Function and Oligomerization Study of the Leucine Zipper-like Domain in P13 from Leucania separata Multiple Nuclear Polyhedrosis Virus

  • The p13 gene is uniquely present in Group II nucleopolyhedroviruses (NPVs) and some granuloviruses, but not in Group I NPVs. p13 gene was first described by our laboratory in Leucania separatamultiple nuclear polyhedrosis virus (Ls-p13) in 1995. However, the functions of Ls-P13 and of its homologues are unknown. When Ls-p13 was inserted into Autographa californica nucleopolyhedrovirus, a Group I NPV, polyhedra yield was inhibited. However, this inhibition was prevented when the leucine zipper-like domain of Ls-p13 was mutated. To determine the cause of this marked difference between Ls-P13 and leucine zipper mutated Ls-P13 (Ls-P13mL), oligomerization and secondary structure analyses were performed. High performance liquid chromatography and yeast two-hybrid assays indicated that neither Ls-P13 nor Ls-P13mL could form oligomers. Informatics and circular dichroism spectropolarimetry results further indicated marked secondary structural differences between Ls-P13 and Ls-P13mL. The LZLD of Ls-P13 has two extended heptad repeat units which form a hydrophobic surface, but it is short of a third hydrophobic heptad repeat unit for oligomerization. However, the mutated LZLD of Ls-P13mL lacks the above hydrophobic surface, and its secondary structure is markedly different. This difference in its secondary structure may explain why Ls-P13mL is unable to inhibit polyhedra yield.

p13 from group II baculoviruses is a killing-associated gene

  • p13 gene was first described in Leucania separata multinuclear polyhedrosis virus (Ls-p13) several years ago, but the function of P13 protein has not been experimentally investigated to date. In this article, we indicated that the expression of p13 from Heliothis armigera single nucleocapsid nucleopolyhedrovirus (Ha-p13) was regulated by both early and late promoter. Luciferase assay demonstrated that the activity of Ha-p13 promoter with hr4 enhancer was more than 100 times in heterologous Sf9 cells than that in nature host Hz-AM1 cells. Both Ls-P13 and Ha-P13 are transmembrane proteins. Confocal microscopic analysis showed that both mainly located in the cytoplasm membrane at 48 h. Results of RNA interference indicated that Ha-p13 was a killing-associated gene for host insects H. armigera. The AcMNPV acquired the mentioned killing activity and markedly accelerate the killing rate when expressing Ls-p13. In conclusion, p13 is a killing associated gene in both homologous and heterologous nucleopolyhedrovirus.

Clinical and molecular characteristics of Korean children with Cornelia de Lange syndrome

  • Purpose: Cornelia de Lange syndrome (CdLS) is a rare genetically heterogeneous disorder caused by genetic variants of the cohesin complex. However, the diverse genetic etiologies and their phenotypic correlations in Korean patients with CdLS are still largely unknown. Hence, this study aimed to clarify the clinical characteristics and genetic background of Korean patients with CdLS. Materials and Methods: The medical records of 15 unrelated patients (3 males and 12 females) genetically confirmed to have CdLS were retrospectively reviewed. All individuals were diagnosed with CdLS using target gene analysis, whole-exome sequencing, and/or chromosomal microarray analysis. The clinical score (CS) was calculated to assess disease severity. Results: The median age at diagnosis was 1.7 (range, 0.0-11.8) years, and median follow-up duration was 3.8 (range, 0.4-11.7) years. Eight (53.3%) patients showed classic phenotypes of CdLS, two (13.3%) showed non-classic phenotypes, and five (33.3%) had other phenotypes sharing limited signs of CdLS. Fifteen causative variants were identified: NIPBL in five (33.3%, including 3 males), SMC1A in three (20.0%), SMC3 in three (20.0%), and HDAC8 in four (26.7%) patients. The CS was significantly higher in the NIPBL group than in the non-NIPBL group (14.2±1.3 vs. 8.7±2.9, P<0.001). Conclusion: We identified the clinical and genetic heterogeneity of CdLS in Korean patients. Patients with variants of NIPBL had a more distinctive phenotype than those carrying variants of other cohesin complex genes (SMC1A, SMC3, and HDAC8). However, further studies are warranted to understand the pathogenesis of CdLS as a cohesinopathy and its genotype-phenotype correlations.

