Effects of Environmental Factors on the Growth of Gametophytes and Young Sporophytes of Eisenia bicyclis (Kjellman) Setchell (대황(Eisenia bicyclis) 배우체와 아포체의 생장에 미치는 환경 인자의 영향)

  • Eisenia bicyclis, a perennial macroalga is a primary producer of in the ocean, It has been identified as a key species that plays a vital role in maintaining the ecosystem stability. Also, it is an important target in marine afforestation projects and useful marine organisms. In addition, E. bicyclis is used as a health food for humans. This study investigated the effect of water temperature, light (photon irradiance), and duration of light (photoperiod) on the growth of gametophytes and young sporophytes of E. bicyclis. The germination and growth of the zoospores of E. bicyclis were examined at five temperatures (5℃, 10℃, 15℃, 20℃ and 25℃), four intensities of photon irradiance (10, 20, 40, and 80 μmol m-2s-1), and photoperiods (14:10 and 10:14 light/dark cycles). The zoospores released from mature plant germinated into the gametophytes under all experimental conditions. The gametophytes were able to grow at water temperature 5℃-25℃ and mature at 10℃-20℃. The optimal range of water temperature for the maturation of the gametophyte was 15℃-20℃. At 25℃, E. bicyclis gametophytes grew rapidly but did not mature. The optimal culture conditions for the growth of young sporophytes grew slowly in low temperature and photon irradiances.

Optimal conditions for adventitious root organogenesis from peony root explant callus cultures (작약(Paeonia lactiflora Pall.) 뿌리절편 유래 캘러스 배양으로부터 부정근발생을 위한 최적 배양조건)

  • The optimal culture conditions for root organogenesis from the callus of peonies (Paeonia lactiflora Pall.) were investigated. Root explants with vascular bundles were cultured in Murashige and Skoog (MS) medium combined with 0.5-4.0 mg/L auxins (indole acetic acid [IAA], naphthalene acetic acid [NAA], indolebutyric acid [IBA], and 2,4-dichlorophenoxyacetic acid [2,4-D]) and 0.0-2.0 mg/L cytokinins (kinetin, zeatin, and benzylaminopurine [BAP]) to induce callus formation. The callus was then cultured in MS medium combined with three concentrations (0.1, 0.5, and 1.0 mg/L) of IAA, NAA, IBA, kinetin, zeatin, and BAP in the dark for 6 weeks. Based on the results, the effects of dark and light conditions on the callus cultured in MS medium with combinations of 0.1-1.0 mg/L IBA and zeatin for 6 weeks were studied. Callus formation was most effective (>+++) in the medium with a combination of 1.0 mg/L NAA and 1.0 mg/L zeatin. A high number of long adventitious roots were observed in the mediums with 0.1 mg/L IBA (6.66 and 4.82 cm) and 0.5 mg/L zeatin (2.32 and 0.72 cm) among auxins and cytokinins, respectively. The highest number (14.06) of adventitious roots were formed from the callus cultured in light in the MS medium combined with 0.1 mg/L IBA and 0.5 mg/L zeatin. This same medium induced the formation of the longest adventitious root (5.45 cm) in the dark. Thus, optimization of in vitro culture conditions may be possible for the mass propagation of adventitious roots in peonies.

  • We investigated the interaction of serotonin and galanin (GA) by a double immunostaining method in the Japanese quail. Serotonin-immunoreactive (IR) cells were located in the paraventricular organ (PVO) and infundibular nucleus (IF). The number of the cells under short-day photoperiod (SD) was less in the dark phase than in the light phase. GA-IR cells were found in the PVO, IF and median eminence. The number of GA-IR cells in SD was significantly greater than that in long-day photoperiod (LD). Numerous GA- IR varicose fibers ran along serotonin- IR cell bodies and nerve fibers in the PVO and IF of the same sections. Very few serotonin-IR fibers ran along GA-IR cell bodies and GA-IR nerve fibers in the ventral part of the IF. The present results suggest that the possibility of functional interaction takes place between serotonin- and GA- IR neurons in the PVO and IF.

