Effects of Organic Composts on Soil and Yield Characteristics of Boxthorn(Lycium chinense Mill.) Organic Cultivation

  • For the development of Boxthorn organic cultivation techniques, we investigated effect of several organic compost as a foundation fertilizer and growing plant fertilizer. And we adopted partly opening rain shelter greenhouse to protect anthracnose. In organic compost applying test yield characteristics of 'Mixed organic compost' treatment was the best but in betaine content measurement of dried fruit, 'Mixed organic compost and nitrogen guano' treatment was the best and it's chlorophyll and 100 fruit weight were also better than the other treatments.

주당 적정줄기수와 번식방법이 구기자의 수량형질에 미치는 영향 (Effect of Stem Number and Propagation Method on Yield Components of Boxthorn(Lycium chines Mill.))

  • 구범자(枸杞子)의 주당(株當) 적정(適正) 줄기수와 번식방법(繁殖方法)을 구명(究明)하고자, $1993\sim1994$년(年)까지 시험(試驗)을 실시(實施)하여 얻은 결과(結果)를 요약(要約)하면 다음과 같다. 1 구기자(拘杞子) 번식방법(繁殖方法)은 종자번식(種子繁殖)에 비해 개화기(開化期)가 15일 빠르고, 건과수량(乾果收量)이 91kg/10a로 30%가 증수(增收)된 지삽(枝揷) 20cm가 가장 좋았다. 2 삽수 굵기는 건과수량(乾果收量)이 227kg/10a로 방임구(放任區)에 비해 28% 증수(增收)된 0.9cm가 좋았다. 3. 주당 적정(適定) 줄기수는 줄기수 5본에서 생육(生育)이 가장 좋았고 100과중도 13.4g으로 가장 무거웠다. 4. 주당(株當) 착과수는 864개로 줄기수 5본에서 증가(增加)되었고 건과수량(乾果收量)도 줄기수 5본에서 104kg/10a로 방임구(放任區)에 비해 16% 증수(增收)되었다.

구기자 추출물의 항비만 및 혈당강하 효과 (Study on Anti-obesity and Hypoglycemic Effects of Lycium chinense Mill Extracts)

  • 구기자는 식용가능하고 제2형 당뇨병 합병증인 지방간분해, 고혈압 및 고혈당에 효능이 있다고 알려져 있다. 하지만 당 대사 및 지질 대사에 작용하는 정확한 기작에 대한 연구는 아직 미비하기에 탄수화물 소화효소인 ${\alpha}$-amylase와 ${\alpha}$-glucosidase의 저해활성 및 정상생쥐를 이용한 혈당강하 효과와 3T3-L1 adipocyte를 통하여 비만축적 억제효과를 보았다. 구기자 추출은 80% MeOH, 70% EtOH 및 열수추출하였으며 이들 중 ${\alpha}$-amylase 50% 저해 활성($IC_{50}$)은 saliva ${\alpha}$-amylase에서 acarbose, 80% MeOH, 70% EtOH, aqueous(7.04, 3.19, 3.20, 3.66 mg/mL), pacreas ${\alpha}$-amylase acarbose, 80% MeOH, 70% EtOH, aqueous(10.97, 4.16, 3.07, 3.55 mg/mL)로 saliva ${\alpha}$-amylase에선 acarbose보다 80% MeOH, 70% EtOH 추출물이 가장 낮은 농도에서 $IC_{50}$값을 나타냈고 pacreas ${\alpha}$-amylase에서는 70% EtOH 추출물이 가장 낮은 농도에서 $IC_{50}$값을 나타냈다. 또한 10 ${\mu}g/mL$ 농도에서 yeast ${\alpha}$-glucosidase 활성저해 $IC_{50}$값은 80% MeOH, 70% EtOH, aqueous(5.01, 4.98, 4.80 ${\mu}g/mL$)로 구기자 추출물 모두 acarbose(5.05)보다 우수한 $IC_{50}$값을 나타냈다. 정상생쥐에게 탄수화물 소화효소의 저해활성이 높은 70% EtOH 추출물을 전분과 함께 경구투여 한 후 내당능을 측정한 결과, 전분과 함께 100, 200 mg/kg body weight를 경구투여 한 경우, 대조군에 비해 투여 후 30, 60, 90, 120분에 혈당 증가가 낮았다. 또한 추출물을 100 mg보다 200 mg 투여하였을 경우 대조군에 비해 각각 19% 및 40%씩 저하되었으며, 120분에는 대조군 수준이 되었다. 그리고 3T3-L1 adipocyte에서 비만축적 억제효과를 측정한 결과 70% EtOH 추출물 농도가 증가함에 따라 leptin, 중성지방 농도가 감소하였고 유리지방산 농도는 증가하였다. 이상의 연구 결과 구기자 추출물 중 70% EtOH는 acarbose보다 높은 ${\alpha}$-glucosidase, ${\alpha}$-amylase 저해활성을 나타냈으며, 정상생쥐를 통해 식후 혈당증가를 저하시키는 효과를 확인하였고 GLUT4 발현 역시 증가함을 확인했다. 이를 통해 구기자 추출물이 지방축적 억제와 인슐린 저항성 억제 및 혈당강하 기전을 확인하여 제2형 당뇨병 개선에 효과적인 것으로 판단, 추후 더 연구하여 보다 더 정확한 성분을 조사할 필요성이 있다고 사료된다.

