Preparation of ZrC/SiC by Carbothermal Reduction of Zircon (지르콘의 탄소열환원에 의한 ZrC/SiC의 합성)

  • The preparation of ZrC/SiC mixed powders from $ZrSiO_4/C$ and $ZrSiO_4/Al/C$ systems was attempted in the temperature range below $1600^{\circ}C$ under Ar or $Ar/H_2$ gas flow(100-500ml/min). The formation mechanism and kinetics of ZrC/SiC were suggested and the resultant powders were characterized. In $ZrSiO_4/C$ system, ZrC and SiC were formed by competitive reaction of $ZrO_2(s)$ and SiO(g) with carbon at temperature higher than $1400^{\circ}C$. The apparent activation energy for the formation of ZrC was approximately 18.5kcal/mol($1400-1600^{\circ}C$). In $ZrSiO_4/Al/C$ system, ZrC was formed by reaction of ZrO(g) with Al(l, g) and carbon at temperature higher than $1200^{\circ}C$, and SiC was formed by reduction-carbonization of SiO(g) with Al(l, g) and carbon at temperature higher than $1300^{\circ}C$. The products obtained at $1600^{\circ}C$ for 5h consisted of ZrC with lattice constant of $4.679{\AA}$ and crystallite size of $640{\AA}$, and SiC with lattice constant of $4.135{\AA}$ and crystallize size of $500{\AA}$. And also, the mean particle size was about $21.8{\mu}m$.

  • High performance liquid chromatography (HPLC) analysis was applied for determination of guanine plus cytosine (G + C) contents of DNA of Butyrivibrio fibrisolvens. By values of G + C contents, a reference strain and 20 wild strains of B. fibrisolvens were classified into at least two distinct subgroups, i.e. G + C contents of 18 strains were 38-40 mol% and those of 3 strains including the reference strain were 43-45 mol%. Clear relationships were not observed between G + C contents and biological properties of 21 strains of B. Fibrisolvens.

High-Temperature Behavior of Ba-Doped Boehmite Hydrothermally Prepared from $Al(OH)_3$ and $Ba(OH)_2$

  • Minute boehmite crystals with high aspect rations, which were hydrothermally synthesized from gibbsite in $Ba(OH)_2$ solution, occluded Ba with the Ba/Al molar ratio of about 0.03 in their interlayers. Their surface areas were about 14$\m^2$/g. The Ba-intercalated bohemite samples were partly used for producing $BaAl_{12}O){19}$ with low sinterability by externally supplementing $Ba(OH)_2$, and for forming transient aluminas. The surface area of $BaAl_{12}O){19}$ obtained by firing at $1500^{\circ}C$ for 3 h was 5.3$\m^2$/g, which was significantly lower than 12$\m^2$/g of the sol-gel origin. While a mixture ${\gamma}$-alumina and BaO is known to from $BaAl_{12}O){19}$ at $1200^{\circ}C$, solid state reaction between η-alumina transformed from the Ba-intercalated boehmite and BaO formed from $Ba(OH)_2$ deposited on the boehmite started above $1300^{\circ}C$. This suggests that large sized $Ba^{2+}$ ion occluded in η-alumina considerably suppresses the diffusion of $Al^{3+}$ ion. The surface area of the Ba-intercalated boehmite fired at $1400^{\circ}C$ for 3h was as high as 14$\m^2$/g indicative of its potential applicability to combustion catalysts. But it was decreased to 5.0$\m^2$/g after firing at $1500^{\circ}C$ for 3 h, accompanied by abrupt formations of $\alpha$-alumina and $BaAl_{12}O){19}$ as main products. The suppression of $\alpha$-alumina formation up to $1400^{\circ}C$ also suggests the significant blocking effect of $Ba^{2+}$ ion on the diffusion of the component ions.

