Optimization of fish oil extraction from Lophius litulon liver and fatty acid composition analysis

  • The Lophius litulon liver was used as raw material for the extraction of fish oil via various extraction methods. The extraction rate by water extraction, potassium hydroxide (KOH) hydrolysis and protease hydrolysis were compared and the results revealed the protease hydrolysis extraction had a higher extraction rate with good protein-lipid separation as observed by optical microscope. Furthermore, subsequent experiments determined neutrase to be the best hydrolytic enzyme in terms of extraction rate and cost. The extraction conditions of neutrase hydrolysis were optimized by single-factor experiment and response surface analysis, and the optimal extraction rate was 58.40 ± 0.25% with the following conditions: enzyme concentration 2,000 IU/g, extraction time 1.0 h, liquid-solid ratio 1.95:1, extraction temperature 40.5℃ and pH 6.5. The fatty acids composition in fish oil from optimized extraction condition was composed of 19.75% saturated fatty acids and 80.25% unsaturated fatty acids. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were 8.06% and 1.19%, respectively, with the ratio (6.77:1) surpassed to the recommendation in current researches (5:1). The results in this study suggest protease treatment is an efficient method for high-quality fish oil extraction from Lophius litulon liver with a satisfactory ratio of DHA and EPA.

Impact of Energy and Access Methods on Extrahepatic Tumor Spreading and the Ablation Zone: An Ex vivo Experiment Using a Subcapsular Tumor Model

  • Objective: To evaluate the impact of energy and access methods on extrahepatic tumor spreading and the ablation zone in an ex vivo subcapsular tumor mimic model with a risk of extrahepatic tumor spreading. Materials and Methods: Forty-two tumor-mimics were created in bovine liver blocks by injecting a mixture of iodine contrast material just below the liver capsule. Radiofrequency (RF) ablations were performed using an electrode placed parallel or perpendicular to hepatic surface through the tumor mimic with low- and high-power protocols (groups 1 and 2, respectively). Computed tomography (CT) scans were performed before and after ablation. The presence of contrast leak on the hepatic surface on CT, size of ablation zone, and timing of the first roll-off and popping sound were compared between the groups. Results: With parallel access, one contrast leak in group 1 (1/10, 10%) and nine in group 2 (9/10, 90%) (p < 0.001) were identified on post-ablation CT. With perpendicular access, six contrast leaks were identified in each group (6/11, 54.5%). The first roll-off and popping sound were significantly delayed in group 1 irrespective of the access method (p = 0.002). No statistical difference in the size of the ablation zone of the liver specimen was observed between the two groups (p = 0.247). Conclusion: Low-power RF ablation with parallel access is proposed to be effective and safe from extrahepatic tumor spreading in RF ablation of a solid hepatic tumor in the subcapsular location. Perpendicular placement of an electrode to the capsule is associated with a risk of extrahepatic tumor spreading regardless of the power applied.

성견에서 관찰된 심막-복막 허니아 증례 (Peritoneopericardial diaphragmatic hernia in adult dog)

  • A two-year-old pointer dog showed a liver failure for long duration. By radiographic examination, barium or gas filled intestine was detected in the pericardium. Although liver function was bad, surgery was tried to repair hernia due to the poor prognosis. Intestine, omentum, and liver with gall bladder were dislocated in the pericadial sac by peritoneopericardial diaphragmatic hernia (PPDH). Intestine was easily removed from pericardial sac but omentum and liver were not because of strong adhesion to the pericardial sac. Postmortem examination was performed because the patient was died after surgery. Grossly, herniated liver whose right medial lobe was strongly adhered to the pericardial sac was severely congested with fibrin adhesion on the surface. Hypoplastic abnormally up-located heart had a fissure in the outside of lower right venticulum bordered apex and depressed large vessels including vena cava and aortic arch into the base of heart. It was suggested that formation of a fissure and depression of vessels might be due to the pressure of herniated organs. In the veterinary literature as far as we knew, PPDH associated with liver failure due to adhesion of liver to the pericardial sac has not been reported.

