광 조사 간격이 Chlorella vulgaris의 성장량 및 질소, 인 대사에 미치는 영향 (Effects of light irradiation interval on the metabolism of nitrogen, phosporus and growth rate of Chlorella vulgaris)

  • This study aimed to investigate growth rate and nutrient consumption of Chlorella vulgaris according to different light irradiation interval. Applied light irradiation intervals were 12 hr, 4 hr, 1 hr, and 1 min. The light source was flexible LED(Blue:Red=1:1), light intensity was 200 PPFD and Light/Dark cycle was 1:1. As a result, growth rate and nutrient removal efficiencies showed no significant differences depending on the light irradiation interval. Considering the reproduction characteristics of applied microalgae cultures of this study, this is thought to be one of the possible reasons of above results. Because Chlorella vulgaris performs an asexual reproduction and it is known that there is no significant relationship between light irradiation interval and growth rate, including nutrient consumption in case of asexual reproduction.

자외선 조사간격이 브로일러 병아리의 중족골 광물질 함량에 미치는 영향 (Influence of UV Irradiation Interval on Mineral Content in Metatarsus of Broiler Chicks)

  • 본 연구는 비타민 D$_{3}$(VD$_{3}$) 결핍 병아리에게 자외선을 상이한 간격으로 조사하여 경과시간에 따라 다리의 중족골을 채취하여 회분, Ca, P의 수준을 조사코자 실시되었다. 육용 Hubbard 계통 199수의 초생추(대조구 10수+3조사간격$\times$9조사 후 경과시간$\times$7반복)를 무창약등 육추사에 넣고 VD$_{3}$ 결핍사료로 3주간 사육한 후, 0.068 mJ/$\textrm{cm}^2$(10분간)의 선량으로 297nm의 UVB 광선을 3회 조사하되 조사간격을 0, 12 또는 24시간 간격으로 하였다. 조사 후 0, 6, 12, 18, 24, 48, 96, 144또는 240시간에 병아리의 중족골을 채취하였다. 중족골은 부착조직을 제거하고 탈지, 건조, 화화하여 회분, Ca, P 함량을 측정하였다. Ca은 원자흡수분광광도법으로, P은 ammonium metavanadate 법으로 비색정량하였다. UVB를 30분간 무간격으로 조사하였을 때 중족골의 Ca함량은 계속 증가되어 240시간에 16.75%에 도달하였다. P의 함량은 UVB 조사후 점점 증가되어 144시간에 최고치 9.75%를 나타내었으며, 회분함량은 UVB 조사후 점점 증가하여 240시간에 42.75%에 이르렀다. 12시간 간격으로 10분간씩 3회 조사하였을 때에는 중족골의 Ca 함량이 12시간에 작은 peak(13.31%), 144시간에 큰 peak (16.91%)를 보였다. P의 함량은 12시간에 작은 peak(7.18%), 240시간에 큰 수준(8.34%)을 보였다. 회분 함량은 UVB 조사후 계속 증가하여 240시간에 46.53%의 높은 값을 나타내었다. 도중의 Ca과 P의 작은 peak는 아마 12시간 및 24시간 전에 조사하였던 UVB의 영향인 것으로 생각된다. 24시간 간격으로 10분간씩 3회 조사했을 때 중족골의 Ca 함량은 점차 증가되어 96시간에 최고치 24.18%를 보였고 P함량은 역시 96시간에 최고치 7.29%를 나타내었으며, 회분 함량은 240시간까지 계속 증가되어 45.73%에 이르렀다. UVB조사 후 경과시간에 따라 살펴보면 중족골의 Ca와 P 함량은 UVB 조사후 96~144시간에 최고치에 도달했으나 회분함량은 240시간까지 계속 증가하는 모습을 보였다. 다음 UVB의 조사방법에 따라 종족골의 Ca함량을 봤을 때 무간격으로 조사시 240시간까지 계속 증가하였고, 12시간 간격으로 조사시 144시간에 최고치를, 24시간 간격으로 조사시에는 96시간에 최고치를 나타내었으며 회분 함량을 봤을 때 12시간 또는 24시간 간격으로 조사하였을 경우가 무간격으로 조사하였을 때보다 더 높은 수준을 보였으므로 24시간 간격으로 10분간씩 조사하는 것이 바람직한 것으로 생각되었다.