Immediate Effects of Muscle Tension and Pain to Myofascial Release and Duoball Assisted Self-Relaxation Techniques in Patients Experiencing Chronic Cervical Pain

  • Objective: This study aims to investigate the immediate effects of myofascial release and Duoball assisted self-relaxation (DASR) techniques on pain and muscle tension in patients experiencing chronic cervical pain. Design: A randomized controlled trial. Methods: This study is a randomized controlled experimental study. Eighteen patients with chronic neck pain who met the selection criteria were randomly assigned to myofascial release group and myofascial release group using Duoball. Results: The frequency results for assessment muscle tension showed a decrease of about 10% in the suboccipital muscle, SCM, Pect m, UT, and LS in both the MFR and DASR groups, and the stiffness results showed a decrease in all muscles except the upper trapezius in the MFR group and the DASR group. All were found to decrease by about 10% in the suboccipital muscle, SCM, Pect m, UT, and LS, and the decrement results showed an increase of about 15% in the suboccipital muscle, SCM, Pect m, UT, and LS in both the MFR and DASR groups(p<0.05). Conclusions: In patients experiencing chronic neck pain, application of MFR and duoball assisted self relaxion was shown to be effective on pain and muscle tension. MFR is a non-pharmacological intervention method with few potential side effects and is considered a universal and easily applicable treatment method.

Analysis of the Dynamic Behavior of Guardrail Posts in Sloping Ground using LS-DYNA (LS-DYNA를 이용한 비탈면에 설치된 가드레일 지주의 동적거동)

  • PURPOSES : This paper presents a finite element model to accurately represent the soil-post interaction of single guardrail posts in sloping ground. In this study, the maximum lateral resistance of a guardrail post has been investigated under static and dynamic loadings, with respect given to several parameters including post shape, embedment depth, ground inclination, and embedment location of the steel post. METHODS : Because current analytical methods applied to horizontal ground, including Winkler's elastic spring model and the p-y curve method, cannot be directly applied to sloping ground, it is necessary to seek an alternative 3-D finite element model. For this purpose, a 3D FHWA soil model for road-base soils, as constructed using LS-DYNA, has been adopted to estimate the dynamic behavior of single guardrail posts using the pendulum drop test. RESULTS : For a laterally loaded guardrail post near slopes under static and dynamic loadings, the maximum lateral resistance of a guardrail post has been found to be reduced by approximately 12% and 13% relative to the static analysis and pendulum testing, respectively, due to the effects of ground inclination. CONCLUSIONS : It is expected that the proposed soil material model can be applied to guardrail systems installed near slopes.

Influence of Berseem and Lucerne Silages on Feed Intake, Nutrient Digestibility and Milk Yield in Lactating Nili Buffaloes

