We investigated the effects of water extracts of unripe black raspberry (UBR) and red ginseng (RG) on cholesterol synthesis, 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase activity, and expression of low-density lipoprotein (LDL) or high-density lipoprotein (HDL)-related genes in HepG2 and Caco-2 (human hepatoma and intestinal cell lines, respectively). Our results showed that cholesterol synthesis and HMG-CoA reductase activity in HepG2 cells were inhibited by UBR and RG. Further, co-treatment with UBR and RG had a greater effect than did treatment with either UBR or RG. In Caco-2 cells, treatment with UBR and RG increased the expression of LDL-regulated genes, such as LDL receptor and SREBP-2, and also upregulated the level of HDL-associated ABCA1. Moreover, co-treatment with UBR and RG appeared to be more effective than treatment with either UBR or RG. Taken together, our results indicate that UBR and RG regulate the level of HDL-associated ABCA1 via signaling pathway, thereby preventing cholesterol synthesis.
Body fat reducing and physical-activity enhancing effects, along with artherosclerosis improving effects, of conjugated linoleic acid (CLA) were elucidated on obese male middle school students with more than 30% body fat. Twenty-four volunteers were randomly divided into control (placebo, n=12) and CLA treatment (n=12) groups. Subjects were daily fed 6 g CLA (6 capsules, twice a day) or a placebo for 12 weeks. At the end of the experiment, body composition, blood lipid composition and exercise capacities of subjects were measured. CLA significantly reduced body fat content and body mass index (BMI) along with body weight, while the placebo did not have any such effects. Similarly, CLA significantly reduced low-density lipoprotein (LDL)-cholesterol, total cholesterol, and triglyceride, but elevated high-density lipoprotein (HDL)-cholesterol content in blood. Meanwhile, in terms of exercise capacity, there were significant enhancements of trunk flexion, closed-eyes foot balance, standing long jump, shuttle run, and sit-up activities in the CLA treatment group. These results indicate that CLA consumption reduced body fats, improved atherosclerosis factors in blood and improved physical activity of young male obese middle school students, and suggest that CLA could be a useful material for the heath care of obese young men.
In humans, N-terminal pro-B-type natriuretic peptide (NT-proBNP) was shown to be inversely related to obesity; in addition, its association with contributing factors for obesity such as insulin, lipids, and glucose profiles has been demonstrated in the literature. However, this association between NT-proBNP and the severity of obesity has not been investigated in veterinary medicine. Our study hypothesis is that plasma levels of NT-proBNP may be related to body condition score (BCS) and contributing factors to obesity in dogs with heart diseases. To achieve our study goal, we collected blood samples from 73 client-owned dogs of small breeds at different stages of heart failure due to chronic mitral valvular insufficiency (CMVI). Fasting glucose concentrations, lipid profiles (i.e., total triglycerides [TG], total cholesterol [TC], high-density lipoprotein cholesterol [HDL-C], low-density lipoprotein cholesterol [LDL-C]), fructosamine, insulin and NT-proBNP concentrations were measured. The insulin/glucose ratio was also determined. NT-proBNP showed not only a significant correlation with the severity of CMVI related heart failure but also an inverse relationship to body condition scores (BCS), insulin plasma levels and fructosamine concentrations. We found the presence of an inverse relationship between plasma levels of NT-proBNP and the severity of obesity. In addition, NT-proBNP was associated with lower levels of contributing factors to obesity such as fructosamine and insulin, creating a possible link between the obesity and NT-proBNP in dogs with heart disease. This is also the first report demonstrating an inverse association between obesity and NT-proBNP in dogs with heart failure.

This is a non-experimental and retrospective study aimed at determining the effects of long-term hospitalization on the body mass index (BMI) and lipid metabolism in long-term hospitalized patients. The study subjects included 120 patients aged 40-65 years who were hospitalized for >3 months in 2 long-term care hospitals in Gyeonggi-do, South Korea. In this study, the BMI and levels of total cholesterol, triglycerides (TG), high-density lipoprotein (HDL), and low density lipoprotein (LDL) at admission and 3 months after hospitalization were compared and analyzed, and the related changes over time were followed up. The general characteristics of the subjects were analyzed by using descriptive statistics and frequency analysis. In addition, logistic regression analysis was performed to determine the effects of the general characteristics on the BMI and Dyslipidemia. The changes in the BMI and blood lipid levels between admission and 3 months after hospitalization were analyzed using the paired t-test. The results showed that with regard to the changes in the blood lipid levels, the triglyceride levels significantly increased 3 months after hospitalization (p<.05). These findings imply that long-term hospitalization for care and rehabilitation after acute-phase treatment should be considered a potential high-risk factor for dyslipidemia, which could be prevented or alleviated by providing the patients with health education, including exercise and dietary education.
