Male Sprageu-Dawely rats fed a cholesterol diet (Control group) or cholesterol diet supplemented with the water-soluble extract of stem bark from Cudrania tricuspidata(CTSB group) at the level of 0.5% for 2 weeks. Concentration of triglyceride in serum of CTSB group was significantly lower than that of control group. However, the other lipids concentrations of liver and serum, body weight gain, and food intake did not show significant difference between CTSB group and control group. Antioxidative activities of water-soluble extract from stem bark of Cudrania tricuspidata on the lipid peroxidation in rat were also studied in vivo by measuring the formation of thiobarbituric acid reactive substance(TBARS). Concentration of TBATS in the liver homogenates of CTSB group was significantly higher than that in control group. Nonheme iron concentration was significantly increased in the liver of CTSB group compared to control group. suggested that enhanced nonheme irom was associated with enhanced peroxidation of liver in CTSB group. These results suggested that water soluble extract from stem bark of Cudrania tricuspidata exert the hypotriglycerolemic effect and might amplify the lipid peroxidation of tissues in cholesterol fed rats.
Dietary protein restriction affects lipid metabolism in rats. This study was performed to determine the effect of a low protein diet on hepatic lipid metabolism and insulin sensitivity in growing male rats. Growing rats were fed either a control 20% protein diet or an 8% low protein diet. Feeding a low protein diet for four weeks from 8 weeks of age induced a fatty liver. Expression of acetyl-CoA carboxylase, a key lipogenic enzyme, was increased in rats fed a low protein diet. Feeding a low protein diet decreased very low density lipoprotein (VLDL) secretion without statistical significance. Feeding a low protein diet down-regulated protein expression of microsomal triglyceride transfer protein, an important enzyme of VLDL secretion. Feeding a low protein diet increased serum adiponectin levels. We performed glucose tolerance test (GTT) and insulin tolerance test (ITT). Both GTT and ITT were increased in protein-restricted growing rats. Our results demonstrate that dietary protein restriction increases insulin sensitivity and that this could be due to low-protein diet-mediated metabolic adaptation. In addition, increased adiponectin levels may influences insulin sensitivity. In conclusion, dietary protein restriction induces a fatty liver. Both increased lipogenesis and decreased VLDL secretion has contributed to this metabolic changes. In addition, insulin resistance was not associated with fatty liver induced by protein restriction.
This investigation was performed to study the antioxidant activities of Cornus officinalis Sieb extracts and the effect of Cornus officinalis Sieb extracts on glucose, lipid metabolism in diabetic rats. DPPH free radical scavanging activity and superoxide anion radical scavenging of Cornus officinalis Sieb extracts were 94.7% and 92.1%, respectively. Streptozotocin (45 mg/kg body weight, i.p.) induced diabetic rats showed a significant increases of plasma glucose, triglyceride and total cholesterol, concomitantly significant decrease of plasma high density lipoprotein. Glutathione level were decrease in cytosol of liver, lung and brain tissue of rats. Lipid peroxide were increase in microsome of liver cells. Group 1 and 2 were treated with Cornus officinalis Sieb extracts 200 mg/kg body weight and 100 mg/kg body weight for 24 days, individually. Group 1 and 2 rats showed decreased plasma glucose, triglyceride, total cholesterol and lipid peroxide in microsome of liver, and increased plasma high density lipoprotein and glutathione in cytosol of liver, lung and brain. The result suggest that Cornus officinalis Sieb extracts may normalize the Impaired antioxiants status in streptozotocin induced diabetic rats. Cornus officinalis Sieb extracts were used to improve the imbalance between free radicals and antioxidant system due to the diabetes.
The Vitamin E (VE) effect was examined on oxidative damage to DNA, lipids, and protein in mice that were fed various levels of lipid diets after total body irradiation (TBI) with X-rays at 2 Gy. No increase of 8-hydroxydeoxyguanosine (8OHdG) by TBI was observed in the +VE group; however, in the case of the -VE group, a significantly higher 8OHdG level was observed in the high-lipid group than in the low- or basal-lipid group. In the groups with TBI, the concentration of thiobarbituric reactive substances (TBARS) only significantly increased in the high-lipid (-VE) group. These changes in TBARS, due to TBI, were not detected in other groups. The contents of protein carbonyls only increased in the (-VE) group. The contents of protein carbonyls was significantly different between the (+VE) and the (-VE) groups, regardless of the lipid levels. The concentrations of GSH, vitamins C and E in the liver were lower, and the concentration of non-heme iron in the liver was higher in the high-lipid group than in the low- and basal-lipid groups. These concentrations in the high-lipid group were significantly different between the (+VE) and the (-VE) groups. These results strongly suggest that mice that are fed a high-lipid diet are susceptible to TBI-induced oxidative damage. Also, decreases in the GSH levels and an increase in the iron level are involved in the mechanism of this susceptibility.
This study was performed to investigate the effect of dietary chitin & chitosan on cadmium (Cd) toxicity and lipid metabolism in rats. Forty-two male rats of Sprague-Dawley strain weighing 137$\pm$2g were blocked into 6 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400ppm in diet and chitin and chitosan were given at the level of 0 or 4%(w/w) of diet. The results are summarized as follow. Chitosan decreased the toxicity of Cd on liver, kidney and femur and increase the Cd content of fecal excretion. Chitosan increased the lipdi & cholesterol content of fecal excretion by combining with lipid and bile acid. Chitosan decreased lipid, cholesterol and TG content in serum and liver by combining with lipid and bile acid. Chitin was less effective than chitosan in decreasing of Cd toxicity and lipid content of rat.
