셀룰로스의 분석을 위한 새로운 시료처리 및 크기배제크로마토그래피 (A New Sampling and SEC Method for Analysis of Underivatized Cellulose)

  • 셀룰로스시료를 치환체로 만들지 않고 분자량 및 분자량분포를 결정하기 위한 새로운 시료처리 및 크기배제크로마토그래피방법을 확립하였다. 먼저 셀룰로스시료를 N-methylmorpholine N-oxide (NMMO)에 녹인 후 dimethyl sulfoxide (DMSO)에 희석함으로써 50/50(w) NMMO/DMSO에 녹은 약 0.1%의 시료용액을 만들었다. 컬럼으로는 글루코오스를 입힌 polydivinylbenzene 겔을 채운 크기배제크로마토그래피용 컬럼을 이용하였으며 용리액으로는 0.05M의 LiBr와 2.5%의 바탕용액을 포함하는 DMSO를 이용하였다.유속은 1 mL/min로 일정하게 유지하였고, DMSO의 점도를 낮추기 위해 컬럼을 포함하는 전체 시스템을 80$^{\circ}C$ 로 유지하였다. 0.05M의 LiBr를 가함으로써 SEC바탕선의 표류를 방지할 수 있었고 2.5%의 바탕용액을 가함으로써 시료와 컬럼충진물질 사이의 상호작용을 감소시키는 효과를 얻었다. 좁은 분포를 가지는 pullulan표준물질로부터 얻은 컬럼보정곡선을 이용하여 두 개의 I 펄프시료와 두 개의 셀룰로스 스펀지 시료의 평균분자량을 결정함으로써 펄프로부터 스펀지를 만드는 서로 다른 두 가지 공정에서의 셀룰로스 분자들의 분해정도를 결정, 비교할 수 있었다.

Role of Cel5H protein surface amino acids in binding with clay minerals and measurements of its forces

  • Our previous study on the binding activity between Cel5H and clay minerals showed highest binding efficiency among other cellulase enzymes cloned. Here, based on previous studies, we hypothesized that the positive amino acids on the surface of Cel5H protein may play an important role in binding to clay surfaces. To examine this, protein sequences of Bacillus licheniformis Cel5H (BlCel5H) and Paenibacillus polymyxa Cel5A (PpCel5A) were analyzed and then selected amino acids were mutated. These mutated proteins were investigated for binding activity and force measurement via atomic force microscopy (AFM). A total of seven amino acids which are only present in BlCel5H but not in PpCel5A were selected for mutational studies and the positive residues which are present in both were omitted. Of the seven selected surface lysine residues, only three mutants K196A(M2), K54A(M3) and K157T(M4) showed 12%, 7% and 8% less clay mineral binding ability, respectively compared with wild-type. The probable reason why other mutants did not show altered binding efficiency might be due to relative location of amino acids on the protein surface. Meanwhile, measurement of adhesion forces on mica sheets showed a well-defined maximum at 69±19 pN for wild-type, 58±19 pN for M2, 53±19 pN for M3, and 49±19 pN for M4 proteins. Hence, our results demonstrated that relative location of surface amino acids of Cel5H protein especially positive charged amino acids are important in the process of clay mineral-protein binding interaction through electrostatic exchange of charges.

  • 산화성 첨가반응을 이용하여 실라트란(분자내 Si-N고리횡단 상호작용을 갖는 5-배위의 규소화합물)의 규소원자와 주족원소간에 Si-M 결합작용이 있는 새로운 이종다핵 화합물을 합성하려고 시도하였다. 1-브로모실라트란(1a)과 $SnBr_2를 반응시켜 노란색(2a)과 흰색(2b)의 고체 혼합물을 얻었고 용해도 차에 의해 각각을 분리하여 ^1H-NMR, ^{29}Si-NMR, ^{119}Sn-NMR, Mass스펙트럼으로 규명한 결과 노란색 화합물만이 Si-Sn결합을 갖는 것으로 판명되었다. SnBr_2를 1-브로모-3,7,10-트리메틸실라트란(1b)과도 반응시켜 Si-Sn 결합을 갖는 화합물, N[CH_2CH(CH_3)O]_3SiSnBr_3(CH_3OH)_2(3)$을 분리하여 여러가지 방법으로 확인하였다.

  • 동일 환경에서 할 수 있는 실험의 크기가 2인 경우 $2^n$요인실험과 그 일부실시법을 설계하려면 교락법을 반복적으로 사용해야한다. $2^n$요인실험 또는 그 일부실시법의 교락법들을 적절히 조합하면 모든 주효과와 2인자 교호작용효과 전부 또는 일부를 검출할 수 있는 실험을 설계할 수 있다. 이런 실험을 설계하기 위해 정의대비를 이용했고 설계된 실험의 처리조합을 제시하였다.