Effect of Explant Types, Auxin Concentration and Light Condition on In Vitro Root Production and Alkaloid Content of Rauvolfia serpentina (L.) Benth. ex Kurz

  • Rauvolfia serpentina (L.) Benth. ex Kurz is a medicinal plant and an endangered tropical rainforest plant species. Since the field cultivation that aims to fulfill the industrial needs is never accomplished, tissue culture appears to be the most feasible way to improve the quality and quantity of R. serpentina. This experiment used two kinds of explants (roots and shoots) to induce optimal root formation in different combinations of auxin and photoperiod. Each explants exhibited different responses on given treatments. Differentiated root could be produced from explants cultured in IBA 20 mg/L with and without light. The highest number of roots, root length and root weight induced from shoot explants were effective on MS medium containing IBA 20 mg/L and incubated under dark condition, while highest total weight (callus and root) from root explants cultured on MS medium supplemented 10 mg/L IBA and 10 mg/L NAA and incubated under day length (11/13 hr). The root induced from shoot explants produced the highest major alkaloid content. The highest content of ajmaline (2.17 ppm fresh weight) and reserpine (1.30 ppm fresh weight) were observed in shoot explants cultured in MS medium containing combination of IBA 10 mg/L and NAA 10 mg/L and incubated under dark condition, yohimbine (1.47 ppm fresh weight) was in the shoot explants cultured in MS medium containing NAA 20 mg/L and incubated under day length, while serpentine was absent.

Effect of Scrapping Aerial Mycelia and Light on the Production of Macroconidia and Chlamydospores of Cylindrocarpon destructans Causing Root Rot of Panax ginseng (기중균사 제거와 광처리가 인삼 뿌리썩음병균 Cylindrocarpon destructans의 대형분생포자 및 후막포자 생성에 미치는 영향)

  • Under the light condition of 25,000 Lux (12 hrs dark and light cycle) with scrapping treatment of aerial mycelia of Cylindrocarpon destructans on potato dextrose agar (PDA), V-8 juice agar, and ginseng extract agar, production of the macroconidia was increased to $3.7\~8.1$ fold over them produced in the dark. They were also produced $7.7\~18.0$ times more in the liquid cultures under the light condition than under the dark as well. PDA and V-8 juice agar among the tested were the best for the macroconidium production. On PDA, 1,585 $macroconidia/mm^2$ were produced under the light of 25,000 Lux with scrapping treatment of aerial mycelia of C. destructans, which is 3.2 and 1.4 times more than those produced under 3,000 and 10,000 Lux, respectively. Meanwhile, $20\~99$ macroconidia/$mm^2$ were produced by the non-scrapping under the light condition between 3,000 Lux and 25,000 Lux. The macroconidia were, however, lysed at $6\~7$ days after being incubated under the above range of the light. They were consisted of $1\~3$ cells in a macroconidium while $69.4\~100\%$ of them were the two-celled and the number did not seem to be affected by either the scrapping or the light. Production of chlamydospore converted from mycelia of C. destructans seemed to be promoted by the light and the scrapping as well. The 1,285 chlamydospres/$mm^2$ were produced with the light (25,000 Lux), which is 2.8 and 1.2 times more than those with 3,000 and 10,000 Lux, respectively. Scrapping the aerial mycelia of the cultures increased the chlamydospore formation to 1.9, 2.5 and 1.4 times more than the non-scrapping under the light intensity of 3,000 Lux, 10,000 Lux, and 25,000 Lux, respectively. On PDA, 1 to 8 chlamydospore(s) per catena were formed by all treatments tested and $34.2\~58.9\%$ of them was a single chlamydospore, However, the numbers was affected by neither the light ($3,000\~25,000$ Lux) nor the scrapping the aerial mycelia.

  • The induction of calli from tissues of a grape, Vitis hybrid, and their suspension cultures were performed and various factors were investigated on cell growth and anthocyanin production. It was shown that light intensity and inorganic nitrogen concentration played an important role on anthocyanin production.1:he contents of anthocyanin produced under 10,000 Iux light irradiation were about twice as much as under the dark. The reduction of inorganic nitrogen concentration of MS medium to one to twenty brought about the increase of approximately five to six-fold in total anthocyanin or sixteenfold in anthocyanin content per dry cell weight and addition of nitrate only as inorganic nitrogen source was shown to be the best for anthocyanin production. Miller medium and Gamborg medium were suitable for the anthocyanin production, as well as high concentrations of Co$^{2+}$ and Fe$^{2+}$. And high yield of 40mg anthocyanins per 200m1 flask was obtained by two stage culture using MS medium for the first stage and the modified MS medium for the second stage.

