기아를 일으킨 흰쥐의 생명연장에 미치는 천연약물의 효과 (Effect of Natural Drugs on the Life Support of Starving Rats)

  • The current study was undertaken to select the crude drug showing life prolonging effect on the starved rats(Sprague-Dawley) and elucidate the reaction mechanism there of. As distilled water extracts prepared from twenty kinds of natural drugs were administered orally to female starved rats, Bupleuri Radix demonstrated the most prominent effect with 34.6% increment in the survival time. To investigate underlying mechanism of life prolonging effect of Bupleuri Radix, the concentrations of adrenocorticosteroids(corticsterone, testosterone and aldosterone) and enzyme activities of superoxide dismutase(SOD), glutamate oxaloacetate transaminase(GOT), glutamate pyrurate transaminase(GPT) were assayed in the serums of rats starved for, 2, 4 and 6 days respectively. The results manifested much elevated values of corticosterone, aldosterone(2 days) and rather decreasing tendency afterwards, specially in the late periods of starvation(6 days). With respect to such a considerable changes according to starvation periods, Bupleur Radix restored these values much near to normal suggesting that Buplerum Radix may play a life prolonging action during starvation probably through reinforcing the homeostatic properties of corticosteroids and some enzyme activities.

이끼 추출물과 절화 보존제 혼용처리가 절화장미 'Enjoy'의 수명에 미치는 영향 (Effect of Moss (Hypnum jutlandicum) Extracts and Mixing Treatments with Preservatives on Vase Life of Rosa hybrida 'Enjoy')

  • 본 연구는 이끼의 추출물을 이용하여 절화장미 'Enjoy'의 수명을 효과적으로 연장시킬 수 있는 천연 절화수명 연장제를 개발하기 위하여 수행하였다. 이끼 추출물중 R. hybrida 'Enjoy'의 절화수명 연장에 가장 효과적이었던 Hypnum jutlandicum 2 $mg{\cdot}L^{-1}$에 sucrose 0, 1, 3, 5%를 첨가한 후 sucrose가 R. hybrida 'Enjoy'의 수명에 미치는 영향을 분석하였다. 연구의 결과, sucrose를 H. jutlandicum 추출물과 함께 사용할 경우에는 R. hybrida 'Enjoy'의 잎이 엽록소를 상실하여 탈색되고 위조현상이 발생하며, 오히려 절화의 수명이 단축되는 것으로 나타났다. 그러나 H. jutlandicum 추출물과 HQS를 함께 사용하는 것은 R. hybrida 'Enjoy'의 수명을 증가시키는 것으로 나타났으며, 특히 HQS를 100 $mg{\cdot}L^{-1}$ 농도로 첨가했을 때 R. hybrida 'Enjoy'의 수명연장 효과가 가장 증가하는 것으로 나타났다. H. jutlandicum 추출물 2 $mg{\cdot}L^{-1}$ 첨가구의 pH를 조절하지 않거나(pH 3.8), pH 3.0, 4.0, 5.0로 조절하여 R. hybrida 'Enjoy'의 절화수명에 미치는 영향을 분석한 결과, pH를 조절하지 않은 pH 3.8인 보존용액을 사용하는 것이 R. hybrida 'Enjoy'의 수명이 오래감을 알 수 있었다. 이상의 연구 결과, H. jutlandicum 추출물(2 $mg{\cdot}L^{-1}$)에 HQS(100 $mg{\cdot}L^{-1}$)를 첨가한 보존용액이 R. hybrida 'Enjoy'의 절화수명을 연장시켰으며, 이끼는 주변에서 쉽게 구할 수 있어 환경오염의 피해가 없으며 천연 보존제로서 개발가치가 있을 것으로 기대된다.

  • The present investigation was undertaken to find out the anticancer activity of monoterpene compounds. Monoterpenes showed generally the inhibitory effect on the proliferation o f L1210 cancer cells (cytotoxicity). Geraniol was found to exibit the most potent cytotoxic effect on L1210 cells with an IC50 values of $0.67{\mu}g/ml$. On the other hand, geraniol proved to be capable of stimulating the macrophage proliferation (135% of control). When the life prolonging activity of geraniol by daily oral administration of 0.1~10${\mu}g/10{\mu}l/20$ g body weight to Sarcoma 180 bearing ICR mouse was examined, there was also a significant elevation of survival (best result of 134% of control). The contradictory effects of geraniol on the proliferation of L1210 cells and macrophages proved to be accompanied by the coincident alterations of RNS (reactive nitrogen species) related enzymes activities such as inducible nitric oxide synthase (Inos) in macrophages and ROS (reactive oxygen species) related enzymes activities such as superoxide dismutase (SOD) in L1210 cells, respectively.

