Effects of grape pomace on the antioxidant defense system in diet-induced hypercholesterolemic rabbits

  • The effects of grape seeds extract and grape peels extract prepared from grape pomace on the activity of antioxidant enzymes, degree of lipid peroxidation in serum and liver tissue were investigated in rabbits fed on high cholesterol diet. New Zealand white rabbits were divided as follows ; 1) NOR (normal group); 2) CHOL (cholesterol group); 3) GSH (cholesterol + grape seed extract group); 4) GPE (cholesterol + grape peel extract); 5) GSP (cholesterol + grape seed powder); 6) GPP (cholesterol + grape peel powder); 7) GE (cholesterol + grape seed and peel extract); 8) GP (cholesterol + grape seed and peel powder). Eight groups of rabbits were studied for 8 weeks. At the end of the experimental period, rabbits were sacrificed and the liver tissue were removed. Then, GSH, GPx, GST, CAT and MDA in the liver were measured. In liver tissues, total glutathione contents (GSH), glutathione peroxidase (GPx) and catalase (CAT) activity, which was significantly higher by grape seed extract supplementation. The level of malondialdehyde (MDA) was lower in the serum of rabbits fed grape seed extract or grape peel powder plus cholesterol than in the serum of rabbits fed cholesterol alone. It is therefore likely that grape seed extract prepared from grape pomace functioned as antioxidants in vivo, negating the effects of the oxidative stress induced by 1% cholesterol diet. The grape seed extract was found effective in converting the oxidized glutathione into reduced glutathione, and in removing $H_2O_2$ that is created by oxidative stress. The grape peel powder was found to have small influence on reduced glutathione content, CAT and GPX activity, but it increased GST activity in liver tissues, resulting in promoting the combination of lipid peroxide and glutathione (GSH), and further, lowering the formation of lipid peroxide in the serum. Therefore, grape pomace (grape seed extract and grape peel powder) supplementation is considered to activate the antioxidant enzyme system and prevent damage with hypercholesterolemia.

Protective Effect and Enzyme Activity of Field Horsetail, Mugwort and Champignon on the Cadmium Poisoning of Rat (일부 식용식물이 랫트의 카드뮴 중독에 미치는 防禦效果와 酵素反應)

  • The concentrations of cadmium, metallothionein(MT), superoxide dismutase(SOD), and lactate dehydrogenase(LDH) were investigated in liver and kidney of rats which were fed the water containing 50 or 100ppm cadmium chloride with basal diet(group A), 5% horsetail diet(group, B), 5% mugwort diet(group C) and 5% champignon diet(group D) for weeks. Cadmium in liver decreased for the first 12 weeks of treatment, but thereafter increased, and was lower in experimental group B,C,D than in control group A. Cadmium in kidney increased linearly during the 16 weeks of treatment, and was lower in group B than in group A. MT in liver decreased for the first 12 weeks of treatment in group A, but increased linearly during the 16 weeks in group B,C,D, higher in group B than in group A. There were significantly higher accumulation of cadmium and MT in liver than in kidney in the beginning of cadmium treatment, but reversed in the ending of treatment. The SOD and LDH activities were not affected during the 16 weeks treatment, and there was no significant difference between groups. Histologic examination revealed moderate to severe hepatic and renal injury in group A compared to horsetail diet group B. These results indicate that the kidney is a major target organ of chronic cadmium poisoning, and suggest that Cd-induced hepatic injury, via release of Cd-MT, may play an important role in the nephrotoxicity. In addition, higher MT concentrations in liver and kidney in the group B constitute a plausible explanation of the protective effects of horsetail diet against the cadmium toxicity in relation to histologic findings.

