Hypoglycemic and Hypolipidemic Effects of Jerusalem artichoke Composites in Streptozotocin induced Diabetic Rats (명월초, 여주 및 울금을 포함하는 돼지감자 복합물의 streptozotocin 유발 당뇨쥐에서 혈당강하 및 체내 지질개선에 미치는 영향)

  • Hypoglycemic and hypolipidemic effects of Jerusalem artichoke composites (JAP) with extracts of G. procumbens (12.5%), M. charantia (12.5%), and C. longa (12.5%) to H. tuberosus concentrate (JA, 50%) were evaluated in streptozotocin (STZ) induced diabetic rats. Rats were divided into seven groups: normal (Normal), diabetic control (Diabetic), group fed G. procumbens extract (0.5 g/kg bw, D-GPE), group fed JAP (0.5 g/kg bw, D-JAP1; 1.5 g/kg bw, D-JAP2), group fed JA (0.5 g/kg bw, D-JA), and group fed Metformin (0.2 g/kg bw, D-MET) as a positive control. The blood glucose levels over 4 weeks were significantly decreased in the D-JAP2 and D-MET groups compared to the other groups after 3 weeks. The serum insulin level was not significant among the groups fed an experimental diet, but the HOMA-IR value was significantly decreased compared to the diabetic control group. AST and ALT activities in the serum were lowest in D-JAP1. Total lipid and triglyceride contents in the serum decreased in the groups fed an experimental diet, and the HDL-C contents of D-GPE, D-JAP1, and D-JAP2 were significantly increased compared to the diabetic control group. Triglyceride contents in the liver tissue were significantly lower in the D-GPE, D-JAP1, and D-JAP2 groups, and hepatic TBARS content was significantly decreased in the D-JAP1 and D-JAP2 groups compared to the diabetic control group. Hepatic antioxidative enzyme levels, such as SOD, catalase, and GSH-Px, were significantly elevated in groups fed an experimental diet compared to the diabetic control group. Therefore, JAP may be more effective than JA in the human body due to its hypoglycemic and hypolipidemic activities.

Study on the Optimum Range of Weight-Age Data for Estimation of Growth Curve Parameters of Hanwoo (한우의 체중 성장곡선 모수 추정을 위한 체중 측정 자료의 최적 범위에 관한 연구)

  • Mature weight (A) and rate of maturing (k) estimated by nonlinear regression were studied to determine the optimum age range over which the estimate of growth curve parameters can be estimated. The weight-age data from 1,133 Hanwoo bulls at Hanwoo Improvement Center of N.A.C.F. were used to fit the growth curve using Gompertz model. All available weight data from birth to the specific age of months were used for the estimation of parameters: the six specific ages used were 12, 14, 16, 18, 20 22 and 24 months of age. The mean estimates of mature weight (A) were 966.5, 1,255.9, 1,126.2, 916.5, 842.2, 780.9 and 767.0kg for ages 12 through 24 months, respectively. The mean estimates of mature weight (A) to 22 and 24 months of age were not different from each other. However, they were different from the estimates based on the data to other ages. Mean estimates of rate of maturing (k) were 3.362, 3.595, 3.536, 3.421, 3.403, 3.409 and 3.411 for ages 12 through 24 months, respectively. The mean estimates of maturing rate (k) for ages 18 through 24 months of age were not significantly different from each other. However, they were different from the estimates based on the data to other ages. Correlations among estimates of A at various ages showed the highest value of 0.93 between 22 and 24 months. Correlations among estimates of k at various ages were highest ranging from 0.91 to 0.99 among 18 to 24 months. The correlations between A and k were positive and tended to decrease with the increase of the age from 0.84 for the age of 12 months to 0.10 for the age of 24 months. Thus, the estimates of growth curve parameters, A and k, suitable for genetic studies can be derived from accumulated Hanwoo bulls after 22 months of age.

