Effect of Biologically Active Fractions from Onion on Physiological Activity and Lipid Metabolism (양파로부터 분리한 생리활성분획이 생리활성 및 지질대사에 미치는 영향)

  • From the 80% ethanol extraction of onion flesh, fraction I,II and III were separated by Sephadex LH-20 column chromatography. Fraction II and III had a high inhibitory effect on ACE and xanthine oxidase. The ACE was about 80% at 50 ppm of fraction IIand III. The xanthine oxidase was about 82.5% at 100 ppm of fraction In. This research was studied to investigate the effects of the fraction In isolated from onion on the reduction of fat accumulation in rats fed high fat diet for 2 months. The metabolism was examined by analyzing the fat in serum and liver. It was shown that cholesterol in serum was remarkably reduced in onion fraction In as compared with those of control.

  • The effect of each azalea(Rhodoendron mucronulatum) pollen extract on phase I enzyme(aniline hydroxylase)was studied in this experiment. Serum aminotransferases were not changed in mice injected each azalea pollen extract, respectively compared to control group. The hepatic micros mal aniline hydroxylase activities in the presence of each azalea pollen extract were not affected in vitro. After treatment with azalea pollen water extract, hepatic microsomal aniline hydroxylase activity was increased with dose-dependent manner as compared to control group. The increment of hepatic microsmal aniline hydroxylase activity was more powerful by the treatment of water extract. As mice received aniline after pollen butanol and water extract-pretreatment once a day for 5 days, the blood and liver levels of aniline were decreased significantly.

Efficacy of Korean Red Ginseng by Single Nucleotide Polymorphism in Obese Women: Randomized, Double-blind, Placebo-controlled Trial

  • This study examined the effects of Korean red ginseng (KRG) on obese women and aimed to confirm that the effects of KRG on obesity differ dependently on a gene. Fifty obese women were recruited and randomized to receive KRG (n=24) or placebo (n=26) for 8 wk. Measurements of blood pressure, height, weight, waist circumference, waist-hip ratio (WHR), total fat mass, percentage of body fat, resting metabolic rate, basal body temperature, and daily food intake (FI), blood test (serum lipid, liver and renal function), Korean version of obesity-related quality of life scale (KOQOL), and a gene examination were performed. Comparisons of subjects before and after the administration of KRG revealed significant improvements of obesity in terms of weight, body mass index (BMI), WHR, FI, and KOQOL. However, in the comparison between KRG group and placebo group, only KOQOL was significantly different. KRG displayed significant efficacy on BMI and KOQOL in the CT genotype of the G protein beta 3 gene, but not in the CC genotype, on blood sugar test in the Trp64/Arg genotype of the beta 3 adrenergic receptor gene, but not in Trp64/Trp genotype, on KOQOL in the DD genotype of the angiotensin I converting enzyme gene, but not in the ID and DD genotypes. The effects of KRG on obesity were confirmed to some extent. However, a distinct effect compared to placebo was not confirmed. KRG is more effective for improving the secondary issues of the quality of life derived from obesity rather than having direct effects on the obesity-related anthropometric assessment and blood test indices.

A comparative study on immune-stimulatory and antioxidant activities of various types of ginseng extracts in murine and rodent models

  • Background: Ginseng (Panax ginseng) is a widely used traditional herbal supplement that possesses various health-enhancing efficacies. Various ginseng products are available in market, especially in the Korean peninsula, in the form of drinks, tablets, and capsules. The different ginseng types include the traditional red ginseng extract (RGE), white ginseng, and black red ginseng extract (BRGE). Their fermented and enzyme-treated products are also available. Different treatment regimens alter the bioavailability of certain compounds present in the respective ginseng extracts. Therefore, in this study, we aimed to compare the antioxidant and immune-stimulating activities of RGE, BRGE, and fermented red ginseng extract (FRGE). Methods: We used an acetaminophen-induced oxidative stress model for investigating the reduction of oxidative stress by RGE, BRGE, and FRGE in Sprague Dawley rats. A cyclophosphamide-induced immunosuppression model was used to evaluate the immune-stimulating activities of these ginseng extracts in BALB/c mice. Results: Our results showed that most prominently, RGE (in almost all experiments) exhibited excellent antioxidant effects via increasing superoxide dismutase, catalase, and glutathione peroxidase activities in the liver and decreasing serum 8-hydroxy-2'-deoxyguanosine, aspartate aminotransferase, and lactate dehydrogenase levels compared with the groups treated with FRGE and BRGE. Moreover, RGE significantly increased the number of white blood cells, especially T and B lymphocytes, and antibody-forming cells in the spleen and thymus, and it also activated a number of immune cell subtypes. Conclusion: Taken together, these results indicate that RGE is the best supplement for consumption in everyday life for overall health-enhancing properties.

