Comparison of an Automated Most-Probable-Number Technique TEMPO®TVC with Traditional Plating Methods PetrifilmTM for Estimating Populations of Total Aerobic Bacteria with Livestock Products (축산물가공품에서 건조필름법과 TEMPO®TVC 검사법의 총세균수 비교분석)

  • We compared between an automated most-probable-number technique $TEMPO^{(R)}$TVC and traditional plating methods $Petrifilm^{TM}$ for estimating populations of total aerobic bacteria in various livestock products. 257 samples randomly selected in local retail stores and 87 samples inoculated with $E.$ $coli$ ATCC 25922, $Staphylococcus$ $aureus$ ATCC 12868 were tested in this study. The degree of agreement was estimated according to the CCFRA (Campden and Chorleywood Food Research Association Group) Guideline 29 and the agreement indicates the difference of two kinds methods is lower than 1 log base 10($log_{10}$). The samples of hams, jerky products, ground meat products, milks, ice creams, infant formulas, and egg heat formed products were showed above 95% in the agreement of methods. In contrast, proportion of agreement on meat extract products, cheeses and sausages were 93.1%, 92.1%, 89.1%, respectively. One press ham and five sausages containing spice and seasoning, two pork cutlets containing spice and bread crumbs, two meat extract product and two natural cheeses and one processing cheese with a high fat content, and one ice cream containing chocolate of all samples showed the discrepancy. Our result suggest that $TEMPO^{(R)}$TVC system is efficient to analyses total aerobic bacteria to compare manual method in time-consuming and laborious process except livestock products having limit of detection.

  • In December 2014, Livestock and Livestock Products Traceability Act was established, allowing consumers to receive livestock traceability information. While the Livestock Traceability System provides consumers with transparent and fair information about their food, it has brought increased workload and penalty burdens to stakeholders in the livestock industry. In this paper, we propose an IoT system for inventory management based on traceability numbers to enable sellers to conveniently provide livestock traceability information to consumers. We analyzed the protocol for managing data from electronic scales and conducted functional testing and verification on mobile devices. Furthermore, we implemented the design and system functionality, taking into account UI/UX on Android OS-based devices to synchronize and interconnect traceability and product information with electronic scales. We anticipate that the proposed approach will minimize user inconvenience and raise production efficiency in the existing market.

Effect of Dietary Chlorella Supplementation on Growth Performance, Immune Response, and Intestinal Micro Flora Concentration of Broiler Chickens (육계 사료 내 클로렐라의 첨가·급여가 생산성, 장내미생물 및 면역력에 미치는 영향)

  • A study was conducted to investigate the effect of dietary supplementation of feedstuff of Chlorella (Chlorella vulgaris) to replace of antibiotic in the diets of broiler chickens. A total of 720 1-d-old straight run broiler chicks (Ross ${\times}$ Ross) was randomly assigned into six treatments with four replicate pens (30 birds/replicate pen) for 5-wk. A corn-soy bean meal basal diet was formulated, the treatment groups were negative group (NC, antibiotic-free diet) and 0.1% virginiamycin in as antibiotic growth promoters (PC), 1.0% fresh liquid Chlorella (T1), 1.0% dried Chlorella powder (T2), 1.0% commercial Chlorella product and 1.0% (T3) and commercial Chlorella product 0.5% (T4) were added to the basal diet to form six dietary treatments. No significant differences were found among the treatments for feed intake and feed conversion of broiler chickens during the whole experimental period, but the BW gain was significantly higher (P<0.05) in commercial Chlorella product supplemental groups than the control group (NC and PC groups). Dietary supplementation of Chlorella significantly (P<0.05) increased the plasma IgA, IgM and IgG concentration of chicks compared to NC and PC groups. Supplemental AGPs and commercial chlorella product did not affect the E. coli and Salmonella concentration in the intestinal microflora of broiler chicks; however, the population of Lactobacillus was significantly increased (P<0.05) when birds were fed commercial Chlorella product groups. It is concluded that commercial Chlorella product supplementation could be used as an alternative of antibiotics to promote growth and immune response by increasing the production of lactic acid bacteria in the intestinal microflora of broiler chickens.

