Aspirin이 출혈성 쇼크로 인한 급성 폐손상에 미치는 효과 (Aspirin Reduces Acute Lung Injury in Rats Subjected to Severe Hemorrhage)

  • 연구배경 : 급성 폐손상을 일으키는 요인중의 하나인 출혈성쇼크시 나타나는 급성 폐손상의 양상과 아스피린이 이에 미치는 효과를 알아보고자 본 연구를 시행하였다. 방법 : 실험동물은 체중 350g 내외의 Sprague-Dawley 종흰쥐를 사용하였고, 혈압측정 및 출혈을 시키기 위하여 catheter를 양쪽 대퇴동맥에 삽입하였다. 수술 후 polygraph를 이용하여 평균동맥압을 기록하였으며, 출혈은 withdrawal pump를 이용하여 5분간 체중 kg 당 20 ml의 혈액을 출혈시켰다. 실험군은 대조군, 출혈군과 아스피린 처치군으로 분류하였다. 대조군은 출혈군과 동일하게 수술하고 출혈은 시키지 않았으며 나머지 과정은 출혈군과 동일하게 처리하였다. 아스피린 처치군은 출혈 30분전 대퇴정맥으로 아스피린 (10mg/kg)을 주입하였고, 출혈군은 체중 당 동일한 양의 생리식염수를 주입하였다. 이상의 처치 후 2시간 또는 24시간 동안 cage에서 자유롭게 활동하도록 하였으며, 출혈후의 폐손상 정도와 아스피린이 이에 미치는 효과를 알아보기 위하여 폐장내 myeloperoxidase 활성도와 폐세척액 내의 단백함량과 백혈구 수를 측정하였다. 결과 : 폐장내 myeloperoxidase 활성도와 폐세척액 내의 단백함량과 백혈구수는 출혈 2시간 및 24시간 후 대조군에 비해 유의하게 증가하였다. 이러한 반응은 아스피린 전처치에 의하여 효과적으로 차단되었다. 결론 : 이상의 결과로 심한 출혈 후에 급성 폐손상이 생길 수 있으며 이는 아스피린 전처치로 효과적으로 예방될 수 있다고 생각된다.

정천화담항기탕(定喘化痰降氣湯)이 ovalbumin으로 유발된 천식(喘息)에 미치는 영향(影響) (The Histopathologic Effects of Jungcheonwhadamgangki-tang on the Experimental Asthma induced by Ovalbumin)

  • Objectives: In order to evaluate the effect of jungcheonwhadamgangki-tang on Balb/c mouse of allergy-sensitive to bronchial asthma induced by ovalbumin. Methods: The changes of diameter lumen of trachea which was upper respiratory organ, weight and gross appearance of lung, histology of lung and trachea, numbers of inflammatory cells in the bronchoalveolar lavage fluld(BALF) were observed. Results : The results are obtained as follows. 1. The diameters of trachea lumen were significantly increased in Jungcheonwhadam gangki-tang treated group as compared with control group. 2. Inflammatory cells including neutrophil and eosinophil in BALF were significantly. in Jungcheonwhadamgangki-tang treated group as compared with control group. 3. Weight of lung and black spots, which resulted from infiltration of inflammatory were significantly decreased in Jungcheonwhadamgangki-tang treated group as compared with control group.4. Hypertrophy of mucous membrane of trachea and bronchus and bronchioles in the lung, peritracheal, peribronchus and peribronchiolar inflammatory cell infiltration, and mucoid exudate deposit in the lumen were significantly decreased in Jungcheonwhadamgangki-tang treated group as compared with control group. Conclusions : It is considered that Jungcheonwhadamgangki-tang has somewhat favorable effect on the bronchial asthma because the bronchial asthma specific series of abnormalities in respiratory system were decreased after oral administration of Jungcheonwhadamgangki-tang in this study. In future, it is needed that the toxicological and dosage specific study of Jungcheonwhadamgangki-tang to use against bronchial asthma with safe.

