate the effect of four pharmacopuncture drugs (scolopendrid, Calculus Bovis-Fel Uris-Moschus (BUM), bee venom 25%, and sweet bee venom 10%) on the cerebral hemodynamics, including changes in the regional cerebral blood flow (rCBF) and in the mean arterial blood pressure (MABP). Methods: The changes in the rCBF and the MABP were determined by using a laser-Doppler flowmeter and a pressure transducer, respectively. Results: Scolopendrid (0.3 ml, 1 ml/kg) caused no significant changes in the rCBF and the MABP, whereas BUM (0.3 ml, 1 ml/kg) decreased the rCBF and the MABP, bee venom 25% (0.3 ml, 1 ml/kg) increased the rCBF and lowered the MABP, and sweet bee venom 10% (0.3 ml, 1 ml/kg) increased the rCBF and had no significant effect on the MABP. Conclusions: The rCBF and the MABP were influenced differently by the administration of various pharmacopunctures. Further studies are needed to elucidate the underlying mechanism.
Objective: The purpose of this study is to examine influences by the order, Reduction and Reinforcement in Acupuncture on cerebral hemodynamics [regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP)] in normal rats. Methods: This experiments was to to investigate eath other changes of rCBF and MABP at Xingjian(LR2)(1st) Shaofu(HT8)(2nd) Reduction, Xingjian(LR2)(1st) Shaofu(HT8)(2nd) Reinforcement, Shaofu(HT8)(1st) Xingjian(LR2)(2st) Reduction and Shaofu(HT8)(1st) Xingjian(LR2)(2st) Reduction in Acupuncture. Results: 1. LR2(1st) HT8(2nd) Reduction in Acupuncture was decreased rCBF and MABP in compared with normal condition. 2. LR2(1st) HT8(2nd) Reinforcement in Acupuncture was significantly decreased rCBF, and was decreased MABP in compared with normal condition. 3. HT8(1st) LR2(2st) Reduction in Acupuncture was decreased rCBF during acupuncture but was recovered rCBF after with-drawing of the neddle. 4. HT8(1st) LR2(2st) Reduction in Acupuncture was decreased MABP during acupuncture and after withdrawing of the neddle. 5. HT8(1st) LR2(2st) Reinforcement in Acupuncture was significantly increased rCBF during acupuncture and 30min after withdrawing of the neddle. 6. HT8(1st) LR2(2st) Reinforcement in Acupuncture was decreased MABP during acupuncture, but was recovered MABP after withdrawing of the neddle. Conclusions: I suggested that LR2 HT8 Reduction in Acupuncture and LR2 HT8 Reinforcement in Acupuncture cause a diverse response of cerebral hemodynas.
Objectives: The present study was carried out to investigate the effects of Yupoongyangyeong-tang(YYT) on changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. In addition, the author also investigated action mechanisms of YYT on changes in rCBF and MABP. Methods and Results : In this study, treatment with YYT elevated rCBF in a dose-dependent manner, but MABP levels were elevated only in the 10 mg/ml treatment group. Pre-treatment with indomethacin. an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. Pre-treatment with methylene blue, an inhibitor of guanylate cyclase, inhibited increase of rCBF induced by YYT, too. In addition. pre-treatment with indomethacin also inhibited increase of MABP. However, pre-treatment with methylene blue did not affect MABP levels. Conclusion : These results suggest that YYT is useful to treat patients with diseases related to cerebral ischemia, because YYT can increase rCBF. In addition, the mechanisms are thought to be related to both cyclooxygenase and guanylate cyclase.
Objective: The aim of this study was to investigate anti-ischemic effect of LR1 HT8 Reduction in Acupuncture Methods: I designed to investigate whether LR1 HT8 Reduction in Acupuncture affects cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP)] in normal rats and to make manifest whether LR1 HT8 Reduction in Acupuncture is mediated by cyclooxygenase or guanylate cyclase. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was de terminated by video microscope and width analyzer. Results: The results were as follows; 1. LR1 Reduction in Acupuncture was increased rCBF and PAD, but decreased MABP. 2. HT8 Reduction in Acupuncture was significantly increased rCBF, but decreased MABP, and increased PAD. 3. LR1 HT8 Reduction in Acupuncture was significantly increased rCBF, PAD, but decreased MABP after withdrawing of the needle. This results suggest that LR1 HT8 Reduction in Acupuncture increased significantly rCBF by dilating PAD. 4. Pretreatment with indomethacin(1mg/kg, i,v.) was significantly inhibited LR1 HT8 Reduction in Acupuncture induced increase of rCBF and PAD, but increased LR1 HT8 Reduction in Acupuncture induced decrease of MABP after withdrawing of the needle. 5. Pretreatment with methylene blue($10{\mu}g/kg$, i,v.) was decreased LR1 HT8 Reduction in Acupuncture induced increase of rCBF and PAD, but accelerated LR1 HT8 Reduction in Acupuncture induced decrease of MABP. Conclusions: I suggest that LR1 HT8 Reduction in Acupuncture has an anti-ischemic effect through the improvement of cerebral hemodynamics, and the mechanism is mediated by cyclooxygenase.
Objectives : Joongjeo($TE_3$) Supplementation Aekmoon($TE_2$) Draining is a method belongs to Ohaeng-acupuncture, using directional supplementation and draining. Methods : This study was designed to investigate the effects of $TE_3$ supplementation $TE_2$ draining on changes in cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in normal rats. For these reasons, the present author investigated rCBF and MABP using laser doppler flowmeter in normal rats. In addtion, the present author also investigated action mechanisms of $TE_3$ supplementation $TE_3$ draining on changes in rCBF and MABP too. Results : In this results, $TE_3$ supplementation $TE_2$ draining elevated rCBF in time-dependent manner, but MABP levels decresed by $TE_3$ supplementation $TE_2$ draining. Pre-treatment with indomethacin (IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. But pre-treatment with methylene blue(MTB), an inhibitor of guanylate cyclase, decreased rCBF levels. In addition, pre-treatment with IDM also decreased MABP levels, but pre-treatement with MTB increased MABP levels. Conclusions : In conclusion, these results suggest that $TE_3$ supplementation $TE_2$ draining is effective to treat patient with disease related to cerebral ischemia, because $TE_3$ supplementation $TE_2$ draining can increase rCBF. In addition, the mechanisms are thought to be related to guanylate cyclase pathways.

