ACAT (Acyl-CoA:cholesterol Acyltransferase) Inhibitory Effect and Quantification of Pyranocurmarin in Different Parts of Angelica gigas Nakai (참당귀(Angelica gigas)에서 분리한 pyranocurmarin 성분의 ACAT 저해활성 및 참당귀 부위별 pyranocurmarin 성분의 함량 분석)

  • Two pyranocoumarin constituents have been isolated from Angelica gigas and were identified as decursinol angelate (1) and decursin (2) by means of NMR analysis, respectively. Human acyl-CoA:cholesterol acyltransferase (hACAT) inhibitory activity of decursinol angelate (1) and decursin (2) was evaluated. Decursin (2) showed significantly inhibitory activity against hACAT1 and hACAT2 with $IC_{50}$ value of 137 and $168\;{\mu}M$, respectively, whereas decursinol angelate (1) exhibited weak ACAT inhibitory activity. These results suggested that decusin from A. gigas might be effective for the prevention and the treatment of hypercholesterolemia or atherosclerosis by inhibitory effect on hACAT. The contents of decursinol angelate (1) and decursin (2) were analyzed in various parts of A. gigas including flower, seed, leaf and root using LC/MS/MS (ESI, positive ion mode, MRM mode). The content of decursinol angelate was increased in order of flower, seed, leaf, and root and decursin content was increased in order of flower, seed, leaf, and root. It was expected that unused parts including leaf and flower of A. gigas might be useful as new functional sources by their high contents of decursin and decursinol angelate.

Anti-Porcine Epidemic Diarrhea Virus (PEDV) Activity and Antimicrobial Activities of Artemisia dubia Essential Oil (참쑥(Artemisia dubia) 오일의 돼지 유행성 설사 바이러스(Porcine Epidemic Diarrhea Virus)에 대한 항바이러스 항균활성)

  • The chemical composition, anti-porcine epidemic diarrhea virus (PEDV) activity and antimicrobial activity of Artemisia dubia essential oil were evaluated in this study. Fifty eight compounds from A. dubia essential oil were identified through analysis by gas chromatography-mass spectrometry (GC-MS). The major constituents of the oil were camphor (17.18 %), germacrene-D (15.70%), trans (${\beta}-$) racaryophyllene (6.79%), ene thujones (6.57%), 1, 8-cineole (5.94%) and camphene (5.08%). The essential oil was evaluated for antiviral activity against PEDV in Vero cells using a cytopathic effect (CPE) reduction method. The oils actively inhibited PEDV replication with a 50% inhibitory concentration ($IC_{50}$) of 43.7 ${\mu}^3/mL$. The 50% cytotoxicity concentration ($CC_{50}$) of the oils was over 100 ${\mu}/mL$ and the derived therapeutic index was >2.3. Similar analysis of the ribavirin revealed that they have a relatively weaker efficacy when compared to the oils. The antimicrobial activity of the essential oil against 5 microorganisms was evaluated by the disc diffusion method. The essential oil exhibited antimicrobial activity against 5 tested microorganisms with a clear zone of 8-22 mm. Among the tested microorganisms, Streptococcus pyogenes was the most sensitive and Candida albicans the least. Therefore, in can be concluded that essential oils of A. dubia may have interesting applications for microbial control or the control of PEDV-derived diseases.

Effects of Rice Hull Addition and Bin Wall Characteristics on Pig Slurry Composting Properties (왕겨 이용 방법과 옹벽이 돈분 퇴비화에 미치는 효과)

