Evaluating phylogenetic relationships in the Lilium family using the ITS marker

  • Lilium is a perennial bulbous plant belonging to the liriotypes genus. Our aim was to study the phylogenetic relationships of the Lilium family. Two varieties of Lilium ledebourii, 44 varieties of the gene bank, and one variety from the Tulipa family served as the out group. In order to study the diversity between lilium masses, ITS regions were used to design the marker. The results showed that the guanine base is the most abundant nucleotide. Relatively high conservation was observed in the ITS regions of the populations (0.653). Phylogenetic analysis showed that sargentiae and hybrid varieties are older than other varieties of the Lilium family. Also, the location of L. ledebourii varieties (Damash and Namin) was identified in a phylogenetic tree by using the ITS marker. Overall, our research showed that ITS molecular markers are very suitable for phylogenetic studies in the Lilium family.

Analysis of Chalcone Synthase and Flavanone 3-Hydroxylase Activity in Lilium Cultivars (Lilium품종의 Chalcone Synthase와 Flavanone 3-Hydroxylase 효소학적 분석)

  • In this work, we analyzed the activity of control enzymes of flower color biosynthesis, chalcone synthase (CHS) and flavanone 3-hydroxylase (FHT) using biochemical and enzymological methods in Lilium longiflorum and 11 Lilium cultivars. The results obtained are as follows ; Naringenin (NAR) was synthesized in all Lilium cultivars tested by the catalytic activity of CHS which used malonyl-CoA and 4-coumaryol-CoA as substrates. Substrate-specific activity of CHS was observed because eridictiol (ERI), which uses caffeoyl-CoA as a substrate, was not detected in tested cultivars. In next step, dihydroflavone product was synthesized by FHT using flavanones as a substrate. FHT synthesized dihydrokaempferol (DHK) by using NAR as substrates. A remarkable activity of FHT was observed in other 11 cultivars.

Effects of Bulblet Formation on Scale Tissue Culture and Growth of Characteretis in Edible Lilium brownii spp. (식용 백합 Lilium brownii의 생육특성과 인편조직배양이 자구 형성에 미치는 영향)

  • This experiment was carried out to establish growth characterestics and number of bulblet, bulb weight, and diameter of bulblet on scale tissue culture in Lilium brownii. Flowering date of Lilium brownii was 25th, May with 50cm hight in field condition. Vimyl house condition was flowered 28th May with 54cm hight. Number of bulblet, bulb weight, diameter of bublet was good position of outer>Middle>inner scale in Lilium brownii. Number of bulblet formation was the best with 3.5 in large scale. Concentration of BA was the best at 1.5mg/L with 3.8 bulblet with bulb weight, weight and diameter of bulblet.

A taxonomic note of the genus Lilium section Martagon in Korea (한국산 백합속(Lilium) 내 말나리절의 분류학적 검토)

  • The genus Lilium comprises about 100 species and is a major element of the family Liliaceae in temperate region of North Hemisphere. In the light of recent studies on the genus, we tried to examine the taxonomy of three Korean species (L. hansonii, L. tsingtauense, and L. distichum) belonging to section Martagon. The four available chloroplast regions (rbcL, matK, ndhF, and atpB) in Lilium species were selected and analyzed for a maximum likelihood phylogeny. As a results, three Korean species of section Martagon formed a monophyletic group. However, they nested within paraphyletic section Sinomartagon. Further, the discussions on taxonomic affinities of Korean species have made based on comparison between topology of phylogenetic tree and morphology.

Induction by Salicylic Acid and Characterization of PR-Proteinases from Bulbs of Lilium formosanum Wallace (흰나리 인편으로부터 PR-Proteinase의 유도 및 특성 규명)

  • Some morphological character were surveyed and PR-proteinases were induced and characterized from Lilium formosanum Wallace endeimc to Cheju island . Its flower characters were similar to those of white trumpet lilies(Lilium longiflorum Thunb) although its flowering period was later than that of white trumpet lilies and it hadd a wide range of variation among individuals. Six PR-proteinases(II-2, III-1, III-2, IV-1, IV-2 and V) were induced from bulbs by 2.5mM salicylic acid and almost excreted into the intercellular spaces. These PR-proteinases were strongly activated by Ca 2+, , whereas they were strongly inhibited by Cu2+ Co2+ and Fe2+ . Three PR proteinases(II-2, IV-1 and IV-2) were strongly inhibited by 1, 10 -phenanthroline, indicating that these enzymes are metallo-proteinases. Three PR-proteinases(III-1, III-2 and V) had a high sensitivity to PMSF and required $\beta$-mercaptoethanol for their activities. These results indicate that these proteinases are cysteine proteinases.