Inhibition of Aminopeptidase N by Two Synthetic Tripeptides

  • MR-387Al (ARPA-Val-Pro) and A2 (AHPA-Val-Hyp) were prepared as aminopeptidase N inhibitors through the synthesis of peptide MR-387A and B analogues which contained 3-amino-2-hydroxy-4-phenyl butanoic acid (ARPA) as a zinc-chelating moiety. They are competitive inhibitors of aminopeptidase N with inhibition constants(Ki) of 4.1 $\times 10^{-7}\;and 1.1 \times 10^{-6}$ M, respectively. MR-387Al also strongly inhibited aminopeptidase B of human myelogenous leukemia K-562 cell with $IC_50$ of 0.35 $\mu$ M. Inhibitions of aminopeptidase N activity by ARPA-bearing inhibitors of various peptide chain lengths also have been studied. $IC_ 50$ values of AHPA-Val (bestatin), ARPA-Val-Pro (MR-387Al) and ARPA-Val-Pro-Leu (MR-387C) compared against porcine kidney aminopeptidase N were 20.1, 0.60 and 0.08 $\mu$ M, respectively. These results support that a multiple interaction between the $S_1\to S'_3$ sites of aminopeptidase N and the $P_1\to P'_3$ of the inhibitor plays a crucial role in stabilizing strongly the enzyme-inhibitor complex.

Interaction between different nitrogen fertilizer levels and maize-bean intercropping patterns

  • In order to investigate the effects of different maize-bean intercropping patterns, and of nitrogen fertilizers on morphological and yield related traits, a factorial study based on Randomized Complete Block Design (RCBD) was performed during the 2010 and 2011 growing seasons in a research filed of Shiraz University, Iran. The first factor of the study was seven different ratios of Maize-Bean intercropping system (Maize sole cropping, Bean sole cropping, and intercropping of maize/bean at the ratios of 1/3, 1/1, 2/3, 3/2 and 3/1) and the second factor was three nitrogen (N) fertilizer application levels (0, 100 and 200 kg N/ha). Results showed that with respect to increasing the levels of N fertilizer, the yield of bean sole cropping decreased but the yield of maize sole cropping increased. On the other hand, in intercropping systems with N fertilizer application, the yield of both crops increased. Results of total land equivalent ratio (LER) for both crops showed that the highest LER value under both 100 and 200 kg N/ha application was that of M1B1 (1 seed of maize after 1 seed of bean, consecutively, on a row with same distance). Under no N fertilizer application the highest LER value was that of M2B3 (2 seeds of maize after 3 seeds of bean, consecutively, on a row with same distance). Overall, it can be concluded that M1B1 is the best intercropping pattern in maize-bean intercropping systems and that the application of N fertilizer can be effective within practical settings of intercropping agriculture, resulting in higher yields.

Genotype by Dietary Lysine Interaction for Growth and Response to Sheep Red Blood Cells and Escherichia coli Inoculation in Commercial Broiler Chicks

  • An experiment was conducted to assess the interaction between genotypes and dietary lysine content in commercial broiler chicks by measuring growth, and response to sheep red blood cells (SRBC) and Escherichia coli (E.coli) inoculation. Female chicks from four genotypes (A=Anak 2000; B=Hubbard; C=Cobb and D=Synthetic broiler) were fed four levels of lysine in diet from d old till the end of experiment. The lysine content of the diet was 9.61, 10.51, 11.41 and 12.31 g/kg. Body weights at 0, 14, 28 and 42 d of age and pen-wise feed intake till 14, 28 and 42 d of age were recorded. Production of antibody against SRBC and resistance to E.coli were measured at 5 d of post inoculation (PI) at 43 d of age. Also, response to phytohemaglutinin-P (PHA-P) was measured at 12 and 24 h of PI at 48 d of age. Genotype by dietary lysine interaction was significant for body weights at 14 and 28 d of age, but not at 42 d of age. Genotype by dietary lysine interaction was not significant for feed efficiency, for antibody titers against SRBC, and for air sac lesion score, relative bodyweight change, and relative weights of bursa and spleen in response to E.coli inoculation. However, a significant interaction was observed between the levels of lysine and dosage of SRBC for antibody titers. There was significant genotype by dietary lysine interaction for cutaneous basophilic hypersensitivity (CBH) response to PHA-P at 12 and 24 h of PI. It may be concluded that to obtain optimum body weight and immunity in commercial broilers the dietary lysine requirement may be recommended specific to the genotype.

