Production of Useful Chemicals from Sunflower Oil by Microorganisms

  • A number of microorganisms have been screened for growth on sunflower oil as a sale carbon source for production of useful chemicals. Rhizopus stolonifer NRRL 1478 was found to transform the lipid contents of sunflower oil into dodecyl ${\beta}$-D-glucopyranoside and dodecanedioic acid in 15 and 25% yield respectively. The produced compounds were isolated and purified by column chromatography and its chemical identity were established using MS, IR, $^1H\;and\;^{13}C\;NMR$ spectroscopy.

  • Dietary protein is the most expensive component in flounder feed because of the high protein requirement of this species. The non-protein energy sources may also influence the protein utilization of fish. The inclusion of adequate levels of non-protein energy sources in diets can minimize use of protein as an energy source. Protein sparing effect by fat and carbohydrate has been studied in other fish.   • Chlorella cells were cultured with M4N media treated with glucose (5 mM) sucrose (10 mM) and raffinose (30 mM). Phospholipids and their fatty acid compositions were analyzed in the chloroplast isolated from cultured Chlorella cells. Growth rate was prominently raised in the treatment with raffinose. Glucose was the most excellent carbon source in the biosynthesis of total lipid, phosphatidylcholine(PC), phosphatidylethanolamine(PE), phosphatidylinositol(PI) of the chloroplast. Also, the major fatty acids were palmitic, linoleic and linolenic acid during the biosynthesis of phospholipid in the control and in the treatment with carbon sources.

(${\omega}-3$ 고도불포화지방산이 흰쥐의 혈청 지질과 cytokines에 미치는 영향 (The effect of ${\omega}-3$ polyunsaturated fatty acids on serumlipid and cytokines of rats)

  • This study was designed to examine the effect of ${\omega}-3$ fatty acid, linlenic acid, EPA, DHA on serum lipid and cytokines of male rats(Sprague-Dawley). Animals of 3 groups were administrated perilla oil, salmoon oil, and tuna oil of 0.4 $m{\ell}/day$ for 8 weeks respectively. These oils were used for a source of linolenic acid, EPA and DHA. ${\omega}-3$ polyunsaturated fatty acid decreases significantly body weight, serum $PGE_2$ content and serum cytokines content of the rat, and increases internal organs weight, specially liver weight and serum HDL-cholesterol level of the rat. In the results, authors propose to use perilla oil for source of effective ${\omega}-3$ poly-unsaturated fatty acid(linolenic acid) to Prevent cardiovascular and immune diseases.

Endophytic Fungi as a Source of Biofuel Precursors

  • Endophytic fungi, isolated from a number of different species of tropical plants, were investigated for lipid biodiesel precursor production. The extracts produced from liquid cultures of these fungi were subjected to acidcatalyzed transesterification reactions with methanol producing methyl esters and then analyzed through chromatographic (GC-FID) and spectrometric techniques (MS, NMR $^1H$). The European Standard Method, EN 14103, was used for the quantification of methyl esters extracted from the fungi of the species and genera studied. Xylariaceous fungi exhibited the highest concentrations of methyl esters (91%), and hence may be a promising source for biofuel.

Advances in microalgal biomass/bioenergy production with agricultural by-products: Analysis with various growth rate models

  • Mass cultivation of microalgae is necessary to achieve economically feasible production of microalgal biodiesel. However, the high cost of nutrients is a major limitation. In this study, corncob extract (CCE) was used as an inorganic and organic nutrient source for the mass cultivation of Chlorella vulgaris (C. vulgaris). Chemical composition analysis of CCE revealed that it contained sufficient nutrients for mixotrophic cultivation of C. vulgaris. The highest specific grow rate of C. vulgaris was obtained at pH of 7-8, temperature of $25-30^{\circ}C$, and CCE amount of 5 g/L. In the analysis using various growth models, Luong model was found to be the most suitable empirical formula for mass cultivation of C. vulgaris using CCE. Analysis of biomass and production of triacyglycerol showed that microalgae grown in CCE medium produced more than 17.23% and 3% more unsaturated fatty acids than cells cultured in Jaworski's Medium. These results suggest that growing microalgae in CCE-supplemented medium can increase lipid production. Therefore, CCE, agricultural byproduct, has potential use for mass cultivation of microalgae.

