한우의 등심과 사태조직 유래 근육위성세포의 성장단계별 유전발현 차이 분석 (Transcriptomic Analysis of the Difference of Bovine Satellite Cell Between Longissimus dorsi and Semimembranosus on Hanwoo Muscle Tissues)

  • 한우의 성장단계별 부위 근육발달을 이해하는 것은 도체율 개선에 따른 소득증대와 증체율 증가에 따른 생산효율 향상에 긍정적인 영향을 미친다. 본 연구에서는 한우의 등심과 사태 유래 근육위성세포를 분리 후 세포단위의 발달 및 분화를 비교하여 transcriptome 단위의 작용기전을 제시하였다. 한우의 부위별 근육 유래 근육위성세포의 근섬유의 양은 4일에 가장 높게 나타났고 이후 감소하였다. 한우의 근육위성세포의 발달 단계에 따라 발현되는 총 전사체 유전자의 종류는 사태근육 위성세포에서 높게 나타났다. 등심과 사태 근육 유래 위성세포의 발달단계에 따라 유의적인 차등 유전자 453개를 찾아냈고 이를 이용한 기능유 전체 분석이 필요하다. 등심과 사태유래 근육위성세포를 이용한 동일조건 분화 비교에서 사태유래 근육 위성세포의 분화 시 myosin complex, skeletal muscle contraction, troponin complex, skeletal muscle tissue development 와 같은 근섬유 형성관련 유전자의 발현이 높게 나타나는 것으로 보아 같은 개체의 근육조직에서도 부위별로 차등 발달이 되고 있다는 것을 알 수 있다. 기존 연구에서는 근육의 성장에 대한 이해를 위해 사양과 영양관련 시험이 많이 이루어졌다. 향후 세포단위의 연구들이 많이 이루어져 작용기작에 대한 생물정보 자료를 추가로 적용한다면 한우의 정밀사양을 적용할수 있는 바탕이 마련될 것이다. 또한 근육위성세포의 연구는 추후 동물실험 윤리제도 강화에 따른 비동물 전임상 screening 시험 활용과 대체단백질 산업의 주요 이슈인 배양육 소재 개발 연구와 같이 축산시험연구의 지속적인 확장성에 많은 영향을 미칠 것으로 생각된다.

Estimation of Correlation Coefficients between Histological Parameters and Carcass Traits of Pig Longissimus Dorsi Muscle

  • The aim of this study was to investigate the histochemical parameters of muscle fibers, and to estimate the correlation between these histological parameters and carcass traits in pigs. A total of 230 crossbred Duroc$\times$(Yorkshire$\times$Landrace) pigs (149 gilts and 81 castrated male pigs) was evaluated. Carcass traits (carcass weight, backfat thickness, and loin eye area), muscle fiber size (crosssectional area, diameter, and perimeter), muscle fiber number (density of fibers/$mm^2$ and total number of fibers), and fiber type composition (percentages of myofibers and relative areas of each fiber type) were evaluated. Mean cross-sectional area (CSA) and type IIB fiber CSA were positively correlated to carcass weight, backfat thickness and loin eye area. Mean fiber CSA was mostly related to type IIB CSA (r=0.98) as a result of the high percentage of type IIB fibers in the longissimus muscle. Correlations between fiber diameters and perimeters were also high, and showed similar results with CSA. Mean fiber density was negatively correlated to carcass weight (r=-0.24), backfat thickness (r=-0.18) and loin eye area (r=-0.27). To the contrary, total fiber number was positively correlated with carcass weight (r=0.27) and loin eye area (r=0.53). Carcass weight and loin eyZe area were not significantly related to muscle fiber composition. For backfat thickness, there was an opposition between type IIA percentage, which was positively related and type IIB percentage, which was negatively related. Fiber type composition of type I and IIA fibers were negatively correlated to that of type IIB fibers (r=-0.67 to -0.74). In the present study, carcass weight and loin eye area were positively correlated to CSA and negatively correlated to fiber density. But, these relationships were generally low. The fiber density was strongly affected by muscle fiber size and the total fiber number was affected either by CSA of muscle fiber and loin eye area. Fiber type composition was much more related to their numerical abundance than their CSA.