리포다당질로 유도된 급성 폐손상 후 섬유화증식에서 Transglutaminase-2의 역할 (The Role of Transglutaminase-2 in Fibroproliferation after Lipopolysaccharide-induced Acute Lung Injury)

  • Background: Transglutaminase-2 (TG-2) has been reported to play an important role in the process of fibrosis. However, TG-2 studies on fibroproliferation of acute lung injury (ALI) are absent. The purpose of this study was to investigate the role of TG-2 in the fibroproliferation of lipopolysaccharide (LPS)-induced ALI. Methods: The male C57BL/6 mice of 5 weeks age were divided into 3 groups; control group (n=30) in which $50{\mu}L$ of saline was given intratracheally (IT), LPS group (n=30) in which LPS 0.5 mg/kg/$50{\mu}L$ of saline was given IT, and LPS+Cyst group treated with intraperitoneal 200 mg/kg of cystamine, competitive inhibitor of TG-2, after induction of ALI by LPS. TG-2 activity and nuclear factor $(NF)-{\kappa}B$ were measured in lung tissue homogenate. Tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, IL-6, myeloperoxidase (MPO), and transforming growth factor (TGF)-${\beta}1$ were measured using bronchoalveolar lavage fluids. Histopathologic ALI score and Mallory's phosphotunistic acid hematoxylin (PTAH) for collagen and fibronectin deposition were performed. Results: The TG-2 activities in the LPS group were significantly higher than the control and LPS+Cyst groups (p<0.05). The TNF-${\alpha}$ and IL-$1{\beta}$ concentrations and $NF-{\kappa}B$ activity were lower in the LPS+Cyst group than the LPS group (p<0.05). The LPS+Cyst group showed lower MPO, ALI score, TGF-${\beta}1$ concentration, and Mallory's PTAH stain than the LPS group, but the differences were not significant (p>0.05). Conclusion: Inhibition of TG-2 activity in the LPS-induced ALI prevented early inflammatory parameters, but had limited effects on late ALI and fibroproliferative parameters.

  • 인터루킨-1을 횐쥐의 기관지 내로 투여하였을 때 급성 부종성 폐손상(acute edematous lung injury)이 유발되며, 폐세척액 내 호중구가 증가되고 surfactant의 양도 증가한다. 대식세포는 증가된 surfactant 기인한 탐식작용 활동이 증가하여 세포 내 환상의 구조물들이 증가하나 정상군과 비교하여 형태적으로 세포기관의 구조에서 차이를 보이고 있다. 이러한 세포구성은 세포간 기능상 의 차이를 나타내는 것이며, surfactant의 순환과 연관성이 있을 것으로 사료된다. 본 연구에서 정상 군에 있어서 폐포강 내 대식세포가 환상의 구조물들을 합성하는 형태를 보였으며, 이것은 surfactant의 재합성과 분비작용과 매우 밀접한 연관성이 있음을 보여 주는 것이다.

Lonicerae Flos Inhibits Cigarette-induced Lung Inflammatory Responses in Animal Model of Chronic Obstructive Pulmonary Disease

  • Objectives: In the present study, we evaluate the anti-inflammatory effect of Lonicerae Flos on cigarette-induced lung inflammatory responses in animal model of chronic obstructive pulmonary diseases (COPD). Methods: To inspect the effects of Lonicerae Flos, we evaluated Lonicerae Flos functions in vivo including immune cell profiles in bronchoalveolar lavage (BAL) fluid, cytokine production and tissue morphological changes. Results: Lonicerae Flos significantly inhibited immune cell infiltrations into the BAL fluid (neutrophils, macrophages, lymphocytes). TNF-${\alpha}$, and interleukin-6 (IL-6) were substantially decreased in the BAL fluid of Lonicerae Flos-treated mice compared with cigarette-exposed control mice. In addition, the hypertrophy of goblet cells in the epithelial cells was reduced in both Lonicerae Flos- and roflumilast-treated mice. Conclusions: The results of this study provide evidence that treatment with Lonicerae Flos exerts strong therapeutic effects against cigarette-induced lung inflammation in vivo. Therefore, this herbal medicine may represent a novel therapeutic agent for lung inflammation in general, as well as a specific agent for the treatment of COPD.