  • The purpose of this study was to measure the free flow of the unpolymerized resin by its weight for 10 minutes by one minute interval, and to measure the dimensional change of composite resins during the irradiation of visible light(Quich light VL-l Kuraray Japan) using visible leser displacement meter(LC-2210 Kerence Japan). The unpolymerized resin was cured by the visible light for 40 seconds, the dimensional change was measured at the begining of irradiation for 5 minutes. The results were as follows : 1. In free flow LFI was the largest, BLI was the smallest at $23^{\circ}C$ and CFP was the largest, and BL was the smallest at $37^{\circ}C$. 2. In dimensional change CFI, LFP, LEI and CFP was excessively contracted flow the begining of irradiation until 15 seconds but BLI and BL was excessively contracted until 30 seconds BL and BLI in dimensional change was much larger than LFI and CFP.

단일 및 분할 방사선조사가 백서 악하선 미세혈관계에 미치는 영향 (EFFECTS OF THE SINGLE AND FRACTIONATED IRRADIATION ON THE MICROVASCULATURE OF THE RAT SUBMANDIBULAR GLAND)

  • The purpose of the study was to investigate the effects of the single and fractionated irradiation on the microvascular structure of the submandibular gland in rats. For this study, 90 Sprague-Dawley strain rats were irradiated to their neck region with equal split doses of 9Gy for a 4 hours interval and 15Gy single dose by 6MV X-irradiation and sacrificed on the 1st, 3rd, 7th, 14th and 27th day after irradiation. The author observed histological changes at Hematoxylin and Eosin staining and PAS staining under a light microscope, and also observed distribution and structural changes of the microvasculature in rat submandibular gland using a scanning electron microscope by forming vascular resin casting. The results were as follows: 1. In the light microscopic examination, the microvasculature was slightly dilated and decreased in number on the 1st day after irradiation, and increase in number of microvasculature was observed on the 3rd day after irradiation. And then distribution of microvasculature was markedly increased on the 7th day after iradiation, but decreased on th 14th day after irradiation again. Such changes were greater in the single irradiated group than in the fractionated irradiated group. 2. The reaction to PAS staining on glandular cell was decreased on the 1st and the 3rd day after irradiation, and recovered on the 7th day after irradiation. The reaction was decreased on the 14th day after irradiation again, and recovered on the 28th day after irradiation. Changes were more apparent in the single irradiated group. 3. In the scanning electron microscopic examination, early changes of microvasculature were decreased capillary density, dilation of conduits and meandering. Increased capillary dentsity or anastomosis due to vascular reproduction and smooth curved running were observed on the 7th and 14th day after irradiation. Decreased capillary and smooth running tendency were observed on the 28th day after irradiation again. Such changes were greater in the single irradiated group than in the fractionated irradiated group.

  • This study was designed to investigate the effects of split irradiation on the salivary ductal cells, especially on the intercalated cells of the rat parotid glands. For this study, 24 Sprague-Dawley strain rats were irradiated on the head and neck region with two equal split doses of 9Gy for a 4 hours interval by Co-60 teletherapy unit, Picker's model 4M 60. The conditions of irradiation were that field size, dose rate, SSD and depth were 12×5㎝, 222 cGy/min, 50㎝ and 1㎝, respectively. The experimental animals were sacrificed 1. 2, 3, 6, 12, hours and 1, 3, 7, days after the irradiation and the changes of the irradiated intercalated cells of the parotid glands were examined under light and electron microscope. The results were as follows: 1. By the split irradiation, the degenerative changes of intercalated cells of the parotid glands appeared at 3 hours after irradiation and the most severe cellular degeneration observed at 6 hours after irradiation. The repair processes began from 12 hours after irradiation and have matured progressively. 2. Under electron microscope, loss of nuclear membrane, microvilli and secretory granules, derrangement of chromosomes, degeneration of cytoplasm, atrophy or reduction of intracytoplasmic organelles were observed in the intercalated ductal cells after split irradiation. 3. Under light microscope, derrangement of ductal cells, widening of cytoplasms and nuclei, hyperchromatism and proliferation of ductal cells were observed in intercalated ducts after split irradiation.