Influence of Feeding Flammuling veluipes Media on Productivity and Egg Quality in Laying Hens (팽이버섯 배지 부산물의 급여가 산란계의 생산성 및 난질에 미치는 영향)

  • This study was conducted to evaluate the feeding value of the Flammulina veluipes media by-product(FMB) in laying hens(Hy-Line Brown). A total of three hundreds sixty were fed corn-soy based experimental diets containing 0(control), 5.0(T1), and $10\%(T2)$ FMB for 12 weeks. There were no significant differences among the treatments in egg production, egg weight, egg mass, feed conversion and viability during the experimental period. Feed intake was significantly(P<0.05) lowered in control(120.5g) than 72(123.9g). There were no significant differences among the treatments eggshell breaking strength, thickness and haugh unit, whereas the yolk color of T1 and T2 were significantly love. than control(p<0.05). In conclusion, the FMB can be used as resource of feed in laying hen feed at $5.0\%$ level without effect on performance and egg qualify. However, dietary pigments must be added when the FMB was used as a feed ingredients more than $10\%$ in diets.

Contamination status of groundwater used as livestock drinking in beef and dairy cattle farms, Korea (국내 소사육농가의 자가용 가축음용수 오염실태 평가조사)

  • In Korea, groundwater is main water source in livestock farms. Most dairy and cattle farms have constructed their own wells for human drinking and livestock farming. However, these private residential wells have not been controlled by government and also there was scant study about livestock drinking water quality. Therefore this study was to monitor of the livestock farms' groundwater quality in Korea. Water samples were collected at 123 dairy and cattle farms and were analysed forty six substances with quality standard for drinking water approved by the Minister of Environment. Seventy eight (63.4%) of 123 samples failed to drinking water stand a test. The most frequent contaminants were nitrate-nitrogen and microbial. 22.8% (n=28) of samples showed nitrate-N concentration of higher than 10 mg/L meant that can't be used drinking water for human and the Nitrate-N concentration analysed in the range of 0.2 to 61.2 mg/L. All of 78 failed to drinking samples had microbial problems, especially 5.7% (n=7) of samples indicated water could be contaminated by feces. Other contaminants detected were zinc and evaporation residue. Especially detected zinc concentration (32 mg/L) was about ten times higher than standard of zinc (3 mg/L). Regression analysis indicated that groundwater pH did not influence to nitrate-N concentration but the hardness and chloride could affect to nitrate-N concentration in the groundwater. Most livestock farms were adjacent to crop farmland in Korea. This could cause contamination of groundwater with nitrate-N and pesticide that could accumulate livestock product. Moreover Heavy metal such as zinc and copper could be released from a corrosive plated water pipe in livestock farm. Put together, Korea livestock system is indoor, not pasture-based, hence livestock could be exposed to potential contaminated water consistently. Therefore on the basis of these data, appropriate livestock drinking water quality standards should be prepared to keep livestock healthy and their product safe. Further, livestock drinking water quality should be monitored continuously in suitable livestock drinking water standards.

Genetic variants of the growth differentiation factor 8 affect body conformation traits in Chinese Dabieshan cattle

  • Objective: The growth differentiation factor 8 (GDF8) gene plays a key role in bone formation, resorption, and skeletal muscle development in mammals. Here, we studied the genetic variants of GDF8 and their contribution to body conformation traits in Chinese Dabieshan cattle. Methods: Single nucleotide polymorphisms (SNPs) were identified in the bovine GDF8 gene by DNA sequencing. Phylogenetic analysis, motif analysis, and genetic diversity analysis were conducted using bioinformatics software. Association analysis between five SNPs, haplotype combinations, and body conformation traits was conducted in 380 individuals. Results: The GDF8 was highly conserved in seven species, and the GDF8 sequence of cattle was most similar to the sequences of sheep and goat based on the phylogenetic analysis. The motif analysis showed that there were 12 significant motifs in GDF8. Genetic diversity analysis indicated that the polymorphism information content of the five studied SNPs was within 0.25 to 0.5. Haplotype analysis revealed a total of 12 different haplotypes and those with a frequency of <0.05 were excluded. Linkage disequilibrium analysis showed a strong linkage (r2>0.330) between the following SNPs: g.5070C>A, g.5076T>C, and g.5148A>C. Association analysis indicated these five SNPs were associated with some of the body conformation traits (p<0.05), and the animals with haplotype combination H1H1 (-GGGG CCTTAA-) had greater wither height, hip height, heart girth, abdominal girth, and pin bone width than the other (p<0.05) Dabieshan cattle. Conclusion: Overall, our results indicate that the genetic variants of GDF8 affected the body conformation traits of Chinese Dabieshan cattle, and the GDF8 gene could make a strong candidate gene in Dabieshan cattle breeding programs.

