This study investigated the biochemical and histological changes associated with the co-administration of cisplatin and methanolic extract of Portulaca oleracea (MEPO) in adult Wistar rats. Twenty-four (24) adult female Wistar rats were randomly divided into six (6) groups (A-F) (n = 4). Group A served as the control group for the experiment and received no treatment. Group B was given a single dose of cisplatin and served as the cisplatin control group. Group C and D received 400 mg/kg and 800 mg/kg of MEPO 6 hours after a single dose cisplatin injection respectively. Group E and F received 400 mg/kg and 800 mg/kg of MEPO 6 hours before cisplatin injection. The cisplatin injection was 2 mL/kg given intraperitoneally for all groups. There was a significant increase in the serum levels of ALT, ALP, AST, total bilirubin, and conjugated bilirubin following cisplatin treatment (p = 0.000, 0.000, 0.039, 0.000, 0.004 respectively) with a consequent reversal due to MEPO administration across all treated groups (p = 0.000, 0.000, 0.000, and 0.000) in a dose-dependent fashion. Cisplatin caused the expansion of the red and white pulp in the spleen which was attenuated by MEPO. MEPO demonstrated a protective effect against cisplatin-induced liver and spleen toxicity.
The focus of this study was to investigate the influences of enzymatic scavengers of active oxygen metabolites and phospholipase $A_2$ inhibitor on hepatic secretory and microsomal function during hepatic ischemia/reperfusion. Rats were pretreated with free radical scavengers such as superoxide dismutase (SOD), catalase, deferoxamine and phospholipase $A_2$ inhibitor such as quinacrine and then subjected to 60 min. no-flow hepatic ischemia in vivo. After 1, 5 hr of reperfusion, bile was collected, blood was obtained from the abdominal aorta, and liver microsomes were isolated. Serum aminotransferase (ALT) level was increased at 1 hr and peaked at 5 hr. The increase in ALT was significantly attenuated by SOD plus catalase, deferoxamine and quinacrine especially at 5 hr of reperfusion. The wet weight-to-dry weight ratio of the liver was significantly increased by ischemia/reperfusion. SOD and catalase treatment minimized the increase in this ratio. Hepatic lipid peroxidiltion was elevated by ischemia/reperfusion, and this elevation was inhibited by free radical scavengers and quina crine. Bile flow and cholate output, but not bilirubin output, were markedly decreased by ischemia/reperfusion and quinacrine restored the secretion. Cytochrome $P_{450}$ content was decreased by ischemia/reperfusion and restored by free radical scavengers and quinacrine to the level of that of the sham operated group. Aminopyrine N-demethylase activity was decreased and aniline p-hydroxylase was increased by ischemia/reperfusion. The changes in the activities of the two enzymes were prevented by free radical scavengers and quinacrine. Our findings suggest that ischemia/reperfusion diminishes hepatic secretory functions as well as microsomal drug metabolizing systems by increasing lipid peroxidation, and in addition to free radicals, other factors such as phospholipase $A_2$ are involved in pathogenes of hepatic dysfunction after ischemia/reperfusion.
Objectives This study was written to report Korean medicine treatment for obese children. Methods This study included obese children who visited the Department of Korean Pediatrics at the Daejeon Korean Medicine Hospital of Daejeon University from September 18, 2020 to March 30, 2023. Height and weight at the first visit and on the day of the last herbal medicine termination were measured, and changes before and after treatment were observed. Liver function tests were performed before and after the treatment. Results Of the nine obese children who received Korean medicine treatment, five were male (55.6%) and four, female (44.4%). In term of age, there were five 8-year-olds (55.6%), two 6-year-olds, and two 10-year-olds (22.2%); the patients' mean age was 8.00 ± 1.414 years. The treatment period was more than three months, and the average treatment period was 177.11 ± 106.013 days. After treatment with Korean medicine, the patients' heights increased, and the body mass index (BMI) percentile and obesity decreased. Liver levels were maintained within the normal range without abnormal liver function after herbal medicine treatment, and no adverse reactions were observed during Korean medicine treatment. As a result of the pattern identification analysis, gastrointestinal damp-heat type and yin deficiency with internal heat were reported at a high frequency. Conclusions Korean medical treatment can reduce the degree of obesity without hindering the growth of childhood obesity.
