In many plant cells, the positions of chloroplasts change in response to changes in light conditions. In the epidermal cells of the aquatic angiosperm Vallisneria gigantea, the avoidance response of chloroplasts is induced specifically by irradiation with blue light of high intensity. Possible roles of actin cytoskeleton in the blue-light-induced avoidance response of chloroplasts were investigated by partial irradiation and phalloidin staining. We showed that the blue-light-dependent redistribution of chloroplasts was induced only in the limited area, where exposed to blue light, even in individual cells. In addition. in the exposed area, the configuration of actin filaments strikingly changed compared with that before the irradiation. Short and thick bundles of actin filaments surrounding the chloroplasts changed to much longer and thinner bundles with a more stretched array. In contrast, in the unexposed area, neither the distribution of chloroplasts nor the configuration of actin filaments exhibited any changes. Cytochalasin D and latrunculin B inhibited the avoidance response of chloroplasts concomitantly with the fragmentation of actin filaments. These results indicate that the reorganization of actin filaments plays a crucial role in the induction of avoidance response of chloroplasts.
Effects of blue and red light on photorespiration in the leaf disks of pea were studied. The rate of total 14CO2 fixation was more or less higher under red light than blue light irradiation of the same quantum (94.8 $\mu$Em-2.S-1/mV). The release of 14CO2 by photorespiration was more stimulated under blue than red light. Among the photorespiratory intermediates, 14C was more incorporated ito serine under blue light than red light. However, 14C was more incorporated into glycine under red light than blue light. The incorporation of 14C into glycolate was very low under both light qualities, but higher under red light than blue light. Among the enzymes related to photorespiration, only glycolate oxidase was activated and/or synthesized by blue light irradiation. Moreover, more 14C2 was released from glycoate-1-14C under blue light than red light irradiation, but 14C2 release from glyoxylate-1-14C and glycine-1-14C showed no difference by the either light qualities. These results suggest that blue light is more effective in the photorespiratory CO2 evolution than red light. The reason is considered that glycolate is easily metabolized under blue light due to the stimulation of the glycolate oxidase activity.
식품 저장 중 효소적 갈변을 일으키는 타이로시네이스의 활성을 저해시킬 수 있는 LED의 파장을 구명하기 위하여 파란색, 초록색, 빨간색 LED와 형광등을 이용하여 타이로시네이스에 7일동안 조사하였다. 또한 강도에 따른 타이로시네이스의 활성을 조사하기 위하여 파란색, 초록색, 빨간색 LED를 선택하여 빛의 강도를 3단계로 구분하여 사용하였다. 파장에 따른 타이로시네이스의 활성 저해율은 형광등에서 가장 컸으며 그 다음으로 파란색 LED에서 높은 저해율을 보였다. 빨간색 LED에서는 대조구와 유사한 저해율을 보였으며 초록색 LED에서 저해율이 가장 낮았다. 상대적 빛의 강도에 따른 타이로시네이스 활성은 초록색과 빨간색 LED 조사 시 효소활성의 변화가 크지 않았고, 반면 파란색 LED에서 강도가 증가할수록 큰 저해효과를 보였다. 이러한 결과를 토대로 볼 때 여러 가지 파장의 LED 중 고강도의 파란색 LED에서 타이로시네이스에 의한 식품의 갈변을 최소화 할 수 있을 것으로 판단되며 이는 갈변 억제를 유도함으로써 과일 및 채소의 저장 중 품질을 향상시킬 수 있을 것이다.