  • This study was conducted to evaluate feeding value of berseem and lucerne silage as a replacement for conventional fodder (berseem fodder) in lactating Nili buffaloes. Fifteen early lactating multi-parous Nili buffaloes, five buffaloes in each group were allotted three experimental diets. Berseem and lucerne fodders were ensiled at 30% DM (wheat straw was used to adjust the DM of fodders) with molasses (at the rate of 2% of fodder DM) in two bunker silos for 30 days. The diets contained 75% DM from berseem fodder (BF), 75% DM from berseem silage (BS) and 75% DM from lucerne silage (LS). Each diet contained 25% concentrate DM. Diets were mixed daily and fed twice a day at ad libitum intakes. Dry matter intake (DMI) was significantly higher (13.8 kg/d) in buffaloes fed BF diet than those fed LS (12.5 kg/d) and BS (11.9 kg/day) diets. The differences in digestible DMI and DMI as percent body weight were significant between fodder and silage based diets but non-significant when BS and LS were compared. Lower DMI with silage-based diets was probably because of low silage pH. Intake of NDF (NDFI) was higher (5.68 kg/d) in buffaloes fed BF diet followed by those fed LS (5.50 kg/d) and BS (5.00 kg/d) diets. The difference was significant (p<0.05) across fodder and silage based diets but NDFI was non-significant across both silage-based diets. The apparent DM digestibility was significantly different (p<0.05) between fodder and silage-based diets but was non-significant between LS and BS diets. Four percent fat corrected milk yield was significantly different (p<0.05) between fodder and silage-based diets but was non-significant between LS and BS diets. Higher milk yield with fodder based diet was because of more digestible nutrient intake (Table 3) compared with silage based diets. Milk CP, TP and NPN and SNF did not show any treatment effects. The present results indicated that the berseem and lucerne fodder ensiled at 30% DM level with 2% molasses could safely replace (75% DM) the conventional leguminous fodder in the diets of lactating Nili buffaloes.

Patterns of Perimenstrual Symptoms and Related Dietary Factors to Premenstrual Syndromes (월경전후기증상의 유형과 월경전증상 관련식이요인)

  • Purpose: The purpose of this study was to differentiate between women with three perimenstrual symptom severity patterns : premenstrual syndrome(PMS), premenstrual magnification(PMM), and low symptom(LS), and to explore the related dietary factors to premenstrual symptoms. Method: Women were asked to keep a diary record of perimenstrual symptoms and food intake for 50 days. Result: Symptom patterns were defined for 26 among 38 women ; Eight(21.1%) demonstrated a PMS pattern, three(7.9%) demonstrated a PMM pattern, and fifteen(39.5%) exhibited a LS pattern. There were significant differences in symptom scores during the premenstrual phase($x^2=19.30$, p=.000), menstrual phase($x^2=13.32$, p=.001), and post menstrual phase($x^2=9.93$, p=.007) for three groups. Protein, vit E, vit C, niacin, folic acid, and phosphorus in the premenstrual phase, and energy, and vit B6 in the menstrual period were significantly different between the three groups. Among dietary compositions, amino acids, lipids, fatty acids, saturated fatty acids, natrium, vit B6, niacin, and vit E were negatively related to PMS symptoms. Conclusion: Pattern of perimenstrual symptoms should be differentiated for individualized PMS management. As a more efficient diet assessment for PMS women, randomized nutritional analysis during the 3 phases of the menstrual cycle should be done and a replication study is necessary with a larger sample.

Use of Dual-Energy X-ray Absorptiometry in Children with Inflammatory Bowel Disease: A Large Single Centre Study

  • Purpose: Low bone mineral density (BMD) is a complication in children with inflammatory bowel disease (IBD). There are limited data evaluating dual-energy x-ray absorptiometry (DXA) as a screening tool for low BMD in children with IBD. We performed a single site retrospective analysis of DXA use. Methods: Children aged 5-18 years with IBD diagnosed between 2013 to 2017 at the Royal Children's Hospital, Australia, were included. Patient demographics, measures of disease activity, DXA scores, and factors related to BMD were collected. Results: Over a median follow up of 5.1 (4-6.4) years, 72/239 (30.1%) children underwent DXA, and 28/239 (11.7%) children had a second DXA. Our DXA practice differed to consensus guidelines regarding initial screening based on height and/or body mass index (BMI) z-score (8/17 [47.1%]), and repeat surveillance (13/42 [31.0%]). Children had a median lumbar spine (LS) z-score -0.80 (-1.65-0.075). Children with LS z-score≤-2.0 (n=14) had lower weight (6.57 [1.78-23.7] vs. 51.1 [26.5-68.7], p=0.0002) and height centiles (3.62 [1.17-17.1] vs. 42 [16.9-67.1], p=0.0001), and higher faecal calprotectin (FCP) (3041 [1182-4192] vs. 585 [139-2419], p=0.009) compared to children with LS z-score>-2.0. No fractures were reported. Of 28 children who underwent a second DXA 1.6 (1.1-2.2) years following initial DXA, no significant change in z-scores occurred. Conclusion: Children with IBD had low BMD. In addition to height centile and weight centile, FCP was associated with lower BMD, and should be considered in DXA screening guidelines. Greater clinician awareness of DXA consensus guidelines is required. Future prospective studies are required.