Effects of Temperature, Illumination, and Sodium Ascorbate on Browning of Green Tea Infusion

  • Browning of tea infusion is an obstructive factor influencing shelf life of ready-to-drink green tea. Effects of temperature and illumination on the browning of green tea infusion were investigated. It was shown that both elevated temperature and illumination led to the browning of green tea infusion, but temperature had greater effect on infusion color and level of catechins than illumination. The levels of unoxidized catechins such as (-)-epigallocatechin gallate (EGCg), (-)-epigallocatechin (EGC), (-)-epicatechin gallate (ECg), (-)-epicatechin (EC), and total catechins remaining in the tea infusion were significantly correlated to color parameters of the tea infusion. Sodium ascorbate inhibited the infusion browning by suppressing the oxidation of tea catechins and it is considered to be a more suitable preservative for prolonging shelf life of ready-to-drink green tea than ascorbic acid because it has less effect on tea taste. The effects of temperature and illumination on the epimerization of catechins were also discussed.

5-HT1A receptors mediate the analgesic effect of rosavin in a mouse model of oxaliplatin-induced peripheral neuropathic pain

  • Oxaliplatin, a third-generation platinum derivative, is the mainstay of current antineoplastic medications for advanced colorectal cancer therapy. However, peripheral neuropathic complications, especially cold allodynia, undermine the life-prolonging outcome of this anti-cancer agent. Rosavin, a phenylpropanoid derived originally from Rhodiola rosea, exhibits a wide range of therapeutic properties. The present study explored whether and how rosavin alleviates oxaliplatin-induced cold hypersensitivity in mice. In the acetone drop test, cold allodynia behavior was observed from days 3 to 5 after a single injection of oxaliplatin (6 mg/kg, i.p.). Cold allodynia was significantly attenuated following rosavin treatment (10 mg/kg, i.p.). Specific endogenous 5-HT depletion by three consecutive pretreatments with parachlorophenylalanine (150 mg/kg/day, i.p.) abolished the analgesic action of rosavin; this effect was not observed following pretreatment with naloxone (opioid receptor antagonist, 10 mg/kg, i.p.). Furthermore, 5-HT1A receptor antagonist WAY-100635 (0.16 mg/kg, i.p.), but not 5-HT3 receptor antagonist MDL-72222 (1 mg/kg, i.p.), blocked rosavin-induced analgesia. These results suggest that rosavin may provide a novel approach to alleviate oxaliplatin-induced cold allodynia by recruiting the activity of 5-HT1A receptors.

Effect of Polylysine on Scrapie Prion Protein Propagation in Spleen during Asymptomatic Stage of Experimental Prion Disease in Mice

  • Prion diseases are incurable neurodegenerative disorders. Our previous study demonstrated that polylysine was effective in prolonging the incubation period in a rodent model and in alleviating the scrapie prion protein (PrPSc) burden in the brain at the terminal stage of the disease. Here, we report that intraperitoneal administration of polylysine suppresses the accumulation of prions in the spleen during the early stages of the disease. This study supports the congruence of PrPSc inhibition by polylysine in both the spleen and brain.

Experimental and numerical investigation on gas turbine blade with the application of thermal barrier coatings

  • The engine parts material used in gas turbines (GTs) should be resistant to high-temperature variations. Thermal barrier coatings (TBCs) for gas turbine blades are found to have a significant effect on prolonging the life cycle of turbine blades by providing additional heat resistance. This work is to study the performance of TBCs on the high-temperature environment of the turbine blades. It is understood that this coating will increase the lifecycles of blade parts and decrease maintainence and repair costs. Experiments were performed on the gas turbine blade to see the effect of TBCs in different combinations of materials through the air plasma method. Three-layered coatings using materials INCONEL 718 as base coating, NiCoCrAIY as middle coating, and La2Ce2O7 as the top coating was applied. Finite element analysis was performed using a two-dimensional method to optimize the suitable formulation of coatings on the blade. Temperature distributions for different combinations of coatings layers with different materials and thickness were studied. Additionally, three-dimensional thermal stress analysis was performed on the blade with a commercial code. Results on the effect of TBCs shows a significant improvement in thermal resistance compared to the uncoated gas turbine blade.