Vascular Plants in the Gyeryongsan National Park (계룡산국립공원의 관속식물상)

  • The vascular plants in the Gyeryongsan National Park were listed 227 taxa; 71 families, 148 genera, 196 species, 1 subspecies, 26 varieties and 4 forms, Among them, trees and herbs were 103 taxa (45.4%), 124 taxa (54.6%), respectively. The rare plants were identified 6 taxa such as Paeonia obovata (Legal protection species), Viola albida, Acanthopanax chiisanensis, Lilium distichum, Lilium callosum and Goodyera schlechtendaliana by the Korea Forest Service. Based on the list of Korean endemic plants, 8 taxa were recorded; Cephalotaxus koreana, Pseudostellaria multiflora, Thalictrum uchiyamai, Indigofera koreana, Acanthopanax chiisanensis, Asperula lasiantha, Weigela subsessilis and Carex okamotoi. Based on the list of approved for delivering overseas of plants, 6 taxa were recorded; Viola albida, Acanthopanax chiisanensis, Asperula lasiantha, Carex okamotoi, Lilium distichum and Lilium callosum. Specific plant species by floral region were total 21 taxa (9.3%); Paeonia obovata in class V, 5 taxa (Betula davurica, Acer palmatum, Asperula lasiantha, Lonicera subhispida, Phaenosperma globosa) in class III, 4 taxa (Viola orientalis, Acanthopanax chiisanensis, Ostericum melanotilingia, Lilium distichum) in class II and 11 taxa (Cephalotaxus koreana, Betula schmidtii, Quercus variabilis, Aphananthe aspera, Ilex macropoda, Meliosma oldhamii, Tilia amurensis, Lilium callosum, Erythronium japonicum, Polygonatum inflatum, Hosta capitata) in class I. It is important that relationship between national and local government will be needed for preservation of natural environment of Gyeryongsan National Park. As well, a long time of monitering and systematical management counterplan will be required.

Influence of virus infection in quality of production of Lilium x fomolongi as cut-flowers (신나팔나리(Lilium x fomolongi) 바이러스 감염이 절화품질에 미치는 영향)

  • Damages caused by infection with Cucumber mosaic virus (CMV), Lily mottle virus (LMoV) and Lily symptomless virus (LSV) were assessed by comparing growth of plants produced from seeds of Lilium x fomolongi cultivar 'Noesan' both infected and free from infection with those viruses. Symptoms and infection rate were investigated in field-grown lily.Dominant viral infection symptom in the field was mottle on leaves, caused by natural infection with LMoV. Incidence of viral disease caused by mixed infection with two of LMoV, CMV and LSV in field-grown Lilium x fomolongi cultivars, reused for more than 6 years consecutively, was 80 percent. In comparison with healthy Lilium x fomolongi cultivar 'Noesan', plants doubly infected with CMV-Li1 and LMoV-Li diminished their plant height by 14 percent, fresh weight by 38 percent, and flower length by 15 percent. Lily plants singly infected with CMV-Li1 or LMoV-Li significantly reduced their freish weight by 21.8% and 28.4% compared to healthy plants, respectively.

A Palynotaxonomic Study on the Genus Lilium in Korea (한국산 나리속(Lilium) 식물의 화분분류학적 연구)

  • Size>; and sculpture pattern of pollen grains in seven species and two varieties of the genus Lilium in Korea were examined by LM and SEM, and their evolutionary trends were discussed. Lilium species in Korea can be divided into two groups based on pollen morphology: one is small reticulate group with small pollen grains and lumens, which includes L. concolor var. pulchellum and L. callosum; the other is large reticulate group with large pollen grains and lumens, which includes remaining taxa. It is thought that small reticulate group is more primitive than large reticulate group on the basis of tectum coverage. According to the above results and some morphological characters of salles and perianth segments, L. concolor var. pulchellum and L. callosum are very closely related, and they would be treated under the same subsection.ection.

An Efficient Micropropagation to Obtain the Disease-free Bulbs from Scales for Cryopreservation in Lilium

  • Cryopreservation is one of the ideal and suitable methods for long-term storage of plant germplasm. The plant contaminated with diseases and pathogens are decreased the multiplication rate, survival rate and high quality of plants after cryopreservation. The aim of this work was to improve a micropropagation method for lily in Korea, which are cultivars and indigenous plant. In the last process of rinsing scales after surface-sterilization, we tried to control the diseases and pathogens lived within the tissue by rinsing in 0.03% sodium hypochlorite (NaClO) instead of sterile distilled water. Bulb scales of Lilium were cultured in vitro on MS medium supplemented with Plant Preservative Mixture (PPM). The newly small bulb formed from bulb-scales was transferred to MS medium. We checked the non-contamination and survival rate after 2 weeks in culture. Non-contamination was shown to be 70 to 90% in formed small bulbs. This study will help to mitigate microbial contamination in Lilium species micropropagation for cryopreservation.

Micropropagation of Lilium longiflorum 'Geogia' by Using Bioreactor. (생물반응기를 이용한 Lilium longiflorum ′Geogil′의 대량번식)

  • Shoot clusters were induced from bulb scales of Lilium longiflorum 'Geogia', and proliferated on medium containing 0.5 mg/L BA and 0.5 mg/L IAA. Thereafter, these shoot clusters were cultured in 5 L air-lift bioreactors to form and grow normal bulblets. Number of bulblets increased on medium with 30 g/L sucrose, but growth of bulblets was effective on medium with 60 g/L sucrose. The number of bulblets from shoot clusters had no differences, though bulblet growth was very effective on medium with between full and double strength of MS salts. The inoculation of 100 g shoot clusters as a cultural material was suitable for formation and growth of bulblets in 5 L bioreactors. Air-lift type was more effective for the formation and growth of bulblets than that in ebb and flood one, and 200∼300 mL$.$min$^{-1}$ injection of air was suitable in growth of bulblets.