시비량과 분시방법이 포기자의 수량형질에 미치는 영향 (The Effects of Fertilizer Application Level and Top-dressing Method on the Yield Component and Fruit Yield of Lycium chinense MILL)

  • 포기자의 시비적량과 알맞는 분류방법을 구명코저 시험을 실시하여 얻은 결과를 요약하면 1. N14P$_2$O$_{5}$7K$_2$O14kg/10a(표준비) 보다 N14P$_2$O$_{5}$14K$_2$O14k/10a 시비시 경장은 4cm길었고, 엽면적은 45~490$\textrm{cm}^2$가 많았으며, 주당건물중은 1.6~5gr이 무거웠다. 2. 1984~1985년 2개년 건과중 표준비의 171kg/10a보다, N14P$_2$O$_{5}$21K$_2$O14kg/10a 시용은 185kg/10a로 8%증수되었으며, 3. 엽의 3요소 흡수량과 건물중은 P$_2$O$_{5}$는 1%, N과 $K_2$O는 5% 수준에서 유의성이 인정되었으며, 경제성을 감안한 10a시비 적량은 N은 13.8kg, P$_2$O$_{5}$는14.2kg, $K_2$O는 14.3kg이었다. 4. 시비방법중 3요소의 분시시용은 3요소 기비시용보다, 주당 엽면적과 건물중의 증대로 인해 건과중이 20% 많았으며, 낙엽은 전혀 되지 않아 합리적인 시비방법으로 사료되었다.

구기자나무의 재식밀도 및 예취높이가 가공용 잎 생산에 미치는 영향 (Effects of Planting Density and Cutting Height on Production of Leaves for Processing Raw Materials in Goji Berry)

  • Background: The leaves of the goji berry (Lycium chinense Mill.) are used as raw materials in processing and by replace fruits to some extent. The reason is that the leaves are cheaper, however, betaine content is higher than in the fruits. These experiments were conducted to determine the planting density and cutting height for producing a large number of leaves. Methods and Results: The cultivar 'Myeongan' with many branches was used. When the shoot height reached 50 cm - 70 cm, harvesting was possible four times a year. The time to next harvest was approximately 38 days after regeneration of new shoots. Leaf quantity was in the order of 1st > 2nd > 4th > 3rd harvest. Insect damage occurred during the third harvest in late July and early August, therefore, eco-friendly control was necessary. The total yield was higher at the planting density 60 cm × 30 cm than that of 60 cm × 20 cm or 60 cm × 40 cm. The yield at cutting for shoot height of 60 cm was increased by 6.3 percent compared to that of 50 cm, At the cutting height of 70 cm, harvest was difficult owing to hardening of stems and thorns. Betaine content, an indicator component of goji berry, was not significantly different according to planting densities and cutting height. Conclusions: The ideal cutting period to produce leaves of goji berry for processing is when the shoots grow to approximately 60 cm, and the leaves can be harvested 4 times a year. The dried-leaf yield was highest at the planting density of 60 cm × 30 cm.