Effects of Muscle Mass and Fiber Number of Longissimus dorsi Muscle on Post-mortem Metabolic Rate and Pork Quality

  • The aim of this study is to investigate the effects of the muscle mass and fiber number on post-mortem metabolic rates and pork quality. Carcass traits, muscle fiber characteristics, and type of fiber composition were evaluated using a sample of 200 cross-bred pigs. The muscle mass was divided into two groups according to carcass weight and loin-eye area measurements (heavy or light). In addition, the muscle histological characteristics were divided into two groups according to the muscle fiber density and total number of muscle fibers (high or low). All the carcass traits were significantly different in the muscle mass groups. Increasing weight significantly affected the cross-sectional area (CSA) of all fibers. The low group, which had a low muscle fiber number indicating a larger CSA of fibers, and especially the heavy-low group had the highest CSA levels of fibers. The fiber number percentage and the area percentage were significantly different in the groups categorized by fiber number. The heavy-high group indicated a normal rate of pH decline and the R-value. In addition, pigs with a heavy muscle mass and high muscle fiber number indicated normal drip loss, lightness, and protein denaturation. The present results suggest that increasing the total muscle fiber number has a beneficial effect on increasing the muscle mass without deteriorating the meat quality.

한우 비육 전·후기의 등심조직에 있어서 지방합성 유전자 발현 (Lipogenesis Gene Expression Profiling in Longissimus dorsi on the Early and Late Fattening stage of Hanwoo)

  • 지방합성이 왕성하게 진행되는 비육후기의 근육내 지방합성에 있어서, 지방산 및 당의 이용경로 및 이에 따른 근육내 지방합성과정을 규명하기 위해서 지방산 및 당 운반 유전자인 FABP4, GLUT4와 근육내 지방대사 주요유전자인 ACL, ACC, LPL 및 SCD 유전자의 mRNA 발현양상을 분석하였다. 한우 비육전기 및 후기 각 3두를 공시하여 total RNA 추출 및 1st cDNA 합성하여 SYBR green을 이용하여 real-time PCR 분석을 각 유전자별로 3반복씩 수행하였다. FABP4 유전자는 비육전기에 비해 비육후기에 있어서, 3배 이상 유전자 발현이 증가함을 확인할 수 있었고, GLUT4 유전자는 비육전기 및 비육후기에서 큰 차이를 보이지 않았다. 또한 근육내 지방합성 주요유전자인 ACL, ACC, LPL 및 SCD 유전자의 발현량은 비육후기에서 ACL 유전자가 3.8배, ACC 유전자는 2.7배, LPL 유전자는 3.5배, 그리고 SCD 유전자는 7.5배 발현량이 증가하였다. 따라서 본 연구를 통하여 한우의 비육후기에서 근육내 지방합성은 FABP4에 의한 지방산 유입의 증가와 더불어 ACL 유전자에 의해서 지방산 합성의 중간대사물인 acetyl-CoA가 합성되고, 이 중간 대사물을 이용하여 ACC 및 SCD 유전자에 의해서 긴 사슬 지방산 합성이 왕성하게 일어남을 알 수 있었다.

Effect of Different Storage-Temperature Combinations on Longissimus dorsi Quality upon Sous-vide Processing of Frozen/Thawed Pork

  • This study investigated the effect of storage state (chilled state on sous-vide, CS; frozen state without thawing on sous-vide, FS; and frozen/thawed states on sous-vide, TS) and sous-vide cooking temperature ($65^{\circ}C$ and $72^{\circ}C$) on the longissimus dorsi muscle quality of pork. FS showed a higher moisture content than that of CS and TS (p<0.001), whereas both FS and CS showed higher expressible moisture loss than that of TS (p<0.001). FS showed a lower cooking loss (p<0.001) than that of CS and TS. FS and TS exhibited significantly higher lipid oxidation than that of CS. Carbonyl and sulfhydryl content were not significantly affected by the storage treatment. FS and TS exhibited lower shear force than that of CS (p<0.001). FS and TS showed higher springiness than that of CS (p<0.001), FS exhibited lower gumminess than that of CS and TS (p<0.01). Sous-vide treatment at $65^{\circ}C$ exhibited significantly higher moisture content and lower expressible moisture loss, cooking loss, and total and sarcoplasmic protein than those at $72^{\circ}C$. Shear force and springiness of $65^{\circ}C$-treated groups were lower than those of $72^{\circ}C$-treated groups (p<0.01). Cooking temperature significantly influenced overall acceptability, whereas the storage state did not affect the overall acceptability. These results indicated that meat quality might be improved upon cooking from the frozen or frozen/thawed state using sous-vide when compared with traditional processing.

