Objectives : This study was conducted to investigate the associations of non alcoholic fatty liver with metabolic syndrome and the serum carotenoids. Methods : This study was conducted in a general hospital in South Korea from November, 2004 to August, 2005. The study subjects were 350 sampled persons who were aged from 40 years and older (males : 180, females : 170). They were grouped into the normal, mild and severe groups according to fat accumulation in their livers, as determined by ultrasonography. We analyzed the association between non alcoholic fatty liver and metabolic syndrome by multiple logistic regression analysis and we analyzed the association between non alcoholic fatty liver and the serum carotenoids by a general linear model(ANCOVA). Results : After adjustment for the effect of potential covariates, the prevalence of metabolic syndrome was associated with fat accumulation in the liver (p trend <0.001). If the odds ratio of normal group is 1.00, then that of the mild group is 2.80 (95% C.I=1.17-6.71) and that of the severe group is 7.29 (95% C.I=2.76-19.30). The prevalence of metabolic alterations fitting the criteria of metabolic syndrome, according to the class of fat accumulation in the liver, was significantly increased, except for criteria of high blood pressure, a large waist circumference and low HDL (high density lipoprotein) cholesterol level (p trend <0.001). The level of serum ${\beta}$-carotene was decreased according to the class of fat accumulation in the liver (p trend=0.036), but the levels of serum ${\alpha}$-carotene, lycopene, ${\beta}$-cryptoxanthin and lutein were not decreased. Conclusions : This study shows that non alcoholic fatty liver was associated with metabolic syndrome and with the serum ${\beta}$-carotene level.
Two feeding experiments were conducted to investigate the effects of dietary paprika (DP) and lipid (DL) levels on growth performance and skin pigmentation of pale chub, Zacco platypus. Six diets (designated as $P_0L_8,\;P_0L_{17},\;P_8L_8,\;P_8L_{17},\;P_{16}L_8\;and\;P_{16}L_{17}$) were formulated to contain 0%, 8% and 16% paprika with 8% and 17% lipid, respectively. For the growth experiment (Exp I), three replicate groups of fish (average weight 2.6${\pm}$0.2 g) were fed one of the six experimental diets for 8 weeks. At the end of the feeding period, survival was above 94% and not significantly different among dietary treatments. Weight gain, feed efficiency and protein efficiency ratio of fish fed the $P_{16}L_8$ diet were lower than for fish fed the $P_0L_8$ diet. The highest total carotenoid (TC) content was observed in fish fed the $P_{16}L_8$ diet. For the pigmentation experiment (Exp II), each experimental diet was fed to two replicate groups of fish (average weight 9.0${\pm}$0.5 g) for 8 weeks. At the end of the feeding period, TC content of the skin was significantly affected by DP (p<0.05). The highest TC content of the skin was observed after 6 weeks of feeding at all dietary treatments. Astaxanthin content of the skin was not affected by DP and DL (p>0.05). The capxanthin and zeaxanthin contents of skin increased significantly with increasing DP, whereas the opposite trend was observed for lutein and ${\beta}$-cryptoxanthin contents. The skin lightness ($L^*$ values) significantly decreased whereas the values of $a^*$ and $b^*$ were significantly increased in fish fed the diets containing paprika (p<0.05). The present results suggest that feeding a diet containing 8% paprika and 8% lipid for 6 weeks could improve skin pigmentation of pale chub without any adverse effects on growth performance.

BACKGROUND/OBJECTIVES: A pivotal role of oxidative stress has been emphasized in the pathogenesis as well as in the disease progression of Parkinson's disease (PD). We aimed at investigating serum levels of antioxidant vitamins and elucidating whether they could be associated with the pathogenesis and progression of PD. MATERIALS/METHODS: Serum levels of retinol, ${\alpha}$- and ${\gamma}$-tocopherols, ${\alpha}$- and ${\beta}$-carotenes, lutein, lycopene, zeaxanthin and ${\beta}$-cryptoxanthin were measured and compared between 104 patients with idiopathic PD and 52 healthy controls matched for age and gender. In order to examine the relationship between antioxidant vitamins and the disease progression, multiple group comparisons were performed among the early PD (Hoehn and Yahr stage I and II, N = 47), advanced PD (stage III and IV, N = 57) and control groups. Separate correlation analyses were performed between the measured antioxidant vitamins and clinical variables, such as Hoehn and Yahr stage and Unified Parkinson's Disease Rating Scale (UPDRS) motor score. RESULTS: Compared to controls, PD patients had lower levels of ${\alpha}$- and ${\beta}$-carotenes and lycopene. ${\alpha}$-carotene, ${\beta}$-carotene and lycopene levels were significantly reduced in advanced PD patients relative to early PD patients and were negatively correlated with Hoehn and Yahr stage and UPDRS motor score in PD patients. No significant differences were found in serum levels of retinol, ${\alpha}$- and ${\gamma}$-tocopherols, and other carotenoids between PD patients and controls. No significant correlations were found between these vitamin levels and clinical variables in PD patients. CONCLUSTIONS: We found that serum levels of some carotenoids, ${\alpha}$-carotene, ${\beta}$-carotene and lycopene, were lower in PD patients, and that these carotenoids inversely correlated with clinical variables representing disease progression. Our findings suggest that decreases in serum ${\alpha}$-carotene, ${\beta}$-carotene and lycopene may be associated with the pathogenesis as well as progression of PD.
