Aloe가 알코올을 섭취한 흰쥐의 간 기능 및 지질대사에 미치는 영향 (Effects of Aloe on Liver Function and Lipid Metabolism in Alcohol-Consuming Rats)

  • The effects of aloe on liver function and lipid metabolic disorders induced by alcohol consumption were studied in rats using aloe power (0.25%, 0.5%, 1%) and 10% ethanol. 35 Sprague-Dawley (male, 4 weeks old) rats were divided into five groups and fed experimental diets for six weeks. Body weights of rats tended to be lower in all alcohol supplemented groups than in the control. Food intakes and dry feces per day were significantly lower in all alcohol supplemented groups than in the control. Atherogenic indices (AI) were highest in the alcohol group and decreased in proportion with aloe amount. Serum triglyceride level was significantly higher in the alcohol group than in the control, but tended to be lower in the aloe supplemented groups. In relation to liver function, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), and alkaline phosphatase (ALP) activities tended to be higher in the alcohol groups than in the control, but lower in the aloe groups, especially in the alcohol+0.5% AO group. The levels of liver cholesterol were significantly lower in the alcohol group than in the control and aloe supplemented groups. In the histochemical evaluation, fat droplets appeared extensively on the liver-lobule in the alcohol group, whereas they decreased slightly in the alcohol+0.25% AO group and apparently disappeared in the alcohol +0.5% AO. On the other hand, fat droplets appeared again on the liver-lobule in the alcohol+1% AO group, but were reduced compared with the alcohol group. Regarding the fatty acid composition of adipose tissue triglycerides, the level of linoleic acid (18:2) was significantly higher in the aloe supplemented group. Regarding the fatty acid composition of liver phosphatidylcholine (PC), the level of linoleic acid was higher in the alcohol group and alcohol+1% AO group than the other groups. In contrast, the level of arachidonic acid was significantly lower in the alcohol group. As a result, arachidonic / linoleic acid ratios were significantly lower in the alcohol group compared to the control group, whereas the ratios of the aloe supplemented groups were similar to that of the control group. Therefore, aloe had some beneficial effects on lipid metabolic disorders induced by alcohol and affected desaturation of fatty acids.

Incorporation of n-3 Long-chain Polyunsaturated Fatty Acids into Duck Egg Yolks

  • The objective of this experiment was to determine the effects of different levels of refined cod liver oil (RCLO) on laying performance, n-3 polyunsaturated fatty acids composition (n-3 PUFAs) and the organoleptic evaluation of duck egg yolks. A total of 180 30 wk old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, supplemented with 2% tallow (control) and graded levels of RCLO at 2, 3, 4, 5, and 6% to a corn-soybean diets, respectively, for 6 wks. All of the experimental diets were formulated to be both isocaloric and isonitrogenous. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the RCLO supplementation levels did not affect (p>0.05) egg production, egg mass, feed intake, feed efficiency or body weight change. Egg weight was the lightest when the ducks received the 6% RCLO diet. The eicosapentaenoic acid, docosahexaenoic acid, and total n-3 PUFAs contents in the yolks increased with increasing RCLO supplementation. The taste and general acceptability of the hard-boiled eggs were not significantly different among the treatments. However, a fishy flavor was much higher when ducks were fed diets supplemented with 5% and 6% RCLO diets.

오리를 이용한 한방식품 추출액이 흰쥐의 지질함량에 미치는 영향 (Effect of Duck Extract on Lipids in Rats)

  • The aim of the present study was to investigate the effect of duck extract on lipids. Rats in the experimental group were orally administered with duck extract for four weeks. The half of the supplementation was composed of duck meat and the other of the supplementation was composed of oriental herbs. Weight gain, diet intake, and food efficiency ratio were compared between control and experimental groups. Organ weight, lengths of small/large intestines, and the following plasma biochemical parameters were also measured: hemoglobin, hemoglobin, GOT, GPT, blood glucose concentration, lipids of plasma, liver, feces. Spleen index (weight/100 g body weight) of the rats in the experimental group was significantly higher than those in the control group due to increase in fat intake from the supplementation of duck meat. The large intestines of the rats in the experimental group was significantly larger due to the supplementation with dietary fiber derived from herbs in the extract. There were significant higher levels of plasma hematocrit, GOT, total cholesterol, and HDL-cholesterol in the rats of the experimental group. From the results, although duck meat is a source of animal food, the fat profile of duck extract, such as the ratio of polyunsaturated fatty acids to saturated fatty acids appears to be beneficial to human health. These findings suggest a possible anti-hyperlipidemic effect of duck extract. (Korean J Nutrition 36(1): 3∼8, 2003)