옥수수 사일리지가 거세한우의 성장, 도체특성 및 등심 유전자 발현에 미치는 영향 (Effects of Corn Silage on Growth Performance, Carcass Characteristics, and Gene Expression of Longissimus dorsi Muscle Biopsy of Hanwoo Steers)

  • 본 연구는 사료비의 절감을 위해 배합사료 비율을 줄이고 조사료 비율을 높이면서 한우 고급육으로 생산할 수 있는 방법을 찾아내기 위해 수행하였다. 대조구(고급육 사양)와 옥수수 사일리지 급여구(대조구 대비 배합사료 66% 급여 및 옥수수 사일리지)의 성장, 도체성적 및 등심근육 내유전자 발현에 미치는 영향에 대하여 분석했다. 옥수수 사일리지 급여는 비육 후기의 일당 증체량 및 출하체중이 수치적으로 낮게 나타났지만 유의미한 차이는 나타나지 않았다. 도체특성에서도 옥수수 사일리지 급여구가 도체중, 등심단면적 및 근내 지방도가 수치적으로 낮게 나타났지만 유의미한 차이는 나타나지 않았다. 육량 A등급 출현은 옥수수 사일리지 급여구가 23% 높게 나타났으며, 육질 1++등급 출현율은 대조구가 17% 높게 나타났다. 이는 옥수수 사일리지 급여구가 대조구 대비 배합사료 급여량이 적음으로 인해 나타난 결과로 보여진다. 등심내 유전자 발현에서는 옥수수 사일리지 급여구가 근육발달 유전자 발현이 높게 나타났으며, 지방발달 유전자 발현에서는 대조구가 높게 나타났다. 본 연구의 결과 배합사료의 고급육 사양 대비 옥수수 사일리지 사양이 육질 1++등급의 출현이 다소 감소하지만 육량 A등급의 출현을 높여주므로 사료비 절감의 생산 방법 중 하나로 될 수 있을 것으로 보여진다.

Detection of Quantitative Trait Loci for Growth and Carcass Traits on BTA6 in a Hanwoo Population

  • The purpose of this study was to detect quantitative trait loci (QTL) for growth and carcass quality traits on BTA6 in a population of Hanwoo cattle. Three hundred and sixty one steers were produced from 39 sires that were sired by 17 grandsires in the two Hanwoo farming branches of the National Livestock Research Institute of Korea, between Spring 2000 and Fall 2002. DNA samples were collected for all of the steers, sires and grandsires, and the phenotypes for six growth and carcass quality traits were measured at 24 months of age. Twelve microsatellite markers were chosen on BTA6 and a linkage map was constructed by using seven of the twelve markers. Then, a chromosome-wide QTL scan was performed by applying an Animal Model, in which effects of QTL alleles within the grand sires were fitted as a random term. Three QTL were detected at the 5% chromosome-wise level for backfat thickness, average daily gain, and final weight. The most likely positions for the QTL were in the proximal region, i.e. 0 cM, 35 cM, and 63 cM, respectively. Also, another QTL for longissimus dorsi muscle area was detected at the 10% chromosome-wise level at 67 cM. These results were, in general, consistent with our previous report, in which candidate gene analyses showed that a SNP near ILSTS035 flanked by BM4621 (62.5 cM) and BMS2460 (81.3 cM) was associated with final weight, carcass weight, average daily gain, and longissimus dorsi muscle area in the same Hanwoo population.