Analysis of Importance, Understanding Level and Needs by Subject of College Students Preparing for Radiological Technologists National Examination (방사선사 국가시험 준비를 위한 대학생들의 과목별 중요도와 이해도 수준 및 요구도 분석)

  • The aim of this study analyzed the important level (IL) and understanding level (UL) including the Borich's need for students preparing of the national examination for radiological technologists at online open chatting room. A total of 254 survey were collected from a total of 1,016 students who used open chatting room from December 13 to December 16, 2022. A general characteristics were the age, gender, curriculum (3 or 4 years), grade and area. The IL, UL, learning satisfaction (LS), learning achievement (LA) and intention to continue using (ICU) were analyzed by using the 5 point Likert scale. There was no significant difference the LS, LA, and ICU according to general characteristics (p>0.05). There was a statistically significant difference a total of sixteen subjects based on the t-test results of the response values from the IL and UL (p<0.05). The total of ten subjects with the highest priority in the Locus for Focus models were the Ultrasonography, Human anatomy, Magnetic resonance imaging, Radiation therapy, Cardiovascular and intervention, Computed tomography, Human physiology, Radiographic imaging, Fluroscopic radiography, and Nuclear medicine) that the Borich's need was also the same as the top 10 ranked subjects. The LS (4.23±0.72), LA (4.18±0.73), and ICU (4.15±0.78) for open chatting room were high. This study identified the subjects most needed by college students by the Borich's need analysis. First, it is necessary to provide intensive education on subjects with high scores that are most needed by college students. Second, it is necessary to improve the teaching method for subjects with low need and low level of understanding.

Comparison of the Diagnostic Performance of $^{14}C$-urea Breath Test According to Counting Method for the Diagnosis of Helicobacter pylori Infection (Helicobacter pylori 감염 진단 시 $^{14}C$-요소호기검사의 계수측정 방법에 따른 진단성능 비교)

  • Purpose: $^{14}C$-urea breath test (UBT) is a non-invasive and reliable method for the diagnosis of Helicobacter pylori (HP) infection. In this study, we evaluated the diagnostic performance of a new and rapid $^{14}C$-UBT (Heliprobe method), which was equipped with $Geiger-M\ddot{u}ller$ counter and compared the results with those obtained by using the conventional method. Materials and Methods: Forty-nine patients with dyspepsia underwent gastroduodenoscopy and $^{14}C$-UBT. A 37 KBq $^{14}C$-urea capsule was administered to patients and breath samples were collected. In Heliprobe method, patients exhaled into a Hellprobe BreathCard for 10 min. And then the activities of the BreathCard were countered using Heliprobe analyzer. In the conventional method, results were countered using liquid scintillation counter. During gastroduodenoscopy, 18 of 49 patients were underwent biopsies. According to these histologic results, we evaluated the diagnostic performance of two different methods and compared them. Also we evaluated the concordant and disconcordant rates between them. Results: In all 49 patients, concordant rate of both conventional and Heliprobe methods was 98% (48/49) and the discordant rate was 2% (1/49). Thirteen of 18 patients to whom biopsies were applied, were found to be HP positive on histologic results. And both Heliprobe method and conventional method classified 13 of 13 HP-positive patients and 5 of 5 HP-negative patients correctly (sensitivity 100%, specificity 100%, accuracy 100%). Conclusion: The Heliprobe method demonstrated the same diagnostic performance compared with the conventional method and was a simpler and more rapid technique.