두뇌 조직의 $\alpha$-Tocopherol에 관한 연구 - I. Saponification 과정 유무에 의한 뇌조직, 간조직 및 혈청 $\alpha$-Tocopherol농도의 비교연구 - (Studies on Concentration of $\alpha$-Tocophero in Rat Tissue and Serum - I. Effect of Saponification on Concentration of $\alpha$-Tocopherol in Rat Brain, Liver and Serum -)

  • The concentrations of $\alpha$-tocopherol in the brain, liver, and serum were studied with and without saponification process between control and vitamin E supplemented rats. Young rats, 80-120g body weight, were fed control and vitamin E supplemented diets, ad libitum, for four weeks. $\alpha$-Tocopherol concentrations were determined by high pressure liquid chromatography. The $\alpha$-tocopherol concentration per wet weight base in the brain tissue was significantly lower than that in the liver. Vitamin E supplementation had no effect on brain $\alpha$-tocopherol levels in contrast to the significant increase in lover $\alpha$-tocopherol concentration with and without saponification is significantly greater in the brain than in the liver or serum. Further study is needed to clarify the nature of interaction or /and binding between $\alpha$-tocopherol and the complex membrane system in brain tissue. It can be speculated from this and other studies that the metabolism and the nature of interaction of $\alpha$-tocopherol with the complex membrane system in brain tissue rich in polyunsaturated fatty acids seems different from that in liver tissue or serum.

Role of stearyl-coenzyme A desaturase 1 in mediating the effects of palmitic acid on endoplasmic reticulum stress, inflammation, and apoptosis in goose primary hepatocytes

  • Objective: Unlike mammals, goose fatty liver shows a strong tolerance to fatty acids without obvious injury. Stearyl-coenzyme A desaturase 1 (SCD1) serves crucial role in desaturation of saturated fatty acids (SAFs), but its role in the SAFs tolerance of goose hepatocytes has not been reported. This study was conducted to explore the role of SCD1 in regulating palmitic acid (PA) tolerance of goose primary hepatocytes. Methods: 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide was examined to reflect the effect of PA on hepatocytes viability, and quantitative polymerase chain reaction was used to detect the mRNA levels of several genes related to endoplasmic reticulum (ER) stress, inflammation, and apoptosis, and the role of SCD1 in PA tolerance of goose hepatocytes was explored using RNA interfere. Results: Our results indicated that goose hepatocytes exhibited a higher tolerant capacity to PA than human hepatic cell line (LO2 cells). In goose primary hepatocytes, the mRNA levels of fatty acid desaturation-related genes (SCD1 and fatty acid desaturase 2) and fatty acid elongate enzyme-related gene (elongase of very long chain fatty acids 6) were significantly upregulated with 0.6 mM PA treatment. However, in LO2 cells, expression of ER stress-related genes (x box-binding protein, binding immunoglobulin protein, and activating transcription factor 6), inflammatory response-related genes (interleukin-6 [IL-6], interleukin-1β [IL-1β], and interferon-γ) and apoptosis-related genes (bcl-2-associated X protein, b-cell lymphoma 2, Caspase-3, and Caspase-9) was significantly enhanced with 0.6 mM PA treatment. Additionally, small interfering RNA (siRNA) mediated downregulation of SCD1 significantly reduced the PA tolerance of goose primary hepatocytes under the treatment of 0.6 mM PA; meanwhile, the mRNA levels of inflammatory-related genes (IL-6 and IL-1β) and several key genes involved in the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT), forkhead box O1 (FoxO1), mammalian target of rapamycin and AMPK pathways (AKT1, AKT2, FoxO1, and sirtuin 1), as well as the protein expression of cytochrome C and the apoptosis rate were upregulated. Conclusion: In conclusion, our data suggested that SCD1 was involved in enhancing the PA tolerance of goose primary hepatocytes by regulating inflammation- and apoptosis-related genes expression.