Antioxidant and Antimicrobial Efficacy of Sapota Powder in Pork Patties Stored under Different Packaging Conditions

  • The present study was undertaken to assess the efficacy of sapota powder (SP) as natural preservatives and its better utilization in food processing with the incorporation of various levels of SP (2, 4, and 6%) by replacing lean meat. Based on the sensory attributes, pork patties with 4% incorporation of SP was found optimum and selected for further storage studies with control under aerobic and modified atmosphere packaging at refrigeration temperature ($4{\pm}1^{\circ}C$) for 42 days for assessing its antioxidant and antimicrobial efficiency. During entire storage period, indicators of lipid oxidative parameters such as thiobarbituric acid reactive substances (TBARS), free fatty acids (FFA) and peroxide value (PV) followed an increasing trend for control as well as treated products; however, treated product showed a significantly (p<0.05) lower value than control. A significantly lower (p<0.05) microbial count in treated patties than control was noted during entire storage. The sensory attributes are better retained in treated product as compared to control and even on $42^{nd}$ day, overall acceptability of treated patties was found to fall in moderately acceptable category (5.95 in aerobic packets and 5.91 in modified atmosphere packets). Therefore SP has potential to enhance antioxidant and antimicrobial properties of pork patties during storage.

Optimization of Processing Conditions of Chinese Smoke-cured Bacon (Larou) with a New Natural Coating Solution during Storage Period

  • The study aims to determine the optimum sterilization rate and water activity of Chinese traditional smoke-cured bacon product (Larou) in the preservation with natural coating solution. With the response surface methodology (RSM), we analyzed 3 factors of processing conditions (the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan) and 2 response factors (sterilization rate and water activity). Sterilization rate and water activity of Larou were largely affected by the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan. The final optimum concentrations of lysozyme, sodium alginate, and chitosan were 0.09, 1.40, and 1.60% and realized the high sterilization rate. Water activity of sliced Larou was significantly correlated with the sterilization rate. Low-field nuclear magnetic resonance analysis verified the optimum processing conditions. The coating resulted in 99.69% rate of reducing bacteria after 30-day storage. The data of the total number of colony, peroxidation value, moisture content, pH, and sensory evaluation provided the theoretical basis for extending the shelf life of Chinese traditional smoke-cured bacon product (Larou) with natural coating solution.

  • Membrane technology has revolutionized the dairy sector. Different types of membranes are used in the industry for various purposes like extending the shelf life of milk without exposure to heat treatment, standardization of the major components of milk for tailoring new products as well increasing yield and quality of the dairy products, and concentrating, fractionation and purification of milk components especially valuable milk proteins in their natural state. In the cheese industry, membranes increase the yield and quality of cheese and control the whey volume, by concentrating the cheese milk. With the advancement of newer technology in membrane processes, it is possible to recover growth factor from whey. With the introduction of superior quality membranes as well as newer technology, the major limitation of membranes, fouling or blockage has been overcome to a greater extent.

  • In Korea, there are a couple of risk assessment organizations: The Animal Plant and Fisheries Quarantine and Inspection Agency(QIA) and the Korea Food and Drug Administration (KFDA). The major food laws include the Food Sanitation Act (FSA), the Livestock Product Processing Act (LPPA), and the Agricultural Products Quality Control Act (APQCA). Milk and dairy products are mostly controlled by the Food Sanitation Act and the Livestock Product Processing Act. This study was carried out to estimate the current standard methods of foodborne pathogens for dairy products, comparing the Livestock Products Processing Act with the Food Sanitation Act. The standard methods of foodborne pathogens for dairy products are composed by growth culture, isolation culture, and identification, however, standard methods of QIA and KFDA are different at the using of medium and inspection stage. Therefore, consolidation of risk management and risk assessment methods are regarded important to provide safe dairy products to consumer.