Objectives: Alcohol abuse is a public issue and one of the major causes of liver disease worldwide. This study was aimed at investigating the protective effect of Ganoderma lucidum pharmacopuncture (GLP) against hepatotoxicity induced by acute ethanol (EtOH) intoxication in rats. Methods: Sprague-Dawley (SD) rats were divided into 4 groups of 8 animals each: normal, control, normal saline pharmacopuncture (NP) and GLP groups. The control, NP and GLP groups received ethanol orally. The NP and the GLP groups were treated daily with injections of normal saline and Ganoderma lucidum extract, respectively. The control group received no treatment. The rats in all groups, except the normal group, were intoxicated for 6 hours by oral administration of EtOH (6 g/kg BW). The same volume of distilled water was administered to the rats in the normal group. Two local acupoints were used: Qimen (LR14) and Taechung (LR3). A histopathological analysis was performed, and the liver function and the activities of antioxidant enzymes were assessed. Results: GLP treatment reduced the histological changes due to acute liver injury induced by EtOH and significantly reduced the increase in the alanine aminotransferase (ALT) enzyme; however, it had an insignificant effect in reducing the increase in aspartate aminotransferase (AST) enzyme. It also significantly ameliorated the superoxide dismutase (SOD) and the catalase (CAT) activities. Conclusion: The present study suggests that GLP treatment is effective in protecting against ethanol-induced acute hepatic injury in SD rats by modulating the activities of ethanol-metabolizing enzymes and by attenuating oxidative stress.
Background: Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease associated with multiple metabolic disorders. The medicinal plant Curcuma longa L. is widely distributed in Asia and has been used to treat a spectrum diseases in clinical practice. To date, there are inadequate reports of the effects of C. longa 50% EtOH extract (CE) on NAFLD. Therefore, in this study, we evaluate the CE on an NAFLD animal and elucidate the mechanism of action. Methods and Results: C57BL/6J mice fed a methionine-choline deficient diet (MCD) were treated with CE or milk thistle, and changes in inflammation and stetosis were assessed. Experimental animals were divided into six group (n = 10); Normal, MCD, MCD + CE 50 mg/kg/day (CE 50), MCD + CE 100 mg/kg/day (CE 100), MCD + CE 150 mg/kg/day (CE 150), and the Control, MCD + Milk thistle 150 mg/kg/day (MT 150). Body weight, liver weight, liver function, and histological changes were assessed in experimental animals. Quantitative real-time polymerase chain reaction and western blot analyses were performed on samples collected after 4 weeks of treatment. We observed that CE administration improved MCD-diet-induced lipid accumulation, and triglyceride (TG) and total cholesterol (TC) levels in serum. Treatment with CE also decreased hepatic lipogenesis through modulation of the sterol regulatory element binding protein-1 (SREBP-1), CCAAT-enhancer binding protein α (C/EBPα), fatty acid synthase (FAS), and peroxisome proliferator-activated receptor γ (PPARγ) expresion. In addition, the use of CE increased adenosine monophosphate-activated protein kinase (AMPK) phosphorylation and inhibited the up-regulation of toll-like receptor (TLR)-2 and TLR-4 signaling and the production of inflammatory mediators. Conclusions: In this report, we observed that CE regulated lipid accumulation in an MCD dietinduced NAFLD model by decreasing lipogenesis. These data suggeste that CE could effectively protect mice against MCD-induced NAFLD, by inhibiting the TLR-2 and TLR-4 signaling cascades.