This study was carried out to investigate the quality characteristics of bread containing sweetpotato (Ipomoea batatas (L.) Lam) leaf powder (0, 2, 3, 5, and 7% of the total flour). We found that the addition of sweetpotato leaf powder decreased the pH of the dough, whereas the total titratable acidity increased and the specific volume and baking loss of bread were decreased. However, the moisture content of the bread did not show any significant differences. The L and a values of the bread inner crumb were decreased by the addition of sweetpotato leaf powder, however, the b value was increased. The 2,2-diphenyl-1-picrylhydrazyl-radical scavenging activity, total polyphenol, lutein and ${\beta}-carotene$ contents were increased significantly by the addition of sweetpotato leaf powder. The taste, color, flavor, chewiness and overall acceptability of bread containing 2~3% sweetpotato leaf powder were better than those of the controls. We found that the sample group with 2~3% sweetpotato leaf powder is the optimum content for making bread.
We investigated photodynamic stress-induced antioxidant responses in transgenic rice overexpressing Bradyrhizobium japonicum 5-aminolevulinic acid synthase (ALA-S) coding sequence lacking plastidal transit sequence. High light of $350{\mu}mol\;m^{-2}\;s^{-1}$ decreased the quantum yield in the transgenic lines, C4 and C5, compared to that of wild-type line. By contrast, non-photochemical quenching (NPQ) levels of C4 and C5 under high light were higher than those of the transgenic lines under low light of $150{\mu}mol\;m^{-2}\;s^{-1}$ as well as wild-type line under low and high light. Greater levels of NPQ in the transgenic lines exposed to high light were in a close correlation with increases in the xanthophyll pigment, zeaxanthin. Under high light, levels of neoxanthin, violaxanthin, lutein, and ${\beta}$-carotene in the transgenic lines were lower than those in wild-type line. Taken together, nonphotochemical energy dissipation and photoprotectant xanthophyll pigments play a critical role to deal with the severe photodynamic damage in the transgenic rice plants, although they could not overcome the photodynamic stress, leading to severe photobleaching symptoms.

Postharvest quality of sweet pepper fruits was mainly defined as external appearance, i.e., shape, fruit weight and surface color. These quality traits tend to involve esthetic appeal, it disregards gustatory properties and nutritional value. However, comparative studies according to the marketability of sweet pepper fruits are insufficient. This study was carried out to compare the physicochemical components, antioxidant activity and carotenoid contents of marketable and unmarketable sweet pepper fruits (Capsicum annuum L.). Physicochemical components (proximate composition, minerals and total phenolic contents) and antioxidant activities using various methods were investigated. The proximate composition values (%) of marketable and unmarketable fruits were: moisture (90.28 and 90.29), ash (0.74 and 0.26), crude protein (0.67 and 0.72), crude lipid (0.38 and 0.32). There were no significant differences in antioxidant activity, while total phenolic content was higher in marketable fruit. Carotenoids contents were 29.3 ± 2.6 and 31.9 ± 2.9 ㎍·g-1 in marketable and unmarketable fruits respectively, and identified β-carotene, violaxanthin, neoxanthin, and zeaxanthin. Lutein and capsaicin were not detected. In this study, potential value of unmarketable sweet pepper fruit could be identified to be applied as a food ingredient and functional food material.
This study examined the growth, fatty acid (FA) content, and carotenoids of a newly isolated freshwater microalga, Mychonastes sp. 246, in various culture media. The appropriate temperature and light intensity for culturing Mychonastes sp. 246 were determined as 18℃-22℃ and 200-250 µmol/m2/s using a high throughput photobioreactor. The microalgal cells were cultivated in 0.5 L bubble column photobioreactors using BG11, Bold's Basal media, and f/2 media. According to the growth results of the microalgae, BG11, among the tested media, showed the highest biomass concentrations (3.5 ± 0.1 g/L in 10 d). To enhance the biomass growth of the microalgae, the N:P ratio in BG11 was manipulated from 45:1 to 7:1 based on the stoichiometric cell composition. The biomass concentrations of Mychonastes sp. 246 grown on the manipulated BG11 (MBG) increased to 38% (4.6 ± 0.3 g/L in d) compared with the original BG11 (3.3 g/L). The FA content of the microalgae grown on the MBG was lower (8.4%) than that of the original BG11 (10.1%) while the FA compositions did not exhibit any significant differences. Furthermore, three kinds of carotenoids were identified in Mychonastes sp. 246, zeaxanthin, lutein, and β-carotene. These results suggest an effective strategy for increasing biomass concentrations, FA content, and carotenoids of microalgae by performing a simple N:P adjustment in the culture media.
