Abundance of Harmful Algae, Cochlodinium polykrikoides, Gyrodinium impudicum and Gymnodinium catenatum in the Coastal Area of South Sea of Korea and Their Effects of Temperature, Salinity, Irradiance and Nutrient on the Growth in Culture (남해안 연안에서 적조생물, Cochlodinium polykikoides, Gyrodinium impudicum, Gymnodinium catenatum의 출현상황과 온도, 염분, 조도 및 영양염류에 따른 성장특성)

  • Three harmful algal bloom species with similar morphology, Cochlodinium polykrikoides, Gyodinium impudicum and Gymodinium catenatum have damaged to aquatic animals or human health by either making massive blooms or intoxication of shellfishes in a food chain. Eco-physiological and hydrodynamic studies on the harmful algae offer useful informations in the understanding their bloom mechanism by giving promising data for the prediction and modelling of harmful algal blooms event. Thus, we studied the abundance of these species in the coastal area of South Sea of Korea and their effects of temperature, salinity, irradiance and nutrient on the growth for the isolates. The timing for initial appearance of the three species around the coastal area of Namhaedo, Narodo and Wando was between Bate July and late August in 1999 when water temperature ranged from $22.8^{\circ}C\;to\;26.5^{\circ}C$ Vegetative cells of C. polykrikoides and G. impudicum were abundant until late September when water temperature had been dropped to less than $23^{\circ}C$. By contrast, vegetative cell of G. catenatum disappeared before early September, showing shorter period of abundance than the other two species in the South Sea. Both G. impudicum and G. catenatum revealed comparatively low density with a maximal cell density of 3,460 cells/L and 440 cells/L, respectively without making any bloom, while C. polykrikoides made massive blooms with a maximal cell density more than $40\times10^6$cells/L, The three species showed a better growth at the relatively higher water temperature ranging from 22 to $28^{\circ}C$ with their maximal growth rate at $25^{\circ}C$ in culture, which almost corresponded with the water temperature during the outbreak of C. polykrikoides in the coastal area of South Sea. Also, they all showed a relatively higher growth at the salinity from 30 to $35\%$. Specially, G. impudicum showed the euryhalic characteristics among the species, On the other hand, growth rate of G. catenatum decreased sharply with the increase of water temperature at the experimental ranges more than $35\%$. The higher of light intensities showed the better growth rates for the three species, Moreover, C. polykrikoides and G. impudirum continued their exponential growth even at 7,500 lux, the highest level of light intensity in the experiment, Therefore, It is assumed that C. polykrikoides has a physiological capability to adapt and utilize higher irradiance resulting in the higher growth rate without any photo inhibition response at the sea surface where there is usually strong irradiance during its blooming season. Although C. poiykikoides and G. impudicum continued their linear growth with the increase of nitrate ($NO_3^-$) and ammonium ($NH_4^-$) concentrations at less than the $40{\mu}M$, they didn't show any significant differences in growth rates with the increase of nitrate and ammonium concentrations at more than $40{\mu}M$, signifying that the nitrogen critical point for the growth of the two species stands between 13.5 and $40{\mu}M$. Also, even though both of the two species continued their linear growth with the increase of phosphate ($PO_4^{2-}$) concentrations at less than the $4.05{\mu}M$, there were no any significant differences in growth rates with the increase of phosphate concentrations at more than $4.05{\mu}M$, signifying that the phosphate critical point for the growth of the two species stands between 1.35 and $4.05{\mu}M$. On the other hand, C. polykrikoides has made blooms at the oligotrophic environment near Narodo and Namhaedo where the concentration of DIN and DIP are less than 1.2 and $0.3{\mu}M$, respectively. We attributed this phenomenon to its own ecological characteristics of diel vertical migration through which C. polykrikoides could uptake enough nutrients from the deep sea water near bottom during the night time irrespective of the lower nutrient pools in the surface water.