실험적 당뇨에 대한 신지골피탕의 면역조직화학적 연구 (Immunohistochemical Study of Sinjigolpy-tang on the Experimental Diabetic Rats)

  • This experiment was performed to investigate the effect of Sinjigolpy-tang on the diabetic rats induced by STZ. After experimental diabets was induced by 55mg/kg of STZ injection, we administered Sinjigolpy-tang extract for 14 days after STZ injection. Glucagon and insulin granules in Langerhans islets were stained by use of immunohistochemical(ABC) method and observed the relative amount of the each granules in Langerhans islet by light microscope and image analysis system. Area % of insulin granules in Langerhans islets in Sinjigolpy-tang increased and showed the statistically significant difference with the control group at 14th day. Area % of glucagon granules in Langerhans islets in Sinjigolpy-tang decreased and showed the statistically significant difference with the control group at 7th and 14th day. It can be inferred that Sinjigolpy-tang has a control effect on glucagon and insulin granules in Langerhans islets of diabetic rats induced by STZ.

Streptozotocin 저용량 반복투여로 유도한 당뇨 흰쥐에서 구기자 추출물의 효능연구 (Effect of the Lycii Fructus on multiple low-dose streptozotocin-induced diabetic rats)

  • Objectives : This study was performed to investigate the anti-diabetic effect of the fruit extract of Lycium chinense Mill(Lycii Fructus, LF) on multiple low-dose streptozotocin-induced diabetic rats.Methods : Male Sprague-Dawley rats were divided into three groups; normal, STZ-control, Lycii Fructus extractorally administrated 300 ㎎/㎏ group (STZ-LF). Diabetes was induced in rats by consecutive injection of streptozotocin (STZ) at doses of 30 ㎎/㎏ for 5 days. After 4 weeks, all rats were sacrificed, Fasting blood glucose, total cholesterol (CHO), triglyceride (TG) and HDL-Cholesterol were measured in sera of rats. Histopathological changes of pancreas, kidney, liver and lung tissues were observed by microscope after H&E, Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured.Results : There were no differences in body, food intake and water intake in LF-administrated groups compared with STZ control group. However, LF extract significantly decreased the levels of serum glucose, CHO, TG and HDL-Cholesterol in diabetic rats. In histopathological analysis of kidney, liver and lung, LF-administrated groups showed the inhibition of morphological damage.Conclusions : These results suggest that LF have a biological action on multi low-dose STZ-induced diabetes in rats via decreasing the serum glucose, TG and TG levels and may protect the morphological changes of kidney, liver and lung.

구기자혹응애(Eriophyes ruro KISHIDA) 및 그 혹(Gall)에 관한 연구 II. 혹의 성장에 따르는 효소활성 및 화학성분에 대하여 (Studies on Eriophyes kuko KISHIDA and its Galls -On the Activity of Enzymes and the Chemical Constituents in Growing Galls-)

