닭에서의 지방산(脂肪酸) 생합성(生合成)에 미치는 급여지방산(給與脂肪酸)의 영향(影響) (Fatty Acid Biosynthesis of Chicken Fed Various Long Chain Fatty Acids)

  • 무지방사료(無脂肪飼料) 및 여기에 17:0, 18:0 혹은 18:2을 첨가(添加)한 사료(飼料)를 병아리에 급여(給與)해서 간장(肝臟) 및 혈장(血漿)의 각지질분획(各脂質分劃)의 지방산조성(脂肪酸組成)에 미치는 급여지방산(給與脂肪酸)의 영향(影響)을 조사(調査)하였다. 그 결과(結果) 간장중(肝臟中)의 총지질함량(總脂質含量) 및 각지질분획(各脂質分劃)의 함량(含量)은 급여지방산(給與脂肪酸)에 의해서 영향(影響)받지 않았다. 그러나 17:0을 급여(給與)하면 간장(肝臟) 및 혈장(血漿)의 각지질분획(各脂質分劃)에 17:0 및 17:1 출현(出現)하고, 혈장(血漿)트리그리세라이드에서는 17:0이 또한 혈장(血漿)콜레스테롤에스텔에서는 17:1이 다른 분획(分劃)보다도 유의(有意)하게 높았다. 더욱이 간장(肝臟)에서는 각분획(各分劃)사이에 양지방산(兩脂肪酸)의 분포치(分布値)에 큰 차이(差異)가 없었다. 18:2의 급여(給與)도 17:0을 급여(給與)한때와 같이 간장(肝臟) 및 혈장(血漿)의 각지질분획(各脂質分劃)에 18:2이 출현(出現)하고, 특(特)히 간장인지질분획(肝臟燐脂質分劃)에서 18:2의 분포치(分布値)는 다른분획(分劃)보다도 유의(有意)하게 높았다. 그러나 18:0을 급여(給與)해도 이것이 간장(肝臟) 및 혈장중(血漿中)에 특별적(特別的)으로 증가(增加)하지는 않았다. 또한 17:0 및 18:2과 같은 외인성지방산(外因性脂肪酸)의 급여(給與)에 의하여 내인적(內因的)으로 합성(合成)되는 16:1 및 18:1의 분포치(分布値)는 간장(肝臟)에서 감소(減少)하는 경향(傾向)을 나타내었다.

Effect of Supplementing the Diet of Male Chickens With Oils Rich in n-6 Polyunsaturated Fatty Acids on the Fatty Acid Profiles of the Testis and Liver

  • Since the n-6 polyunsaturated fatty acid, docosatetraenoic acid (22:4n-6), is a major functional constituent of avian spermatozoa, the effects of two dietary oils rich in fatty acids which are metabolic precursors of 22:4n-6 on the fatty acid profiles of testicular lipids were investigated during a 39 week period of supplementation from 21 to 60 weeks of age. The effects on liver lipids were determined for comparison. Dietary supplementation of male chickens with Arasco Oil, which provides a large amount of arachidonic acid (20:4n-6), increased the proportion of 20:4n-6 in liver phospholipid by almost 2.5-fold. Although liver phospholipid normally contains very little 22:4n-6, this proportion was significantly increased as a result of Arasco feeding, indicating that the conversion of 20:4n-6 to 22:4n-6 was occurring. The phospholipid of the testis contains much higher proportions of 20:4n-6 and particularly of 22:4n-6 than the liver; supplementation with Arasco Oil significantly increased the proportions of both these polyunsaturates in testis phospholipid but the magnitude of this effect was much lower than that which occurred in the liver. Dietary supplementation with Evening Primrose Oil which contains ${\gamma}-linolenic $ acid (18:3n-6) resulted in significant increases in the proportions of 20:4n-6 and 22:4n-6 in liver phospholipid, although the extent of this increase was less than that produced by the Arasco Oil. By contrast, the feeding of Evening Primrose Oil did not alter the fatty acid composition of phospholipid in the testis. The findings raise the possibility that dietary supplementation with Arasco Oil may modulate the fatty acid profile of avian spermatozoa in a way which could potentially be beneficial for fertility. Moreover, the weights of the testes were almost doubled as a result of supplementation with Arasco Oil or Evening Primrose Oil.