Assessment of Internal Dose by $^3H\;&\;^{14}C$ of Total Diet for Inhabitants near Wolsung Nuclear Power Plants

  • To assess the internal dose by $^3H\;&\;^{14}C$ in total diet of inhabitants near Wolsung Nuclear Power Plants, TFWT, OBT and $^{14}C$ concentration in total diet was analyzed for collection region and time. TFWT, OBT and $^{14}C$ concentrations were in the range of 3.19-42.2 Bq/L, 1.00-39.4 Bq/L, and 0.230-0.855 Bq/gC, respectively. The calculated annual effective dose with TFWT, OBT and $^{14}C$ is $6.10{\times}10^{-5}mSv/y,\;3.71{\times}10^{-5}mSv/y\;and\;7.08{\times}10^{-3}mSv/y$, respectively. And then annual internal dose with total diet for inhabitants near Wolsung NPPs is about $7.18{\times}10^{-3}mSv/y$, which is about 0.72% of annual effective dose limit 1 mSv/y.

Optimization of γ-Aminobutyric Acid (GABA) Production Using Immobilized Lactobacillus plantarum K154 in Submerged Culture of Ceriporia lacerata (Ceriporia lacerata 배양액과 고정화 Lactobacillus plantarum K154를 이용한 감마아미노뷰티르산 생산 최적화)

  • The production of GABA was optimized by co-cultivation of immobilized Lactobacillus plantarum K154 (ILK) with Ceriporia lacerata cultures. The mycelial culture of C. lacerata was performed in a defined medium containing 3% glucose, 3% soybean flour, and 0.15% $MgSO_4$ in a submerged condition for 7 days at $25^{\circ}C$, resulting in the production of 29.7 g/L mycelia, 3.1 g/L exopolysaccharides, 2% (w/w) ${\beta}$-glucan, 68.96 unit/mL protease, and 10.37 unit/mL ${\alpha}$-amylase. ILK in C. lacerata culture showed viable cell counts of $3.13{\time}10^9CFU/mL$ for immobilized cells and $1.48{\time}10^8CFU/mL$ for free cells after 1 day. GABA production in the free and immobilized cells was 9.96 mg/mL and 6.30 mg/mL, respectively, after 7 days. A recycling test of ILK in the co-fermentation was consequently performed five times at $30^{\circ}C$ for 15 days, resulting in the highest production of GABA. GABA could also be efficiently overproduced by co-cultivation with the produced polysaccharides, ${\beta}$-glucan, peptides, and probiotics.

Antioxidant and Acetylcholinesterase Inhibitory Activities of Subcritical Water Extracts from Houttuynia cordata Thunb (어성초의 아임계수 추출물의 항산화 활성 및 Acetylcholinesterase 저해 활성)

  • Houttuynia cordata Thunb powder (0.05 g) was placed in a stainless vessel containing 10 mL of water, and subcritical water (SCW) extraction was carried out at various temperatures (50, 100, 200, and $300^{\circ}C$) for 10, 30, and 60 min. SCW treatment significantly affected physiologically important compounds, antioxidant activity, and acetylcholinesterase inhibitory activity of H. cordata Thunb extracts. H. cordata Thunb extracted at $300^{\circ}C$ for 30 min had the highest quercetin content, DPPH radical scavenging activity, total phenolic content, and acetylcholinesterase inhibitory activity, whereas the extract at $100^{\circ}C$ for 10 min possessed the highest flavonoid content. The highest ABTS radical scavenging activity was found in H. cordata Thunb extracted at 200 and $300^{\circ}C$ for 60 min. These results indicate that SCW extraction might be a useful processing method for obtaining valuable materials from H. cordata Thunb.

  • $Li_4Mn_5O_{12}$ was synthesized by combustion method using $LiNO_3$, $Li(CH_3COO){\cdot}2H_2O$ and $Mn(CH_3COO)_2{\cdot}4H_2O$. $Li_4Mn_5O_{12}$ was obtained over $400^{\circ}C$, however, the sample calcined at $400^{\circ}C$ for any time was mixed phases of $Li_4Mn_5O_{12}$ and $Mn_2O_3$. $Li_4Mn_5O_{12}$ calcined at $400^{\circ}C$ for 5 h had larger first discharge capacity (41.5mAh/g) at 1C-rate for 3.7~4.4V than other calcined samples. Moreover, applying to hybrid capacitor, it had good discharge capacity (24.74 mAh/g or 10.46 mAh/cc) at 100 mA/g for 1~2.5 V and higher energy density (39Wh/kg or 16.49Wh/cc) at same condition.