Purpose: The purpose of this study was to estimate dietary vitamin A intake and examine major food sources of vitamin A in Korean adults. Methods: Using data from the 2007~2012 Korea National Health and Nutrition Examination Survey, a total of 33,069 subjects over 19-years-old were included in this study. We estimated individual daily intakes of retinol, carotenoids such as ${\alpha}$-carotene, ${\beta}$-carotene, ${\beta}$-cryptoxanthin, lutein/zeaxanthin, and lycopene, and vitamin A by linking food consumption data with the vitamin A database of commonly consumed foods. We compared individual vitamin A intakes with the reference value of Dietary Reference Intakes for Koreans. Results: Average dietary vitamin A intakes of study subjects were $864.3{\mu}g$ retinol equivalent/day ($495.7{\mu}g$ retinol activity equivalent/day) in men and $715.0{\mu}g$ retinol equivalent/day ($403.6{\mu}g$ retinol activity equivalent/day) in women. Exactly 42.9% and 70.6% of total subjects consumed less vitamin A than the Estimated Average Requirement (EAR) based on retinol equivalent and retinol activity equivalent, respectively. The major food sources of vitamin A were Korean radish leaves, carrot, red pepper, and laver, and the top 20 foods provided about 80% of total vitamin A intake. Conclusion: This study provides basic data for estimation of vitamin A intake in Korean adults. Further research will be needed to analyze the association of insufficient or excess intakes of vitamin A and health problems in the Korean population.
Protective effects of natural components including genistein (4',5,7-trihydroxyisoflavone) from Glycine max MERRILL on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. Genistein $(10{\sim}100\;{\mu}m)$ suppressed photohemolysis in a concentration-dependent manner, and was more effective than the lipid peroxidation chain blocker, ${\alpha}$-tocopherol (Vit. E). Glycoside of genistein, genistin, the water-soluble antioxidant, L-ascorbate, and the iron chelator, myo-inositol hexaphosphoric acid dodecasodium salt (sodium phytate) did not exhibit protective effect against photohemolysis. L-Ascorbate and sodium phytate stimulated photohemolysis at high concentration $(500\;{\mu}m)$. ${\alpha}$-Carotene 3,3'-diol (lutein), a singlet oxygen $(^1O_2)$ quencher, exhibited pronounced protective effect, an indication that $^1O_2$ is important in photohemolysis sensitized by rose-bengal. Reactive oxygen scavenging activities $(OSC_{50})$ of natural antioxidants including genistein on reactive oxygen species (ROS) generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were in the order of sodium phytate > L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin. $OSC_{50}$ value of genistein, genistin, ${\alpha}$-tocopherol, L-ascorbate, and sodium phytate were 41.0, 109.0, 9.0, 5.2, and $0.56{\mu}m$ respectively. The order of free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity $(FSC_{50})$ was L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin. These results indicate that genistein can function as an antioxidant in biological systems, particularly skin exposed to solar UV radiation by scavenging $^1O_2$ and other ROS, and to protect cellular membranes against ROS.

Purpose: We prospectively investigated clinical changes and long-term outcomes after administration of the drugs recommended by the Age-Related Eye Disease Study-2 to patients with intermediate age-related macular degeneration (AMD). Methods: This prospective multicenter study enrolled 79 eyes of 55 patients taking lutein and zeaxanthin. The primary endpoint was contrast sensitivity; this was checked every 12 months for a total of 36 months after treatment commenced. The secondary endpoints were visual acuity, central macular thickness, and drusen volume; the latter two parameters were assessed using spectral domain optical coherence tomography. Results: The mean patient age was $72.46{\pm}7.16years$. Contrast sensitivity gradually improved at both three and six cycles per degree. The corrected visual acuity was $0.13{\pm}0.14logMAR$ and did not change significantly over the 36 months. Neither the central macular thickness nor drusen volume changed significantly. Conclusions: Contrast sensitivity markedly improved after treatment, improving vision and patient satisfaction. Visual acuity, central retinal thickness, and drusen volume did not deteriorate. Therefore, progression of AMD and visual function deterioration were halted.
