Optimization of liquid-liquid extraction conditions for paclitaxel separation from plant cell cultures (식물세포배양으로부터 Paclitaxel 분리를 위한 액-액 추출 조건의 최적화)

  • In this study, the process parameters of liquid-liquid extraction were optimized to obtain a high purity and yield of paclitaxel in a pre-purification step. The optimal solvent ratio (methylene chloride/concentrated methanol extract ratio), extraction times, mixing time, and standing time for liquid-liquid extraction were 0.28 (v/v), 3(times), 30 min, and 40 min, respectively. The polar impurities from the biomass extraction were efficiently removed by liquid-liquid extraction. The complete concentration of liquid-liquid extract by rotary evaporator was reliable enough to obtain a high purity and yield of paclitaxel for subsequent purification steps.

Operation Characteristics of the SBR Process with Electro-Flotation (EF) as Solids-liquid Separation Method (전해부상을 고액분리 방법으로 적용한 SBR 공정의 운전 특성)

  • Electro-flotation (EF) was applied to a sequencing batch reactor process (SBR) in order to enhance solids-liquid separation. Solids-liquid separation was good enough in the SBR coupled with EF (EF-SBR) and it was possible to maintain the concentration of mixed liquor suspended solids (MLSS) high in the EF-SBR. Under moderate organic loading condition (COD loading rate: 6 g/day), control SBR (C-SBR) showed similar treatment efficiencies with the EF-SBR. Under high organic loading condition (COD loading rate: 9.6 g/day), the solids-liquid separation in the C-SBR was deteriorated due to proliferation of filamentous bulking organisms at high F/M ratio. However, the EF-SBR was operated stably and with the high MLSS concentration (above 4,000 mg/L) regardless of the organic loading conditions during overall operating period leading to the satisfactory effluent quality. Gas production rate of the electrodes was gradually decreased because of anodic corrosion and scale build-up at the surface of cathode. However it could be partially overcome by use of corrosion-proof electrode material (SUS-316 L) and by periodic current switching between the electrodes.

  • This study was performed to investigate the characteristics and performance of model Decanter for separating swine wastewater to solid and liquid which is slurry state with 12.6% TS. Swine wastewater of the slurry tank was pumped into model Decanter which capacity was $2m^3$/hr in 10% TS Slurry inside of bowl was separated to solid-liquid by centrifugal acceleration. Sampling was done in the section of slurry feed pipe, supernatant outflow pipe, cake discharge pipe. After solid-liquid separation TS, $COD_cr$ and slurry volume reduction effect represented 38%, 40%, 19.6% respectively. Relation factor of model Decanter operation slurry concentration, optimum retention time of slurry, overflow velocity of supernatant, supernatant concentration, sludge removal rate etc. Optimal operation conditions can be set and evaluated efficiency based on the experimental results in the case of Decanter adopted for solid-liquid separation in highly concentrated swine wastewater.

  • Effects of liquid-crystal droplets size on electro-optical properties of the polymer dispersed liquid crystals are investigated experimentally The liquid crystal droplets size was varied by controlling the Laser power for phase separation and LC/oligomer composition ratio. Electro-optical switching voltage is dependent on the LC/oligomer composition ratio and liquid crystal droplet size. The experimental results showed droplet size will be small and saturation voltage is increased but rise time will be short at high power illumination laser light for phase separation.

  • We conducted simultaneous, small-angle, X-ray scattering/differential scanning calorimetry (SAXS/DSC) and simultaneous, wide-angle, X-ray scattering (WAXS)/DSC measurements for a polymer blend of poly($\varepsilon$-caprolactone)/poly(ethylene glycol)(PCL/PEG). The time-dependent SAXS/DSC and WAXS/DSC results, measured while the system was quenched below the melting temperature of PCL from a melting state, revealed the competitive behavior between liquid-liquid phase separation and crystallization in the polymer blend. The time-dependent structural evolution extracted from the SAXS/WAXS/DSC results can be characterized by the following four stages in the PCL crystallization process: the induction (I), nucleation (II), growth (III), and late (IV) stages. The influence of the liquid-liquid phase separation on the crystallization of PCL was also observed by phase-contrast microscope and polarized microscope with 1/4$\lambda$ compensator.

