Gamma-linolenic acid (GLA, C18:3 ${\Delta}^{6,9,12}$) is synthesized by a delta-6 fatty acid desaturase using linoleic acid (LA, C18:2 ${\Delta}^{9,12}$) as a substrate. To enable the production of GLA in the conventional yeast Pichia pastoris, we have isolated a cDNA encoding the delta-6 fatty acid desaturase from Cunninghamella echinulata MIAN6 and confirmed its function by heterogeneous expression in P. pastoris. Sequence analysis indicated that this cDNA sequence has an open reading frame of 1,404 bp, which encodes a 52 kDa peptide of 468 amino acids. This sequence has 64% identity to the previously reported delta-6 fatty acid desaturase from Rhizopus oryzae. The polypeptide has a cytochrome b5 domain at the N-terminus including the HPGG motif in the heme-binding region, as reported for other delta-6 fatty acid desaturases. In addition, this enzyme differs from other desaturases by the presence of three possible N-linked glycosylation sites. Analysis of the fatty acid composition demonstrated the accumulation of GLA to the level of 3.1% of the total fatty acids. Notably, the amounts of ginkgolic acid (C17:1) and palmitic acid (C16:0) were increased from 1.3% to 29.6% and from 15% to 33%, respectively. These results reveal that the modification of the fatty acid biosynthetic pathway by genetic manipulation in order to produce specific polyunsaturated fatty acids in P. pastoris is a promising technique.
The chemical components of the wild vegetables Solidago virgaurea spp. gigantea, Aster glehni var. hondoensis, and Aruncus dioicus var. kamtschaticus grown on Ulleung island, Korea were investigated. In dried powders of these three wild vegetables, the contents of moisture, crude protein, crude lipid, crude fiber, and crude ash were 3.77-5.72%, 15-29%, 3.50-6.68%, 4.00-6.01%, and 8.70-10.54%, respectively. There were differences in the levels and nature of organic acids in the vegetables; the major organic acids were succinic acid, citric acid, and malonic acid. The major free amino acids in the vegetables were glutamic acid, aspartic acid, arginine, and phenylalanine. Riboflavin contents were 90 mg% in Aruncus dioicus var. kamtschaticus and $2{\sim}3$ mg% in the other vegetables. The thiamin contents were 113 mg% in Aster glehni var. hondoensis, 85 mg% in Aruncus dioicus var. kamtschaticus, and 71 mg% in Solidago virgaurea spp. gigantea. The major fatty acids in the three vegetables were linolenic acid, linoleic acid, and palmitic acid; 60% of total fatty acid was linolenic acid. Catechins were present at 5.37 mg% in Aruncus dioicus var. kamtschaticus, 2.46 mg% in Solidago virgaurea spp. gigantea, 1.29 mg% in Aster glehni var. hondoensis.(-) ECC was detected in only Aruncus dioicus var. kamtschaticus. The contents of Ca, P, and Na in the three vegetables were higher than the contents of other minerals.
In order to develop new materials for the functional food, the components of safflower(Carthamus tinctorius L.) sprout was studied. Chemical composition, minerals, amino acids, and fatty acids of the leaf and the stem parts of 2 weeks grown safflower sprout were analyzed. The chemical composition of safflower spout was 12.7% (w/w) moisture, 28.5% crude protein, 10.1% crude fat, and 5.3% crude ash. Mineral contents of the leaf part were P 49%, K 22%, Mg 15%, Ca 10% whereas these of the stem part were K 51%, P 27%, S 10%, Ca 7%, Mg 4%. Other mineral contents were less than 2% in both parts. Especially, Pt was 0.18 ppb and 0.17 ppb in the leaf and the stem parts, respectively. The composition of the amino acid were approximately as follow, the major amino acid in the leaf and the stem were aspartic acid and glutamic acid, the contents of these were 10.7mg/g, 10mg/g in the leaf, 11.3 mg/g, 8.4 mg/g in the stem, respectively. The major fatty acids in the leaf and the stem parts were linoleic acid and $\alpha$-linolenic acid. The linoleic acid (C18:2) in the leaf and the stem parts were 67% (w/w) and 47% whereas the $\alpha$-linolenic acid. The linoleic acid (C18:2) in those parts were 14% (w/w) and 11%, respectively. On the basis of chemical analysis, the safflower sprout showed to have relatively high contents of crude protein and crude fat, minerals including small amount of Pt, polyunsaturated fatty acid as linoleic acid and $\alpha$ -linolenic acid. These results suggested that safflower sprout was found to be a useful material of natural health food for the functional food development.