  • This work was carried out to investigate the effects of rice hull continuously utilized and/or replenished on the composting properties and to obtain the fundamental data between an unsupported wall and a soil supported wall during the period of composting with pig slurry in winter season. There were no the temperature holding effects in soil supported wall. New compost facility design for the temperature holding effects from a soil supported wall was required. The results were as follows; 1. Composting 1㎥ of pig slurry caused to save on 0.31㎥ of bulking agent in the unsupported wall in comparison with a soil supported wall in the rice hull single addition, and 0.45㎥ in the rice hull gradual addition. 2. The pile in the rice hull single addition had a high temperature in 4 days of composting indicating $71^{\circ}C$ and had a tendency in repeating periodically between $40^{\circ}C$ and $65^{\circ}C$ till 43 days of composting. And also the temperature of the pile was maintained between $48^{\circ}C$ and $28^{\circ}C$ after 50 days of composting. The pile of a rice hull gradual addition had the lower point of the temperature high increasingly according to adding up rice hull during the 35 days of composting. 3. The pH recorded in the rice hull single addition was higher(8.35∼10.02) compared to the rice hull gradual addition(8.6∼9.8). The pile of a rice hull single addition had a tendency in abruptly decreasing pH of the unsupported wall during the period of between 0.363$\textrm m^3$ and 0.537$\textrm m^3$ as a unit of pig slurry per rice hull. EC depending upon the way in adding rice hull was changed between 1.10 mS/$\textrm {cm}^3$ and 1.87 mS/$\textrm {cm}^3$. 4. The organic matter in an unsupported wall of the hull single addition was maintained the level of 55% during the period between 0.119㎥ and 0.363㎥ as a unit of pig slurry per rice hull while in the soil supported wall between 48 and 70. Water soluble C:N ratio was maintained between 1 and 2 in the rice hull single addition, while between 1 and 3 in the rice hull gradual addition. 5. Fertilizer constituents were detected higher level in the unsupported wall than in the soil supported wall in all treatments. This was dependant upon the input of pig slurry.

Behaviour of the Soil Residues of the Acaricide-Insecticide, [$^{14}C$]Acrinathrin;I. Behaviour during Crop(Maize) Cultivation (살비살충제 [$^{14}C$Acrinathrin 토양 잔류물의 행적 규명;I. 농작물(옥수수) 재배시의 행적)

  • In order to elucidate the fate of the residues of the pyrethroid acaricide-insecticide, acrinathrin in soil, maize plants were grown for one month on the specially-made pots filled with two different types of soils containing fresh and one-month-aged residues of [$^{14}C$]acrinathrin, respectively. The mineralization of [$^{14}C$]acrinathrin to $^{14}CO_2$ during the one-month period of aging and of maize cultivation amounted to $23{\sim}24%$ and $24{\sim}33%$, respectively, of the original $^{14}C$ activities. At harvest after one-month growing, the shoots and roots contained less than 0.1% and 1% of the originally applied $^{14}C$ activity, respectively, whereas the $^{14}C$ activity remaining in soil was $65{\sim}80%$ in both soils. Three degradation products with m/z 198(3-phenoxybenzaldehyde), m/z 214(3-phenoxybenzoic acid), and m/z 228(methyl 3-phenoxybenzoate) besides an unknown were identified from acetone extracts of both soils without and with maize plants after treatment of [$^{14}C$]acrinathrin, by autoradiography and GC-MS, and those with m/z 225(3-phenoxybenzaldehyde cyanohydrin) and m/z 198 (3-phenoxybenzaldehyde) from acetone extract of the Soil A treated with 50 ppm acrinathrin and grown with maize plants for 30 days were identified by mass spectrometry. These results suggested that the hydrolytic cleavage of the ester linkage adjacent to the $^{14}C$ with a cyano group, forming 3-phenoxybenzaldehyde cyanohydrin. The removal of hydrogen cyanide therefrom leads to the formation of 3-phenoxybenzaldehyde as one of the major products. The subsequent oxidation of the aldehyde to 3-phenoxybenzoic acid, followed by decarboxylation would evolve $^{14}CO_2$. Solvent extractability of the soils where maize plants were grown for 1 month and/or [$^{14}C$]acrinathrin was aged for 1 month was less than 31% of the original $^{14}C$ activity and over 95% of the total $^{14}C$ activity in soil extracts was distributed in the organic phase. Accordingly, acrinathrin turned out to be degraded rapidly in both soils and be bound to soil constituents as well, not being available to crops.