Pathogenicity of Bacillus Strains to Cotton Seedlings and Their Effects on Some Biochemical Components of the Infected Seedlings

  • Pathogenicity of eight Bacillus strains to seedlings of four cotton cultivars was evaluated under greenhouse conditions. Each of the tested cultivars was individually treated with powdered inoculum of each bacterial strain. Untreated seeds were planted as control treatments in autoclaved soil. Effects of the tested strains on levels and activities of some biochemical components of the infected seedlings were also assayed. The biochemical components included total soluble sugars, total soluble proteins, total free amino acids, peroxidase, polyphenol oxidase, phenols, and lipid peroxidation. ANOVA showed that Bacillus strain (B) was a very highly significant source of variation in damping-off and dry weight. Cotton cultivar (V) was a nonsignificant source of variation in damping-off while it was a significant source of variation in dry weight. B × V interaction was a significant source of variation in damping-off and a nonsignificant source of variation in dry weight. Bacillus strain was the most important source of variation as it accounted for 59.36 and 64.99% of the explained (model) variation in damping-off and dry weight, respectively. The lack of significant correlation between levels and activities of the assayed biochemical components and incidence of damping-off clearly demonstrated that these biochemical components were not involved in the pathogenicity of the tested strains. Therefore, it was hypothesized that the pathogenicity of the tested strains could be due to the effect of cell wall degrading enzymes of pathogenic toxins. Based on the results of the present study, Bacillus strains should be considered in studying the etiology of cotton seedling damping-off.

사람에서 n-3계 불포화지방산이 Serum Lipoprotein과 지질조성에 미치는 영향 (Effect n-3 Polyunsaturated Fatty Acids on Serum Lipoprotein and Lipid Compositions in Human Subjects)

  • Ten college women were divided into 5 groups and treated in randomized block design for 5 weeks with 1 interval between treatments and subjects serving as their own controls. The experimental diets were corn oil diet as a source of n-6 linoleic acid, perilla oil diet as a source of n-3 $\alpha$-linolenic acid, and fish oil diet as a source of n-3 EPA and DHA. Dietary fat was supplied at 30% Cal and modified to give the total amount of saturated fatty acids and monoenoic acids at constant level. There was no significant effect on serum cholesterol level by different PUFA. However, on a gram-for-gram basis, there was a trend that the decrease in serum cholesterol was proportionate to the degree of fat unsaturation. On the other hand, only fish oil diet significantly decreased TG level but no significant effect on the relative proportion of TG in VLDL. The degree of hypotriglyceridemia did not corrleate with the degree of unsaturation. The relative proportion of CE in LDL was reduced by all PUFA diets but significant only by perilla oil diet. The relative amount of apoprotein in LDL was significantly reduced by n-3 PUFA. HDL-Chol content was significantly increased only in fish oil diet but no change in the relative proportion of its chemical components of HDL.

Effect of Perilla Oil in Diet on the Biochemical Property of Cultured Sweet Smelt Plecoglossus altivelis

  • The effect of perilla oil added in diet on the biochemical properties of cultured sweet smelt, Plecoglossus altivelis, was investigated. The cultured fish were fed two different diets for 8 weeks; a control diet was a commercial diet, which was low in the content of docosa­hexaenoic acid (DHA, 22: 6n-3) and eicosapentaenoic acid (EPA, 20: 5n-3) less than approximately $2\%$ (CO group) and an experimental diet (PO group) was added perilla oil as a lipid source in the diet of the CO group. The PO group was superior in growth rate and feed efficiency compared with CO group. This trend showed markedly in female of both groups. The fatty acid composition in the muscle of PO group was closely related with those of the diet, while those of CO group were not. For plasma components, total cholesterol (CHOU of PO group was higher than that of CO group. Thiobarbituric acid-reactive substances (TBARS), hydroxyl (OH) radical levels and superoxide dismutase (SOD) activity of plasma were higher in PO group than CO group. The intensity of watermelon-like or cucumber-like aroma was much stronger in PO group with higher level of TBARS and OH radical in plasma compared CO group. Survival rate was also high in PO group with high levels of phagocytic rate, CHOL and SOD activity. These results suggest that perilla oil might be usefulness as a lipid source of the cultured sweet smelt diet, in which result in high quality of the cultured fish.

  • This study was conducted to investigate the contents of dietary fiber (DF), compositions of fatty acids in lipid fraction and amino acids in salt-soluble protein from the functional food source such as Platycodi radix, perilla seed, evening primrose seed and aloe vera. The contents of dietary fiber, neutral detergent fiber (N.D.F) , acid detergent fiber( A.D.F) . kignin, hemicellulose an dcellulose in evening primorse seed were higher than those of other samples, except the content of cellulose .The ration of polyunsaturated /saturated (P/S) fatty acid in total lipids was 6.31 in perilla seed, which was higher than those of other samples. The content of linolenic acid (n-3) in perilla seed was 55.47%. The content of linoleic acid (n-6) in evening primrose seed was 71.88% , which was higher than those of other samples. The fatty acid composition in neutral lipids were the same as those of total lipids. The PUFA contents of fatty acid in glycolipids were 61.76% in perilla seed. And also, the ratio of n-6/n-3 in evening primrose seed was 15.19. The fatty acid compositions in phospholipids were the sameas those of glycolipids. The contents of PUFA in Platycodi radix were 62.96% . The essential amino acid contents of salt-soluble protein were 47 mole % in Platycodi radix , which was slightly higher than those other samples. The ration of essential amino acid /nonessential amino acid (E/N) was 0.9 and 0.66 in Platycodi radix and aloe vera, respectively.