Purification of Complement System-Activating Polysaccharide from Hot Water Extract of Young Stems of Cinnamomum cassia Blume (계지(桂枝) 열수추출물로부터 보체계 활성화 다당의 정제)

  • A complement system-activating (anti-complementary) polysaccharide was purified from the hot water extract of young stems of Cinnamomum cassia Blume. Crude polysaccharide fraction (CC-1) was prepared from the hot water extract of the young stems followed by methanol-reflux, precipitation with ethanol, dialysis, and lyophilization. The anti-complementary activity of CC-1 was decreased greatly by periodate oxidation, but was not changed by pronase digestion. These suggest that carbohydrate moiety may be related to the activation of complement system. According to its ionic strength CC-1 was fractionated first using cetavlon to give 4 fractions, CC-2, 3, 4 and 5. Among them CC-2 fraction was found to retain the highest activity and yield. CC-2 was separated to an unabsorbed neutral sugar portion (CC-2-I) and seven absorbed acidic sugar fractions $(CC-2-II{\rightarrow}CC-2-VIII)$ on DEAE-Toyopearl 650C (Cl-). CC-2-III showing higher anti-complementary activity and yield than those of other fractions, was further purified on the gel permeation of Sephadex G-100 and Sepharose CL-6B to CC-2-IIIa-3. CC-2-IIIa-3 was determined to have a homogeneity hy GPC (Sepharose CL-6B) and HPLC. Gel chromatography using standard dextrans gave a value of $2.4{\times}10^5$ for the molecular weight. The purified polysaccharide, CC-2-IIIa-3 consisted of arabinose, xylose, glucose, galactose, galacturonic acid and glucuronic acid in a molar ratio of 5.56 : 3.77 : 1.87 : 1.00 : 5.12 : 3.13 and contained no nitrogen.

Effects of Wilting and Additives on the Fermentation Chrateristics , Quality and Aerobic Stability of Rye Silage (예건 및 첨가제가 호밀사일리지의 발효특성과 사료가치 및 호기적 안정성에 미치는 영향)

  • This experiment was conducted to evaluate effects of wilting and additives on fermetation characteristics, quality and aerobic stability of silage of rye(Seca1e cereale L.) harvested at the early heading stage. Harvested rye was wilted or mixed with beet pulp and then treated with formic acid or lactic acid bacteria(LAB) inoculant. Treatments were consisted of direct cut(DC), wilted(WT), beet pulp added(BP), direct cut and formate applied(DF), wilted and formate applied(WF), beet pulp and formate applied(BF), direct cut and LAB inoculated(DL), beet pulp added and LAB inoculated(BL), and wilted and LAB inoculated(WL) silages. Afler 70 days of ensiling period, the silages were opened and exposed to air for 7 days, and the silages of opening day(0 day) were compared with the silages exposed to air for 7 days(7 day). 1. LAB inoculated rye silages(DL, WL, BL) lowered pH values effectively eom the third day of ensiling and showed most stable pattern of pH changes during the initial fermentation process. Direct cut and formate applied(DF), beet pulp and formate applied(BF) and beet pulp added(BP) silages were also effective in lowering pH from the seventh day thai direct cut treatment(DC). 2. pH was low below 3.7 in all treatments of 0 day. After 7-day period of aerobic exposure, DL and WL showed considerable increases in pH from 3.5 and 3.4 to 8.3 and 6.4, respectively. 3. Direct cut rye silages(DF, DC, DL) and beet pulp and formate treatment(BF) produced effluent of 121.2, 85.9, 80.3 and 34.2 mlkg, respectively and these were greater than others(P<0.01). Beet pulp retained 1.61 I/kg of effluent at the application rate of 50 kg/t in comparing BP with DC, and formate application increased the amount of effluent in comparing DF and BF with DC and BP, respectively(P<0.01). 4. Beet pulp and LAB treatment(B1) was highest in content of lactate as 3.1% and formate treated silages(DF, WF, BF) showed lower content of lactate and total acid than others. LAB inoculated(D1, WL, BL) and wilted (WT) silages were graded to be good quality as a second group by the Flieg's score. Formate applied silages (DF, WF, BF) were graded as a foum group lower than DC by one. 5. Residual content of WSC of rye silage was higher in BP, BF, DL and BL than direct cut treatment(M3) (P< 0.01). Formate application had a tendency to increase the content of residual content of WSC. 6. For the ratio of NH3-N to Total N, wilted and LAB inoculated(W1) and direct cut and LAB inoculated(D1) silages were lowest on 0 day as 8.9 and 9.3% respectively. But after 7-day period of aerobic exposure, WL and DL showed largest increase of the ratio of $NH_3-N$ to Total N from 0 day(P<0.01). 7. On 0 day wilted(WT), beet pulp and formate treatment(BF), and beet pulp and LAB treatment(B1) were lower than direct cut treatment(DC) for NDF and ADF concentrations consistently. Formate applied silages (DF, WF, BF) made little change in ADF and NDF concentrations during 7 day period of aerobic exposure. DL and WL showed a large increase in ADF and NDF concentrations under aerobic condition(P<0.01). 8. IVDMD values of wilted(WT), beet pulp added(BP), wilted and LAB inoculated(W1) and beet pulp added and LAB inoculated(B1) silages were higher than direct-cut treatmentPC) as 84.7, 84.7, 84.4 and 83.0%, respectively on 0 day. But during 7-day period of aerobic exposure, a great decreae in IVDMD of WL was showed(P<0.01). The experimental results indicate that wilted silage(WT) could be recommended as the most effective treatment for reducing efluent and increasing quality and feed value of rye silage without deteriorating aerobic stability more than direct cut treatment(DC). Additionally, under unfavorable weather condition beet pulp added and LAB inoculated treatment(T3L) might be the possible alternative for successful ensiling of forage rye.