Artificial Radical Generating and Scavenging Systems: Synthesis and Utilization of Photo-Fenton Regent in Biological Systems

  • A photo-labile compound which is bioinactive but, upon irradiation with light, yields bioactive species is called as "caged compound". Photolysis of caged compounds generating bioactive species, has become a general method to produce a desired amounts of bioactive species in the specific time interval at the desired place or area of the target biological systems. For this purpose, we designed and synthesized caged hydroxyl radical., "Photo-Fenton Reagent" NP-IIl. NP-IIl has a strong absorption maximum at 377 nm and yields hydroxyl radicals upon UV light irradiation. The antioxidant activity of the ${\alpha}$ -lipoic acid and other naturally occurring compounds has been examined by using NP-IIl as a molecular probe. For example, upon photoirradiation of NP-lII with BSA or apolipoprotein of human low density (LDL), the significant oxidative modifications were observed in both cases. The oxidation was completely suppressed in the presence of ${\alpha}$-lipoic acid, which clearly demonstrates the strong hydroxyl radical scavenging activity of ${\alpha}$-lipoic acid.   • The author studied the histopathologic changes according to a single or a split dose and the time after irradiation on the acinar cells of rat parotid gland. 99 Sprague Dawley rats, weighing about l20gm, were divided into control and 3 experimental groups. In experimental groups, GroupⅠ and Ⅱ were delivered a single dose of l5Gy, 18Gy and Group Ⅲ and Ⅳ were delivered two equal split doses of 9Gy, 10.5Gy for a 4 hours interval, respectively. The experimental groups were delivered by a cobalt-60 teletherapy unit with a dose rate of 222cGy/min, source-skin distance of 50㎝, depth of l㎝ and a field size of l2×5㎝. The animals were sacrificed at 1, 2, 3, 6, 12 hours, 1, 3, 7 days after irradiation and examined by light and electron microscopy. The results were as follows: 1. As the radiation dose increased and the acinar cells delivered a single dose exposure were more damaged, and the change of acinar cells appeared faster than those of a split dose exposure. 2. The histopathologic change of acinar cells appeared at 1 hour after irradiation. The recovery from damaged acinar cells appeared at 1 day after irradiation and there was a tendency that the recovery from damage of a split dose exposure was somewhat later than that of a single dose exposure. 3. Light microscope showed atrophic change of acinar cells and nucleus, degeneration and vesicle formation of cytoplasm, widening of intercellular space and interlobular space. 4. Electron microscope showed loss of nuclear membrane, degeneration of nucleus and nucleoli, clumping of cytoplasm, widening and degeneration of rough endoplasmic reticulum, loss of cristae of mitochondria, lysosome, autophagosome and lipid droplet. 5. Electron microscopically, the change of rough endoplasmic reticulum was the most prominent and this appeared at 1 hour after irradiation as early changes of acinar cells. The nuclear change appeared at 2 hours after irradiation and the loss of cristae of mitochondria was observed at 2 hours after irradiation in all experimental groups.

단일 및 분할 방사선조사에 의한 백서절치의 급성반응에 관한 연구 (ACUTE RESPONSE OF THE RAT INCISOR BY SINGLE AND FRACTIONATED IRRADIATION)

