In Vivo $^{13}C$-NMR Spectroscopic Study of Polyhydroxyalkanoic Acid Degradation Kinetics in Bacteria

  • Polyhydroxyalkanoic acid (PHA) inclusion bodies were analyzed in situ by $^{13}C$-nuclear magnetic resonance ($^{13}C$-NMR) spectroscopy. The PHA inclusion bodies studied were composed of poly(3-hydroxybutyrate) or poly(3hydroxybutyrate-co-4-hydroxybutyrate), which was accumulated in Hydrogenophaga pseudoflava, and medium-chain-length PHA (MCL-PHA), which was accumulated in Pseudomonas fluorescens BM07 from octanoic acid or 11-phenoxyundecanoic acid (11-POU). The quantification of the $^{13}C$-NMR signals was conducted against a standard compound, sodium 2,2-dimethyl-2-silapentane-5-sulfonate (DSS). The chemical shift values for the in vivo NMR spectral peaks agreed well with those for the corresponding purified PHA polymers. The intracellular degradation of the PHA inclusions by intracellular PHA depolymerase(s) was monitored by in vivo NMR spectroscopy and analyzed in terms of first-order reaction kinetics. The H. pseudoflava cells were washed for the degradation experiment, transferred to a degradation medium without a carbon source, but containing 1.0 g/l ammonium sulfate, and cultivated at $35^{\circ}C$ for 72 h. The in vivo NMR spectra were obtained at $70^{\circ}C$ for the short-chain-length PHA cells whereas the spectra for the aliphatic and aromatic MCL-PHA cells were obtained at $50^{\circ}C\;and\;80^{\circ}C$, respectively. For the H. pseudoflava cells, the in vivo NMR kinetics analysis of the PHA degradation resulted in a first-order degradation rate constant of 0.075/h ($r^{2}$=0.94) for the initial 24 h of degradation, which was close to the 0.050/h determined when using a gas chromatographic analysis of chloroform extracts of sulfuric acid/methanol reaction mixtures of dried whole cells. Accordingly, it is suggested that in vivo $^{13}C$-NMR spectroscopy is an important tool for studying intracellular PHA degradation in terms of kinetics.

Fc fusion to Glucagon-like peptide-1 inhibits degradation by human DPP-IV, increasing its half-life in serum and inducing a potent activity for human GLP-1 receptor activation

  • The short in vivo half-life of GLP-1 prevents it from being used clinically. This short half-life occurs because GLP-1 is rapidly degraded by dipeptidyl peptidases such as DPP-IV. To overcome this obstacle, a GLP-1/Fc was constructed and evaluated to determine if it was degraded by DPP-IV and in serum. When the degradation of GLP-1/Fc by human DPP-IV and rabbit serum was compared with that of GLP-1 it was found to be reduced by approximately 5- and 4-fold, respectively. Furthermore, GLP-1/Fc showed a potent activity for human GLP-1 receptor activation ($EC_{50}$ approximately 6 nM). Taken together, these results indicate that GLP-1/Fc may have an extended half-life in vivo that occurs as a result of inhibition of degradation by human DPP-IV. Due to the extended half life, GLP-1/Fc may be useful for clinical treatments.

  • Our studies diagnose insulation degradation using the method of computer sensing system, which has the advantages of PD(partial discharge) and AE(acoustic emission) sensing system. To use advantages of these two methods can be used effectively to search for treeing location and PD in some materials. In analysis method of degradation, using statically operator such as the center of gravity (G), the gradient of the discharge distribution(C), we have analyzed for the prediction of life which we can be obtained the time, occurred of many pulse of small discharge amplitude.

A practical approach to handling protein samples under degradation

  • Protein structure determination using NMR spectroscopy requires a suite of heteronuclear 3-D NMR experiments that can take a couple of weeks for completion. During the experiments, protein samples may suffer from slow degradation due to co-purifying proteases, which complicates and slows down the assignment procedure. Here we describe a practical protocol to avoid unwanted proteolysis during the experiment.