Effect of Korean Red Ginseng on Artificial Sand Dust (ASD) Induced Allergic Lung Inflammation

  • Asian sand dust is known to promote various respiratory symptoms or disorders. For the prevention of harmful health effects by Asian sand dust, the best strategy is known to avoid or reduce exposure to the Asian sand dust. Several studies have shown that Korean red ginseng (RG) has anti-inflammatory and anti-allergic effects. The study aimed to clarify the effect of Korean red ginseng intake on lung inflammation responses to artificial sand dust (ASD) similar to Asian sand dust. BALB/c mice were divided into five groups (n=12) of control (saline), ovalbumin (OVA), OVA with ASD, OVA plus RG with ASD, and OVA plus dexamethasone (DEXA) with ASD. Histopathologic evaluation of lung was conducted. Interleukin (IL)-5, IL-12, interferon (IFN)-${\gamma}$, IL-13, monocyte chemotactic protein (MCP)-1, and eotaxin within bronchoalveolar lavage (BAL) fluid were measured by ELISA. OVA+ASD group significantly increased concentrations of IL-5, IL-13, MCP-1, and eotaxin (P<0.01) compared to the control. OVA+ASD+RG group showed significant decreased levels of IL-2, IL-13, MCP-1 and eotaxin (P<0.01) compared with OVA+ASD. Between RG and DEXA treatment groups, there was no significant difference in all cytokines and chemokines. The inflammatory cells were significantly decreased in treatment groups with RG or DEXA compared to OVA+ASD group. This study suggests a beneficial effect of Korean RG administration in preventing inflammation of lung resulting from Asian sand dust.

Free-Living Amoeba Vermamoeba vermiformis Induces Allergic Airway Inflammation

  • The high percentage of Vermamoeba was found in tap water in Korea. This study investigated whether Vermamoeba induced allergic airway inflammation in mice. We selected 2 free-living amoebas (FLAs) isolated from tap water, which included Korean FLA 5 (KFA5; Vermamoeba vermiformis) and 21 (an homolog of Acanthamoeba lugdunensis KA/E2). We axenically cultured KFA5 and KFA21. We applied approximately 1×106 to mice's nasal passages 6 times and investigated their pathogenicity. The airway resistance value was significantly increased after KFA5 and KFA21 treatments. The eosinophil recruitment and goblet cell hyperplasia were concomitantly observed in bronchial alveolar lavage (BAL) fluid and lung tissue in mice infected with KFA5 and KFA21. These infections also activated the Th2-related interleukin 25, thymic stromal lymphopoietin, and thymus and activation-regulated chemokines gene expression in mouse lung epithelial cells. The CD4+ interleukin 4+ cell population was increased in the lung, and the secretion of Th2-, Th17-, and Th1-associated cytokines were upregulated during KFA5 and KFA21 infection in the spleen, lung-draining lymph nodes, and BAL fluid. The pathogenicity (allergenicity) of KFA5 and KFA21 might not have drastically changed during the long-term in vitro culture. Our results suggested that Vermamoeba could elicit allergic airway inflammation and may be an airway allergen.

