  • A lymphosarcoma was diagnosed in a 4-year-old female Formosan sika deer presented with persistent reproductive failure, anorexia, depression and diarrhea. Characteristic pathological findings were infiltration of neoplastic lymphoid cells and cancer emboli in the lymph nodes, heart, lung, kidney, urinary bladder, ovary, uterus and peritoneal fat tissue.

Implications of NQO1 in cancer therapy

  • NAD(P)H:quinone oxidoreductase (NQO1), an obligatory two-electron reductase, is a ubiquitous cytosolic enzyme that catalyzes the reduction of quinone substrates. The NQO1- mediated two-electron reduction of quinones can be either chemoprotection/detoxification or a chemotherapeutic response, depending on the target quinones. When toxic quinones are reduced by NQO1, they are conjugated with glutathione or glucuronic acid and excreted from the cells. Based on this protective effect of NQO1, the use of dietary compounds to induce the expression of NQO1 has emerged as a promising strategy for cancer prevention. On the other hand, NQO1-mediated two-electron reduction converts certain quinone compounds (such as mitomycin C, E09, RH1 and β-lapachone) to cytotoxic agents, leading to cell death. It has been known that NQO1 is expressed at high levels in numerous human cancers, including breast, colon, cervix, lung, and pancreas, as compared with normal tissues. This implies that tumors can be preferentially damaged relative to normal tissue by cytotoxic quinone drugs. Importantly, NQO1 has been shown to stabilize many proteins, including p53 and p33ING1b, by inhibiting their proteasomal degradation. This review will summarize the biological roles of NQO1 in cancer, with emphasis on recent findings and the potential of NQO1 as a therapeutic target for the cancer therapy.

Anti-cancer Activities of Ginseng Extract Fermented with Phellinus linteus

  • In the present study, the anti-cancer effects of ginseng fermented with Phellinus linteus (GFPL) extract were examined through in vitro and in vivo assays. GFPL was produced by co-cultivating ginseng and Phellinus linteus together. Ginsenoside Rg3, Rh1 and Rh2 are important mediators of anti-angiogenesis and their levels in GFPL were enriched 24, 19 and 16 times, respectively, more than that of ginseng itself through the fermentation. GFPL exhibited distinct anti-cancer effects, including growth inhibition of the human lung carcinoma cell line A549, and promotion of immune activation by stimulating nitric oxide (NO) production in Raw 264.7 cells. Further evidence supporting anti-cancer effects of GFPL was its significant prolongment of the survival of B16F10 cancer cell-implanted mice. These results suggest that the GFPL may be a candidate for cancer prevention and treatment through immune activation and anti-angiogenic effects by enriching Rg3, Rh1 and Rh2.

  • Naturally-ocurring short-lived decay products of radon gas in indoor air are the dominant source of ionizing radiation exposure to the general public. It is written in BEIR VI Report(l999l the radon progeny were identified as the second cause of lung cancer next to cigarette or 10 % to 14 %(15,400 to 21,800 persons p.a.) of all lung cancer deaths in USA. Indoor radon concentrations in houses typically result from radon gaining access to houses mainly from the underlying soil. In the States, they have "Indoor Radon Abatement Act" which was converted from "Toxic Substance Control Act" in 1988 to establish the national long-term goal that indoor air should be as free of radon as the ambient air outside of buildings. To review and study techniques for controlling radon, two test cells were constructed for a series of tests and are under measuring indoor and soil gas (underneath of floor slab)radon concentrations according to EPA's measurement protocol. In this paper, important theoretical studies are previewed and the following paper will explain the test results and confirm the theories reviewed to find out suitable coefficients. On the basis of test analysis, it will be described and evaluated various techniques that can be used to mitigate elevated indoor concentration of radon including the control of radon and its decay products.

  • Formaldehyde (FA) is an important industrial chemical, but it can cause allergic reactions, sick building syndrome and so on. It has also been observed to cause cancer in scientific studies using laboratory animals, and it even causes cancer in humans. Natural products such as ginseng and pine needle containing complicated mixtures of organic chemicals are widely used in the world, because their effective components are responsible for some pharmacological activities including antioxidative effect, anticancer effect. We investigate the effect of Korean ginseng (GE), pine needle extract (PE) and combined GE and PE (cNPE) on mouse lung injury by FA exposure. GE, PE and cNPE was directly transported to pulmonary cells through respiratory organ by nebulizer inhalation. In the case of FA exposure, the pulmonary structure was damaged and its function became abnormal. However, cNPE-FA, GE-FA, and PE-FA treated groups showed similar with the control group compared with FA group. Among them, GE was proved to be more effective than any other extracts. These results demonstrate that natural product extracts could protect pulmonary structure and function against FA exposure.

Induction of G1 Arrest by Methanol Extract of Lycopus lucidus in Human Lung Adenocarcinoma A549 Cells (택란 메탄올 추출물에 의한 인체 폐암 세포주 A549의 G1 arrest 유발)

  • Induction of G1 Arrest by Methanol Extract of Lycopus lucidus in Human Lung Adenocarcinoma A549 Cells Lycopus lucidus, a herbaceous perennial, is used as a traditional remedy in East Asia, including China and Korea. It has been reported that L. lucidus has anti-allergic effects, inhibitory effects on cholesterol acyltransferase in high glucose-induced vascular inflammation, and anti-proliferative effects in human breast cancer cells. However, the molecular mechanisms of the anti-cancer effects of L. lucidus have not yet been fully determined. In this study, we evaluated the anti-cancer effect and the mechanism of action of L. lucidus in human lung adenocarcinoma A549 cells using methanol extracts of L. lucidus (MELL). MELL treatment showed cytotoxic activity in a dose-dependent manner and induced G1 arrest in A549 cells. The induction of G1 arrest by MELL was associated with the up-regulation of phospho-CHK2 and the down-regulation of Cdc25A phosphatase. In addition, MELL treatment induced decreased expression of G1/S transition-related proteins, including CDK2, CDK4, CDK6, cyclin D1 and cyclin E. MELL also regulated the mRNA expression of CDK2 and cyclin E. On the other hand, the expression of p53 and the cyclin-dependent kinase inhibitor p21 was not induced by MELL. Collectively, these results suggest that MELL may exert an anti-cancer effect by cell cycle arrest at G1 phase through the ATM/CHK2/Cdc25A/CDK2 pathway in A549 cells.

  • Signet ring cell carcinoma of lung is an unique variant of mucin producing adenocarcinoma which is characterized by abundant intracellular mucin accumulation. Only a few cases of primary signet ring cell carcinoma of lung have been reported in the world wide literature. And we have, recently experienced one case of primary signet ring cell carcinoma of lung. A 55 years old man was evaluated for paralysis of lower extremities and was found to have lung cancer in the left upper and lower lobe with pleural, multiple spinal, bone and liver metastases. Signet ring tumor cells were revealed by cytologic examination of pleural fluids. And there were no evidence of signet ring cell carcinoma of other organs. Primary signet ring cell carcinoma of lung seems to have an aggressive behavior and therapeutic modalities could be different from those for signet ring cell carcinomas from other organs. Therefore it is important to separate primary signet ring cell adenocarcinoma of lung from metastatic tumors.