  • Chromated copper arsenate (CCA), a wood preservative, has been widely used to protect wood products from attacks by bacteria, fungi and insects. However, the use of CCA is currently forbidden or limited to some applications in many countries because the toxic elements (Cr, Cu, and As) of CCA are released into the environments during outdoor uses, which may cause adverse health effects on humans and ecological systems. This study was conducted to investigate the distributions of chromium, copper and arsenic in soils adjacent to two CCA-treated wood structures. In a 7 month old pond entry structure, ten surface soil samples (0-2.5 cm) were collected at lateral distances of 0, 0.5, and 1 m from the stairway, and nine surface soil samples were collected beneath the deck. Nine top soil samples were taken from a 2 year old sound barrier structure at lateral distances of 0, 1, and 2 m. Background surface soil samples were also collected from each structure. Samples were analyzed for some physicochemical properties such as pH, electrical conductivity, organic matter content, and soil texture. Following the extraction of the elements with a microwave digestion system, samples were analyzed for Cr, Cu, and As. The concentrations of the three elements in soils adjacent to the structures were significantly elevated compared to the background levels, indicating that the elements have been leached out of the structures. Released e1ements showed lateral concentration gradients within 1 m. The elevations of the three elements in soils underneath the deck did not seem different (background-corrected concentrations: Cr, 5.01 mg/kg; Cu, 5.50 mg/kg; As, 4.91 mg/kg), while the elements in soils near the sound barrier were elevated in the order of As>Cu>Cr with measured concentrations of 49.7, 44.7 and 52.5 mg/kg, respectively. Background As, Cu, and Cr concentrations near the sound barrier were 9.88, 30.8, and 46.5 mg/kg, respectively. These results showed that CCA constituents are released into the environment and it is suggested that risk assessment need to be conducted to investigate harmful effects of the released elements on humans and ecological systems.

  • Commercially pure titanium (CP Ti) and Ti-6Al-4V alloys have been widely used for biomedical applications. However, the use of the Ti-6Al-4V alloy in biomaterial is then a subject of controversy because aluminum ions and vanadium oxide have potential detrimental influence on the human body due to vanadium and aluminum. Hence, recent works showed that the synthesis of new Ti-based alloys for implant application involves more biocompatible metallic alloying element, such as, Nb, Hf, Zr and Mo. In particular, Nb and Hf are one of the most effective Ti ${\beta}-stabilizer$ and reducing the elastic modulus. Plasma electrolyte oxidation (PEO) is known as excellent method in the biocompatibility of biomaterial due to quickly coating time and controlled coating condition. The anodized oxide layer and diameter modulation of Ti alloys can be obtained function of improvement of cell adhesion. Manganese(Mn) plays very important roles in essential for normal growth and metabolism of skeletal tissue in vertebrates and can be detected as minor constituents in teeth and bone. Radio frequency(RF) magnetron sputtering in the various PVD methods has high deposition rates, high-purity films, extremely high adhesion of films, and excellent uniform layers for depositing a wide range of materials, including metals, alloys and ceramics like a hydroxyapatite. The aim of this study is to research the Mn coatings on the micro-pore formed Ti-29Nb-xHf alloys by RF-magnetron sputtering for dental applications. Ti-29Nb-xHf (x= 0, 3, 7 and 15wt%, mass fraction) alloys were prepared Ti-29Nb-xHf alloys of containing Hf up from 0 wt% to 15 wt% were melted by using a vacuum furnace. Ti-29Nb-xHf alloys were homogenized for 2 hr at $1050^{\circ}C$. Each alloy was anodized in solution containing typically 0.15 M calcium acetate monohydrate + 0.02 M calcium glycerophosphate at room temperature. A direct current power source was used for the process of anodization. Anodized alloys was prepared using 270V~300V anodization voltage at room. Mn coatings was produced by RF-magnetron sputtering system. RF power of 100W was applied to the target for 1h at room temperature. The microstructure, phase and composition of Mn coated oxide surface of Ti-29Nb-xHf alloys were examined by FE-SEM, EDS, and XRD.

Characteristics of Tidal Current and Tidal Residual Current in the Chunsu Bay, Yellow Sea, Korea based on Numerical Modeling Experiments (수치모델링 실험을 통한 서해 천수만의 조류와 조석잔차류 특성)

  • This study is based on a series of numerical modeling experiments to understand the circulation and its change in the Chunsu Bay (CSB), Yellow Sea of Korea. A skill analysis was performed for the tidal height and tidal current of the observation data using the amplitude and phase of the 4 major tidal constituents respectively for verification of modeling experimental results. As a result, most of the skill score was seen to be over 90%, so numerical model experiment results can be said to be in good agreement with the observed tidal height and tidal current. Tidal wave proceeded from the entrance of the CSB towards inside, and the tidal range gradually increased to the north. It took about 10 to 30 minutes for the tidal wave to reach to northern end. The tidal wave showed a characteristic to rotate counter-clockwise in the southern part. The tidal current flowed to the north-south direction along the bottom topography; the angle of the major axis appeared alongside the isobath. It showed the characteristics of reversing tidal current with the minor axis less than 10% of the major axis. The strength of the tidal residual current that is influenced by geographical factors including bathymetry and coastline showed the range of 1~30 cm/sec, greater in the south channel and smaller in northern Bay. Two pairs of cyclonic/anti-cyclonic eddies around Jukdo and 3~4 pairs of strong eddies at the southern part of CSB in hundreds of m to a few km size by relative vorticity derived from the tidal residual current.