  • Liquid column separation occurs when the valve on the pipeline is closed rapidly in an oil hydraulic system. In this case two-step pressure rise is sometimes observed in a comparatively short pipeline. In this study the two-step pressure rise phenomenon was investigated experimentally and theoretically. The experiments showed that maximum pressure values during two-step pressure rise might exceed extremely the values computed by the theory of rigid-liquid-column separation. So the two-step pressure rise phenomenon appears one of important factors of pipe strength design. From the theoretical considerations based on the experimental and numerical results, the mechanism of two-step pressure rise phenomenon could be explained clearly.

  • The separation of tryptophan enantiomers was carried out with medium-pressure liquid chromatography using BSA (bovine serum albumin)-bonded silica as a chiral stationary phase. The influence of various experimental factors such as pH and ionic strength of mobile phase, separation temperature, and the presence of organic additives on the resolution was studied. In order to expand this system to preparative scale, the loadability of sample and the stability of stationary phase for repeated use were also examined. The separation of tryptophan enantiomers was successful with this system. The data indicated that a higher separation factor (a) was obtained at a higher pH and lower temperature and ionic strength in mobile phase. Addition of organic additives (acetonitrile and 2-propanol) in mobile phase contributed to reduce the retention time of L-tryptophan. About $30\%$ of the separation factor was reduced after 80 days of repeated use.

  • The bunsen reaction, part of IS(Iodine-sulfur) cycle that one of the hydrogen production by the thermochemical water splitting, was investigated. It was observed that $H_2SO_4$ was uniformly generated and generation of $H_2SO_4$ was independent of iodine input. However, generation of HI was decreased with increasing iodine input. It was thought that HI and unreacted iodine were formed complex compound such as $HI_3$ $HI_5$ or $HI_7$. The complex compound accelerated liquid-liquid separation properties in the product. It was also revealed that reaction kinetics was increased with increasing iodine input. Liquid-liquid separation properties were improved with increasing iodine input and reaction temperature. Moreover, no side reaction was occurred at all reaction conditions.

  • A bulk liquid membrane system was introduced for selective separation of an amino acid mixture. We confirmed p-diamethylaminobenzaldehyde (DAB), sulfosalicylic acid (SSA) and 1-naphtol were very useful carriers for selective separation of an amino acid mixture. As a result, Ala, Leu, Val, Phe and Ile were successfully separated by SSA, 1-naphtol in basic condition, 1-naphtol in weak acidic condition, DAB in strong acidic condition and DAB in strong basic condition. The separation mechanism was proposed by ion pair mechanism in the case of SSA and 1-naphtol and Imine bond formation mechanism was also introduced for DAB.

Treatment of High Concentration Organic Wastewater with a Sequencing Batch Reactor (SBR) Process Combined with Electro-flotation as a Solids-liquid Separation Method

  • Operation characteristics of the sequencing batch reactor (SBR) process with electro-flotation (EF) as a solid liquid separation method (EF-SBR) were investigated. EF-SBR process showed excellent solid-liquid separation performance which enabled to separate biosolids from liquid phase within 30 min and to extend cyclic reaction time. Although influent organic loading rate was increased stepwise from 5 to 15 g COD/day, food to microorganisms (F/M) ratio could be maintained about 0.3 g COD/g VSS/day in EF-SBR because biomass concentration could be easily controlled at desired level by EF. However, it was impossible to increase biomass concentration at the same level in control SBR (C-SBR) process because solid-liquid separation by gravity settling showed a limitation at higher mixed liquor suspended solids (MLSS) concentration with 60 min of settling time. Total chemical oxygen demand (TCOD) removal efficiency of EF-SBR process was not decreased although influent organic loading rate became 3 times higher than initial value. However, it was seriously deteriorated in C-SBR process after increasing the rate over 10 g COD/day, which was accounted for insufficient organic removal by relatively higher food to microorganisms (F/M) ratio as well as biosolids wash-out by a limitation of gravity sedimentation.