${\omega}$-3 및 ${\omega}$-6계 지방산 첨가 사료의 급여가 어린 병아리에서 면역기관 내 지방산 조성에 미치는 영향 (Effects of Dietary ${\omega}$-3 and ${\omega}$-6 Polyunsaturated Fatty Acids on Fatty Acid Composition of Immune Organs in Young Chicks)

  • 본 연구에서는 ${\omega}$-6 및 ${\omega}$-3계 지방산을 함유한 유지의 첨가비율을 달리한 실험사료를 어린 병아리에서 급여했을 때 면연 기관의 지방산조성에 미치는 영향을 조사하였다. 5개의 처리구에 각 처리당 3반복을 두고 총 75수의 1일령 수평아리를 공시하였다. 옥수수유(CO)와 들깨유(PO)를 중량비로 각각 8%+0%, , 6%+2%, 4%+4%, 2%+6%, 0%+8% 수준으로 첨가한 반정제 실험 사료를 7주간 급여하였다. 실험 종료시 증체량 및 사료섭취량은 처리 간에 큰 차이가 보이지 않았으며 체중 100g당 간장 중량 및 면역 기관 중량에서도 유의한 차이가 인정되지 않았다. 면역조직 내 지방산 조성은 EPA와 AA(arachidonic acid)를 포함하여 섭취한 사료 지방산의 조성을 잘 반영하는 경향을 보였는데, 면역기관 간에 그 정도에 차이가 있었다. 들깨유의 첨가수준이 증가함에 따라 linoleie acid LA)의 비율이 점진적으로 감소하였고 반면 a-linolenic acid(LNA) 비율은 현저하게 증가하는 경향이었다. 흉선에는 .LA와 LNA의 함량이 다른 조직보다 현저히 더 많아서 섭취하는 지방산의 조성이 가장 잘 반영되었다. 비장 조직에는 HPA 및 AA 농도와 EPA/LNA, AA/LA의 비율이 다른 조직에 비해 현저히 더 높았는데 이는 비장이 LA나 LNA로부터 AA나 EPA로 전환하는 능력이 높음을 의미한다. 실험사료 급여후 2, 4주후에 조사된 anti-BSA 항체가 유의한 차이는 인정되지 않았으나 CO 8%구에 비해 PO를 첨가한 모든 구에서 더 많이 생성되는 경향이 관찰되었고 특히 PO를 6% 및 8% 첨가한 실험구에서 20~30%나 더 높아졌다. 결론적으로 섭취하는 지방산 조성에 따라 면역기관내 모든 지방산 조성이 변하였으며, 면역기관에 따라 그 반응정도가 달라지는 것이 관찰되었다. 이는 eicosanoids 합성에도 영향을 미칠 수 있기 때문에 가금에서 질병에 대한 면역력 향상이라는 관점에서 앞으로 더 세밀한 연구가 필요하다.

오징어(Omnastrephes bartrami) 간지질(肝脂質)에 존재(存在)하는 Ethyl Acylates의 구조(構造)와 그 구성(構成) 지방산(脂肪酸) 조성(組成)에 관한 연구(硏究) (The Presence of Ethyl Acylates in the Liver Lipids of A Squid, Omnastrephes bartrami and Their Fatty Acid Composition)

  • Lipid levels in the tissues of liver and intestines of O.bartrami amounted to 40.0% and 1.5%. The new compounds was found to be ethyl acylates, from a deduction of their detailed $^1H-nuclear$ magnetic resonance(NMR) and $^{13}C-NMR$ as well as infra red spectra (IR). The fatty acid composition of total lipids were mainly composed of C16:0(19.0%), C18:1(16.2%) and $C22:6{\omega}3$(15.7%), followed by C20:1(9.4%), C22:1(6.4%) and C18:0(5.4%). New compound A and B were seemed to derived from the cleavage of glycerol moieties of triglycerides by microbial activities during storage in a frozen state. Compound A contained C16:0(38.2%), C18:1(13:4%), C20:1(13.3%) and C22:1(11.7%) as major components, while compound B predominantly comprised polyunsaturated fatty acid such as $C20:5{\omega}3$ (41.2%) and $C22:6{\omega}3$(36.1%). In both compounds small amounts of odd numbered fatty acids were also detected ($3.8{\sim}2.2%$).