Immunomodulatory Effects of Ambroxol on Airway Hyperresponsiveness and Inflammation

  • Ambroxol is used in COPD and asthma to increase mucociliary clearance and regulate surfactant levels, perhaps through anti-oxidant and anti-inflammatory activities. To determine the role and effect of ambroxol in an experimental model of asthma, BALB/c mice were sensitized to ovalbumin (OVA) followed by 3 days of challenge. Airway hyperresponsiveness (AHR), lung cell composition and histology, and cytokine and protein carbonyl levels in bronchoalveolar lavage (BAL) fluid were determined. Ambroxol was administered either before the first OVA challenge or was begun after the last allergen challenge. Cytokine production levels from lung mononuclear cells (Lung MNCs) or alveolar macrophages (AM) were also determined. Administration of ambroxol prior to challenge suppressed AHR, airway eosinophilia, goblet cell metaplasia, and reduced inflammation in subepithelial regions. When given after challenge, AHR was suppressed but without effects on eosinophil numbers. Levels of IL-5 and IL-13 in BAL fluid were decreased when the drug was given prior to challenge; when given after challenge, increased levels of IL-10 and IL-12 were detected. Decreased levels of protein carbonyls were detected in BAL fluid following ambroxol treatment after challenge. In vitro, ambroxol increased levels of IL-10, IFN-γ, and IL-12 from Lung MNCs and AM, whereas IL-4, IL-5, and IL-13 production was not altered. Taken together, ambroxol was effective in preventing AHR and airway inflammation through upregulation of Th1 cytokines and protection from oxidative stress in the airways.

The storage mite Tyrophagus putrescentiae induces greater lung inflammation than house dust mites in mice

  • Exposure to storage mite (SM) and house dust mite (HDM) allergens is a risk factor for sensitization and asthma development; however, the related immune responses and their pathology have not been fully investigated. The HDMs Dermatophagoides farinae and Dermatophagoides pteronyssinus and SM Tyrophagus putrescentiae are potent allergens that induce asthma. Most SM-related studies have focused on the allergic reactions of individuals by measuring their immunoglobulin (Ig)E expression. Considering the limited research on this topic, the present study aims to investigate the differences in the immune responses induced by HDMs and SMs and histologically analyze lung tissues in a mouse asthma model to understand the differential effects of HDM and SM. The results revealed that all mite species induced airway inflammation. Mice challenged with T. putrescentiae had the highest airway resistance and total cell, eosinophil, and neutrophil counts in the bronchoalveolar lavage fluid (BALF). The SM-sensitized groups showed more severe lesions and mucus hypersecretions than the HDM-sensitized groups. Although the degree of HDM and SM exposure was the same, the damage to the respiratory lung tissue was more severe in SM-exposed mice, which resulted in excessive mucin secretion and increased fibrosis. Furthermore, these findings suggest that SM sensitization induces a more significant hypersensitivity response in mucosal immunity than HDM sensitization in asthma models.

급성(急性) Ethyl 알콜 중독(中毒) 흰쥐의 폐용압률(肺容壓率)과 폐포활성물질(肺胞活性物質)의 변화(變化) (Effect of Acute Ethanol Intoxication on the Pulmonary Compliance and Surfactant in Rats)

