구기자 분획물이 Streptozotocin으로 유발 된 당뇨 흰쥐에 대한 항당뇨 및 항산화작용에 미치는 효과 (Antidiabetic and Antioxidative Effect of Lycii fructus in Streptozotocin-Induced Diabetic Rats)

  • This study was carried out to investigate the antidiabetic and antioxidative effect of Lycii fructus in the Streptozotocin(STZ)-induced diabetic rats. The effective fractions were prepared as a form of organic solvents of $CH_{3}(CH_{2})_{4}CH_{3}$ $CHCI_{3}$, EtOAc, BuOH and $H_{2}O$ fractions prepared from the EtOH extract of Lycii fructus and The diabetes were induced by an tail-intravenous injection of STZ with a dose of 45 mg/kg dissolved in citrate buffer. The various fractions of Lycii fructus were orally administrated once a day for 7 days. The contents of serum glucose, and triglyceride in the $CHCI_{3}$ fraction and hepatic lipid peroxidation in the EtOAc, BuOH and $H_{2}O$ fractions treated rats were significantly decreased when compared to those of the STZ-control group In addition, an activity of hepatic GST in the BuOH fraction treated rats was significantly increased compared to that of the STZ-control group. whereas, activities of hepatic catalase, GSH-Px in the BuOH fraction treated rats were significantly decreased compared to those of the STZ-control group. Meanwhile, The content of hepatic glycogen and avtivity of hepatic glucokinase in $CHCI_{3}$ fraction treated rats were significantly increased, but activity of glucose-6-pase was significantly decreased in the $CHCI_{3}$ fraction treated rats. In conclusion, these results indicated that the BuOH fraction of Lycii fructus was effective for the antioxidation, and also the $CHCI_{3}$ fraction of Lycii fructus was effective for the antidiabetes in the STZ-induced diabetic rats.

  • 구기자 음료의 풍미 개선을 위하여 구기자를 가열 및 볶음 처리하여 추출물을 제조하고, 추출물의 성분, 색도, 향기성분을 분석하여 다음과 같은 결과를 얻었다. 1. 건조구기자의 성분은 수분 21.8%, 전당 27.6%, 환원당 15.2% 조단백질 14.3% 조지방 5.7%, 조섬유 7.5%, 회분 8.0% 이었다. 2. 추출물의 고형물을 기준으로 추출수율은 건조 구기자와 용매비를 높임에 따라 증가하였다. 3. 건조구기자 추출물 제조에 있어 고형물은 물추출이 가장 높았고, ethanol의 농도를 높임에 따라 감소하였다. 4. 구기자는 $140^{\circ}C$에서 8분간 볶음처리하고 $120^{\circ}C$에서 1시간 가열 처리 하였을 때 수율, 색도 등에 있어서 가장 양호 하였다. 5. 생구기자의 향기성분은 hexadecanoic acid, methyl linoleate, benzyl alcohol, dimethal benzene 이었으며 가열 및 볶음처리에 의하여 2-methyl-2-butenal, 1.4-dimethyl-benzene, benzyl alcohol 등은 감소 하였고 methyl thiopropanol, benzene acetaldehyde, ethyl linoleate는 증가 하였다.

Anti-inflammatory and Anti-allergic Effects of Lycii Fructus Extract

  • Lycii Fructus (LF) has been reported to possess various biological activities. In this study, we investigated the anti-inflammatory and anti-allergic effects of LF extract in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells and rat peritoneal mast cells(RPMC). LF extract inhibited NO production and the expressions of inducible NO synthase (iNOS) mRNA and enzyme protein, determined by Griess reactions, RT-PCR and Western blotting, respectively, in LPS-stimulated RAW 264.7 macrophages cells. LF extract significantly inhibited histamine release in compound48/80 stimulated rat peritoneal mast cells compared with the compound48/80 only treated cells. It is suggested that the LF extract can improve the inflammation status that involves the histamine release.

구기자(Lycium chinence Miller) 첨가에 따른 요구르트의 발효 특성과 기능성 (Fermentation Properties and Functionality of Yogurt added with Lycium chinence Miller)

