Biochemical properties and antimicrobial susceptibility of Pasteurella multocida isolated from pneumonic lungs in slaughtered animals (도축장 출하축의 폐병변에서 분리한 Pasteurella multocida의 생화학적 특성 및 약제감수성)

  • This study was carried out to investigate isolation of Pasteurella multocida from pneumonic lungs of slaughtered animals in Kyungsan abattoir to examine the antimicrobial susceptibility and biochemical properties. The results were summarized as follows ; P multocida was isolated from 50(29.2% ) of the 171 pneumonic lungs collected individually from cattle(3/40), goats(0131) and pigs(47/100), All of the isolated P multocida showed biocemical and cultural properties similiar to reference strains. All isolates were very susceptible to AK, AM, ENR, CF, GM,07, whereas resistant to SDM.

  • Heat stroke can lead multi-organ damage with hemorrhage and necrosis in the lungs, heart, liver, kidneys, brain and Ut. Heat stroke occurs when the elevation of core body temperatures induce a failure of thermoregulatory mechanism. A four-year-old male African Lion(Panthera leo) showed clinical signs such as panting, tachycardia, hyperthermia, unconsciousness and mydriasis under He hish humidity and hot weather. Clinical treatment and pouring cool water was unsuccessful. Grossly, congestion of lungs and pleura was observed. Yellowish discoloration was observed in the renal cortex. Microscopically, the coagulative necrosis in kidney and congestion of lungs and spleen were observed. In our knowledge, this case was closely associated with acute heat stroke.

Metal Concentrations in Lungs of Coal Workers' Pneumoconiosis Patients (탄광부 진폐증자의 폐조직내 금속농도)

  • The metal concentrations in lungs from 12 coal workers' pneumoconiosis(CWP) patients and 6 controls, who were not exposed occupationally to coal mine dust and metals during their life time, were analyzed by atomic absorption spectrophotometry. 1. Copper, lead, nickel, magnesium, manganese, zinc and iron concentrations in lungs of CWP patients were $1.10{\pm}0.088$, $1.12{\pm}0.068$, $0.22{\pm}0.020$, $113.7{\pm}1.31$, $0.19{\pm}0.012$, $10.2{\pm}1.54$, $426.7{\pm}2.63{\mu}g/g$ wet weight. 2. Copper, lead, nickel, magnesium, manganese, zinc and iron concentrations in lungs of controls were $1.10{\pm}0.013$, $0.85{\pm}0.007$, $0.10{\pm}0.008$, $87.6{\pm}1.29$, $0.18{\pm}0.005$, $10.6{\pm}1.44$, $164.9{\pm}3.29{\mu}g/g$ wet weight. 3. The ratios of concentrations for copper, lead, nickel, magnesium, manganese, zinc, and iron in lungs for CWP patients and controls were 1 : 1, 1.32 : 1, 2.20 : 1, 1.30 : 1, 1.06 : 1, 0.92 : 1, 2.58 : 1, respectively. There were significant differences in concentrations of lead, nickel, magnesium, iron by group(p<0.05). 4. There was no significant difference in metal concentrations of right upper lobe, right lower lobe, left upper lobe and left lower lobe for both CWP patients and controls (p>0.05, p>0.05). 5. In CWP patients lead was well correlated with nickel showing a rank correlation coefficient of 0.533, and zinc was correlated with copper showing a rank correlation coefficient of 0.476. 6. The concentrations of copper, nickel, maganese, and zinc in Korean CWP patients were lower than those in foreign CWP patients.