  • Alcohol 의 급성중독(急性中毒)으로서 폐기능중(肺機能中) 특(特)히 폐용압률(肺容壓率)과 폐포활성물질(肺胞活性物質)(surfactant)의 변화(變化)를 살펴봄으로써 alcohol 이 폐기능(肺機能) 또는 폐대사(肺代謝)에 미치고 효과(效果)의 일단(一端)을 알기 위(爲)하여 흰쥐를 재료(材料)로 하여 50% alcohol 을 체중(體重) 100 gm 당(當) 1.5 ml 씩 경구적(經口的)으로 단회투여(單回投與)하여 급성(急性) Ethyl alcohol 중독(中毒)을 일으켰다. Alcohol 중독과정(中毒過程)에서 24 시간(時間)까지 경시적(經時的)으로 혈중(血中) alcohol 농도(濃度)와 hematocrit(Hct)치(値)를 측정(測定)하고, 폐용압률(肺容壓率) 및 surfactant 중(中)의 단백량(蛋白量)과 무기인(無機燐)(Pi)를 측정(測定)하여, 정상군(正常群)과 비교(比較)하여 다음과 같은 결과(結果)를 얻었다. Alcohol 혈중농도(血中濃度)는 정상치(正常値)인 $0.17{\pm}0.01\;gm%$(평균(平均)${\pm}$표준오차(標準誤差) 이하동일(以下同一))에 비(比)하여 중독(中毒) 1시간(時間)에서 $0.71{\pm}0.02\;gm%$로 최고치(最高値)에 달(達)하고, 그후(後) 차차(次次) 낮아져서 24 시간(時間)에서는 $0.36{\pm}0.02\;gm%$이었으나, 전실험치(全實驗値)는 정상(正常)에 비(比)해서는 유의(有意)하게 높았다. Hct 치(値)는 1시간(時間)에서 $64.86{\pm}2.45%$로서 최고치(最高値)를 나타내었고, 그후(後) 계속(繼續) 정상치(正常値)인 $48.87{\pm}0.28%$에 비(比)하여 유의(有意)하게 높은 값을 유지(維持)하였다. 폐중량대(肺重量對) 체중비(體重比)(L/B 비(比))는 중독(中毒) 3시간(時間)부터 $5.49{\pm}0.61$로서 정상치(正常値)인 $7.57{\pm}0.56$에 비(比)하여 유의(有意)하게 낮아지고, 24시간(時間)까지 유의(有意)한 감소(減少)를 나타내었다. 폐용압률(肺容壓率)은 확대(擴大)(inflation)와 축소(縮小)(deflation)에서 모두 3시간(時間)까지 정상(正常)과 큰 차이(差異)가 없었으나, 6및 12 시간(時間)에서는 증가(增加)된 값을 나타내었고, 특(特)히 12 시간(時間)에서는 유의(有意)하게 높은 값을 나타내었다가, 24시간(時間)에는 정상(正常)에 가까워지는 경향(傾向)을 나타내었다. 폐세척액(肺洗滌液)의 단백량(蛋白量)은 정상(正常)의 $10.65{\pm}0.62\;mg/gm$에 비(比)하여 3시간(時間)에서 $12.36{\pm}0.35\;mg/gm$, 12시간(時間)에서 $12.70{\pm}0.74\;mg/gm$ 및 24시간(時間)에서는 $12.65{\pm}0.88\;mg/gm$로서 유의(有意)하게 높은 값을 나타내었고, Pi치(値)도 중독(中毒) 6시간(時間)부터는 높은 값을 나타내었으며, 특(特)히 12 시간(時間)에서는 $7.65{\pm}0.63\;{\mu}mol/gm$, 24시간(時間)에서는 $6.70{\pm}0.36\;{\mu}mol/gm$로서, 정상(正常)의 $5.32{\pm}0.20\;{\mu}mol/gm$에 비(比)하여 유의(有意)하게 높은 값을 나타내었다. 한편, 폐추출액(肺抽出液)의 단백량(蛋白量)은 1및 3시간(時間)에서 낮은 값을 나타내었으나, 대체(大體)로 정상(正常)과 비슷한 경향(傾向)을 나타내었고, Pi 치(値)는 대체(大體)로 높은 양상(樣相)을 나타내었으며, 특(特)히 중독(中毒) 6, 12 및 24 시간(時間)에서는 각각(各各) $17.77{\pm}1.54\;{\mu}mol/gm$, $13.92{\pm}0.78\;{\mu}mol/gm$$14.57{\pm}5.53\;{\mu}mol/gm$로서, 정상(正常)의 $10.34{\pm}0.37\;{\mu}mol/gm$에 비(比)하여, 모두 유의(有意)하게 높은 값을 나타내었다. 이상(以上)의 결과(結果)로서 alcohol 급성중독(急性中毒)에서는, surfactant 의 활성도(活性度)나 폐용압률(肺容壓率)이 증가(增加)되어 폐기능(肺機能)에 변화(變化)가 초래(招來)된다고 하겠다.