  • Six to eight-month-old female albino rats were used as experimental animals. As an irradiation equipment, a Co-60 was used. The experimental animals were divided to; 6 of the control group, 12 of the 500cGy single irradiation group, 12 of the 1000cGy fractionated irradiation group, and 12 of the 1500cGy fractionated irradiation group. From the first week to the forth, 3 rats were picked from each group every week to be sacrificed and fixed with formalin. Those rats were observed by means of H-E stain after being taken radiograph and decalcified. The analysis of radiographic findings and light microscopic findings gives results as follows: 1. The delay of dental eruption rate was found in every group which underwent the irradiation experiment. Dentin niche, osteodentin, and dentin island were formed in the parts which were damaged by the irradiation. 2. The longer the observation period was, the more deposit of osteodentin and dentin island was formed. 3. In the single irradiation group, the damage effect was in proportion to the increase of radiation dose, whereas the damage was much less in the fractionated group receiving the same dose. 4. The 500cGy single irradiation group got temporary repairable damage, while the 1000cGy single irradiation group got considerable damage and showed much slower eruption rate than the 500cGy single irradiation group. The basal portion of the 1500cGy single irradiation group, whose growth was arrested, was destroyed. 5. The fractionated group were irradiated 500cGy everyweek. Repair was visible during the interval periods. The damage was accumulated as irradiation repeated, but degree of damage was lower than that of the 1000cGy and 1500cGy single irradiation group.

소형동물 전임상실험을 위한 하이퍼스펙트럼 영상장비 연구 (Research about Hyperspectral Imaging System for Pre-Clinical testing of Small Animal)

  • In this study we have developed a hyperspectrum imaging system for highly sensitive and effective imaging analysis. An optical setup was designed using acoustic optical tunable filter (AOTF) for high sensitive hyperspectrum imaging. Light emitted by mercury lamp gets split in to diffracted and undiffracted beams while passing though AOTF. GFP transfected HEK-293 cell line was used as a model for in vitro imaging analysis. Cells were first, analyzed by fluorescence microscope followed by flow cytometric analysis. Flow cytometric analysis showed 66.31% transfection yield in GFP transfected HEK-293 cells. Various images of GFP transfected HEK-293 cell were grabbed by collecting the diffracted light using a CCD over a dynamic range of frequency of 129-171 MHz with an interval of 3 MHz. Subsequently, for in vivo image analysis of GFP transfected cells in mouse, a whole-body-imaging system was constructed. The blue light of 488 nm wavelength was obtained from a Xenon arc lamp using an appropriate filter and transmitted through an optical cable to a ring illuminator. To check the efficacy of the newly developed whole-body-imaging system, a comparative imaging analysis was performed on a normal mouse in presence and absence of Xenon arc irradiation. The developed hyperspectrum imaging analysis with AOTF showed the highest intensity of green fluorescent protein at 153 MHz of frequency and 494 nm of wavelength. However, the fluorescence intensity remained same as that of the background below 138 MHz (475 nm) and above 162 MHz (532 nm). The mouse images captured using the constructed whole-body-imaging system appeared monochromatic in absence of Xenon arc irradiation and blue when irradiated with Xenon arc lamp. Nevertheless, in either case mouse images appeared clearly.

방사선조사가 설의 개방성창상치유에서 섬유아세포의 재형성에 미치는 영향에 관한 연구 (ULTRASTRUCTURAL STUDY OF THE FIBROBLAST REMODELING IN THE RADIATION- IMPAIRED WOUND HEALING IN THE TONGUE OF THE RAT)

  • Radiation-impaired wound healing in animal experiments was believed to be an another logical experimental model to understand the wound healing mechanism in patients. The purpose of this study was to reveal the block point which would result in impaired healing. Twenty four rats(Sprague-Dawley strains) were divided into two groups according to the time interval between irradiation and wounding. Group I, observing the healing effect on the 1st day and Group II are the healing effects on the 7th days after irradiation to the wound of the rat tongue. Experimental animals were sacrificed 3, 6, 12, and 24 hours after wounding. The specimens were examined by the light microscope and transmission electron microscope. The following results were obtained 1. Fibroblasts in both groups showed degenerative changes which were dilated mitochondria and rER, reduced microorganelle, vacuoles and little cytoplasmic process. 2. Average length between bands and Quantity of the newly produced collagen fibers around fibroblasts remained unchanged against control group. 3. The severity of degenerative change of the fibroblast and impairment of wound healing including shortening of the thickness of collagen fibers were more severe in the group II than in the group I.