It failed occasionally to take a reasonable emergence rate since tobacco (Nicotiana tabacum L.) seeds were planted on late Jan. showing lowest temperature. This experiment was done to measure the effect of GA3 (concentration ; period) , light quality (red ; white ; dark) during or after its treatment, daily irradiation hours( 0 ; 8 ; 12 ;16) and germination termperature (20 or 10 $^{\circ}C$ ocnstant ; 20/1$0^{\circ}C$ alternating) on the germination rate. Red and white light given during grmination showed no differences between the other daily irradiation hours except that 8 hours red light delayed germination although their 12 hours irradiation had the gratest rate. The rate was increased with increased concentration to GA3 0.01 mM or increased imbibition period to 3 days although the rate of cv. NC 82 was less than that of cv. Burley 21 in the case of dark imbibition of GA3 but daily 12 hours irradiation during germination. Light quality forced during GA3 imbibition eliminated such effect of GA3 shown in the darkness so that only light quality pretreatment and termperature during germinition were affected on the rate. The germination rate of thecultivars was decreased in the order of red, white light, darkness meaning that it was highly influenced by the light quality during GA3 treatment. Regardless of GA3 or light quality treatment,on the other hand, the rate was greater in 20 $^{\circ}C$ constat than 1$0^{\circ}C$ constant and 20/1$0^{\circ}C$ alternating germination temperature having similar germinative patterns.
식물공장 내 적색광, 청색광, 적청 혼합광, 자외선, 적외선, 형광등의 광원을 달리하여 어린잎 상추의 생육과 무기성분 흡수를 검토하였다. 잎의 형태는 Red 파장에서 초장 및 하배축의 길이신장이 촉진되어 도장하였고 Far red에서는 생장이 불가능하였으나 Red + Blue의 혼합광원에서는 초장이 짧고 엽수가 많고 생체중이 증가하여 상추의 형태 및 발달 차원에서 유의적으로 좋았다. 광질에 따른 어린잎 상추의 색차 및 상대 엽록소 함량을 조사한 결과, 적색도를 나타내는 Hunter $a^*$ 값은 Red + Blue 혼합광 및 형광등에서 높았고 적색광 및 자외선에서는 낮게 나타났는데 상대 엽록소 함량을 나타내는 SPAD도 같은 경향이었다. 특히 상대엽록소 함량은 형광등의 10.5에 비해 혼합광에서 1.8배 향상된 적색도를 나타내었다. 광원별 상추의 무기물 함량을 조사한 결과, pH 및 K 함량은 모든 처리에서 차이가 없었으나 N, Ca, Mg, Mn, Fe, Ascorbic acid 함량은 LED 처리구에서 많았고 P 및 Mn 함량은 형광등 처리구에서 많았다. 이상의 결과를 요약하면 단색광에 비하여 Red + Blue 혼합광에서 상추의 생육이 우수하고 무기물 함량이 증대되어 식물공장 내 생산성 향상을 위해서는 혼합광 조절로 상품성 있는 고품질의 상추 생산이 가능 할 것으로 생각되었다.
본 연구에서는 비정상조직(파라핀)을 가진 생체조직에 광을 조사하고 그에 따른 조직의 표면온도와 비정상조직 주위에서의 온도분포를 실험과 해석적 방법을 통해 분석하였다. 파라핀을 이용하여 비정상 조직을 모사한 후 조사하는 광의 파장과 시간을 변화시키면서 조직 주위에서의 온도를 K형 열전대를 사용하여 측정하였다. 또한 전산열전달 기법을 이용하여 해석적으로 조직에 대한 온도분포를 예측하였다. 정상조직과 비정상조직의 주위에서의 온도는 차이가 있었으며, 비정상조직이 있는 경우 표면과 조직 주위의 온도가 높게 나타났다.
We investigated the generation of high pretilt angle for nematic liquid crystal (NLC) in the cell with slanted non-polarized ultraviolet (UV) light irradiation on two kinds of the polyimide (PI) film. It was shown that the monodomain alignment in NLC is obtained in the cell with slanted non-polarized UV light irradiation on PI surface. The pretilt angle of NLC is generated about 3 degrees in the cell with slanted non-polarized UV light irradiation with 70 degrees on PI surface without side chain. But, the pretilt angle of NLC is generated about 1 degree in the cell with slanted non-polarized UV light irradiation with 80 degrees on PI surface with side chain. We consider that the pretilt angle generation in NLC is attributted to anisotropic dispersion force between the LC molecular and the PI surface.