  • Objectives : The purpose of this study was to investigate the anti-inflammatory effect of Cobrotoxin on binding affinity of cobrotoxin with P50, $IKK{\alpa}$ and $IKK{\beta}$, activities of NF-${\kappa}B$, Cell viability of astrocyte, expressions of protein molecules of NF-${\kappa}B$ such as P50, P-$1{kappa}B$, $1{\kappa}B$ and iflammation related genes such as Cox-2, iNOS, cPLA2 in the SNP or LPS induced Inflammatory pathway of Rats' astrocytes. Methods : In this study, The expression of cytosolic phospholipase A2, Nitric oxcide, Cyclooxygenase-2 and inducible nitrogen oxide synthase was determined by western blotting with corresponding antibodies, and the generation of NF-${\kappa}B$ was assayed by EMSA method in astrocytes of rats. The Cell viability of astrocytes was determined by MTT assay, and Binding affinity of Cobrotoxin with P50, $IKK{\alpha}$ and $IKK{\beta}$ was assayed by Surface plasmon resonance analysis, and NF-${\kappa}B$ dependent luciferase activity was determined by luciferase analysis, and Uptake of cobrotoxin in astrocytes was identified by Confocal laser scanning microscope Results : 1. Compared with control, LPS-induced NF-${\kappa}B$ DNA binding activity was decreased significantly by 0.1, $0.5{\mu}g/m{\ell}$ of Cobrotoxin in Astrocyte. 2. Compared with control, LPS-induced NF-kB dependent luciferase expression was decreased significantly by 0.1, 0.5 and $1{\mu}g/m{\ell}$ of Cobrotoxin in Astrocyte. 3. Compared with control, SNP induced P50, $I{\kappa}B$ expressions in astrocyte were decreased significantly by 0.1, 0.5 and $1{\mu}g/m{\ell}$ of Cobrotoxin and P-$1{\kappa}B$ expression was decreased significantly by 0.5 and $1{\mu}g/m{\ell}$ of Cobrotoxin. 4. Compared with control, LPS induced P50, $1{\kappa}B$ expressions in astrocyte were decreased significantly by 0.5 and $1{\mu}g/m{\ell}$ of Cobrotoxin. 5. Compared with control, SNP induced Cox-2, iNOS, CPLA2 expressions in astrocyte were decreased significantly by $1{\mu}g/m{\ell}$ of Cobrotoxin. 6. Compared with control, LPS induced Cox-2, cPLA2 expressions in astrocyte were decreased significantly by 0.1, 0.5, $1{\mu}g/m{\ell}$ of Cobrotoxin and iNOS expression was decreased significantly by 0.5, $1{\mu}g/m{\ell}$ of Cobrotoxin. 7. Compared with $0.5{\mu}g/m{\ell}$ of Cobrotoxin, SNP-induced NF-${\kappa}B$ DNA bindins activity in astrocyte was increased significantly by Cobrotoxin $0.5{\mu}g/m{\ell}$ with DTT 1mM and Cobrotoxin $0.5{\mu}g/m{\ell}$ with DTT 5mM. 8. Compared with $0.5{\mu}g/m{\ell}$ of Cobrotoxin, LPS-induced NF-${\kappa}B$ DNA binding activity in astrocyte was increased significantly by Cobrotoxin $0.5{\mu}g/m{\ell}$ with DTT 1mM, Cobrotoxin $0.5{\mu}g/m{\ell}$ with DTT 5mM, Cobrotoxin $0.5{\mu}g/m{\ell}$with GSH 1mM and Cobrotoxin $0.5{\mu}g/m{\ell}$ with GSH 5mM 9. Compared with $0.1{\mu}g/m{\ell}$ of cobrotoxin, SNP induced P50 expressions in astrocyte were increased significantly by Cobrotoxin $0.5{\mu}g/m{\ell}$ with DTT 1mM, Cobrotoxin $0.5{\mu}g/m{\ell}$ with DTT 5mM Cobrotoxin $0.5{\mu}g/m{\ell}$ with GSH 1mM and Cobrotoxin $0.5{\mu}g/m{\ell}$ with GSH 5mM. 10. The uptake of the labeled cobrotoxin into the cells was shown under a confocal laser scanning microscope. cobrotoxin was uptaken into the membrane and nucleus of astrocytes. Conclusions : In summary, the present results demonstrate that cobrotoxin directly binds to sulfhydryl group of p50 and IKKS resulting In the reduction of translocation of p50 and IkB release, thereby inhibits activation of NF-${\kappa}B$, and suggest that pico to nanomolar range of cobrotoxin could inhibit the expression of genes in the NF-${\kappa}B$ signal pathway.