  • Effects of seed coating on establishment, early growth and herbage production were investigated in Pot, Soil box and Field experiments. Seed was coated with 60% Lime, 20% Phosphate and 20% Peatmoss(W/W), also finely sprayed 13% Arabic gum and 2% Methyl cellulose(W/V). Coating were generally 1:20, seed: coat(W/W). 1. In the general growth of the growing stages and different oversown species at 80 days, seed coated was promoted than none in early growth. (Table 2 and 3) 2. Stand counts at 60 days after oversown, seed coated was higher 15% than none, especially, there was advantage due to coating on the medium and large size seeds such as Orchardgrass, Perennial ryegrass and Kentucky bluegrass, but did not assist establishment of the large (Tall fescue) and small (Ladino clover) size seed. 3. Variation of sward composition was not quite different between two treatments, but seed coated was slightly increased the Percentage of sown grasses with advance of cutting time. 4. Total dry metter yield per unit area with seed coated was higher 17% than none in establishment year. 5. Variations in the chemical composition of the herbage and soil were influenced by coating materials in the coating regime (Table 8). 6. From these facts, seed coated also appeared that adequate seedling stands, early growth and herbage production could be obtained with higher than conventional overseeding method under a poor conditions.

평행식 진동탄환 암거 천공기의 연구 (IV)(V)-실기 설계 제작 및 보장실험-Development of Balanced-Type Oscillating Mole Drainer(IV)(V)

  • This paper is the forth and fifth one of the study on balanced type oscillating mole drainer. In the light of the results from previous reports about the model tests, some design criteria were established and a prototype machine was set up for experimental purpose. Motion characteristics and functionof the each parts of the machine were checked and analyzed. After that, performance tests of the prototype machine were carried out in thefield. Obtained results are summarized as follows ; 1. Ten centimeter of the bullet diameter was determined so as to be able to attach it to the tractors with capacity of 30 PS to 40 PS. 2. To maintain the balance between the moments of the front shank and rear shank, the oscillating amplitude of the rear bullet was determined to be larger than that of the front bullet. At the same time , the oscillating direction of the rear bullet was designed with the inclines of ten to thirty degrees. 3. An octagonal dynamo transduced was developed for measuring the compressive force of the upper link is measuring the draft force of the machine. Acceptable linear relationship between forces and strain responses from O.D.T. was obtained. 4. Analysing the balancing mechanism of the acting part of the machine , it was found that the total draft force of the machine was equal to the difference between the sum of the draft force produced from the right and left side bending moments of the lower drawber and the compressive force on the upper link. 5. There are acceptable linear relationship between the strain and twisting moment by driving shaft, and between strain and shank moment. Above results enable us to carry out the field experiment with prototype machine. 6. When the test machine was used in the field, it was possible to reduce the oscillating acceleration by forty percent in average as compared it with the single bullet mole drainer. 7. When the test machine was used under the oscillating condition, the dratt torce was reduced by 27 percent to 59 percent as compared it with the test machine under non-oscillating condition, while the draft force was increased by 7 percent to 20 percent as compared it with the mole drainer having oscillating single bullet. The reasoning behind this fact was considered as the resistance force due to the rear shank and bullet. 8. As the amplitude and frequency of the bullet were increased, the torque was increased accordingly. This tendency could be varied with the various characteristics of the given soils. And the larger frequency and amplitute, the more increasing oscil\ulcornerlating power but decreasing draft brce were needed, and draft force was increased as the velocity was increased.9. When the amplitude of the rear bullet was designed to be larger than that of the front bullet, the minimum value of the moment was lowered and oscillating acceleration was reduced. And when the oscillating direction of the rear bullet was declined back\ulcornerwards, oscillating acceleration was increased along with the increasing angle of decli\ulcornernation. When the test machine was operated in high speed, the difference between maximum moments and minimum ones became narrow. This varying magnitude of moments appeared on the moment oscillogram seems to be correlated to the oscillating acceleration and draft force. 10. From the analysis of variance, it was found that those factors such as frequency, amplitude, and operating velocity significantly affected in the oscillating acceleration, the draft resistance, the torque, the moment, and the total power required. And interaction between frequency and amplitude affected in the oscillating acceleration. 11. Within the given situation of this study, the most preferable operating conditions of the test machine were 7 Hz in oscillating frequency, 0.54 m/sec in operating velocity, and 39.1 mm in oscillating amplitude of front and rear bullets. However, it is necessary to select the proper frequency and magnitude of oscillation depending on the soil properties of the field in which the mole drainer is practiced by use of a bal1nced type oscillating mole drainer. 12. It is recommended that a comparative study of the mole drainers would be performed in the near future using two separate balanced oscillating bullet with the one which is operated by oscillating the movable bullet in a single cylinder or other balanced type which may be single oscillating bullet with spring, damper or balancing weight, and that of thing. To expand the applicability of the balanced type oscillating mole drainer in practical use, it is suggested to develop a new mechanism which perform mole drain with vinyl pipe or filling material such as rice hull.