한우문맥(韓牛門脈)의 분지(分枝)에 관한 해부학적(解剖學的) 연구(硏究) (Anatomical studies on pattern of branches of portal veins in Korean native cattle)

  • The distribution of portal veins within the liver in 30 Korean native cattle were observed. Vinylite solution was injected into portal veins of eighteen specimens for cast preparation. The angiography was prepared in twelve specimens by injecting 30% barium sulfate solution into portal veins, and then radiographed on a X-ray apparatus(Shimadzu 800MA 120Kvp). The results were summarized as follow: 1. The Vena portae was divided immediately upon entering the liver into a very short Truncus dexter venae portae($14.75{\pm}4.86$ : 6.9~23.1mm) and a long Truncus sinister venae portae($94.16{\pm}9.62$ : 110~150mm). 2. The Truncus sinister venae portae runs of first in the long axis of the liver from the Porta hepatis toward the left lobe. At the boundary between the quadrate and left lobes it bends sharply 50 to 80 degrees toward the Incisura ligamentum teretis, and after a course of 36. 5 to 54.mm between the quadrate and left lobes, ends abruptly. The Truncus sinister venae portae is divided for description into the Pars transversa, from the Porta hepatis to the flexure, and the Pars umbilicalis, from the flexure to the end. 3. The branches of Venae portae were Ramus ventralis lobi sinistri, Ramus intermedius lobi sinistri, Ramus dorsalis lobi sinistri, Ramus lobi quadratii, Ramus ventralis lobi dextri, Ramus intermedius lobi dextri, Ramus dorsalis lobi dextri, Rami processus caudatorum and Rami processus papillarum. 4. The Ramus intermedius lobi sinistri was arised from the left surface of the Pars umbilicalis, and was origined on the common trunk with Ramus dorsalis lobi sinistri(3 cases, 10%) or Ramus ventralis lobi sinistri(3 cases, 10%). 5. The Rami lobi quadratii consisted of the vein(15 cases, 50%) or two veins(15 cases, 50%), and was observed on the arched-shaped at 2 cases (6.6%) of the liver. 6. The Rami processus caudatorum consisted of one vein(28 cases, 93.3%) or two veins(2 cases, 6.6%). The former were formed common trunk with R, dorsalis lobi dextri(7 cases, 23.3%) or R. ventralis lobi dextri (2 cases, 6.6%). 7. The Rami processus papillarum were arised from the dorsal border of Pars transversa, and also gave off many small branches supplied papillary process of the caudate lobe. 8. The anastomosis on the branches of Vena portae was observed in the intralobar and interlobar areas. 9. The Truncus dexter venae portae and Truncus sinister venae were ramified many secondary branches that were radiated within the liver. 10. On the diaphragmatic surface, small vessels of the portal veins were observed, while there were big ones on the visceral surface. 11. The ramified angles at Ramus dorsalis lobi dextri, Rami processus papillarum, Ramus dorsalis lobi sinistri, Ramus intermedius lobi sinistri, Ramus ventralis lobi sinistri, Rami lobi quadratii, Rami processus caudatorum, Ramus ventralis lobi dextri and Ramus intermedius were 10~50, 70~110, 100~150, 140~170, 185~220, 270~330, 240~300, 270~320 and 340~10 degrees, respectively.

  • In attempt to develop a drug delivery system using serum albumin microspheres, bovine serum albumin microspheres containing antitumar agent. Cytarabine, were prepared. The shape, surface characteristics, size distribution, behavior of in vivo distribution, drug release behavior, and degradation of albumin microsphers in animal liver issue homogenate and proteolytic enzyme were investigated. The shape of albumin microspheres was spherical and the surface was smooth and compact. The size distribution of the albumin microspheres was effected by dispertion forces during emulsification and albumin concentration. Distribution of albumin microspheres after imtravenous administration in rabbit was achieved immediately. In vitro, albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin microsphere matrix was so hard that it retained most of cytarabine except initial burst during the first 10 minutes, and the level of drug release during the initial burst was affected by heating temperature, drug/albumin concentration ratio and size distribution. After drug release test, the morphology of albumin microspheres was not changed. Albumin microsphere matrix was degraded by the animal liver issue homogenate and proteolytic enzyme. The degree of degradation was affected by heating temperature.