Liposoluble portion of the red alga Pyropia yezoensis protects alcohol induced liver injury in mice

  • The hepatoprotective effect of liposoluble portion of Pyropia yezoensis (PYLP) was investigated against alcohol-induced liver injury in mice. Fatty acids were predominant in PYLP obtained from hexane fraction of 70% EtOH extract after ultrasonication. In particular, polyunsaturated fatty acids such as eicosapentaenoic acid and linoleic acid accounted for 56.91% of the total lipids. PYLP significantly reduced liver damage induced by the alcohol treatment in mice. PYLP treatment increased the activity of antioxidant enzymes including superoxide dismutase, catalase, and glutathion peroxidase by reducing thiobarbituric acid reactive substances. Histological observations showed that PYLP minimizes damage to living tissue induced by alcohol treatment by modulating the expression level of proteins involved in the anti-apoptotic signaling pathway. Our results suggest that PYLP, rich in polyunsaturated fatty acids extracted from the red alga P. yezoensis, will be useful as a potential liver protectant in the hangover industry.

Omega-3 Polyunsaturated Fatty Acid for Cholestasis due to Bile Duct Paucity

  • Omega (${\omega}$)-3 polyunsaturated fatty acids appear to be effective in preventing and treating parenteral nutrition-associated liver disease, and several mechanisms were proposed for this observation. An 8-week-old male infant with cholestasis and acholic stool was diagnosed non-syndromic intrahepatic interlobular bile duct paucity by open-wedge liver biopsy. Initially he was treated with usual supportive medical therapy, including ursodeoxycholic acid. However, the clinical status and laboratory tests did not improve. Omega (${\omega}$)-3 polyunsaturated fatty acids (initially intravenous administration and oral administration later), were started and his liver function, including aminotransferase level and bilirubin levels normalized, and the ivory stool color turned green. We report the possible effectiveness of ${\omega}$-3 polyunsaturated fatty acids as a potent choleretic agent for non-syndromic intrahepatic interlobular bile duct paucity, a very rare structural pediatric hepatic disease.

지방산의 P/S비와 항산화영양소의 보충이 흰쥐의 혈청 지질 농도 및 간의 효소 활성도에 미치는 영향 (Effects of P/S Ratio of Fatty Acids and Antioxidants Supplement on Serum Lipids Levels and Hepatic Antioxidants Enzyme Activities in Rats)

  • The aim of this study was to investigate the effects of P/S ratio of fatty acid and antioxidant (vitamin E, selenium) supplements on the serum lipid levels and hepatic antioxidant enzyme activity in rats. Female 16-week-old Sprague-Dawley rats were fed 6 different experimental diets for 4 weeks. While the peroxidizability index (PI) levels of fatty acids in the experimental diets were fixed at 81.22, the levels of P/S ratio of fatty acids were formulated at 0.38, 1.00, 4.81 (LP, MP, HP). These diets were supplemented with vitamin E (1,000 mg/kg diet) and selenium (2.5 mg/kg diet) (LP-S, MP-S, HP-S). This study showed that the serum concentrations of total-cholesterol and HDL-C increased with the increasing of the P/S ratio in the diet (p <0.05). Antioxidant supplementation significantly lowered the concentrations of triglyceride (TG) and VLDL-C of serum (p<0.05). Levels of thiobarbituric acid reactive substance (TBARS) in the liver tended to decrease with the increasing of the P/S ratio in the diet (p<0.001), but antioxidant enzyme activity in the liver was not significantly different. In addition, antioxidant supplementation significantly lowered TBARS level in the liver (p<0.05), but had no effect on antioxidant enzyme activity except for glutathione reductase (p<0.05). In conclusion, it is necessary to consider the properties of dietary fatty acids and antioxidants supplementation for the prevention of cardiovascular diseases.