  • Objectives : The aim of this study was to investigate the effect of Asthma-depression and Immunoregulation with PR-HAS(Herbal-acupuncture with Platycodi Radix infusion solution) injection at Joksamni(St36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week). The experimental group(OVA-PR-HA) wase treated with concentrations(1%) of PR-HAS at Joksamni(St36) for the later 8 weeks(3times/week). The second experimental group(OVA-Needle prick) was treated with Needle-Prick at Joksamni(St36) for the later 8 weeks(3times/week). Results : 1. The weight and total cells in the mice lung treated with PR-HA decreased significantly compared with that of control group. 2. Total leukocytes and eosinophils in BALF of the mice group treated with PR-HA decreased remarkably compared with those of control group. 3. The sticking of collagen on histological analysis of lung sections, the mice group treated with PR-HA decreased significantly compared with those of control group. 4. The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. 5. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The numbers of $CCR3^+\;cells,\;CD4^+\;cells\;and\;CD8^+\;cells$ in the lungs, and $CD3e^+/CD69^+$ in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The mRNA expression of ${\beta}-actin,\;TNF-{\alpha}$, IL-4, IL-5,IL-13 in the mice group treated with PR-HA with RT-PCR decreased significantly compared with those of control group. Conclusion : The concentrations of IL-4, IL-5, IgE in BALF, and IL-4, IL-5, IL-13 in serum of the mice group treated with PR-HA decreased significantly compared with those of control group. The number of $Gr-1^+/CD11b^+\;and\;CD11b^+$ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with that of control group.   • Ex vivo lung perfusion (EVLP) is a technique that enables active metabolism of the lung by creating an environment similar to that inside the body, even though the explanted lungs are outside the body. The EVLP system enables the use of lung grafts that do not satisfy the acceptance criteria for lung transplantation (LTx) by making it possible to evaluate the function of the lung grafts and repair lungs in poor condition, thereby reducing the waiting time of patients requiring LTx and consequently mortality.

Curcumin Attenuates Radiation-Induced Inflammation and Fibrosis in Rat Lungs

  • A beneficial radioprotective agent has been used to treat the radiation-induced lung injury. This study was performed to investigate whether curcumin, which is known to have anti-inflammatory and antioxidant properties, could ameliorate radiation-induced pulmonary inflammation and fibrosis in irradiated lungs. Rats were given daily doses of intragastric curcumin (200 mg/kg) prior to a single irradiation and for 8 weeks after radiation. Histopathologic findings demonstrated that macrophage accumulation, interstitial edema, alveolar septal thickness, perivascular fibrosis, and collapse in radiation-treated lungs were inhibited by curcumin administration. Radiation-induced transforming growth factor-${\beta}1$ (TGF-${\beta}1$), connective tissue growth factor (CTGF) expression, and collagen accumulation were also inhibited by curcumin. Moreover, western blot analysis revealed that curcumin lowered radiation-induced increases of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), TNF receptor 1 (TNFR1), and cyclooxygenase-2 (COX-2). Curcumin also inhibited the nuclear translocation of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) p65 in radiation-treated lungs. These results indicate that long-term curcumin administration may reduce lung inflammation and fibrosis caused by radiation treatment.

Ameliorating Effects of Moxifloxacin on Endotoxin-Induced Acute Lung Injury in Rats (흰쥐에서 내독소로 유도된 급성 폐손상에서 moxofloxacin의 개선효과)

  • The immunomodulating effects of moxifloxacin seem to be effective in downregulating inflammatory reactions. This presumed effect was tested in endotoxin (ETX)-induced acute lung injury (ALI) in rats. After moxifloxacin treatment (10 mg/kg) of ETX-given rats, lung myeloperoxidase (MPO) activity, bronchoalveolar-lavage (BAL) protein, and the number of neutrophils in the BAL cells were measured. Light and electron microscopic structures were also examined. Electron microscopic $CeCl_3$ histochemistry for the detection of hydrogen peroxide in the lungs and immunohistochemistry of cytosolic phospholipase A2 (cPLA2) in the lung tissues and BAL cells were performed. To examine the expression of TNF${\alpha}$ in the lungs, western blotting was carried out with the lung tissues. ETX had accumulated neutrophils in the lungs, which was followed by lung leak. Oxidative stress occurred, and increased expression of cPLA2 in the lung tissues and BAL cells was observed in the ETX-given rats. Simultaneously, the expression of TNF${\alpha}$ was enhanced by ETX. Moxifloxacin, however, decreased all these parameters, indicating that ALI may have been ameliorated. Moxifloxacin appears to ameliorate ETX-induced ALI partially through the suppression of cPLA2 in the lungs of rats.