Composition and unsaturation ratio of fatty acids composing phospholipid which is the main constituent of biological membranes are known to be related to the response of plant to cold stress. In order to investigate a relationship between fatty acid unsaturation ratio of phospholipid and the range of cold tolerance of rice cultivars obtained in the field experiment, the lipid from the leaves of 32 rice cultivars were isolated and the fatty acid composition of phospholipid was determined by gas chromatography. In this experiment, composition and unsaturation ratio of fatty acids composing phopholipid on rice plants grown under natural condition and treated with chilling temperature were compared with the cold tolerance in the field for rice cultivars. The results obtained were summarized as follows ; 1. The phospholipids isolated from the leaves of 32 rice cultivars both grown under natural condition and chilling treatment contained palmitic, linoleic and linolenic acid as major components and palmitoleic, stearic and oleic acid as minor components. 2. The content of palmitic acid, a saturated fatty acid, was reduced in rice seedlings subjected to chilling treatment for 3 days at $15^{\circ}C$, and contents of linoleic and linolenic acid, unsaturated fatty acids, were increased. 3. The unsaturation ratio of fatty acid was increased by chilling treatment, and was high in cold-tolerant cultivars but low in cold-susceptible cultivars and the results were correlated with the range of cold tolerance investigated in the field. 4. The content of palmitic acids among fatty acids composing phospholipid was reduced and that of linolenic acid was increased by hardening treatment. 5. The unsaturation ratio of fatty acid was increased 18~24% by hardening treatment compared to control.

This study was designed to examine the effect of ${\omega}-3$ fatty acid, linlenic acid, EPA, DHA on serum lipid and cytokines of male rats(Sprague-Dawley). Animals of 3 groups were administrated perilla oil, salmoon oil, and tuna oil of 0.4 $m{\ell}/day$ for 8 weeks respectively. These oils were used for a source of linolenic acid, EPA and DHA. ${\omega}-3$ polyunsaturated fatty acid decreases significantly body weight, serum $PGE_2$ content and serum cytokines content of the rat, and increases internal organs weight, specially liver weight and serum HDL-cholesterol level of the rat. In the results, authors propose to use perilla oil for source of effective ${\omega}-3$ poly-unsaturated fatty acid(linolenic acid) to Prevent cardiovascular and immune diseases.

This research was performed to observe the effect on aggregation of rat platelet treated with ginkgo and perilla oil which contain much linolenic acid. The numbers of platelet treated with ginkgo and perilla oil were $2.7{\times}10^{8}/m{\ell}\;and\;4.7{\times}10^{8}/m{\ell}$, respectively. These numbers were much less than control group(this group was $7.5{\times}10^{8}/m{\ell}$). The ability of platelet aggregation treated with Perilla oil and ginkgo oil was 1.4 folds less than control group. Concentrations of total cholesterol and free fatty acid in serum of rat treated with ginkgo and perilla oil group were almost equal to those of control group. group.