Si in particular has been found to be essential for normal bone and cartilage growth and development. In vitro studies have shown that Mg plays very important roles in essential for normal growth and metabolism of skeletal tissue in vertebrates and can be detected as minor constituents in teeth and bone. The aim of this study is to research Si and Mg doped hydroxyapatite film formation by plasma electrolytic oxidation. Vulnerability Analyses of Wave Overtopping Inundation by Synthesized Typhoons with Sea-Level Rise (해수면 상승과 빈도 합성태풍이 고려된 월파범람 위험성 분석)

  • Storm surges caused by a typhoon occur during the summer season, when the sea-level is higher than the annual average due to steric effect. In this study, we analyzed the sea-level pressure and tidal data collected in 1 h intervals at Incheon, Kunsan, Mokpo, Seogwipo stations on the Yellow Sea coast to analyze the summer season storm surge and wave overtopping. According to our analyses, the summer mean sea-level rise on the west and south coasts is approximately 20 cm and 15 to 20 cm higher than the annual mean sea-level rise. Changes in sea-level rise are closely related to changes in seasonal sea-level pressure, within the range of 1.58 to 1.73 cm/hPa. These correlated mechanisms generates a phase difference of one month or more. The 18.6 year long period tidal constituents indicate that in 2090, the amplitude of the $M_2$ basin peaks on the southwest coast. Therefore, there is a need to analyze the target year for global warming and sea-level rise in 2090. Wave overtopping was simulated considering annual mean sea-level rise, summer sea level rise, the combined effect of nodal factor variation, and 100-year frequency storm surge. As a result, flooding by wave overtopping occurs in the area of Suyong Bay, Busan. In 2090, overtopping discharges are more than doubled than those in Marine City by the recent typhoon Chaba. Adequate coastal design is needed to prepare for flood vulnerability.

Feasibility Study of Wetland-pond Systems for Water Quality Improvement and Agricultural Reuse (습지-연못 연계시스템에 의한 수질개선과 농업적 재이용 타당성 분석)

  • A pilot study was performed from September 2000 to April 2004 to examine the feasibility of the wetland-pond system for the agricultural reuse of reclaimed water. The wetland system was a subsurface flow type, with a hydraulic residence time of 3.5 days, and the subsequent pond was 8 $m^3$ in volume (2 m ${\times}$ 2 m ${\times}$ 2 m) and operated with intermittent-discharge and continuous flow types. The wetland system was effective in treating the sewage; median removal efficiencies of $BOD_5$ and TSS were above 70.0%, with mean effluent concentrations of 27.1 and 16.8 mg $L^{-1}$, respectively, for these constituents. However, they did often exceed the effluent water quality standards of 20 mg $L^{-1}$. Removal of T-N and T-P was relatively less effective and mean effluent concentrations were approximately 103.2 and 7.2 mg $L^{-1}$, respectively. The wetland system demonstrated high removal rate (92 ${\sim}$ 90%) of microorganisms, but effluent concentrations were in the range of 300 ${\sim}$ 16,000 MPN 100 $mL^{-1}$ which is still high for agricultural reuse. The subsequent pond system provided further treatment of the wetland effluent, and especially additional microorganisms removal in addition to wetland-pond system could reduce the mean concentration to 1,000 MPN 100 $mL^{-1}$ from about $10^5$ MPN 100 $mL^{-1}$ of wetland influent. Other parameters in the pond system showed seasonal variation, and the upper layer of the pond water column became remarkably clear immediately after ice melt. Overall, the wetland system was found to be adequate for treating sewage with stable removal efficiency, and the subsequent pond was effective for further polishing. This study concerned agricultural reuse of reclaimed water using natural systems. Considering stable performance and effective removal of bacterial indicators as well as other water quality parameters, low maintenance, and cost-effectiveness, wetland- pond system was thought to be an effective and feasible alternative for agricultural reuse of reclaimed water in rural area.