고지방식이 및 운동훈련이 흰쥐 간조직의 총지방산 및 인지질 지방산패턴에 미치는 영향 (High Fat Diet or Exercise Training Alters Hepatic Total and Phospholipid fatty Acid Compositions in Rats)

  • Effects of high fat diet and/or endurance exercise training on hepatic total and phospholipid(PL) fatty acid compositions were evaluated in rats fed one of the following diets for 31 days. control diet(CD, 5 wt% corn oil) or high fat diet(HFD, 35 wt% corn oil). Half of the rats in each group were exercise-trained regularly on a treadmill for 90 minutes/day during the entire feeding period. Total and PL fatty acid compositions of hepatic lipid extracts were determined by a gas-liquid chromatograph),. Endurance exercise training did not change the daily food intake, but significantly reduced body weight gain and feed efficiency ratio of rats, which were most prominent in animals fed HFD. Exercise training did not significantly change the percentages of ∑saturated fatty acids (SFA) and ∑polyunsaturated fatty acids(PUEA), but decreased the percentage of ∑monounsaturated fatty acids(MUFA) in hepatic total fatty acids, which might be associated with the decrease in (equation omitted) 9-desaturation index of hepatic total fatty acid metabolism. Exercise training significantly lowered the percentages of 16 : 0 and 22 : 5$\omega$3, and increased the percentages of 20 : 1 and 20 : 3$\omega$3 in both total and PL fatty acid compositions in rat liver. Both total fatty acid and PL fatty acid compositions of rat liver responded more sensitively to changes in dietary fat content than to endurance exercise training in this study. Feeding HFD, whoch contains high level of linoleic acid(LA, 18 : 2$\omega$6), significantly decreased the percentages of ∑SFA and $\Sigma$MUFA, and increased the percentages of ∑PUFA and ∑$\omega$6 fatty acids of hepatic total fatty acids. Hepatic total fatty acid composition was affected by dietary fat content and dietary fatty acid composition more sensitively than those found in hepatic PL fatty acid composition. HFD significantly decreased most of desaturation indices, while exercise training significantly decreased elongation index(20 : 5$\omega$3⇒22 : 5$\omega$3) of hepatic total and PL fatty acid metabolism in rats. 택사(澤瀉)가 유리지방산으로 유발된 HepG2 cell의 lipoapoptosis에 미치는 영향 (The Effect of Alisma orientale Extract on Free Fatty Acid-induced Lipoapoptosis in HepG2 Cells)

  • Objectives : This study was designed to investigate the effect on lipoapoptosis of Alisma orientale extract against free fatty acid-induced cellular injury. Methods : HepG2 cells were used in an vitro model. HepG2 cells were treated with free fatty acids to generate a cellular model of nonalcoholic fatty liver disease (NAFLD). Using this cellular model, the anti-apoptotic effect and reducing steatosis of Alisma orientale extract against free fatty acid-induced cellular injury was evaluated by measuring steatosis and apoptosis. Results : Alisma orientale extract significantly attenuated free fatty acid-induced intracellular steatosis. Alisma orientale extract inhibited free fatty acid-mediated activation of pJNK, PUMA, BAX, caspase-3, and -9, and apoptotic kinases that are correlated with NAFLD. Alisma orientale extract also promoted Bcl-2, a anti-apoptotic protein. Conclusions : From the above, the Alisma orientale extract decreased the hepatocyte steatosis and showed the hepatocelluar protective effect by the regulation of apoptosis-related protein. It proposes the possibility of Alisma orientale extract to the treatment of nonalcoholic fatty liver disease in clinics.

고지방식이에서 서로 다른 불포화 지방산 섭취가 쥐의 혈청과 조직의 Tocopherol함량과 지질과산화물 형성에 미치는 영향 (Effects of Dietary Polyunsaturated Fatty Acids on Tocopherol Contents and Lipid Peroxidation of Plasma and Tissues in Rats Fed High Fat Diet)