This research employed a senescence-accelerated mouse(SAM) to explore the possibility that differences exist among the major antioxidatns, lipid peroxidation in terms of ability to protect such animal treatment PQ, SAM-R/1 and SAM-P/8 were administered with PQ(200ppm/Kg) orally. The toxicity of PQ on SAM was determined as a bioassays of SOD, catalase and lipid peroxidation in the mouse liver. The data show that the SOD activity was induced by paraqwuat terement in both SAM-R/1 and SAM-P/8. The degree of lipid peroxidation was increased with PQ treatment. This means that SOD rather than catalase may protect against oxygen radical toxicity. Finally, over data lead to the toxicity of PQ and its function may efect to the antioxidants including SOD, catalase and lipid peroxidation in both SAM-R/1 and SAM-P/8 .
This study was conducted to compare the effect of dietary sucrose and starch on the lipid metabolism of Rat fed by low protein diet for 3 and 6 weeks periods. Forty male weanling Spargue-Dawley rats weighing 52.7 grams each, after being adopted for 2 days with standard diet, were blocked into 8 groups and fed experimental diet as designed. Experimental diet were composed of two different levels of proteins, 6% designed as low protein diet and 15% designed as standard protein diet. In each group, the content of serum lipid, serum cholesterol and liver lipid were measured. The results of this experiment were summerized as follows. 1. liver lipid content was tended to be high in the sucrose group of low protein fed animal. 2. the content of total serum lipid was tended to be high in the sucrose group, and this tendency was showed statistical significance in the animals fed by low and standard protein after 6 weeks of experimental period. 3. The difference in the total serum cholesterol content between the sucrose and starch group was not significant
The aim of this study was to examine whether administration of glutamine are able to prevent the NSAID induced bacterial translocation and lipid peroxidation in the rats. The an imals with glutamine were fed with L-glutamine for 5 days before diclofenac administration (100 mg/kg orally). 48 hour after diclofenac administration, intestinal permeability, serum biochemical profiles, and malondialdehyde levels of ileum were measured for evaluation of gut damage. Also, enteric aerobic bacterial counts, number of gram-negatives in mesenteric Iymph nodes, liver, spleen and kidney and malondialdehyde levels in liver, spleen, kidney and plasma were measured. Diclofenac caused the gut damage, enteric bacterial overgrowth, increased bacterial translocation and increased lipid peroxidation. Co-administration of glutamine reduced the gut damage, enteric bacterial overgrowth, bacterial translocation and lipid peroxidation induced by diclofenac. This study suggested that glutamine might effectively prevent non-steroidal anti-inflammatory drug induced bacterial translocation and lipid peroxidation in the rat.
We have studied the effect of 31 extracts and 10 components from some edible and medicinal plants on the formation of lipid peroxide in the liver homogenate of rat in vitro. The 70% acetone extracts of Allium tuberosum, Beta vulgaris var. cicla and Brassica juncea var. integrifolia, and methanol extract of Capsicum annuum decreased the formation of lipid peroxide by 33%, 58%, 62% and 56% at the concentration of 1mg/ml, respectively. And these four extracts inhibited the lipid peroxidation at the concentration of $10^{-1}$mg/ml by 17%, 46%, 49% and 45% respectively. Among the component tested, quercetin, quercitrin and isorhamnetin reduced the formation of lipid peroxide by 45%, 15% and 28% respectively at the concentration of $10^{-2}$mg/ml.
This study was designed to find out the effects on the lipid metabolism of two herbs used in folk treatment in Korea The two herbs were A: Equisetum arvense L. (Seddugi), B: Eunymus alatus Siebold (Hwasalnamoo). The experiment was performed with the male rats of Sprague-Dawley which fed on high calorie and animal fat supplemented diets from which 30% of the total calories can from lard. Experimental groups were divided into standard (ST) and two other(A, B) groups for the each herb. Each of the two experimental groups consisted of three subgroups (I, II, III) according to the three different doses of the folk remedy used, which were five times and ten times the dose commonly used in a folk remedy. During the experimental period, total food intake and body weight gains were measured, and FER, CER, PER were calculated at the same times. Serum levels of triglyceride, total lipid, ttal cholesterol, HDL-cholesterol, and total lipid as well as cholesterol contents of the liver were measured at the time of sacrifice. Epididymal fat pads were weighted and an indes was calculated. The results of the effects on the lipid metabolism were as follows : 1.Total food intake and body weight gains increased in group A & B. 2. The serum levels of the total lipid and triglyceride decreased in AIII, BI and BIII.3. The serum level of cholesterol decreased in group B, while level of HDL-Cholesterol were increased in all groups and most effectively in group B. 4. The contents of total liver lipid increased in groups A&B. cholesterol decreased in groups A&B. In conclusion, effects on the lipid metabolism associated with the kinds of herbs a diet were the increase of body weighta and the decrease of total lipid and cholesterol levels in serum and liver.