Marine Algal Flora and Community Structure in Daejin on the Mid-East Coast of Korea (동해안 중북부 대진 해역의 해조상 및 군집구조)

  • Seaweed biomass was estimated in the rocky subtidal zone of Daejin on the mid-east coast of Korea using a destructive sampling method from February to November 2007. Monthly sampling was conducted at depths of 5, 10 and 15 m using 50 $\times$ 50 cm quadrats. A total of 73 species, including 5 chlorophytes, 25 phaeophytes, 43 rhodophytes and one seagrass were identified. Average seaweed biomass was 2,852.3 g wet wt. $m^{-2}$ and biomass values varied monthly from 1,189.8 g to 7,523.7 g. The species dominant in biomass were Laminaria japonica (481.7 g), Sargassum confusum (470.85 g), Undaria pinnatifida (422.57 g), Costaria costata (282.16 g), Odonthalia corymbifera (174.46 g), S. sagamianum (163.22 g) and S. horneri (122.18 g). The vertical distribution of algae was characterized by S. confusum and U. pinnatifida at 5 m, L. japonica and U. pinnatifida at 10 m, and O. corymbifera and L. japonica at 15 m depth. The C/P, R/P and (R+C)/P values were 0.20, 1.72 and 1.92, respectively.

  • Karyotypes are described for Juniperus rigida Sieb. et zucc, in two provenances of Gyeong-nam and Choong-puk. Chromosome numbers of two provenances, are 2n=22. The most common feature of mitotic chromosomes was shown at the chromosome 7, which has secondary constriction on the short arm. And the most differential chromosome was shown at chromosome 9 from Gyeong-nam and chromosome 5 from Choong-puk provenance which bore secondary constriction. The karyotype formulae are as follows; Gyeong-nam, Jinyang provenance race is $$K(2n)=22=2A^m+2B^m+2C^m+2D^{sm}+2E^{st}+2F^m+2^{sc}G^m+2H^m+2^{sc}I^t+2J^{st}+2K^m$$ Choong-puk, Jechun provenance race is $$K(2n)=22=2A^m+2B^m+2C^m+2D^{st}+2^{sc}E^{sm}+2F^m+2^{sc}G^m+2H^m+2I^m+2J^{st}+2K^{sm}$$.

The Characteristics of Holocellulose Aerogel (홀로셀룰로오스 에어로겔의 특성)

  • This study was carried out to investigate the characterization of aerogel made by holocellulose, the thermal properties of the aerogel, and its shapes and porous structures. The aerogel was made by holocellulose through the gelation in alkali hydroxide-urea solution and freeze drying processes. Holocellulose aerogel had porous structure such as net or sponge. The density of holocellulose aerogel was 0.04 g/$cm^3$, and the specific surface area 145.3 $m^2$/g. Although thermal degradation occurred in the range of $210{\sim}350^{\circ}C$, significant thermal degradation occurred at low temperature with low heating rate, Micropore volume was sharply increased with low heating rate. Holocellulose aerogel char obtained by carbonization with $900^{\circ}C$ and $0.5^{\circ}C$/min. heating rate had the highest surface area, 656.7 $m^2$/g. The deformed and irregular structures of holocellulose aerogel chars due to the thermal degradation were observed in SEM.

Identification of Phospholipase C Activated by $GTP{\gamma}S$ in Plasma Membrane of Oat Cell

  • In order to investigate whether phospholipase C (PLC) activity in oat celIs is regulated by Gprotein, we have characterized PLC in plasma membranes of oat tissues. To identify the purified plasma membrane, $K^+$-stimulated, $Mg^{2+}$-dependent ATPase activity was measured. The activity of ATPase was shown to be proportional to the concentration of membrane protein. To examine the PLC activity regulated by G-protein, we used the inside-out and outside-out plasma membrane mixture isolated from the oat cells. The plasma membrane mixture showed higher PLC activity than the one of the outside-out plasma membrane. This suggests that PLC activity is located at the cytoplasmic surface of plasma membrane. PLC activity in plasma membrane mixture was dependent on $Ca^{2+}$ with maximum activity at 100 ${\mu}m$ $Ca^{2+}$ and it was inhibited by 1 mM EGTA. Using Sep-pak $Accell^{TM}$ Plus QMA chromatography, we found that inositol 1,4,5-trisphosphate ($IP_3$) was produced in the presence of 10 ${\mu}m$ $Ca^{2+}$. The PLC activity in the membrane was enhanced by an activator of G-protein ($GTP{\gamma}S$) and not by an inhibitor ($GDP{\beta}S$). This indicates that a G-protein is involved in the activation of PLC in the plasma membrane of oat cells.