Effect of Natural Functional Mixture on the Descent of Blood Glucose Level in Streptozotocin-Induced Diabetic(type I) Rats(II) (Streptozotocin 유발 제1형 당뇨 쥐의 혈당강하에 대한 천연 기능성 소재 혼합물의 효과(II))

  • The aim of this study was designed to evaluate the effects of natural functional mixture(FM) on plasma BUN and lipid levels, hepatic lipid levels, hepatic antioxidant enzyme activities and plama aminotransferase activity in streptozotocin (STZ) induced diabe1ic rats. Total cholesterol (TC) level in the diabe1ic rats supplemented with FM(70.69 mg/dL) was reduced comparing to groups without FM(87.12 mg/dL). This results caused the increase of the ratio of HDL-cholesterol to TC (42.60 to 51.49 %). However, superoxide dismutase (SOD), cytosol catalase (CAT), glutathione peroxidase (GSH-Px), GSH and lipoperoxide (LPO) activities were not significantly changed, which indicated the supplementation with FM could not reduce the oxidative stress in diabetic rats. In addition, asperate aminotransferase (AST) activity in FM-diabetic rat was lower than that in diabetic group. This results showed supplementation with FM in rats could improve the hepatic function damaged by STZ-induced diabetes.

  • Rice(Oryza stiva L.) is a major cereal food providing nourishment to over half of the world's populations and was considered only as a source of energy. However, The recent many researches have been made to suggest that rice may relate to prevention chronic disease and health-promoting properties such as postprandial glucose response, hypocholesterolemic effect and blood pressure-lowering effect. There has been numerous observation supporting that rice has hypocholesterolemic effect. Rice, rice bran, rice bran oil and unsaponifiable matter of rice bran oil reduced plasma cholesterol in rat, hamster as well as human. Components of rice showing hypocholesterolemic effect include dietary fiber(hemicellulose, phytic acid). protein, ${\gamma}$-oryzanol, $\beta$-sitosterol, and tocotrienols. Crapo et al has been studied that the effect of various of starchy foods on the postprandial blood glucose and insulin responses in healthy and diabetic humans. The results showed that rice had lower blood glucose and insulin responses compared to potato, bread and dextrose. The different physical forms in the same starch also produce the different postprandial glucose and insulin responses. In recent years, several studies have shown that some components of rice have potent antioxidant activity against Fe$^{2+}$ -ascorbate induced lipid peroxidation in rat liver microsomal membranes. Cell culture and animal studies have shown that some components of rice have inhibitory effect on the growth and proliferation of several types of human cancer cell. It was also reported that the methanol extract of brown rice has antimutagenic activity against various mutagens. In addition, the pepsine hydrolysate from rice protein is reported to inhibit angiotensin converting enzyme activity. GABA (${\gamma}$ - aminobutyric acid) and GABA enriched rice germ is also effective for lowering blood pressure and triglyceride levels.s.

Perceived Stress, Life Style, Health Status Indicatiors in Medical Center Employees (일 의료기관 직원들의 지각된 스트레스, 생활습관 및 건강상태 지표)

  • Purpose: The purpose was to examine the level of perceived stress, lifestyle, and health status indicators and identify the relationships among them in medical center employees. Method: This study utilized data from Cohen's Perceived Stress Scale, health survey developed by National Health Insurance Corporation, and laboratory tests (such as liver enzyme or lipid profile) on 873 medical center employees. The data were analyzed using the SPSS 12.0 program through descriptive statistics, $x^2-test$, independent t-test, ANOVA and ANCOVA. Results: Perceived stress was relatively high, but lifestyle and health status indicators were good. Age, gender, and job were significantly related to perceived stress, life style, and health status indicators: younger employees, men, nurses reported higher stress; older employees, men and medical technicians reported worse life style; older employees, men and doctors showed more abnormal health status indicators. There was a significant relationship between perceived stress and life style: the higher the level of stress was, the more drinking or the more smoking they had. However, there was no clear relationship between perceived stress and health status indicators. Conclusion: Although the subjects' lifestyle and health status indicators were relatively good, their perceived stress was seriously high. Therefore, programs for reducing stress are necessary for medical center employees.