The Research of Comparison Evaluation on the Decline in Artifact Using Respiratory Gating System in PET-CT (PET-CT 검사 시 호흡동조 시스템을 이용한 인공물 감소에 대한 비교 평가)

  • Purpose Among various causes that influence image quality degradation, various methods for decrease in Artifact occurred by respiration of patients are being used. Among them, this study intended to evaluate CTAC Shift correction method and additional scan compare to the Scan(Q static scan) using respiratory gated system. Materials and Methods This study was conducted on 10 patients, and used PET-CT Discovery 710 (GE Healthcare, MI, USA) and Varian's RPM system. 5.18 Mbq per kg of $^{18}F$-FDG was injected on patients, asked them to take a rest for 1 hour in the bed, and conducted test after urination. Images were visualized through Q static scan, CTAC Shift correction method, Additional scan based on the Whole body scan(WBS) with Artifact. Decrease in Artifact was compared in each image, conducted Gross Evalution, and measured changes of SUVmax. Results For image obtained through the CTAC Shift correction method through WBS with Artifact, 12~56%, Q static scan image showed 17~54% of change rate and Additional Scan showed -27~46% of change rate. In Blind Test, the CTAC Shift correction image showed the highest point with 4 points, Q static scan image showed 3.5 points, and Additional scan image showed 3.4 points. The standardized WBS scan through Oneway ANOVA and three types of Scan method showed significant difference(p<0.05), and did not show significant difference between the three Scan methods(p>0.05). However, the three Scan methods showed significant difference in Blind test. Conclusion Additional scan and Q static scan require more time than the CTAC Shift correction method, there is concern about excessive exposure to patients by CT rescan and Q static scan is difficult to apply on patients with inconsistent respiration or irregular respiration cycle due to pain. For CTAC Shift correction method, limited correction is possible and the range is limited as well. It is considered as a useful method of improving diagnostic value when hospitals use the system appropriately and develop various advantageous factors of each method.

Control Efficacy of Bacillus thuringiensis against Herpetogramma luctuosalis on 'Campbell Early' Organic Vineyard (비티제를 이용한 포도 '캠벨얼리' 유기과수원 포도들명나방 방제효과)

  • The experiment of grape leafroller (Herpetogramma luctuosalis) was carried out at Okcheon area from 2007 to 2009 and 2015. The grape leafroller had been occurred at a campbell early' organic vineyard in Okcheon. It's larva was rolling the leaf of grape and ate the leaf. So the leaf of grape decreased. In organic vineyards, adult grape leafroller' generation rate per year showed the first peak in mid-June, the second peak in early -August and the third in mid-September. The larva showed the first peak in early July and the second peak in late August-early September. The grape leaf roller had three generations per year. And it took $60.9{\pm}1.09days$ from egg to adult in growth chamber (VS-91G09M-1300) which the relative humidity conditions was $60{\pm}10%$, temperature $25{\pm}2^{\circ}C$ and photoperiod 16L:8D (The egg : $12{\pm}0days$, larvae : $22.2{\pm}0.22days$, pupa : $10.6{\pm}0.75days$ and adult : $16.1{\pm}0.45days$). It was conducted to find out the effect of microbial pesticide treatments to control H. luctuosalis. The 4 microbial pesticides (Bacillus thurigiensis) were treated twice on the grape leaves in June 11 and 21 at an organic vineyard in Okcheon. On 10 days after last treatment, the control value of all microbial pesticides were more than 95%. When the dates of spraying to the grape leaves were on May 22, June 12 and July 2 each, the effects of microbial pesticide were 73.9%, 93.5% and 43.6% respectively. As a result, it was effective that Bt was sprayed to grape leaves on mid and late June for controling the H. luctuosalis in organic vineyard. And microbial pesticide Bt was thought to be useful to control the grape leafroller in organic vineyard.