  • The impurities in isoprene were determined using GC/MS. There were good linearities (above $R^2=0.996$) in the range of the $1.0-50{\mu}g/mL$ and 0.5-5.0%, and the detection limits of the method were below $0.1{\mu}g/mL$ for each impurities. Reagent-grade isoprene contained isopropenyl acetylene and 2-butyne at $10-50{\mu}g/mL$ and the concentrations of cis-piperylene, trans-piperylene, cyclopentadiene and dicyclopentadiene were below $1{\mu}g/mL$. The analysis confirmed that the reagent grade isoprene could be obtained from the distillation of C5 fractions.

Effects of Heat Treatments on the Antioxidant Activities of Fruits and Vegetables (과채류의 항산화 활성에 미치는 열처리 효과)

  • The effects of heat treatments on the antioxidant activities of selected fruits and vegetables were investigated by heating at various temperatures (110, 120, 130, 140, and 150$^{\circ}C$) for 2 hr. The examined fruits and vegetables included tomatoes (Lycopersicon esculentum), oriental melon (Cucumis melon var. makuwa), apples (Malus pumila Miller var. domestica Schneider), melon (Cucumis melon), watermelon (Citrullus vulgaris), and banana (Musa sapientum). The total polyphenol and total flavonoid contents of the juices from heated samples were quantified spectrophotometrically, and their antioxidant activities were determined using DPPH and ABTS radicals. As the heating temperature increased, antioxidant activity also increased. The highest total polyphenol content (2.80 mg/g) occurred in the oriental melon with heating at 150$^{\circ}C$, and this value was 7 times higher than that of the untreated oriental melon (0.40 mg/g). The highest total flavonoid content (148.80 ${\mu}$g/g) occurred in the melon heat treated at 150$^{\circ}C$, and this value was 37 times higher than that of the untreated melon (4.54 ${\mu}$g/g). DPPH radical-scavenging activity was the highest in the watermelon treated at 150$^{\circ}C$ (84.37%, 0.50 mg/g), and this value was 40 times higher than that of the untreated watermelon. Finally, the highest ascorbic acid (AA) equivalent antioxidant capacity (AEAC) value (239.50 mg AA eq/g) was obtained in the watermelon heat treated at 150$^{\circ}C$ for 2 hr (control = 18.35 mg AA eq/g).

Effects of Aqueous Chlorine Dioxide and UV-C Irradiation on Decontamination and Growth of Microbes during Chilled Storage of Celery and Cherries (이산화염소수, Ultraviolet-C 또는 병합처리가 샐러리와 체리에서의 살균 및 냉장저장 중 미생물 성장에 미치는 효과)

  • The effects of a combined treatment of aqueous chlorine dioxide ($ClO_2$) and ultraviolet-C (UV-C) irradiation on microbial growth in celery and cherries were investigated. Celery and cherry samples were treated with 50 ppm $ClO_2$, UV-C at dose of 10 kJ/$m^2$, and a combination of $ClO_2$ and UV-C. The changes in the counts of Escherichia coli O157:H7 inoculated in the celery and cherries as well as those of total aerobic bacteria, yeast and molds in the celery and cherries were investigated after each treatment. After the combined treatment of aqueous $ClO_2$ and UV-C irradiation, the populations of E. coli O157:H7 in the inoculated celery and cherries were reduced by 2.8 and 3.0 log CFU/g, respectively, compared to those of the control. For the un-inoculated celery and cherries, the populations of total aerobic bacteria were reduced by 2.9 and 1.8 log CFU/g, respectively, compared to the control. In addition, the populations of yeast and molds were decreased by 1.8 and 1.2 log CFU/g, respectively. These results suggest that the combined treatment of 50 ppm $ClO_2$ and UV-C at a dose of 10 kJ/$m^2$ would be an effective technology for decontamination and improving the microbiological safety in celery and cherries during chilled storage.