  • 젓새우(Acetes chinensis), 피둥어꼴뚜기 (Todarodes Pacifcus, 속칭 동해안산의 오징어), 젓꼴뚜기 (Loligo beka)를 1 Mrad까지의 각 성량의 ${\gamma}$선을 조사하여 이것들을 2~4$^{\circ}C$에서 저장하여 방사선에 의한 저장효과를 시험하였다. 원료와 저장중의 어체선도는 관능검사, 화학적 검사 (VRS, TMA-N량), 생화학적검사 (Hypo-xanthine 량) 및 세균학적검사 (생균수)에 의거하여 판정하였다. VRS 값은 3어종에 있어서 모두 관능검사치와 잘 부합되지는 않으나 TMA-N, Hypoxanthine 식 및 생균수는 관능검사치와 잘 부합되며, 각 어종의 선도판정치로서 이용할수 있다. 비조사 젓새우는 3일간의 저장이 가능하나 최적선량 0.12-0.20 Mrad에서 8일간까지 저장할 수 있고, 피둥어꼴뚜기는 8일간의 저장을 최적선량 0.20-0.30 Mrad 에서 14-21일간의 저장이 가능하였으며, 젓꼴뚜기는 5-6일의 저장기간이 최적선량 0.12-0.20 Mrad에서 약 10일간의 저장이 가능하였다.

어성초 추출물의 ICR생쥐와 L1210 세포에 대한 항암작용 및 SOD, GPx 효소활성변화 (Anticancer Effect of Houttuynia cordata Extract on Cancered ICR Mouse and L1210 Cells With Changes of SOD and GPx Activities)

  • The present investigation was undertaken to examine the anticancer activity of the methanol extract from Houttuynia cordata on ICR mouse with induced abdominal cancer and L1210 cancer cells. When the methanol extract of Houttuynia cordata (10∼200 $\mu\textrm{g}$/$m\ell$) was administered orally to ICR mouse with abdominal cancer, 47.8% of the best life prolonging effect was obtained. In case of cytotoxicity study (inhibition of cell proliferation) of Houttuynia cordata extract against L1210 cells, $IC_{50}$/ was found to be 62.8 $\mu\textrm{g}$/$m\ell$. In contrast to such considerable toxicity against cancer cell line, the toxicity demonstrated by the identical extract against normal lymphocytes was very meagre as shown to be < 5% compared with 86.5% in case of L1210 cells at the same condition. To get an insight into the reaction mechanism undelying the anticancer activity, $O_2$ion quantity and antioxidant enzyme activities such as superoxide dismiutase (SOD) and glutathione peroxidase (GPx) of L1210 cells in the presence of Houttuynia cordata extract were measured. The increased values of SOD and GPx enzyme activities in addition to the augmented generation of $O_2$ ion in L1210 cells implied that the reactive oxygen species induding $O_2$ion which were presumably induced by Houttuynia cordata extract might have participated in the process of L1210 cells cytotoxicity.

Effect of Modified Starches on Caking Inhibition in Ramen Soup

  • The effect of the addition of 2 kinds of chemically modified starches (the anti-caking agents; tapioca starch and com starch) on caking of ramen soup was observed using a low-resolution proton-pulsed nuclear magnetic resonance (NMR) technique. After storing ramen soup samples with diverse compositions of modified starch at 20-40% relative humidity for 4 weeks, changes in the spin-spin relaxation time constant ($T_2$) were measured as a function of temperature. $T_2-Temperature$ curves for ramen soup containing modified starches showed that the caking initiation temperature (glass transition temperature) was increased by $5^{\circ}C$ following the addition of only 0.5% modified cornstarch. The results indicate that the modified com starch used in this study would be an effective anti-caking agent for ramen soup, thus prolonging the shelf life of the product.