Effect of Chicory Extract on Triglyceride Metabolism in Rats (흰쥐의 중성지질 대사에 미치는 치커리 추출물의 영향)

  • We investigated the effect of chicory (Chicorium intybus) extract on triglyceride concentration and microsomal triglyceride transfer protein (MTP) activity in rats. The effect of water-soluble extract of chicory fed at the $2.0\%\;and\;4.0\%(w/w)$ levels for 2 weeks on the concentration of serum triglyceride and the activity of hepatic microsomal riglyceride transfer protein (MTP) was investigated in male Sprague-Dawley rats. The triglyceride concentrations in serum of the chicory extract fed groups were significantly lower than in the control group. MTP activity, known to be essential for the assembly/secretion of apolipoprotein B-containing lipoproteins, was also significantly lower in the chicory extract groups than in the control group. The concentrations of other lipids in serum and liver and the activity of phosphatidate phosphohydrolase, the rate-limiting enzyme in triglyceride synthesis, showed no significant differences among in the chicory fed groups. These results indicate that dietary chicory extract decrease hepatic MTP activity and serum triglyceride concentration, and therefore reduces hepatic lipoprotein assembly and secretion.

사료중 어유가 급성기 반응중인 육계 병아리에서 에너지 대사와 항산화효소 활성에 미치는 영향

  • Three experiment were designed to determine the effects of dietary(n-3) fatty acids on the growth-suppressive effects of the inflammatory response and anti-oxidant enzyme activity and balances of nitrogen and energy. Day-old chicks were fed diets containing 1.0 % soybean oil in experiment 1, or diets containing 1.0 % of fish oil in experiment 2, and then on 7-d of age chicks were fed diets containing 1.0 % of either soybean oil or fish oil. In experiment 3, day-old chicks were fed on diets containing 1.0 % of either soybean oil or fish oil. In chicks of half pens within each dietary treatments of each experiments, the acute phase response were induced by injecting with Salmonella typhymurium lipopolysaccaride(LPS) i.p. 3 times at 8, 10 and 12-d of age. During acute phase response, chicks fed dietary fish oil from 1-d of age enhanced energy balances and did not give negative effect on nitrogen balance and uric acid excretion, but elevated MnSOD and Cu/ZnSOD activity In liver and erythrocyte cytosols compared with those fed dietary soybean oil. The results indicated that enhanced daily gain and metabolizable energy value of diet in birds fed fish oil from one-d of age seems due to change on the antioxidants systems during acute phase response.

Changes of Biochemical Composition and Activity of Enzyme in the Serum Exposed to Carbon Tetrachloride in Mice (사염화탄소를 투여한 MICE 혈청내 효소 활성 및 생화학적 조성의 변화)

  • Carbon tetrachloride ($CCl_4$) has been used to treat a variety of parasitic infection in both large and small animals, including Fasciola hepatica. Recently, we can easily contest with carbon tetrachloride in air, food, water, rain and Industry area etc. and it is using in order to induce liver injury in laboratory. In this study. we investigated activitis of LDH, ALP, AST and ALT and amount of cholesterol, triacylglycerol, glucose and BUN in mice serum exposed to $CCl_4.$ The mice divided Into a Naive control(A), corn oil control(B) and experimental group(C, D). Naive control group(A) was given feed and water only. Corn oil control group(B) was given corn oil 1ml /100g of body weight(B.W). Experimental group(C) was given carbon tetrachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:20(V /V) ), Experimental group(D) was given carbon teterachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:10(V /V) ). The results obtained were summarized as follows : 1) The body weight was declined after the l0th day In mice exposed to carbon tetra-chloride. 2) The total protein level in serum was significantly in mice exposed to carbon tetra-chloride($P{\leq}0.05$). The albumin and A /G ratio was decresed significantly in mice exposed to carbon tetrachloride($P{\leq}0.01$) 3) All of the activity of LDH, AST, ALT and ALP in mice serum exposed to carbon tetra chloride inclosed significantly activity of LDH ($P{\leq}0.05$), inclosed significantly activity of AST($P{\leq}0.05, \;p {\leq}0.01$), inclosed significantly activityof ALT 3nd ALP($P {\leq}0.05,\;p {\leq}0.01$). 4) The amount of cholesterol and triacylglycerol, lipid metabolite products in serum was inclosed in case of cholesterol but did not change in case of triacylglycerol.