This study was done to investigate the effect of Ganyangsanghang-bang(GYSHB) on hypertension. After administering GYSHB extract to SHR for 5 weeks, changes in blood pressure, pulse rate, aldosterone and catecholamine levels in plasma were examined, and immunohistochemical changes were observed, and liver function test was done. The following results were obtained; blood presure decreased significantly as well as levels of aldosterone and norepinephrine in SHR. But levels of dopamine were unaffected. No capillary vessel dilation in adrenal cortex was observed. Safety against hepatic toxicity was showed. These results support a role for GYSHB might be usefully applied in treatment of hypertension.

No capillary vessel dilation in adrenal cortex was observed. Safety against hepatic toxicity was showed. These results support a role for GMSMT might be usefully applied in treatment of hypertension.
To identify the changes in professional services pattern after introducing the deligated system of claims review started in 1982, a university hospital under this system was examined. For comparison, claims of the hospital to Federation of Korean Medical Insurance Societies, where this system is not accepted, were reviewed. A total of 600 cases each were studied operated at the Departments of General Surgery & Orthopedic Surgery in 1981 and 1983. The results are summarized as follow: 1. Percentages of hospital charges for basic care was decreased by 10.2% and that for medical service increased by 8.4% in 1983. 2. After the introduction of the deligated review system, percentages of cutting off the claims was decreased by 12.4% for basic care and increased by 3.8% for medical services. 3. Percentage of testing liver function, and the frequency of administering high cost intravenous fluid injection, applicating Robinul as anesthetic premedication were decreased respectively after introducting the deligated services system.
Background and Objectives: Allergic rhinitis is an allergic reaction characterized by sneezingm itchy nose, mouth and throat, congestion and/or nasal discharge. The offending allergenes are usually pollens, molds, dust mites and animal allergen. Recently, the incidence of infectious nasal diseases tend to decrease. However, allergic rhinitis has increased and treatment in most cases has only deat with the symptom. Tongkyu-tang was composed of sixteen crude drugs. The Oriental Medical References mention therapeutic effects of Tongkyu-tang on nasal obstruction, watery nasal discharg. And Tongkyu-tang has clinically been used for the treatment of common cold, headache, sternutation, rhinitis etc. Speacially Tongkyu-tang is one of the most frequently used medical treatment for the allergic rhinitis. Experimental studies were conducted to investigate for the effect of Tongkyu-tang on the changes of neutrophil segment, lymphocyte, total IgE and nassal tissue in allergic rhinitis of ovalbumin-inhalation rat. Meterial and Methods: Fifteen Sprague-Dawley rats were divided into three group: normal group, control group, experimental group. To induce the allergic rhinitis in control group and experimental group, rats were sentitized intraperitoneally with 0.1 % ovalbumin solution 3 times at intervals of I week. Then intranasal sensitization was performed by diffusing 0.1 % ovalbumin solution 3 times at intervals of 2 days. After that time, rat in the experimental group were oral administration treated by Tongkyu-tang for 28 days. We observed changes in nasal tissue; changes in the number of white blood cell, red blood cell and total Ig E; also changes in the segment of neutrophil and lymphocyte in blood. And we observed the changes of AST, ALT of three group. We used anova test statistically. Result: The number of leucocyte remained unchanged between three group. The number of erythrocyte was increased in experimental group and control group when compared with the normal group. The segment of neutrophil, in blood was decreased in experimental group when compared with the control group but, that was not significant statistically(p<0.05). The promotion of lymphocyte in blood was significantly decreased in experimental group when compared with the Control group(p<0.05). Total IgE was decreased in experimental group when compared with the control group but, that was not significant statistically(p<0.05). The cilium be well preserved in experimental group: the nasal tissue in experimental group was similar to in the normal group. Congestion and expantion of grandular cell in nasal submucosa, hypertropy of epithelium in nasal mucosa, acid mucus in epithelium and neutral mucus in subepithelium were decreased in experimental group when compared with the control group. Effect of Tongkyu-tang on the liver function were also studies in rats. Treatment of Tongkyu-lang did not affected on AST and ALT. Conclusion: Considering the above experimental results, it is suggested that oral administration treatment using Tongkyu-lang, without worry about liver function injury, decreased response on an Animal model with Allergic Rhinitis.