Lycopene supplementation suppresses oxidative stress induced by a high fat diet in gerbils

  • The effect of lycopene supplementation on the antioxidant system was investigated by analyzing lipid peroxide levels, glutathione contents, and antioxidant enzyme activities in Mongolian gerbils fed a high fat diet. Gerbils were fed on each experimental diet for 6 weeks; normal diet (NC), normal diet with 0.05% lycopene (NL), high fat diet (HF), and a high fat diet with 0.05% lycopene (HFL). Dietary supplementation of lycopene increased hepatic lycopene level in gerbils fed a normal or high fat diet (P < 0.05). Liver and erythrocyte concentrations of lipid peroxide increased in gerbils fed a high fat diet, whereas lycopene supplementation decreased liver and erythrocyte concentrations of lipid peroxide (P < 0.05). Hepatic total glutathione content was higher in the NL group than that in the NC group (P < 0.05). Total antioxidant status in plasma increased following lycopene supplementation compared with that of the non-lycopene supplemented groups (P < 0.05). Hepatic catalase activity increased following dietary lycopene supplementation (P < 0.05). Superoxide dismutase activity in liver remained unchanged with lycopene supplementation, but erythrocyte superoxide dismutase activity increased in NL group compared with NC group (P < 0.05). Glutathione-S-transferase activity increased in the NL group compared to NC group (P < 0.05). Liver and erythrocyte glutathione peroxidase activity increased significantly in the NL group compared to that in the HF group (P < 0.05). Liver glutathione reductase activity was higher in the NL group than that in the NC group (P < 0.05). These results suggest that lycopene supplementation may be efficient for preventing chronic diseases induced by oxidative stress related to high fat diet.

The Effect of the Hot Water Extract from Taraxacum coreanum Nakai on Hepatocarcinogenesis Induced by N-nitrosodiethylamine in Rats (흰민들레 열수추출물이 N-nitrosodiethylamine로 유발된 흰쥐의 간암 발생에 미치는 효과)

  • The present study was carried out to investigate the hepatoprotective effect of hot water extract of Korean white dandelion (Taraxacum coreanum Nakai) on liver cancer induced in rats by a single dose of N-nitrosodiethylamine (NDEA). The animals were divided into three groups. The animals in the normal (Nor) group were fed basal diet. Control (Con) group were administered with NDEA (200 mg/kg, i.p.). Hot water extract of Korean white dandelion (KDDA, KDDB) group treated with hot water extracts of Korean white dandelion (30, 60 mg/kg/day) for 8 weeks after NDEA. Serum AST and ALT were measured, enzyme activity of liver were examined by spectrophotometer and histological alternation of liver were examined by light microscopy. KDDA and KDDB group were decreased the increase of serum AST, ALT level induced by NDEA. The activities of SOD and CAT were significantly increased in KDDA, KDDB then in the Con. In the histological observation, KDDA and KDDB group were structure of liver lobula and nuclear shape was relatively cleared. In addition, the cytoplasm staining was similar to those of the Nor group. vacuolization of cytoplasm was not observed. These results suggest that administration of hot water extract of Korean white dandelion suppress or retard liver cancer induced by NDEA in rats.

The Effect of Stevia (Stevia. Rebaudian Bertoni) Extract Supplementation on Lipid Metabolism and Liver Function of Rats Administered with Ethanol (스테비아(Stevia. Rebaudian Bertoni)추출물 보강이 만성 알코올 섭취 흰쥐의 지질대사 및 간기능에 미치는 영향)

  • To investigate the effects of the supplementation of Stevia. Rebaudian Bertoni on serum and hepatic lipid levels and enzyme activities in rat administered with ethanol chronically. Sprague-Dawley male rats were orally treated with either an AIN-76 diet(C, control), a control diet plus ethanol(E, 4g/kg bw), E plus stevia extract-1(ES1, 1ml/kg bw), or E plus stevia extract-2(ES2, 2ml/kg bw) for 7 weeks. Serum triglyceride levels were increased in the E group and were decreased in the ES 1 group. Liver triglyceride levels were decreased significantly in the ES2 groups and Total-cholesterol were decreased in the ES1, ES2 groups compared with the E group. Liver $\gamma-GTP$ levels were decreased significantly in the ES1, ES2 groups compared with the E group. In addition, we have evaluated the serum or liver carnitine levels in those groups. Liver TCNE levels were increased significantly in the ES1, ES2 compared with the E group. These results may suggest that supplementation of Stevia. Rebaudian Bertoni has effects on the recovery of chronic ethanol-related diseases.