This study was designed to determine whether low-density lipoporoteins (LDL) extracted from egg yolk in extender improve the function of Korean Jeju Black Bull semen. The semen was cryopreserved with 5% ethylene glycol (EG) or 7% glycerol (G) extenders containing 10% egg yolk (EY), 4% LDL and 5% EY or 8% LDL. Frozen-thawed sperm were evaluated sperm motility, viability, membrane integrity and acrosome integrity. Post-thawed sperm motility has been significantly higher (p<0.05) in 4% LDL + 5% EY ($69.00%{\pm}4.18$; EG and $63.00%{\pm}9.75$; 7% G) than 8% LDL ($57.00%{\pm}5.70$; EG and $52.00%{\pm}4.47$;G). Treatment of 4% LDL + 5% EY-EG ($66.85%{\pm}5.06$) has been significantly improved sperm viability compared to other treatments except 10% EY - EG. Moreover, in membrane integrity, swollen sperm ratio has been only significantly increased (p<0.05) in 4% LDL + 5% EY - EG ($64.65%{\pm}6.10$) among all treatments. In assess to detect acrosome integrity, especially, AR pattern ratio has been significantly decreased (p<0.05) in 4% LDL + 5% EY - EG among all treatments. In sperm viability as time passes, between 4% LDL + 5% EY and 10% EY, there was no significant difference, but 8% LDL was significantly decreased sperm viability in EG (1 and 2 hrs) and G (30 min, 1, 2, 5 and 12 hrs) extender. However, there were no significant differences among all treatments except 8% LDL-G in sperm membrane integrity. 8% LDL-G has been significantly decreased swollen sperm ratio at 5 hrs after thawed. It is concluded from these results that 4% LDL + 5% EY to the freezing extender showed more positive effect on the frozen-thawed spermatozoa in Korean Jeju Black bull.

In hypertriglyceridaemic individuals, atherosclerogenesis is associated with the increased concentrations of very low density lipoprotein (VLDL) and VLDL-associated remnant particles. In vitro studies have suggested that VLDL induces foam cells formation. To reveal the changes of the proteins expression in the process of foam cells formation induced by VLDL, we performed a proteomic analysis of the foam cells based on the stimulation of differentiated THP-1 cells with VLDL. Using two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser-desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis, 14 differentially expressed proteins, containing 8 up-regulated proteins and 6 down-regulated proteins were identified. The proteins are involved in energy metabolism, oxidative stress, cell growth, differentiation and apoptosis, such as adipose differentiation-related protein (ADRP), enolase, S100A11, heat shock protein 27 and so on. In addition, the expression of some selected proteins was confirmed by Western blot and RT-PCR analysis. The results suggest that VLDL not only induces lipid accumulation, but also brings about foam cells diverse characteristics by altering the expression of various proteins.
The purpose of this study was to investigate the effect of Lycii fructus on the serum lipid in rats fed high fat diet. We compared the effects of L. fructus and L.fructus water extract both adminstered with high fat diets on rats that had previously been on high fat or standard diets. Two separate experiments were conducted for 6 weeks. respectively. In experiment I, 4 groups of rats were fed experimental diets consisting of either \circled1 6 weeks of a standard diet(control), \circled2 6 weeks of a high-fat diet(HHC), \circled3 3 weeks of a high-fat diet followed by 3 weeks of a high-fat diet containing L. fructus(HHL) or \circled4 6 weeks of a high-fat diet with L. fructus extract in place of water for the last 3 weeks (HHT). In the second set of experiments, a high-fat diet (SHC), high-fat diet containing L.fructus(SHL) or high-fat diet with L. fructus extract in place of water (SHT) were fed for 3 weeks after 3 weeks of standard diet feeding. Rats fed L. fructus diet consumed more diets than high-fat diets. THe results of experiment I showed significant decreases(p<0.05) in serum triglyceride(TB) and low density lipoprotein-cholesterol (LDL-C) levels with L. fructus feedings, but did not show andy changes in total cholesterol (TC) level. High density lipoprotein-cholesterol (HDL-C) level was increased significantly(p<0.05) with L. fructus feedings. Therefore, the ration of LDL-C to HDL-C(LDL-C/HDL-C) which is used as an atherosclerosis index was significantly (p<0.05) low, while the HDL-c/TC ration was significantly(p<0.05) high with L.fructus intake. However, no significant were found in serum cholesterols and TG levels in experimentII. The results of these experiments indicate that , regardless of the feeding from, L. fructus can be beneficial in lowering serum TG and LDL-C levels for habitual high-fat diet intakers. L.fructus also seems to be effective in elevating serum HDL-C level, theregy having beneficial effects on atherosclerosis by influencing the serum lipoprotein profile.