  • 요구르트의 제조 시 기능성 성분을 함유한 첨가제로서 구기자, 구기엽 및 지골피를 기질로 하여 요구르트를 제조하여 젖산균주에 따른 발효특성을 시험하였고, 각 부위를 첨가한 요구르트의 cholestol 저하 효과 및 병원성 균주의 성장 억제 효과를 시험하였다. 상업용 starter를 사용한 모든 처리구에서 발효 후 pH가 4.06 이하로 나타나 발효 촉진 효과가 있었고, 산 생성은 S. salivarius ssp. thermophilus와 L. delbrueckii ssp. bulgaricus에서 높아 발효촉진 효과가 우수하였으며 젖산균수는 S. salivarius ssp. thermophilus와 L. delbrueckii ssp. bulgaricus를 이용한 구기자 extract 첨가구에서 9시간째 2.62${\times}$$10^9$ cfu/$m\ell$로서 최대 균수를 나타내었다. 유당분해율은 S. salivarius ssp. thermophilus와 L. dilbrueckii ssp. bulgaricus를 이용한 구기자, 구기엽, 지골피 extract 처리구에서 36.11%, 37.76% 및 32.70%를 나타내어 높았고, 유기산 생성은 S. salivarius ssp. thermophilus와 L. delbrueckii ssp. bulgaricus 균주를 stsrter로 사용한 요구르트의 경우에만 isobutylic acid가 34.39${\sim}$37.72 mM로 높은 유기산을 생성하였다. 점도는 L. acidophilus, B. longum과 S. salivarius ssp. thermophilus를 starter로 사용하였을 때 1,615${\sim}$2,030 cP로서 모든 첨가구에서 높은 점도를 나타내었고, 관능검사 결과 대조구가 3.27${\sim}$3.44인데 비하여 구기자와 구기엽 extractffm 첨가한 요구르트의 관능검사 결과는 1.77${\sim}$2.58로서 상당히 낮은 기호도를 나타내었다. 그러나 지골피 extract 첨가구에서 L. casei와 L. acidophilus. B. longum 및 S. salivarius ssp. thermophilus를 starter로 사용한 요구르트 기호도의 경우는 3.34${\sim}$3.77로 나타나 기호성이 좋았다. 구기자, 구기엽 및 지골피 extract 첨가에 의한 cholesterol 저하 효과는 인체 유래의 각 젖산균과 상업용 starter 균주를 이용한 모든 처리구에서 17.38${\sim}$32.08%의 콜레스테롤 저하 효과를 나타내었다. 그 중 L. acidophilus KCTC315.와 L. salivarius subsp. salivarius CBU27 젖산균은 25.75${\sim}$32.08%의 높은 콜레스테롤 저하 효과를 보였다. 구기자, 구기엽 및 지골피 extract 첨가 요구르트의 식중독 유발 살모넬라 균주에 대한 성장 억제 효과는 S. typhimurium M-15 균주에서 약간의 억제 효과를 보였고, 설사와 식중독을 유발하는 것으로 알려진 대장균 중 E. coli KCTC1021와 리스테리아증을 유발하는 L. monocytogenes에 대해서는 높은 성장 억제 효과를 보여 구기자, 구기엽, 지골피를 기질로 한 요구르트가 살모렐라, 대장균 및 리스테리아 유발 병원성 균주에 대한 탁월한 억제 효과가 있었다.

  • Objectives : This study was conducted to 9% compared with the previous experimental obesity-induced metabolic function in rats that it was a mixture of pear extract and herbal drugs(Lycii Fructus, Coicis Semen, Alimatis Rhizoma and Astragali Radix) on high fat diet-induce obesity rats. Methods : The animals were used in male rats. Control group fed high-fat diet only. Experimental groups were divided four, ExpI group was fed high-fat diet with a mixture pear extract and Lycii Fructus 9%, Exp II group was fed high-fat diet with a mixture pear extract and Coicis Semen 9%, Exp III group was fed high-fat diet with a mixture pear extract and Alimatis Rhizoma 9%, and Exp IV group was fed high-fat diet with a mixture pear extract and Astragali Radix 9% for 4 weeks. Results : The body weight gain increased in all groups, but attenuated gradually in the experimental groups compared to the control group. The food intakes were significantly lower in all Exp groups than the control group. The concentrations of serum total cholesterol (TC), LDL-cholesterol, and free fatty acid were significantly lower in Exp II than in the control group. Also the concentration of serum free fatty acid was significantly lower in the Exp IV group than in the control group. In inflammatory activities, the Exp II, IV group was significantly lower than the control group. Conclusion : The results indicated that Exp I group (administered a mixture of pear extract and Lycii Fructus) reduced fat accumulation, body weight and the highest elevated lipid metabolism, while the Exp II group (administered the mixture of pear extract and Coicis Semen) and Exp IV group (administered the mixture of pear extract and Astragali Radix) had the highest elevated immune activity.

갱년기 장애 유도 시 구기자의 혈중지질 농도 저하 및 골 중 collagen 합성 효과 (Effect of Lycii fructus Extracts on Serum Lipids and Bone Collagen Contents in Ovariectomized Rats)

  • The aim of this study was to evaluate the effect of Lycii fructus (LF) on serum lipid and the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rat (Sham), ovariectomized control rat (OVX-control), and ovariectomized rats supplemented with the LF 50mg/kg bw/day (OVX-LF). The LF ethanol extract were orally administrated 1ml per day. Body weight gain was not significantly different in groups. Although total-cholesterol and triglyceride were increased in the ovariectomized control, supplementation with the LF extract decreased the levels. Moreover, the serum HDL-cholesterol levels were significantly increased after supplementation with the LF extract(p<0.05). Supplementation with the LF extract prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of LF. Therefore, it may be used to possibly improve the quality of life in menopausal women.