One of the leading causes of night time automobile accidents is the darkness of surroundings. Headlamps play a critical role in casting light and providing drivers with visibility. Headlamp design and new technology have been developed recently as research has been actively carried out to increase headlamp recognition. This study statistically analyzes irradiation distance using computer simulation by categorizing headlamps applied in domestic automobiles in the last decade by year, light source, form, vehicle type, and height of installation. After analyzing results of irradiation distance, it appears irradiation distance has been increased by approximately 10m in the last decade. This increase in irradiation distance is predicted to decrease night time accidents by allowing more time to recognize potential causes of accidents.
Photobiomodulation (PBM) using organic light emitting diodes (OLEDs) surface light sources have recently been claimed to be the next generation of PBM light sources. However, the differences between light emitting diodes (LEDs) and OLED mechanisms in vitro and in vivo have not been well studied. In vivo mouse models were used to investigate the effects of OLED irradiation on cellular function and cutaneous wound healing compared to LED irradiation. Mice in the LED- and OLED-irradiated groups were subjected to irradiation with 6 J/㎠ LED and OLED (630 nm), respectively, for 14 days after wounding, and some mice were sacrificed for the experiments on days 3, 7, 10, and 14. To evaluate wound healing, we performed hematoxylin-eosin and Masson's trichrome staining and quantified collagen density by computerized image analysis. The results showed that the size of the wound, collagen density, neo-epidermis thickness, number of new blood vessels, and number of fibroblasts and neutrophils was significantly influenced by LED and OLED irradiation. The tissue levels of interleukin (IL)-β, IL-6 and tumor necrosis factor (TNF)-α were investigated by immunohistochemical staining. LED and OLED irradiation resulted in a significant increase in the tissue IL-β and IL-6 levels at the early stage of wound healing (P < 0.01), and a decrease in the tissue TNF-α level at all stages of wound healing (P < 0.05), compared to the no-treatment group. The expression levels of the genes encoding vascular endothelial growth factor and transforming growth factor-beta 1 were significantly increased in LED and OLED-irradiated wound tissue at the early stage of wound healing (P < 0.01) compared to the no-treatment group. Thus, OLED as well as LED irradiation accelerated wound healing by modulating the synthesis of anti-inflammatory cytokines and the expression levels of genes encoding growth factors, promoting collagen regeneration and reducing scarring. In conclusion, this suggests the possibility of OLED as a new light source to overcome the limitations of existing PBMs.
BACKGROUND: Photoreversibility, a reversion of the inductive effect of a brief red light pulse by a subsequent far red light pulse, is a property of photo responses regulated by the plant photoreceptor phytochrome B. Plants use photoreceptors to sense photo signal and to adapt and modify their morphological and physiological properties. Phytochrome recognizes red light and far red light and plays an important role in regulating plant growth and development. METHODS AND RESULTS: The reversal responses of growth and fruiting characteristics were investigated to increase the yield of oriental melon (Cucumis Melo L. var. Kumsargakieuncheon) by means of controlling light quality in a plastic house. Red (R:660nm) and far red (FR:730nm) lights were subsequently irradiated on the whole stems and leaves of the oriental melon plant during growing periods, using red and far red LEDs as light sources, from 9:00 PM daily for 15 minutes. The intensities of R and FR light were 0.322-0.430 μmol m-2s-1 and 0.250-0.366 μmol m-2s-1, respectively. Compared to R light irradiation, combination of R and FR light irradiation increased the length of internode, number of axillary stems, number of female flowers, and fruit number of oriental melons. The results of treatment with R were similar to R-FR-R light irradiation in terms of length of internode, number of axillary stems, number of female flowers, and number of fruits. When FR treatment was considered, R-FR and R-FR-R-FR light irradiation had similarities in responses. These reversal responses revealed that oriental melon showed a photoreversibility of growth characteristics, flowering, and fruiting. CONCLUSION: These results suggested the possibility of phytochrome regulation of female flower formation and fruiting in oriental melon. The fruit weight of the oriental melon was the heaviest with the R light irradiation, while the number of fruits was the highest with the FR light. With the FR light irradiation, the fruit weight was not significantly higher compared to that of the control. Meanwhile, the yield of oriental melon fruits increased by 28-36% according to the intensities of the FR light due to the increases of the number of fruits.