  • 구기자혹응애(Eriophyes kuko KISHIDA)의 침입기에 의하여 구기자나무(Lycium chineens MILL.)외 잎형성된 혹(mite gall)의 생화학적 성질을 구명할 목으로 몇 가지 효소의 활성도, 화학성분의 함량 및 호량 등에 관하여 실험한 바 다음과 같은 결과를 얻었다. Catalase와 peroxidase 활성도는 건전한 잎의 직보다 혹 조직에서 높았으며 혹이 점차로 커짐에 따라 활성도도 높아졌다. 2. 조효소의 phosphorylase 활성도는 큰 혹(matured gall) 조직과 건전한 잎의 조직에 있어서 차이가 없었으나 활성 측정에 미치는 다른 효소의 영향을 고려하면 건전한 잎의 조직이 높은 것 같으며, phosphomonoesterase의 활성도는 건전한 잎의 조직보다 큰 혹 조직이 5배 가까이 높았으며 $\beta-amylase$와 invertase 등의 활성도는 큰 혹 조직이 2배 가까이 높았다. 3. 조단백질 함량은 혹의 발생 형성초기인 작은 혹일때는 $20\%$ 정도 건전한 잎의 조직보다 증가하였으나 혹이 점차 커 짐에 따라 감소하는 경향을 나타내었으며, 환원당은 자은 측일 에는 약간 감소하는 경향이었으나 혹이 점차 커짐에 따라 증가하였다. 인과 tannin의 함량도 혹이 점차커짐에 따라 증가하여를 혹이 건전한 잎보다 2배정도 많았다. 4, 호흡에 있어서 $QO_2$는 혹 조직이 건전한 잎보다 높았으나, $QCO_2$는 반대로 건전한 잎이 높았으며, RQ는 혹 조직이 낮았다.

  • This research was planned and executed to evaluate how the composition of Yack-sun tea can affect the health conditions of people who are suffering from diet-related such as being overweight, are obese and have hyperlipidemia, by taking Yack-sun tea in a form of a nutritional supplement with our daily meals. We produced Kangjieum with Lycium chinense Mill., Polygonum multflorum Thunb, Cassia tora L., Crataegus pinnatifida Bge and Salvia miltiorrhiza Bge. Thus, we approach of oriental diet therapy area research of Kangjieum and analysis proximate composition, water soluble antioxidant content. The content(%, dry basis) of total carbohydrate was 60.23%, crude protein was 18.18%, crude ash was 11.36% and crude fat was 10.23% in Kangjieum. Total water soluble antioxidant content was 1.027 ${\mu}g/m{\ell}$ of Kangjieum. We think that scientific and objective evaluation was done on the components of the Kangjieum prescription. This basic data could help guide the application of oriental medicinal resources into other foods and serve as a stepping-stone for use of Kangjieum in the burgeoning field of nutraceutical foods. Last, the scientific effects of oriental medicinal foods developed according to oriental medicinal theory. This theory is believed to be essential for government policy development concerning validation of medicinal effects and assessment, with the aim of fostering systematic development and providing guidance to food development in the interest of national health.

지골피, 산수유의 streptozotocin 유발 당뇨흰쥐에서의 효과 비교 연구 (The comparisons of Lycii Radicis Cortex and Corni Fructus water extract effects on streptozotocin-induced diabetes in rats)

  • Objectives : This study aimed to compare the anti-diabetic efficacy of Lycii Radicis Cortex (Lycium chinense Mill.) and Corni Fructus (Cornus officinalis) on streptozotocin (STZ)-induced diabetes in rats. Methods : Male Sprague-Dawley rats were divided into four groups; normal, STZ-control, Lycii Radicis Cortex extract-administrated group (LRC) and Corni Fructus extract-administrated group(CF). Diabetes in rats was induced by intraperitonal injection with streptozotocin (STZ) at doses of, 30 mg/kg (body weight) for 5 days (once per a day). STZ-induced diabetic rats were orally administrated LRC and CF extract daily for 4 weeks at doses of 300 mg/kg. Fasting blood glucose, total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN) and creatinine were measured in sera of rats. Histopathological changes of kidney, liver and lung tissues were observed by microscope after H&E staining. Results : There were no differences in body and kidney weights, food intake and water intake in LRC- and CF-administrated groups compared with STZ control group. However, glucose, TC and TG levels in serum were significantly decreased in LRC-administrated groups compared with STZ-control group. In histopathological analysis of kidney, liver and lung, both LRC- and CF-administrated groups showed the inhibition of morphological damage. Conclusions : These results suggest that LRC and CF have a biological action on STZ-induced diabetes in rats via decreasing the serum TG and TG levels and may protect the morphological changes of kidney, liver and lung.