Excessive Dietary Conjugated Linoleic Acid Affects Hepatic Lipid Content and Muscular Fatty Acid Composition in Young Chicks

  • The effects of dietary conjugated linoleic acid (CLA) on lipid concentrations and fatty acid composition of various tissues were studied in young chicks. From 7 days of age, a total of 160 chicks were divided into 4 groups, placed into 4 pens per group (10 birds per pen) and fed one of four experimental diets containing 6% tallow (TO 6%), 4% tallow plus 2% CLA (TO 4%-CLA 2%), 2% tallow plus 4% CLA (TO 2%-CLA 4%) or 6% CLA (CLA 6%) for 3 weeks. There were no significant differences in growth performances and the relative weights of various organs, but relative liver weight of chicks fed dietary CLA at 4 and 6% levels was significantly higher (p<0.05) than that of TO 6% group. The chemical compositions of leg muscle were not affected by CLA feeding. However, hepatic total lipid of chicks fed 6% CLA diet was significantly higher (p<0.05) than those of TO 6% and TO 4%-CLA 2% groups. The concentrations of various lipid fractions in serum were not affected by CLA feeding. With the increase in dietary CLA levels, cis 9-trans 11 CLA, trans 10-cis 12 CLA and total CLA of leg muscle increased linearly. The relative proportions of C18:1 $\omega$ -9 and C20:4 $\omega$-6 fatty acids in the leg muscles of chicks fed the CLA containing diets were significantly lower (p<0.05) than those of TO 6% group. These results indicate that the levels of CLA isomers were increased linearly in dose-dependent manner after feeding of synthetic CLA source. But it was also observed that excessive amount of dietary CLA resulted in the possible adversely effects, such as increase of liver weight, hepatic lipid accumulation and serum GOT level.

  • To evalulate the antioxidant effect of flavonoid(+)-catechin on n-3 polyunsaturated fatty acids in vivo, rats were fed with diets containing $5\%$ corn oil(CO), $5\%$ corn oil and $15\%$ purified fresh fish oil(FO) or peroxidized fish oil(PFO) for 10 days. To accerelate lipid peroxidation, all of them were injected with 60mg phenobarbital(a day per kg body weight), and treated with phorone(diisopropylidene acetone) before the rats were killed. Contents of triglyceride, phospholipid, cholesterol and lipid peroxide and the activities of GOT, GPT in serum and total lipid and cholesterol content in liver of PFO group rats were significantly higher than those of the FO one. Contrary to our expectations, the activities of superoxide dismutase (SOD), catalase and glutathione peroxidase(GSH-Px) in liver of the FO group were ]ewer than those of the PFO group. These results might be explained as the results of homeostasis. Even though the hepatic glutathione were depleted, catechin and a-tocopherol inhibited production of lipid peroxide effectively. These results suggested that catechin be considered an antioxidative and hepatoprotective agent.

Streptozotocin 유발 당뇨병 쥐에서의 인삼 지용성분획의 혈당 강하작용 (Hypoglycemic Action of the Fat Soluble Fraction of Panax Ginseng C.A. Meyer in Streptozotocin Induced Diabetic Rats)

  • This study was made to understand a hypoglycemic action of the fat soluble fraction of Panax ginseng C.A. Meyer in streptozotocin induced diabetic rats by determining the activities of several enzymes related to carbohydrate and lipid metabolism as well as several blood component levels such as glucose and ketone bodies, and non-esterified fatty acids. Albino rats (Sprague Dawley, 170-200g, 3) were injected once with 70mg streptozotocinhg body weight intraperitoneally and fed with ordinary diet for 7 days, and then the fat soluble fraction (5 mg~20 mg/day/rat) was injected intraperitoneally once a day for three days to rats having high blood glucose level over 340 mg/100ml. After a final injection of the fat soluble fraction, rats u.ere starved for 16 hours followed by the analysis of blood serum and liver enzymes. It was found that increased levels of glucose, ketone bodies and free fatty acids in streptozotocin induced rats were decreased appreciably by administration of the fat soluble fraction. However, the amount of administered fat soluble fraction did not show any significantly different hypoglycemic action. Decreased activities of glucokinase, phosphofructokinase, pyruvate kinase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of the liver of streptozotocin induced diabetic rats were greatly modified suggesting that a hypoglycemic action of the fat soluble fraction was also appreciable as ginseng saponin fraction. We also compared a hypoglycemic action of the fat soluble fraction prepared from American ginseng and Chinese ginseng with that of Korean pain ginseng. 핀o significant difference of the hypoglycemic activity was observed between the above ginseng fat soluble fractions, suggesting that a study of the fat soluble fraction might be one of the most interesting subjects relating to diabetic hyperglycemia in the near future.