Effects of Dietary Restriction on the Body Weight and Antioxidant Enzymes in Various Organs of Diabetic Rats (당뇨병 흰쥐에서 식이 제한 급여가 장기의 항산화효소 활성도 및 체중에 미치는 영향)

  • The effects of dietary restriction (DR) on antioxidant enzymes were studied in liver, lung and erythrocytes of diabetic rats. Experimental animals used Sprague-Dawley (SD; body weight 350$\pm$20g) male rats and Otsuka Long Evans Tokushima fatty (OLETE; body weight 5--$\pm$30g) male rats, as a model of type 2 diabetes mellitus. Type I diabetes was induced in SD rats by intramuscular injection of alloxan (80 mg/kg BW). Animals were randomly assigned either to continue the ad libitum diet or 40% DR (60% intake of ad libitum diet) groups. The body weight was measured at every 2 weeks to 4 months following DR. The activities of antioxidant enzymes (superoxide dismutase (SOD), catalase, glutathione peroxidase (GSHPx) were measured in liver, lung and erythrocytes and the concentration of TBARS as a marker of reactive oxygen species-induced tissue injry was also measured in rats after 4 months 40% DR. The body weight 4 months after 40% DR of control SD, alloxian-diabetid SD and OLETE rats were 80%, 98% and 75% of each control groups, respectively. The activities of SOD, catalase and GSHPx in lung and erythrocytes of rats were not change by 40% DR but in 4 month 40% DR rat liver, the activities of SOD and catalase were increased in control SD, alloxan-diabetic SD, and OLETF groups. The concentration of TBARS in lung and erythrocytes was also not changed by 40% DR, while liver TBARS concentration was decreased in OLETF and control SD rats compared to each non-DR control rats. These results suggested that the body weight changes in diabetic rats by DR was more prominent in type 2 diabetes and changes of antioxidant enzymes is most prominent in liver by DR either type 1 and 2 diabetic rats.

An Effect of Methanethiol Treatment on the Liver Function of Rats Previously Fed Low or High Protein Diet (단백식이 조건을 달리하여 성장한 흰쥐에 Methanethiol 투여가 간기능에 미치는 영향)

  • To evaluate an effect of dietary protein on the intoxication of methanethiol in rats, the methanethiol was intraperitoneally injected to the rats fed a low or high protein diet and then the liver weight per body weight and seurm levels of alanine aminotransferase (ALT) activities were determined to investigate the differences in liver damage between the animal groups fed low protein diet and that fed high protein diet. On the other hand, the hepatic glutathione content and its conjugating enzyme, glutathione S-transferase (GST) activity were determined to clarify the cause of difference in liver function between the two groups. The increasing rate of liver weigh/body wt., serum levels of ALT to its control group were higher in methanethiol-treated rats fed low protein diet than those fed high protein diet. The hepatic content of glutathione and GST activity were higher in rats fed high protein diet than those fed low protein diet and the decreasing rate of hepatic glu-tathione content to its control group was higher in rats fed low protein diet than those fed high protein diet. Furthermore, the hepatic GST activity in methanethiol-treated rats was higher in rats fed high protein diet than those fed low protein diet. In case of control group, the GST activity was also higher in rats fed high protein diet than those fed low protein diet.

Tosufloxacin Tosylate Increased the Liver Enzyme Levels in a Bladder Cancer Patient with Normal Liver Functions: a Case Report (정상 간기능을 가진 방광암환자에서 간효소 수치를 올리는 Tosufloxacin Tosylate: 증례보고)

  • Summary: We report the first hepatic adverse effect of tosufloxacin tosylate in a muscle invasive bladder cancer patient with normal liver functions and with scheduling to undergo a surgical operation for a neobladder. Tosufloxacin tosylate 150 mg was administered to a 57-year-old man who maintained transurethral resection of bladder tumor (TUR-BT) postoperative multiple medications. His labs presented significant increases in alanine amino transferase (ALT) and aspartate amino transferase (AST) levels with 2-week compliance of 150 mg tablet three times a day. After discontinuing tosufloxacin tosylate, the levels slowly decreased and completely returned to normal ranges without any intervention in a few weeks. The Naranjo Causality Algorithm indicates a probable relationship between increased ALT and tosufloxacin. The patient was to have the second surgical operation as scheduled after getting normal range of ATL level. Therefore, tosufloxacin should be avoided in patients at risk for having liver dysfunctions or diseases if the patients have a schedule for any operation. Background: Tosufloxacin tosylate has been shown to have favorable benefits as an antibiotic. Tosufloxacin tosylate may be considered to have the adverse effects such as nauseas, vomiting, diarrhea, abdominal pain, stomatitis, tendonitis, tendon rupture, headache, dizziness, drowsiness, insomnia, weakness, agitation including hemolysis in the event of glucose-6-phosphate dehydrogenase deficiency as other fluoroquinolones. More severe adverse reactions of tosufloxacin tosylate over the above common adverse effects of fluoroquinolones were thrombocytopenia and nephritis. It also is not well known that tosufloxacin can cause hepatic problem. Here the study reports the first hepatic reaction from tosufloxacin and might arouse heath care providers' attention to appropriate drug choice for patients.