Novel Phosphotungstate-titania Nanocomposites from Aqueous Media

  • We report a novel method to synthesize nanocomposites composed of titania nanoparticles and phosphotungstate ions with various composition ratios ranging from W/Ti = 12/10 to 12/500 by inducing the electrostatic interaction between the positively charged protonated titania sol-particles and the negatively charged phosphotungstate anions to flocculate and precipitate. The precipitates showed varied features depending on the composition. The precipitate from the tungsten-richest W/Ti = 12/10 reaction is amorphous in its powder X-ray diffraction (XRD), transmission electron microscopy (TEM), and Raman spectroscopy data. This material shows the Type II adsorption characteristics in its N2-adsorption isotherm, but with quite low surface area of 34 m2/g. To the contrary, the precipitates from the titanium-richer reactions (W/Ti = 12/50- 12/500) are composed of anatase nanoparticles of 2-6 nm by XRD, TEM and Raman and show the Type I adsorption characteristics. The surface area linearly increases with the titanium content from 131 m2/g for W/ Ti = 12/50 to 228 m2/g for 12/500. The precipitate from the reaction with the intermediate composition W/Ti = 12/20 is composed of anatase nanoparticles and does not have any pore accessible to N2. With the wide variety of the physical properties of the precipitates, the present method can be a novel, viable means to tailor synthesis of nanocomposite materials. A formation mechanism of the precipitates is based on the electrostatic interactions between the titania nanoparticles and phosphotungstate ions.

질소시비량이 쇠무릎의 생육 및 건물수량에 미치는 영향 (Influence of Nitrogen Application Rate on Growth and Dry Matter Yield of Achyranthes japonica Nakai)

  • 제주도에서 쇠무릎의 뿌리와 포과의 생산을 위한 적정질소시비량을 구명하고자 재식밀도(50, 100주/$m^2$, 1주 2본)와 질소시비량(0, 6, 12, 18, 24, 30 kg/10a)에 따른 생육, 건물수량, 지상부 질소 함량 및 수량 등을 조사하였던 바 그 결과를 요약하면 다음과 같다. 조사한 모든 형질에 대하여 재식밀도와 질소시비량 간에는 상호작용이 없었다. 100주/$m^2$ 재식구에 비하여 50주/$m^2$ 재식구에서 주근장 및 뿌리수는 각각 5%와 52%증가되었고 경엽의 질소함량은 다소 낮았으나 다른 형질에서는 차이가 없었다. 질소시비량은 주경직경, 주당 화서수, 화서당 포과수, 주근장, 주근직경, 포과 질소함량에 영향을 미치지 않았다. 질소시비량이 10a당 0 kg에서 30kg으로 증가함에 따라 SPAD 값은 35.0에서 40.5로, 경엽의 질소함량은 1.09%에서 1.38%로 직선적으로 증가되었다. 질소시비량은 초장, 주당 분지수, 포과 뿌리 건물수량, 주당 근수, 경엽질소수량에 대해서는 2차함수식으로, 주당 화서수에 대해서는 3차함수적인 관계로 분석되었다. 이러한 분석결과 포과 및 뿌리 건물수량이 최대가 되는 질소시비량은 21kg/10a로 추정되었다.

Apolipoprotein A5 3'-UTR variants and cardiometabolic traits in Koreans: results from the Korean genome and epidemiology study and the Korea National Health and Nutrition Examination Survey

  • BACKGROUND/OBJECTIVES: This study aimed to test the association between APOA5 3'-UTR variants (rs662799) and cardiometabolic traits in Koreans. SUBJECTS/METHODS: For this study, epidemiological data, Apolipoprotein A5 (APOA5) genotype information, and lymphoblastoid cell line (LCL) biospecimens from a subset of the Ansung-Ansan cohort within the Korean Genome and Epidemiology study (KoGES-ASAS; n = 7,704) as well as epidemiological data along with genomic DNA biospecimens of participants from a subset of the Korea National Health and Nutrition Examination Survey (KNHANES 2011-12; n = 2,235) were obtained. APOA5 mRNA expression was also measured. RESULTS: APOA5 rs662799 genotype distributions in both the KoGES-ASAS and KNHANES groups were 50.6% for TT, 41.3% for TC, and 8.1% for CC, which are similar to those in previous reports. In both groups, minor C allele carriers, particularly subjects with CC homozygosity, had lower high-density lipoprotein (HDL) cholesterol and higher triglyceride levels than TT homozygotes. Linear regression analysis showed that the minor C allele significantly contributed to reduction of circulating HDL cholesterol levels [${\beta}=-2.048$, P < 0.001; ${\beta}=-2.199$, P < 0.001] as well as elevation of circulating triglyceride levels [${\beta}=0.053$, P < 0.001; ${\beta}=0.066$, P < 0.001] in both the KoGES-ASAS and KNHANES groups. In addition, higher expression levels of APOA5 in LCLs of 64 healthy individuals were negatively associated with body mass index (r = -0.277, P = 0.027) and circulating triglyceride level (r = -0.340, P = 0.006) but not significantly correlated with circulating HDL cholesterol level. On the other hand, we observed no significant difference in the mRNA level of APOA5 according to APOA5 rs662799 polymorphisms. CONCLUSIONS: The C allele of APOA5 rs662799 was found to be significantly associated with cardiometabolic traits in a large Korean population from the KoGES-ASAS and KNHANES. The effect of this genotype may be associated with post-transcriptional regulation, which deserves further experimental confirmation.