Formation of Al0.3Ga0.7As/GaAs Multiple Quantum Wells on Silicon Substrate with AlAsxSb1-x Step-graded Buffer (AlAsxSb1-x 단계 성분 변화 완충층을 이용한 Si (100) 기판 상 Al0.3Ga0.7As/GaAs 다중 양자 우물 형성)

  • The $AlAs_xSb_{1-x}$ step-graded buffer (SGB) layer was grown on the Silicon (Si) substrate to overcome lattice mismatch between Si substrate and $Al_{0.3}Ga_{0.7}As$/GaAs multiple quantum wells (MQWs). The value of root-mean-square (RMS) surface roughness for 5 nm-thick GaAs grown on $AlAs_xSb_{1-x}$ step-graded buffer layer was ~1.7 nm. $Al_{0.3}Ga_{0.7}As$/GaAs MQWs with AlAs/GaAs short period superlattice (SPS) were formed on the $AlAs_xSb_{1-x}$/Si substrate. Photoluminescence (PL) peak at 10 K for the $Al_{0.3}Ga_{0.7}As$/GaAs MQW structure showed relatively low intensity at ~813 nm. The RMS surface roughness of the $Al_{0.3}Ga_{0.7}As$/GaAs MQW structure was ~42.9 nm. The crystal defects were observed on the cross-sectional transmission electron microscope (TEM) images of the $Al_{0.3}Ga_{0.7}As$/GaAs MQW structure. The decrease of PL intensity and increase of RMS surface roughness would be due to the formation of the crystal defects.

Improving Diagnostic Accuracy for Malignant Nodes and N Staging in Non-Small Cell Lung Cancer Using CT-Corrected FDG-PET (비소세포폐암에서 CT-보정 양전자단층촬영술을 이용한 악성 림프절 평가 및 N 병기 결정 성적 향상)

  • Purpose: We investigated prospectively whether the interpretation considering the patterns of FDG uptake and the findings of unenhanced CT for attenuation correction can improve the diagnostic accuracy for assessing malignant lymph node (LN) and N stage in non-small cell lung cancor (NSCLC) using CT-corrected FDG-PET (PET/CT). Materials & Methods: Subjects were 91 NSCLC patients (M/F 62/29, age: $60{\pm}9$ yr) who underwent PET/CT before in dissection. We evaluated the maximum SUV (maxSUV), patterns of FDG uptake, short axis diameter, and calcification of LN showing abnormally increased FDG uptake. Then we investigated criteria improving the diagnostic accuracy and correlated results with postoperative pathology. In step 1, in was classified as benign or malignant based on maxSUV only. In step 2, LN was regarded as benign if it had lower maxSUV than the cut-off value of step 1 or it had calcification irrespective of its maxSUV. In step 3, LN regarded as malignant in step 2 was classified as benign if they had indiscrete margin of FDG uptake. Results: Among 432 LN groups surgically resected (28 malignant, 404 benign), 71 showed abnormally increased FDG uptake. We determined the cut-off as maxSUV=3.5 using ROC curve analysis. The sensitivity, specificity, and accuracy for assessing malignant LN were 64.3%, 86.9%, 85.4% in step 1, 64.3%, 95.0%, 93.1% in step 2, and 57.1%, 98.0%, 95.4% in step3, respectively. The accuracy for assessing N stage was 64.8% in step 1, 80.2% in step 2, and 85.7% in step 3. Conclusion: interpreting PET/CT, consideration of calcification and shape of the FDG uptake margin along with maxSUV can improve the diagnostic accuracy for assessing malignant involvement and N stage of hilar and mediastinal LNs in NSCLC.