  • This study was observed the effect of n6 and n3 polyunsaturated fatty acids of dietary corn oil and fish oil which was supplemented with similar levels of tocopherol in high fat diet on the levels of tocopherol, malondialdehyde ( MDA) productions of plasma and tissues of rats. Also RBC hemolysis, superoxide dismutase (SOD) and glutathione peroxidase (GSH Px) activities In liver were determined. Male Sprague Dawley rats were fed high fat (40%Cal) diet which was different only In fatty acid composition for 6 weeks. Dietary (arts were beef tallow(BT) as a source of saturated fatty acid, corn oil(CO) for n6 linoleic acid (LA) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) + docosahexaenoic acid (DHA). Plasma and liver tocopherol levels were lowered by n3 PUy4 but there was no difference in malondialdehyde(MDA) level by different dietary PUFA. However, MDA content of RBC and hemolysis were increased by n3 PUFA. MDA content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in liver were increased in more unsaturated dietary fat groups. Especially, SOD activity was increased in proportion to the degree of fat unsaturation.

Growth Performance, Meat Quality and Fatty Acid Metabolism Response of Growing Meat Rabbits to Dietary Linoleic Acid

  • An experiment was conducted to determine the effects of different amounts of dietary linoleic acid (LA) on growth performance, serum biochemical traits, meat quality, fatty acids composition of muscle and liver, acetyl-CoA carboxylase (ACC) and carnitine palmitoyl transferase 1 (CPT 1) mRNA expression in the liver of 9 wks old to 13 wks old growing meat rabbits. One hundred and fifty 9 wks old meat rabbits were allocated to individual cages and randomly divided into five groups. Animals in each group were fed with a diet with the following LA addition concentrations: 0, 3, 6, 9 and 12 g/kg diet (as-fed basis) and LA concentrations were 0.84, 1.21, 1.34, 1.61 and 1.80% in the diet, respectively. The results showed as follows: the dietary LA levels significantly affected muscle color of LL included $a^*$ and $b^*$ of experimental rabbits (p<0.05). The linear effect of LA on serum high density lipoprotein cholesterol was obtained (p = 0.0119). The saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs) contents of LL decreased and the polyunsaturated fatty acids (PUFAs) content of LL increased with dietary LA increase (p<0.0001). The PUFA n-6 content and PUFA n-3 content in the LL was significantly affected by the dietary LA levels (p<0.01, p<0.05). The MUFAs content in the liver decreased and the PUFAs contents in the liver increased with dietary LA increase (p<0.0001). The PUFA n-6 content and the PUFA n-6/n-3 ratio in the liver increased and PUFA n-3 content in the liver decreased with dietary LA increase (p<0.01). The linear effect of LA on CPT 1 mRNA expression in the liver was obtained (p = 0.0081). In summary, dietary LA addition had significant effects on liver and muscle fatty acid composition (increased PUFAs) of 9 wks old to 13 wks old growing meat rabbits, but had little effects on growth performance, meat physical traits and mRNA expression of liver relative enzyme of experimental rabbits.

Conjugated Linoleic Acid Changes fatty Acid Composition by Decreasing Monounsaturated fatty Acids in Rabbits and Hep G2 Cells

  • Conjugated dienoic derivatives of linoleic acid(CLA) are a mixture of positional and geometric isomers of linoleic acid(LA). We previously found that CLA changes the fatty acid profile in chicken eggs and serum by decreasing monounsaturated fatty acids. Studies were conducted to explore the effects of CLA on fatty acid composition. Rabbits were fed a semisynthetic diet with or without CLA(0.5g CLA/rabbit/day) for 22 weeks. Compared to the control, rabbits fed CLA had significantly lower monounsaturated fatty acid levels(palmitoleic acid Cl6 : 1 by 50% and oleic acid Cl8 : 1, by 20%) in plasma lipids. We found similar differences in fatty acid composition in the liver and the aorta. The inhibitory effect of CLA on $\Delta$9 desaturation was confirmed in a human hepatoma cell line, Hep G2. CLA significantly decreased $\Delta$9 desaturation in 4-5 hours as shown by an increase in the ratio of Cl6 : 0 to C 16 1, This is apparently due to a decrease in $\Delta$9 desaturase(stearoyl-CoA desaturase, SCD) activity ; it was decreased more than 50%. These results, along with our previous findings, indicate that CLA is an inhibitor of $\Delta$9 desaturase in the liver.