This study investigated the effects of chromium propionate on growth performance, serum traits and immune response in weaned pigs. Twenty-four 4 wk-old crossbred weanling pigs (initial body weight about 9.52${\pm}$0.48 kg) were randomly allotted into one of two groups, a control group (basal diet), chromium propionate group (diet supplemented with 200 ${\mu}g$$kg^{-1}$ (ppb) of chromium propionate). This experiment was conducted over nine weeks. Escherichia coli lipopolysaccharide (LPS) 100 ${\mu}g$$kg^{-1}$ BW was used as the stress-inducing agent in the middle (4 wks) and final (8 wks) periods. The experimental results indicated that chromium propionate had no effect on growth performance (p>0.05). Chromium propionate supplementation reduced the percentage of LDL+VLDL (low and very low-density lipoprotein) and increased HDL (high-density lipoprotein), but did not affect other serum traits. Pigs supplemented with chromium propionate had higher antibody titers specific for sheep red blood cells (SRBC) and serum total globulin relative to the control during the final period (p<0.05). A challenge with LPS increased white blood cells in the chromium propionate group in both experimental periods (p<0.05). The chromium propionate group exhibited higher IgG and $\gamma$-globulin than the control during the middle experimental period (p<0.05). Moreover, the PHA (phytohemagglutinin) challenge result in the chromium propionate group was better than the control group (p=0.056). Greater neutrophil activity was displayed than in the control (p<0.05). This suggests that chromium propionate supplementation benefited the weaned pigs in lipoprotein and immune response.

Purificationi of egg yolk immunoglobulin(IgY) was performed to understand the property of egg immunoglobulin. IgY differs from mammalian IgY in the molecular size(larger), isoelectric point(more acidic), and binding ability with mammalian complement and protein A(nonbinding ability). IgY is also known as ${\gamma}$-livetin and exists in egg yolk together with other two water-solubel proteins, ${\alpha}$-livetin(chicken serum albumin) and ${\beta}$-livetin(${\alpha}_2$-glycoprotein) and various lipoproteins(Low density lipoprotein, LDL and High density lipoprotein, HDL) which are the major components of egg yolk. The first step of isolation of IgY is to separate the water-solube proteins from lipoproteins. We report a simple method for separation of water soluble proteins using k-carrageenan and sedimentation. k-carrageenan was found to be effective for removal of yolk lipoprotein as a precipitate. IgY remained supernatant, and was isolated by chromatography on columns of DEAE-Sephacel and G 75 gel filtration chromatography.
This study compared the lipid metabolism of white Leghorn layers between the growing and the laying periods. The study was conducted on thirty layers in the growing period (14 weeks old) and in the initial laying period (36 weeks old; 14 weeks from the onset of laying). Results indicated that all plasma lipid traits were significantly different (p<0.05) between the two periods. The estrogen concentration in the laying period was about 3 fold that in the growing period. Triacylglycerol (TG) concentration in the laying period was about 12 fold that in the growing period. The phospholipids, cholesterol, glycerol and non esterified fatty acid (NEFA) in the laying period were also higher than those in the growing period (p<0.05). Lipogenic enzyme activities in the laying period were higher than in the growing period (p<0.05). High-density lipoprotein (HDL) was the largest lipoprotein portion in growing layers. In laying hens, the major lipoprotein portion was very low-density lipoprotein (VLDL). It was also shown that protein is the primary component of HDL and TG is the largest portion of VLDL in both periods. In laying hens, VLDL contained more TG than that in growing layers (p<0.05). The VLDL particle size in laying hens was larger than that in growing layers (p<0.05). This study indicates that the lipid metabolism traits of a layer in the laying period were different from those in the growing period. The lipogenesis related traits in the laying period were markedly greater than in the growing period.