지골피, 산수유의 streptozotocin 유발 당뇨흰쥐에서의 효과 비교 연구 (The comparisons of Lycii Radicis Cortex and Corni Fructus water extract effects on streptozotocin-induced diabetes in rats)

  • Objectives : This study aimed to compare the anti-diabetic efficacy of Lycii Radicis Cortex (Lycium chinense Mill.) and Corni Fructus (Cornus officinalis) on streptozotocin (STZ)-induced diabetes in rats. Methods : Male Sprague-Dawley rats were divided into four groups; normal, STZ-control, Lycii Radicis Cortex extract-administrated group (LRC) and Corni Fructus extract-administrated group(CF). Diabetes in rats was induced by intraperitonal injection with streptozotocin (STZ) at doses of, 30 mg/kg (body weight) for 5 days (once per a day). STZ-induced diabetic rats were orally administrated LRC and CF extract daily for 4 weeks at doses of 300 mg/kg. Fasting blood glucose, total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN) and creatinine were measured in sera of rats. Histopathological changes of kidney, liver and lung tissues were observed by microscope after H&E staining. Results : There were no differences in body and kidney weights, food intake and water intake in LRC- and CF-administrated groups compared with STZ control group. However, glucose, TC and TG levels in serum were significantly decreased in LRC-administrated groups compared with STZ-control group. In histopathological analysis of kidney, liver and lung, both LRC- and CF-administrated groups showed the inhibition of morphological damage. Conclusions : These results suggest that LRC and CF have a biological action on STZ-induced diabetes in rats via decreasing the serum TG and TG levels and may protect the morphological changes of kidney, liver and lung.

The Biochemical and Histological Studies of the Oriental Medicine Extract on Hair Growth Effect

  • The hair growth effect of the oriental medicine extract which is composed of Polygoni multiflori Radix, Angelica gigantis Radix and Lycii Fructus was studied biochemically and histologically. The study was conducted dividelly into three groups, control, propecia and oriental medicine extract and three groups were compared each other in skin androgen, testosterone (T) and dihydrotestosterone (DHT), content biochemically and the number of hair follicular unit and hair follicle histologically. The oriental medicine extract and propecia decreased skin DHT conetent and increased skin total (T+DHT) content compared with control. In transverse sections of skin specimens the oriental medicine extract and propecia increased total counts of follicular units and follicles compared with control. On the basis of the result, the oriental medicine extract has same hair growth effect as propecia and it is suggested that the oriental medicne extract is capable of a therapeutic agent of alopecia.

The Clinical Study Using Oriental Medicine Extract on Male Pattern Hair Loss

  • In order to examine the clinical effects on male pattern hair loss, the oriental medicine extract composed of Polygoni multiflori Radix, Angelica gigantis Radix and Lycii Fructus was tested to balding people. 40 males and 10 females were participated in this experiment, and they had spread the extract on their scalp themselves one time a day for 18 months. After 18 months, all participants submitted the reports that had questionnaires to determine the effect of topical oriental medicine extract on hair growth and/or hair loss preventing in balding people. In the results, ir was found that hair density on the entire scalp was gradually increased during the spreading period of the extract in most of balding people. The hair loss was observed about 1 week or 2 months after extract treatment and hair growth was observed about 6 months after extract treatment. In the reports, 74% of participants answered that the extract was effective on hair growth and hair loss preventing, and improvement of scalp condition. From these results, it is suggested that the extract composed of Polygoni multiflori Radix, Angelica gigantis Radix and Lycii Fructus may be an effective medical care to hair growth and hair loss preventing in human.

당귀와 구기자 에탄올 추출물의 피부 미백작용 (Skin Whitening Effects of Extracts from Angelicae Gigantis Radix and Lycii fructus Ethanol Extracts)

  • Objective : The purpose of this study was to investigate the synergistic effect of Angelicae gigantis Radix (AG) and Lycii fructus (LF) ethanol extracts on skin-whitening effects. Method : LFAG extracts were prepared by extracting with 80% ethanol. The efficacy of LFAG was judged by measurement of cell viability, tyrosinase activity, melanin production, tyrosinase and microphthalmia-associated transcription factor (MITF) expression in B16F10 murine melanoma cells by lipopolysaccharides (LPS) treatment. Results : Each extract (LF or AG) inhibited the tyrosinase activity in a dose-dependent manner. The co-treatment of LFAG extracts ($25{\mu}g/mL$ LF plus $25{\mu}g/mL$ AG) markedly suppressed the LPS-induced cellular tyrosine activity, melanin production, tyrosinase and MMP-1 expression in B16F10 murine melanoma cells. These suppressive effects were synergistically increased by their combination. Conclusions : With these observations, we suggest that the extracts from Lycii fructus and Angelica gigantis Radix could be potent natural materials for whitening skin.
