Biochemical Characterization of an Extracellular ${\beta}$-Glucosidase from the Fungus, Penicillium italicum, Isolated from Rotten Citrus Peel

  • A ${\beta}$-glucosidase from Penicillium italicum was purified with a specific activity of 61.8 U/mg, using a chromatography system. The native form of the enzyme was an 88.5-kDa tetramer with a molecular mass of 354 kDa. Optimum activity was observed at pH 4.5 and $60^{\circ}C$, and the half-lives were 1,737, 330, 34, and 1 hr at 50, 55, 60, and $65^{\circ}C$, respectively. Its activity was inhibited by 47% by 5 mM $Ni^{2+}$. The enzyme exhibited hydrolytic activity for p-nitrophenyl-${\beta}$-D-glucopyranoside (pNP-Glu), p-nitrophenyl-${\beta}$-D-cellobioside, p-nitrophenyl-${\beta}$-D-xyloside, and cellobiose, however, no activity was observed for p-nitrophenyl-${\beta}$-D-lactopyranoside, p-nitrophenyl-${\beta}$-D-galactopyranoside, carboxymetyl cellulose, xylan, and cellulose, indicating that the enzyme was a ${\beta}$-glucosidase. The $k_{cat}/K_m\;(s^{-1}mM^{-1})$ values for pNP-Glu and cellobiose were 15,770.4 mM and 6,361.4 mM, respectively. These values were the highest reported for ${\beta}$-glucosidases. Non-competitive inhibition of the enzyme by both glucose ($K_i=8.9mM$) and glucono-${\delta}$-lactone ($K_i=11.3mM$) was observed when pNP-Glu was used as the substrate. This is the first report of non-competitive inhibition of ${\beta}$-glucosidase by glucose and glucono-${\delta}$-lactone.

A Study for the Optimum pH of Hydrogen Production in Anaerobic Batch Reactor (혐기성 회분반응기에서 수소생산 시 최적 pH 산정에 관한 연구)

  • The influences of pH were investigated for anaerobic hydrogen gas production under the constant pH condition ranged from pH 3 to 10. Carbon dioxide and hydrogen gas were main components of the gas but methane was not detected in the produced gas when sucrose was added in enrichment medium. When the modified Gompartz equation was applied for the statistical analysis of experimental data, a hydrogen production potential and maximum gas production rate at pH 5 were 1,182 mL and 112.46 mL/g dry wt biomass/hr. The hydrogen conversion ratio was 22.56%. The butyrate/acetate ratios at pH 5 and pH 6 are 1.63 and 0.38. Higher butyrate/acetate ratio produced more hydrogen gas generation. The Haldane equation model was used to find the optimum pH and fitted well with the experimental data$(r^2=0.98)$. The optimum pH and specific hydrogen production were 5.5 and 119.61 mL/g VSS/h.

  • Superluminescent diodes (SLD) with the emitting wavelength of $1.55{\mu}m$ was fabricated on InGaAs quantum dot structure grown by MOCVD. The output power and 3-dB bandwidth at room temperature and continuous wave operation were 3 mw and 55 nm, respectively.

Microbial Conversion of Ginsenoside from the Extract of Korean Red Ginseng (Panax ginseng) by Lactobacillus sp.

  • Thirty-four strains of Lactobacillus species were isolated from soil and eight of these isolates (M1-4 and P1-4) were capable of growing on red ginseng agar. The M1 and P2 strains were determined to be L. plantarum and other strains (M2, M3, M4, P1, P3 and P4) were determined to be L. brevis. Fermentation of red ginseng extract (RGE) with strains M1, M2, P2 and P4 resulted in a low level of total carbohydrate content (174.3, 170.0, 158.8 and 164.8 mg/mL, respectively). RGE fermented by M3 showed a higher level of uronic acid than the control. The polyphenol levels in RGE fermented by M1, P1 and P2 (964.9, 941.7 and $965.3\;{\mu}g/mL$, respectively) were higher than the control ($936.8\;{\mu}g/mL$). Total saponin contents in fermented RGE (except M1) were higher than the control. RGE fermented by M2 and M3 had the highest levels of total ginsenosides (31.7 and 32.7 mg/mL, respectively). The levels of the ginsenoside Rg3 increased from 2.6 mg/mL (control) to 3.0 mg/mL (M2) or 3.1 mg/mL (M3). RGE fermented by M2 and M3 also had the highest levels of Rg5+Rk1 (7.7 and 8.3 mg/mL, respectively). Metabolite contents of ginsenoside (sum of CK, Rh1, Rg5, Rk1, Rg3 and Rg2) of M2 (13.0 mg/mL) and M3 (13.9 mg/mL) were also at a high level among the fermented RGE. Protopanaxadiol and protopanaxatriol content of ginsenoside of M2 (10.9 and 5.4 mg/mL, respectively) and M3 (11.0 and 5.7 mg/mL, respectively) were at higher levels than other fermented RGE.