Thermal Degradation Kinetics of Antimicrobial Agent, Poly(hexamethylene guanidine) Phosphate

  • The thermal degradation of poly(hexamethylene guanidine) phosphate (PHMG) was studied by dynamic thermogravimetric analysis (TGA) and pyrolysis-GC/MS (p-GC). Thermal degradation of PHMG occurs in three different processes, such as dephosphorylation, sublimation/vaporization of amine compounds and decomposition/ recombination of hydrocarbon residues. The kinetic parameters of each stage were calculated from the Kissinger, Friedman and Flynn-Wall-Ozawa methods. The Chang method was also used for comparison study. To investigate the degradation mechanisms of the three different stages, the Coats-Redfern and the Phadnis-Deshpande methods were employed. The probable degradation mechanism for the first stage was a nucleation and growth mechanism, $A_n$ type. However, a power law and a diffusion mechanism, $D_n$ type, were operated for the second degradation stage, whereas a nucleation and growth mechanism, $A_n$ type, were operated again for the third degradation stage of PHMG. The theoretical weight loss against temperature curves, calculated by the estimated kinetic parameters, well fit the experimental data, thereby confirming the validity of the analysis method used in this work. The life-time predicted from the kinetic equation is a valuable guide for the thermal processing of PHMG.

A Study on the Physical Characteristics of the Low-voltage Circuit Breaker Based on the Accelerated Degradation Test (가속 열화 시험에 따른 저압용 차단기의 물리적 특성에 관한 연구)

  • This study analyzed the characteristics of insulation resistance and operating time based on an accelerated degradation test of a low-voltage circuit breaker. The experimental sample used a molded case circuit breaker (MCCB) and an earth leakage circuit breaker (ELCB). After measuring the insulation resistance of the circuit breakers, the leakage current was affected by an external rather than an internal structure. Furthermore, the insulation resistance of the circuit breakers with accelerated degradation was measured using a Megger insulation tester. In the accelerated degradation test, aging times of five, ten, 15, and 20 years were applied according to a temperature derived using the Arrhenius equation. Circuit breakers with an equivalent life of ten, 15, and 20 years had increased insulation resistance compared to those with less degradation time. In particular, the circuit breaker with an equivalent life of ten years had the highest insulation resistance. Component analysis of the circuit breaker manufactured through an accelerated degradation test confirmed that the timing of the increase in insulation resistance and the time of additive loss were the same. Finally, after analyzing the operating time of the circuit breakers with degradation, it was confirmed that the MCCB did not change, but the ELCB breaker failed.

  • In recents years, the statistical properties has become an important quantity for reliability based design of a component. The effects of the materials and test conditions for parameter estimation in residual strength degradation model are studied in carbon/epoxy laminate. It is shown that the correlation between the experimental results and the theoretical prediction on the fatigue life distribution using the life distribution convergence method is very reasonable.

  • Electrical equipment for railway is always experiencing wear and degradation by mechanical, electrical and environmental stress in service and the fault or the accident of high voltage main circuit directly causes operation interruption. Particularly propulsion drive of high speed switching inverter takes the form of specific degradation mechanism such as fast rising transient surge, reflective overvoltage and harmonic stress, and it is known that it threatens the long life and the reliability of electrical equipment. In this paper, statistics of fault and accident on main electrical equipment for railway are presented and also insulation degradation mechanism, which governs end life of electrical device, is analyzed. Finally the method of fault respondence and reliability improvement on the main electrical equipments will be reviewed in order to prevent operation interruption.

  • In this paper, we observed degradation characteristics of luminous flux for new light source. Because degradation tests can be a useful tool for assessing the reliability when few or even no failures are expected in a life tests. And we use a simple random coefficient degradation model to induce most suitable equation of degradation. As a result, exponential distribution and equation is best suitable model for new light source.

  • The life of elevator wire ropes is one of the most important characteristics of an elevator, which is closely related to the safety of users and its maintenance policy. It is not cost effective to measure the lifetime of elevator wire ropes during their use. In this study, the life estimation of elevator wire ropes (8x19W-IWRC) is considered using accelerated degradation test data. A bending fatigue tester is used to perform the accelerated degradation tests, incorporating the acceleration factor of tensile force. Assuming that the life of wire ropes is log-normally distributed, two life estimation methods are suggested and their results are compared. The first method estimates the life of wire ropes utilizing the accelerated life model with pseudo lives obtained from a linear regression model. The second method estimates the life using a logistic model based on failure probability.