  • In this study, a polymer-carbon composite material is prepared for environmental-friendly organic anode. On poly(styrenesulfonate)(PSS)-carbon composite anode, the carbon is coated by PSS as a core-shell structure and the PSS-carbon composite anode has $524mAh\;g^{-1}$ theoretical capacity with <0.6V voltage. Moreover, the PSS-carbon composite anode shows $519.6mAh\;g^{-1}$, $461.2mAh\;g^{-1}$, $411.8mAh\;g^{-1}$ and $315.9mAh\;g^{-1}$ discharge capacities at 0.1, 0.5, 1 and 10 C, respectively, and stable cycle performance up to 30 cycles. The PSS-carbon composite anode, containing polystyrene and sulfonate functional groups, is suitable for high electrochemical properties organic rechargeable battery.

Effect of Water Temperature and Stocking Density on Growth of Juvenile Red Drum Sciaenops ocellatus (사육수온과 밀도가 홍민어 Sciaenops ocellatus의 성장에 미치는 영향)

  • Fed on commercial flounder diet at 20, 23 and $26^{\circ}C$ in semiclosed culture system for 32 weeks, the juvenile red drum (1.2 g) showed linear increase in daily feeding rate (DFR), growth (g/fish) and specific growth rate (SGR) with increasing temperature. In the second experimental series, the young red drum (214 g), cultured at densities of 2.16, 4.24 and 6.40 kg/$m^3$ in flow-through tanks at water temperatures from 12.3 to $27.2^{\circ}C$ for 25 weeks, grew faster at the stocking density of 2.16 kg/$m^3$ than at the densities of 4.24 and 6.40 kg/$m^3$ the difference in growth observed at the stocking densities of 4.24 and 6.40 kg/$m^3$ was not significant. The DFR and SGR were also significantly higher for the density group of 2.16 kg/$m^3$. Briefly, growth of the red drum increased with increasing tested range of temperature and was also faster with decreasing stocking density. However, the total growth (g/tank) increased with increasing stocking density.

Dust Spatial Distribution and Emission of Tunnel Ventilated Windowless Broiler Building (터널환기 무창육계사의 분진크기별 분포와 배출농도에 관한 연구)

  • This study investigated the spatial distribution of dust originating from tunnel-ventilated windowless broiler building measuring 12 m wide, 61 m long, with a side wall height of 3 m and a capacity of 16,982 birds. Dust concentrations in terms of total suspended particles (TSP), and particulate matter of sizes $10\;{\mu}m$(PM10), $2.5\;{\mu}m$(PM2.5), and $1\;{\mu}m(PM1)$ were measured at 30 minutes interval by using GRIMM Aerosol Monitor (GRIMM AEROSOL). The spatial distribution of dust showed the lower dust concentration in the inlet than in the outlet of the tunnel ventilation, and dust concentration decreasing as the dust size decreased, as follows: $317.9\;{\mu}g/m^3$ TSP; $74.7{\mu}m/m^3$ PM10; $9.7\;{\mu}g/m^3$ PM2.5; and $6.2\;{\mu}g/m^3$ PM1 in the inlet; and $2,678.5\;{\mu}g/m^3$ TSP; $555.5\;{\mu}g/m^3$ PM10; $33.3\;{\mu}g/m^3$ PM2.5; and $10.2\;{\mu}g/m^3$ PM1 in the outlet. The dust concentration emitted from the tunnel ventilated fan was $446.6\;{\mu}g/m^3$ TSP; $129.1\;{\mu}g/m^3$ PM10; $15.8\;{\mu}g/m^3$ PM2.5; and $6.1\;{\mu}g/m^3$ PM1 in the 3 meters from the fan and $25.1\;{\mu}g/m^3$ TSP; $8.8\;{\mu}g/m^3$ PM10; $5.6\;{\mu}g/m^3$ PM2.5; and $4.9\;{\mu}g/m^3$ PM1 in the 50 meters from the fan.