This experimental study was designed to investigate the effects of Sagunja-tang(SGJT), Ijin-tang(IJT), Yukgunja-tang(YGJT) on the change of cerebral hemodynamics [regional cerebral blood f1ow(rCBF), mean arterial blood pressure(MABP), and pial arterial diameter (PAD)] in normal rats, and further to determine the mechanism of action of YGJT. And, this Study was designed to investigate whether YGJT inhibit lactate dehydrogenase(LDH) activity in neuronal cells. The results were as follows ; 1. SGJT significantly increased rCBF but MABP was not changed comparing with normal MABP(l00 %). This results were suggested that SGJT significantly increased rCBF by dilating PAD. 2. IJT significantly decreased rCBF in a dose-dependent, but significantly increased MABP in a dose-dependent. This results were suggested that IJT significantly decreased rCBF by contracting PAD. 3. YGJT significantly increased rCBF and PAD in a dose-dependent, and YGJT increased MABP compared with normal MABP(100 %). This results were suggested that YGJT significantly increased rCBF by dilating PAD. 4. The YGJT-induced increase in rCBF was significantly accelerated by pretreatment with indomethacin (IDN, 1 mg/kg, i.p.), an inhibitor of cyclooxygenase but was significantly inhibited by methylene blue (MTB, 10 ㎍/㎏ i.p.), an inhibitor of guanylate cyclase. 5. The YGJT-induced increase in PAD and MABP were accelerated by pretreatment with IDN but was significantly inhibited by MTB. This results suggested that the mechanism of YGJT is mediated by guanylate cyclase. 6. YGJT inhibited significantly LDH activity in neuronal cells. This results were suggested that YGJT prevented the neuronal death. I thought that YGJT should have improvement of cerebral hemodynamics and inhibitive effect on the brain damage.

Pretreatment with indomethacin(1 mg/kg, i.v.) was significantly inhibited Haenggan(LR2) Reduction in Acupuncture induced increase of rCBF, but was increased Haenggan(LR2) Reduction in Acupuncture induced increase of MABP. 4. Pretreatment with methylene blue$(10\;{\mu}g/kg,\;i.v.)$ was significantly decreased Haenggan(LR2) Reduction in Acupuncture induced increase of rCBF and MABP. This results suggest that Haenggan(LR2) Reduction in Acupuncture increased rCBF by dilating pial arterial diameter, and the mechanism of Haenggan(LR2) Reduction in Acupuncture is mediated by guanylate cyclase.

The study was designed to investigate the effects of Mixture of Yukgunja-tang and Bambusae Caulis in Liquamen (YTBCL) on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats, and further to determine the mechanism of action of YTBCL. The results in rats were as follows ; YTBCL 25 ${\mu}l$ significantly decreased rCBF and MABP compared with basal condition. YTBCL 100 ${\mu}l$ significantly increased rCBF compared with basal condition, but decreased MABP compared with basal condition. YTBCL 50 ${\mu}l$ significantly increased rCBF compared with basal condition, but MABP was somewhat decreased compared with basal condition. The VTBCL 50 ${\mu}l$-induced increase in rCBF was significantly inhibited by pretreatment with methylene blue (10 ${\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase and indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. The YTBCL 50 ${\mu}l$-induced decreased MABP significantly increased by pretreatment with methylene blue but was inhibited by indomethacin. This results were suggested that the mechanism of YTBCL was mediated Dy guanylate cyclase.