Soybean [Glycine max(L.) Merr.] oil is versatile and used in many products. Modifying the fatty acid profile would make soy oil more functional in food and other products. The ideal oil with the most end uses would have saturates(palmitic + stearic acids) reduced from 15 to < 7%, oleic acid increased from 23 to > 55%, and linolenic acid reduced from 8 to < 3%. Reduced palmitic acid(16:0) is conditioned by three or more recessive alleles at the Fap locus. QTLs for reduced palmitic acid have mapped to linkage groups(LGs) A1, A2, B2, H, J, and L. Genes at the Fad locus control oleic acid content(18:1). Six QTLs($R^2$=4-25%) for increased 18:1 in N00-3350(50 to 60% 18:1) explained four to 25% of the phenotypic variation. M23, a Japanese mutant line with 40 to 50% 18:1 is controlled by a single recessive gene, ol. A candidate gene for FAD2-1A can be used in marker-assisted breeding for high 18:1 from M23. Low linolenic acid(18:3) is desirable in soy oil to reduce hydrogenation and trans-fat accumulation. Three independent recessive genes affecting omega-3 fatty acid desaturase enzyme activity are responsible for the lower 18:3 content in soybeans. Linolenic acid can be reduced from 8 to about 4, 2, and 1% from copies of one, two, or three genes, respectively. Using a candidate gene approach perfect markers for three microsomal omega-3 desaturase genes have been characterized and can readily be used in for marker assisted selection in breeding for low 18:3.
To compare the hypolipidemic effects of n6 linoleic acid. n3 $\alpha$-linolenic acid and n3 eicosapentaenoic acid in rats fed high carbohydrate(70% Cal) diet. male Sprague Dawley rats were fed different experimental diets for 6 weeks. which were different only in fatty acid composition. The dietary fats were beer tallow(BT) as a source of saturated fatty acid (SFA), corn oil(CO) for n6 linoleic acid(LA), perilla oil(PO) for n3 $\alpha$-linolenic acid(LL) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) and docosahexaenoic acid (DHA) Plasma total cholesterol(Chol) level was increased by n6 LA but decreased by n3 LL and n3 EPA and most effectively reduced by n3 EPA. HDL-Chol level was raised by n6 LA, but there was no significant change in HDL-Chol levels by n3 LL and was lowered by n3 EPA. Plasma TG level was reduced by n6 LA, but lipogenesis in liver was not affected by n6 LA. However, plasma TG level was lowered by n3 LL and EPA. Both lipogenic enzyme activity and liver TC level were also decreased by n3 PUFA. The relative proportions of TG in VLDL was significantly lowered by n3 EPA. but the proportions of Apo B in VLDL was not changed by n3 EPA. Overall. the hypolipidemic effect was in the order of EPA+ DHA(n3) >LL(n3) >LA(n6) and fish oil and perilla oil rich in n3 PUFA may have important nutritional applications in the prevention and treatment of hypertriglyceridemia.
This experiment was carried out to investigate the changes in phospholipid and glycolipid of Kaesojoo and Kaesojoo added medicinal herbs during storage(30 days) at 4$^{\circ}C$and -18$^{\circ}C$. Two dogs with 12kg live weight(♀, The Korean Jindo dog Hy-breed, 11∼12 month) were slaughtered to obtain samples. The result obtained were as follows: The saturated fatty acids found in phospholipid of Kaesojoo were palmitic acid, stearic acid, myristic acid, while the unsaturated fatty acid found in phospholipid of Kaesojoo with medicinal herbs were palmitic acid, stearic acid, arachidonic acid, while the unsaturated fatty acid were mostly oleic acid, increased during the period of storage. The saturated fatty acid found in glycolipid obtained from Kaesojoo were mostly oleic acid, linoleic acid and linolenic acid. The saturated fatty acid obtained from Kaesojoo with medicinal herbs were palmitic acid, stearic acid, while the unsaturated fatty acid were mostly oleic acid, linoleic acid and linolenic acid. The rates of changes in unsaturated and saturated fatty acid were higher at 4$^{\circ}C$ than that of -18$^{\circ}C$ during storage.
The biosynthesis of phospholipid and the composition of fatty acid in C. ellipsoidea mitochondria treated with antibiotics(cycloheximide, nalidixic acid) during the culture analyzed. The growth of Chlorella and the contents of total lipid in mitochondria treated with antibiotics were lower than those of the control. The synthesis of PC (phosphatidylcholine) and PI(phosphatidylinostiol) were inhibited in the nalidixic acid treatment and also the contents of PC(phosphatidylcholine), PE (phosphatidylethanolamine), PG(phosphatidylglycerol) and PI(phosphatidylinositol) in the cycloheximide treatment were also inhibited.