Study on Growth Curves of Longissimus dorsi Muscle Area, Backfat Thickness and Body Conformation for Hanwoo (Korean Native) Cows

  • The objective of this study was to estimate the parameters of Gompertz growth curves with the measurements of body conformation, real-time ultrasound longissimus dorsi muscle area (LMA) and backfat thickness (BFT) in Hanwoo cows. The Hanwoo cows (n = 3,373) were born in 97 Hanwoo commercial farms in the 17 cities or counties of Gyeongbuk province, Korea, between 2000 and 2007. A total of 5,504 ultrasound measurements were collected for the cows at the age of 13 to 165 months in 2007 and 2008. Wither height (HW), rump height (HR), the horizontal distance between the top of the hips (WH), and girth of chest (GC) were also measured. Analysis of variance was conducted to investigate variables affecting LMA and BFT. The effect of farm nested in location was included in the statistical model, as well as the effects of HW, HR, WH, and GC as covariates. All of the effects were significant in the analysis of variance for LMA and BFT (p<0.01), except for the HR effect for LMA. The two ultrasound measures and the four body conformation traits were fitted to a Gompertz growth curve function to estimate parameters. Upper asymptotic weights were estimated as $54.0cm^2$, 7.67 mm, 125.6 cm, 126.4 cm, 29.3 cm, and 184.1 cm, for LMA, BFT, HW, HR, WH, and GC, respectively. Results of ultrasound measurements showed that Hanwoo cows had smaller LMA and greater BFT than other western cattle breeds, suggesting that care must be taken to select for thick BFT rather than an increase of only beef yield. More ultrasound records per cow are needed to get accurate estimates of growth curve, which, thus, helps producers select animals with high accuracy.

고품질 한우를 위한 여러 경제형질에서의 주요 SNP 규명 (Important SNPs Identification from the Economic Traits for the High Quality Korean Cattle)

  • 고품질 한우를 만들기 위해 여러 경제형질에 영향을 주는 유전자 즉 single nucleotide polymorphisms(SNPs)를 규명하려고 한다. 이미 Lee 등 (2008a)에 의해 SNP(19_1)$^*$SNP(28_2)가 등심단면적 (LMA: longissimus muscle dorsi area)에 주요한 유전자로 규명되었다. 여기에 추가로 도체중 (CWT: carcass cold weight)과 일당증체량 (ADG: average daily gain)을 선형 모형에 적용하였으며 또한 상호작용에 더 유리하고 연속형 데이터에도 사용할 수 있는 expanded multifactor dimensionality reduction (expanded MDR)을 이용하여 주요한 SNP를 파악하였다. Expanded MDR 적용결과 등심단면적과 같은 결과인 SNP(19_1)과 SNP(19_1)$^*$SNP(28_2)의 상호작용 형태가 가장 좋은 SNP로 선정되었으며, 최종적으로 SNP(19_1)*SNP(28_2) 마커가 한우의 여러 경제형질에 우수 유전자임을 규명하였다.

Evaluation of the Activities of Antioxidant Enzyme and Lysosomal Enzymes of the Longissimus dorsi Muscle from Hanwoo (Korean Cattle) in Various Freezing Conditions

  • This study was conducted to evaluate the activities of antioxidant enzyme (glutathione peroxidase (GSH-Px)) and lysosomal enzymes (alpha-glucopyranosidase (AGP) and beta-N-acetyl-glucosaminidase (BNAG)) of the longissimus dorsi (LD) muscle from Hanwoo (Korean cattle) in three freezing conditions. Following freezing at -20, -60, and $-196^{\circ}C$ (liquid nitrogen), LD samples (48 h post-slaughter) were treated as follows: 1) freezing for 14 d, 2) 1 to 4 freeze-thaw cycles (2 d of freezing in each cycle), and 3) refrigeration ($4^{\circ}C$) for 7 d after 7 d of freezing. The control was the fresh (non-frozen) LD. Freezing treatment at all temperatures significantly (p<0.05) increased the activities of GSH-Px, AGP, and BNAG. The $-196^{\circ}C$ freezing had similar effects to the $-20^{\circ}C$ and $-60^{\circ}C$ freezing. Higher (p<0.05) enzymes activities were sustained in frozen LD even after 4 freeze-thaw cycles and even for 7 d of refrigeration after freezing. These findings suggest that freezing has remarkable effects on the activities of antioxidant enzyme and lysosomal enzymes of Hanwoo beef in any condition.