자카스펭귄에서 발생한 Aspergillus fumigatus 감염증 (Aspergillus fumigatus infection in Jackass penguin)

  • A three years old male Jackass penguin(Spheniscus demersus) showed anorexia, depression, respiratory problem for a few days, followed by clinical treatment for 20 days resulted in death. Grossly, multiple white nodules measuring 2~3 mm in diameter were observed in the surface of and inside lung, liver, kidney, thoracic wall. Microscopically, granuloma formations were observed in the lung and liver. The margin of granuloma surrounded by connective tissue barrier and infiltrated lymphocytes, and also observed giant cell near the granuloma. By Periodic acid Schiff reaction, hyphae were detected in granuloma of lung and liver. This case was a chronic and systemic aspergillosis caused by Aspergillus fumigatus infection in a Jackass penguin at a zoo.

Systemic Candidiasis of a Wild Slaty-Backed Gull (Larus schistisagus) in Jeju, Korea

  • A young slaty-backed gull was found dead nearby Seongsan harbor, Seogwipo-si, Jeju. Grossly, focal ulceration and a protuberant nodule were observed in intermediated zone between proventriculus and gizzard. Numerous discrete or confluent white-yellowish nodules were scattered on the surface of liver. Caudal portion of left kidney was replaced by a white-yellowish nodule. Three raised masses were observed in the spleen. Histologically, there were severe ulceration with hemorrhage in the gizzard and multifocal granulomatous inflammations in liver, kidney, spleen, and lungs. Pink to dark-red, round to oval fungal spores and pseudohyphae in granulomatous lesions were confirmed by periodic acid-Schiff stain. Candida albicans were isolated from the lesions of liver, kidney, spleen and gizzard using fungal culture and confirmed by VITEK 2 system. Based on the gross, histopathological findings, and fungal examination, this case was diagnosed as systemic candidiasis of a wild slaty-backed gull. In our best knowledge, this is the first report for systemic candidiasis characterized by multifocal granulomatous lesions in internal organs of a wild gull in Korea.

  • The livers of 300 cases of Korean native cattle which were collected at slaughted house in Pusan were examined by gross and histopathological means to study patterns of pathological changes of the organ. 1. The most dominated lesion of the liver was those of fascioliasis showing incidence rate as 907 per cent of inspected cattle. 2. On age distribution of the fascioliasis lesion in the liver, higher incidence was seen in animals that were from 6 to 9 years of age. 3. Significant changes of the hepaticparenchyma caused by immature flukes included thrombosis, migrational tracks such as hemorrhagic foci with massive eosinophilia and hepatic necrosis. The changes of chronically infected livers of cattle were recognized as proliferation of periductal eosinophilic granuloma of various degrees in each portal triad. And this easential cholangiohepatitis might develop into multiple focal cirrhosis or diffuse perilobular cirrhosis. 4. Most cases with gross lesion of swelled bile ducts on visceral surface of the liver showed histological evidences of fascioliasis lesion throughout hepatic lobes.

Catechin reduces liver inflammation by regulating Kupffer cell activation in rats

  • The liver is a defense against infections due to its strategic location between the gastrointestinal and systemic circulations. In dogs and cats, infectious hepatitis encompasses a range of contagious diseases affecting the liver either directly or as part of a broader systemic infection, including bacterial, mycobacterial, viral, fungal, protozoal, parasitic, and rickettsial diseases. Catechins possess well-recognized natural antioxidant properties. This study investigated their therapeutic potential for applications in hepatology, evaluating whether catechins reduce hepatic inflammation in rats repeatedly exposed to carbon tetrachloride (CCl4). Sprague-Dawley rats were given catechin 50 (C50) or 100 (C100) mg/kg body weight orally daily for 3 days. This treatment was given with or without concurrent intraperitoneal injections of CCl4. Phosphate-buffered saline served as the vehicle control, while silymarin administered at 100 mg/kg was used as the positive control. Gross examination revealed significant enlargement, edema, and darker tissue in CCl4-induced livers treated with vehicle. Additionally, spotty discoloration was observed on the surface. Kupffer cell activation suppressed the expression of inflammatory mediators, including inducible nitric oxide synthase (iNOS), in groups co-treated with catechin and CCl4; this effect was reversed when catalase replaced catechin in CCl4-injured rats. Catechin alleviates hepatic inflammation in rats repeatedly exposed to CCl4; it also modulates the activation of Kupffer cells and monocytes. These results should lead to new treatments for liver inflammation in veterinary practice.