Growth and changes in the biochemical composition of Isochrysis galbana under different light-emitting diode conditions

  • The marine microalgae Isochrysis galbana was cultured under various light-emitting diode (LED) light conditions with different wavelengths to examine changes in growth and in amino acid and fatty acid profiles. The culture conditions for the microalgae were Conway medium, salinity of 33 psu, temperature of $24^{\circ}C$, and a 16/8 h light/dark photoperiod. Six light sources, including 5 units of 180W LED lamps (peak wavelength: blue [LB] 470 nm; green [LG] 525 nm; yellow [LY] 595 nm; red [LR] 636 nm; white [LW] 442 nm) and 1 unit of a 175W metal halide (MH) lamp, were used for the experiment. The dry cell weights ($gL^{-1}$) of I. galbana under different light conditions were in the order of LW>LB${\geq}$MH>LR>LG>LY. Levels of essential amino acids were revealed to be significantly higher under LW, LG, and MH than under the other wavelengths (P<0.05). The fatty acid, unsaturated fatty acid, and DHA contents of I. galbana were higher under MH, LW, and LG. In addition, the carotenoid content was higher under MH, LW, and LG than under the other wavelengths (P<0.05). The fucoxanthin content was highest under MH (0.28%) and lowest under LY (0.2%), and it was 0.26% under LW and LG. The results indicate that the combined use of LW and LG is effective when using LED lamps for I. galbana cultivation.

  • The purpose of this research is to find out the effect of a chromatic Korean jacket and an achromatic Korean skirt on the visual image of a traditional Korean dress wearer. This experiment was designed to the 3 factors which were composed of 3 colors(red, yellow, green), 4 jacket tones(vivid, light, dull, dark), and 4 skirt colors(N9, N7, N4, N2). The 288 students in Gyeongsang National University assessed the 48 stimuli which wear variously incorporated with colors and tones by a semantic differential scale of 7-point: The results of the study were as follows. Image factors of stimuli are youth and activity, gracefulness, visibility, and tenderness. Among these, youth and activity factors, and gracefulness factors proved to be important. The colors and tones of a jacket and a skirt used in this experiment showed that the colors and tone had an effect on dimension of 4 images by interacting of two valuables or working independently. While a yellow jacket with vivid and a white(N9) skirt had a significant effect on youth and activity factors, a red jacket and a dark- gray(N4), -black(N2) skirt were evaluated as a graceful image. In the red jacket case, a visible factor was considerably different according to skirt tones. Only jacket tones worked as a striking clue in a tenderness factor.

  • In this research, I compared and analyzed the 'factors of style' that expresses and visualizes the 'characteristics of characters' and the characters from three movies each that was filmed with Kang Ho Song and Keong Ku Seol who acted in many movies both. As the result, for the factors for actors' makeup design, since they have limited choices of textures, colors, and the range of changes, changing the tone of skin to light to dark implied the images of characteristics. Somewhat dark skin tone that was like tanned skin expressed the life style of the character who works for an active profession not for an intellectual position. For the factors of hair style, medium sized wavy hair that goes straight down forehead expressed the character's familiar and informal personality while short cut sized all back style hair expressed confidence, sociality, and logical personality of the character. The actors' costume was important to express the characteristics of characters such as changes of their mind, especially the costumes delivered symbolic meanings of the role of their social class, profession, and their financial state. In addition, there were common factors to create and design a character in terms of the changes of the actors' body shape; when the actor acted the character who has conflicts inside and sarcastic personality, they lost weight on purpose while they gained weight and made a barrel shaped body to act the character who is positive in every occasion and does not realize the real world or who is greed.

Effect of Plant Growth Regulators on Bulblet Formation and Plant Regeneration in Fritillaria thunbergii Miq. (패모조직배양에서 생장조절 물질이 자구형성 및 식물체 재생에 미치는 영향)

  • To improve the efficiency of mass propagation in vitro of Fritillaria thunbergii, bulb scale and nodes were cultured in LS medium supplemented with the combination of 2, 4-D and kinetin or NAA and BA. The number and size of bulb, the number of adventitious shoot, the ratio of callus formation, rooting, and the effects of light and dark on the culture, plant regeneration from calli, and the gelling substances were investigated. The combination of 2, 4-D and kinetin in media was more effective than the media of NAA and BA for the bulblet formation. The media supplemented with 2 mg/L 2, 4-D and 1 mg/L kinetin, $1{\sim}2\;mg/L$ 2, 4-D without kinetin and $1{\sim}3\;mg/L$ BAA only were effective in the adventitious shoot development. Callus formation and root formation, respectively were effective in the medium supplemented with 2mg/L 2, 4-D and 1mg/L kinetin. In bulb formation, the medium with 5 mg/L kinetin was effective, and the most of bulbs were formed from the axillary bud of node part. In bulb formation, shoot growth, callus and root formation, the light culture for 16 hours per day was better than that in the dark culture. Bulb was nicely formed in the medium with 0. 2 mg/L 2, 4-D, 1 mg/L kinetin. The medium without hormone was most effective for plant regeneration. The phytagel was more effective than agar in the medium as the gelling agent.