Interactions between Hyaluronic Acid, Lysozyme, Peroxidase, and Glucose Oxidase in Enzymatic Activities at Low pH

  • Purpose: Many substances in saliva or oral health care products interact with each other. The aim of this study was to investigate interactions between hyaluronic acid (HA), lysozyme, peroxidase, and glucose oxidase (GO) in enzymatic activities at low pH levels. Methods: HA (0.5 mg/mL), hen egg-white lysozyme (HEWL, $30{\mu}g/mL$), bovine lactoperoxidase (bLPO, $25{\mu}g/mL$), and GO ($50{\mu}g/mL$) were used. The influences of HA, bLPO, and GO on HEWL activity were determined by measuring the turbidity of a Micrococcus lysodeikticus suspension. The influences of HA and HEWL on bLPO activity were determined by the NbsSCN assay, measuring the rate of oxidation of 5-thio-2-nitrobenzoic acid (Nbs) to 5,5'-dithiobis(2-nitrobenzoic acid) $(Nbs)_2$. The influences of HA and HEWL on GO activity were determined by measuring oxidized o-dianisidine production. All experiments were performed at pH 4, 5, and 6. Results: HA and GO did not affect the enzymatic activity of HEWL at pH 4, 5, and 6. bLPO enhanced the enzymatic activity of HEWL at pH 5 (p<0.05) and pH 6 (p<0.05) significantly. The enzymatic activity of bLPO was not affected by HA and HEWL at pH 4, 5, and 6. HA and HEWL did not affect the enzymatic activity of the GO at pH 4, 5, and 6. Conclusions: Peroxidase enhances lysozyme activity at low pH, otherwise there were no significant interactions in enzymatic activities between HA, lysozyme, peroxidase, and GO at low pH levels.

  • The sequential separation of Ru-103, Cs-137 and Ce-144 was carried out by organic cation exchanger, Amberite CG-120, and inorganic ion exchangers, silica gel and montmorillonite. The optimum conditions of Ru-103, Cs-137 and Ce-144 on Amberite CG-120 are 0.01M-, 0.01M- and 0.1IM- hydrochloric acid for the adsorption, and 3M-, 3M- and 5M-hydrochloric acid for the desorption, respectively. The optimum conditions of Ru-103, Cs-137 and Ce-144 on silica gel are pH 8, pH 8 and pH 8 for the adsorption. and 3M-, 1M- and 1M-hydrochloric acid for the desorption. respectively. The optimum conditions of Ru-103, Cs-137 and Ce-144 on montmorillonite are pH 8, 0.01M-hydrochloric acid and pH 4 for the adsorption, and 1M-, 5M- and 3M-hydrochloric acid for the desorption. respectively. The adsorption which occurs at lower ionic strength and the differences in desorption ionic strength are utilized for the separation of tracer mixture in continuous experiments. The individual separation of Ru-103, Cs-137 and Ce-144 can be carried out more efficiently with montmorillonite than with silica gel and Amberite CG-120.