Effect of Quartz Porphyry on the Functional and Morphological Changes of Liver and Kideny in Common Finch and white Java Sparrow (맥반석 식이가 십자매와 백문조 간장 및 신장의 형태학적 변화에 미치는 영향)

  • Effect of Quartz porphyty(QP) on functional and morphological changes of the liver and kidney was studied in male common finch and white java sparrow fed with the basal diet(Control group) or experimental diet containing 3.0% QP(QP group) for 14 days. There was not significantly different morphological change of the liver upon light microscopic examination in common finch and white java sparrow between control group and QP group. Morphological change of renal tissue upon light microscopic examination in common finch and white java sparrow was not also significantly different between control group and QP group. The concentrations of serum creatinine, blood urea nitrogen, and nric acid as renal functional parameters of common finch and white java sparrow were not significantly different in the both groups. The activity of glutamic oxaloacetic transaminase(GOT) as hepatic functional parameter in common finch was significantly higher in the QP group($\rho$<0.05), whereas the activity of glutamic pyruvic transaminase(GPT) as hepatic functional parameter in common finch was not significantly different in the both groups. The activities of GOP and GPT in white java sparrow were not significantly different in the both groups. The morphologic findings and functional parameters of the liver and kidney observed in common finch and white java sparrow fed with 3.0% QP diet showed evidence of slightly liver damage accompanied with increased release of enzyme and fatty change of the hepatocytes in common finch, suggested that the tissues in some animals can be damaged by feeding a diet supplemented with 3.0% QP.

Effects of Insamyangyoung-tang Aqueous Extracts on the Hypothyroidism Induced by Propylthiouracil in Rats (인삼양영탕(人蔘養榮湯)이 PTU로 유발된 Rat의 갑상선기능저하증에 미치는 영향)

  • Objectives : The object of this study was to evaluate the effect of Insamyangyoung-tang aqueous extracts (ISYYT) on the propylthiouracil (PTU) induced rat hypothyroidism. Methods : The rats were divided into 6 groups : intact control, PTU control, Levothyroxine (LT 4 ), 500, 250 and 125 mg/kg ISYYT treated groups. In ISYYT treated groups, PTU and ISYYT were administered for 4 weeks after 500, 250 and 125 mg/kg ISYYT were administered for 2 weeks. In LT 4 group, PTU and LT 4 were administered for 4 weeks. The changes were observed : the weigh, serum thyroid hormone levels, serum sex hormone levels, serum lipid profiles, serum liver enzyme levels, liver and testis antioxidant defense system, histopathology of thyroid gland, liver, epididymis, prostate and testis. Results were compared with PTU control group in this experiment. Results : In comparison with PTU control group, 500 and 250 mg/kg ISYYT treated groups showed significant increase of body, thyroid, liver, testis, epididymis and prostate weights, decrease of serum TSH levels with increase of serum T3 and T4 level, increase of serum testosterone and DHT levels with decrease of serum FSH levels, decrease of serum HDL with increase of TG and increase of serum AST levels. Histopathological inspections of hepatic and male reproductive organ damage induced by PTU were improved. And the changes of antioxidant defense system of Liver and testis induced by PTU were improve. There was no significant difference between 125 mg/kg ISYYT treated group and PTU control group in this experiment. Conclusions : The results obtained in this study considered that Insamyangyoung-tang may be effective in hypothyroidism and related organ damages.