So a remedy about lung heat is clearing away lung heat, this lung heat makes inevitably bleed in lungs, therefore a remedy in this case is clearing the lung to stop bleeding, or moistening the lung. Exterior heat sign means that exterior syndrome coexists with heat syndrome and it means that a remedy of this syndrome need to mix prescriptions for relieving exterior syndrome and heat-clearing prescriptions to treat this syndrome.

Escherichia coli septicemia concurrent with mycotic infection in captive salt water crocodiles in Bangladesh

  • Crocodile farms are getting popular in Bangladesh in an economic point of view. In one of the farms, some crocodiles were found sick and three of them died between May and July in 2006. This investigation was performed to diagnose the cause of the death. Routine postmortem examination was conducted. Samples were collected in 10% neutral buffered formalin for histopathology and in falcon tube for microbiological study. Additional swabs were collected in nutrient broth. Histopathological and microbiological studies were conducted using routine procedures. In addition Giemsa, Gram and PAS stains were performed to detect the organism in tissues. Grossly, esophagus, trachea, lungs, liver, spleen, heart and kidney were congested. Intestine, rectum and colon were hemorrhagic. Clay colored material was found in colo-rectum. Purulent exudates in lungs and thick and cloudy pericardial fluid in pericardial sac were found. Histologically, multifocal granulomatous inflammation was evident in lung, liver, kidney, intestine and colon with bacterial colonies, fungal spores and hyphae. These bacteria were appeared as Gram negative. Fungal hyphae and spores were detected in liver, lungs and colon by using PAS stain. Bacteriologically, E. coli were isolated from lungs exudates, pericardial fluids and intestinal fluids. Therefore, it can be concluded that 3 crocodiles died due to E. coli septicemia concurrent with mycotic infection.

Biochemical characteristics and serotypes of Actinobacillus pleuropneumoniae isolated from pneumonic lungs of pigs (돼지 폐렴병소에서 분리한 Actinobacillus pleuropneumoniae의 특성에 관한 연구)

  • The present study was conducted to investigate the biochemical and serologic characteristic of Actinobacillus pleuropneumoniae isolated from pneumonic lungs of pigs during the period from January 1992 to April 1993. A pleuropneumoniae was isolated from 17(27.0%) of 63 growing pigs with respiratory signs and 21(6.4%) of 330 pneumonic lungs of slaughtered pigs. The seasonal isolation frequency of A pleuropneumoniae was higher in winter and spring than that in summer or fall. The biochemical and cultural properties of A pleluropneumoniae isolated from the pneumonic lungs of pigs were identical to those of the reference strains used. The isolates were highly susceptible to ampicillin, cephalothin, ceftiofur, ciprofloxacin(MIC : ${\leq}0.39{\mu}g/ml$) and moderately susceptible to amikacin, chloramphenicol, erythromycin, kanamycin, methicillin, penicillin-G, streptomycin(MIC : 0.78~25IU or ${\mu}g/ml$), respectively. Sulfadimethoxine, sulfamerazine, tylosine showed no response to the isolates(MIC : ${\geq}100{\mu}g/ml$). Among the 38 isolates, 21(55.3%) and 13(34.2%) were resistant to oxytetracycline aid lincomydn, respectively(MIC : ${\geq}50IU$ or ${\mu}g/ml$). The majority of 38 A pleuropneumoniae isolates were turned out as serotype 2(47.4%) or serotype 5(54.7%) and the remaining 3 isolates were evenly classified to serotype 7, 10 or 12. It was noted A pleuropneumonine serotype 5 isolates were more resistant to oxytetracycline than serotype 2 isolates.