  • A series of homobinuclear complexes of the type [$M_2L(NO_3)_n(H_2O)_m$] where M=$UO_2$(VI), ZrO(IV), Th(IV); L=3-benzylidine-imino-5-p-sulphonamido phenyl azo-2-thiohyatoin(bispt), 3-furfurylidine-imino-5-p-sulphonamido phenyl azo-2-thiohydantoin(fispt),3-pyridyl-2'-methylene-imino-5-p-sulphonamido phenyl azo-2-thiohydantoin(pmispt) and 3-thienyl-2'-methylene-imino-5-p-sulphoanamido phenyl azo-2-thiohydantoin(tmispt); n=8 for Th(IV) and 4 for others, m=4 for bispt and 3 for others have been synthesized and characterized on the basis of elemental analysis, thermal analysis, molar conductance, magnetic moment and spectroscopic data (IR, electronic and $^1H$-NMR). In the light of this information, the ligands can be visualized as tetradentate co-ordinating through azomethine nitrogen, carbonyl oxygen to one metal centre where as azo nitrogen and thioimido nitrogen to the other metal centre yielding homo binuclear complexes of the above composition. The fungi toxicity of the ligands & their zirconyl complexes against some fungal pathogen has been studied.

Properties of Acid Tolerance of Acid-Resistant Mutant Leuconostoc mesenteroides Which Was Improved as Kimchi Starter. (김치 starter용으로 개량된 내산성 변이주 Leuconostoc mesenteroides의 내산성 특성)

  • To investigate the increased acid tolerance of a acid-resistant mutant Leuconostoc mesenteroides M-100 improved as a kimchi starter, proton permeability, ATPase acitivity, glycolysis activity, $Mg^{2+}$ releasement, and membrane fatty acid composition were studied and comprised with its wild type Leuconostoc mesenteroides Mw. In the proton permeability experiment, the mininum values of the average half time ($t_{1/2}$) of pH equilibration through the cell membrane of the Mw and the M-100 were about 8.6 min and 9.2 min in 150 mM KCI solution, respectivily. In the 3% NaCl solution, the $t_{1/2}$ values of the Mw and the M-100 were 6min and 8.6 min, respectivily. The values and pHs of maximal specific activities of ATPase originated from the Mw and the M-100 were 0.6U at pH 5.5 and 0.8U at pH 5.5, respectivily. The result of pH dependence of glycolysis showed that the M-100 had higher activities than that of Mw except at pH 5.0. The releases of magnesium from the Mw and the M-100 were observed about 36.5% and 13% at pH 4.0 after 2 hours, respectivily. The results of comparison of membrane fatty acid composition of the Mw with the M-100 showed that $C_{12}$, $C_{14}$, $C_{18:1}$, and $C_{19:0cyclo}$ were major different fatty acids between two strains and the content of $C_{18:1}$, and $C_{19:0cyclo}$ were 23.4%, 10.2% in the Mw and 15.1%, 12.2% in the M-100. These results indicated that acid tolerance of the M-100 was significantly improved in comparison with its wild type Mw.

Effects of Polymannuronate Feeding on Compositions of Serum and Liver Lipids in the High-Cholesterol Fed Rats (Polymannuronate의 급이가 고콜레스테롤 급이 흰쥐의 혈청 및 간 지질 조성에 미치는 영향)

  • The cholesterol and fatty acid levels in serum and liver were compared in 4-week-old Sprague-Dawley male fed by addition of polymannuronate (M, 5.0%), polyguluronate (G, 5.0%), and polymannuronate and polyguluronate (MG, 2.5% to each) with by addition of cholesterol (1.0%). Feed efficiency by the addition of M, MG, and G was lower than cholesterol fed group (p<0.01). The liver weights were less in M, MG, and G fed groups than in cholesterol fed group (p<0.01). Triglyceride levels in serum and liver were 58.2∼77.4% and 51.5∼65.5% lower in M, G and MG fed groups than cholesterol fed group, respectively. Total-, LDL-, and free-cholesterol levels in serum and liver in M, MG, and G fed group were significantly lower than cholesterol fed group. The cholesterol levels were the most reduced in M fed group. However, HDL- cholesterol level in serum was increased in M, MG, and G fed group (p<0.01). The of polyene levels were 47% higher in serum and 76% in liver in M fed group than in cholesterol fed group. The activities of GOT and GPT were lower in M, MG, and G fed group than in cholesterol fed group (p<0.01). Above the results demonstrate that supplementation of low molecular polymannuronate in diets improve physiologically lipid composition in serum and liver.
