인간과 감정적 상호작용을 위한 '감정 엔진' (Engine of computational Emotion model for emotional interaction with human)

  • 지금까지 로봇 및 소프트웨어 에이전트들을 살펴보면, 감정 모델이 내부에 종속적으로 존재하기 때문에 감정모델만을 별도로 분리해 새로운 시스템에 재활용하기란 쉽지 않다. 따라서 어떤 로봇 및 에이전트와 연동될 수 있는 Engine of computational Emotion model (이하 EE로 표시한다)을 소개한다. 이 EE는 어떤 입력 정보에도 치중되지 않고, 어떤 로봇 및 에이전트의 내부와도 연동되도록 독립적으로 감정을 담당하기 위해, 입력 단계인 인식과 출력 단계인 표현을 배제하고, 순수하게 감정의 생성 및 처리를 담당하는 중간 단계인 감정 발생만을 분리하여, '입력단 및 출력단과 독립적인 소프트웨어 형태, 즉 엔진(Engine)'으로 존재한다. 이 EE는 어떤 입력단 및 출력단과 상호작용이 가능하며, 자체 감정뿐 아니라 상대방의 감정을 사용하며, 성격을 활용하여 종합적인 감정을 산출해낸다. 또한 이 EE는 로봇 및 에이전트의 내부에 라이브러리 형태로 존재하거나, 별도의 시스템으로 존재하여 통신할 수 있는 구조로 활용될 수 있다. 감정은 Joy(기쁨), Surprise(놀람), Disgust(혐오), Fear(공포), Sadness(슬픔), Anger(분노)의 기본 감정을 사용하며, 문자열과 계수를 쌍으로 갖는 정보를 EE는 입력 인터페이스를 통해 입력 신호로 받고, 출력 인터페이스를 통해 출력 신호로 내보낸다. EE는 내부에 감정마다 감정경험의 연결 목록을 가지고 있으며, 이의 계수의 쌍으로 구성된 정보를 감정의 생성 및 처리하기 위한 감정상태 목록으로 사용한다. 이 감정경험 목록은 '인간이 실생활에서 경험하는 다양한 감정에 대한 이해를 도모'하는 감정표현어휘로 구성되어 있다. EE는 인간의 감정을 탐색하여 적절한 반응을 나타내주는 상호작용 제품에 이용 가능할 것이다. 본 연구는 제품이 '인간을 공감하고 있음'을 인간이 느낄 수 있도록 유도하는 시스템을 만들고자 함이므로, HRI(인간-로봇 상호작용)나 HCI(인간-컴퓨터 상호작용)와 관련 제품이 효율적인 감정적 공감 서비스를 제공하는데 도움이 될 수 있을 것으로 기대한다.

Osteonectin Interacts with Human Nebulin C-terminus in Skeletal Muscle

  • Nebulin is a giant actin binding protein (600-900 kDa) which is specific to skeletal muscle. This protein is known to regulate thin filaments length in sarcomere as a molecular template. The C-terminus of nebulin is located in the Z-disc of muscle sarcomere and is bound to other proteins such like myopalladin, titin, archvillin, and desmin. The N-terminus of nebulin binds to tropomodulin at the pointed ends of the thin filaments. In recent research, nebulin not only found in brain but also expressed in heart, stomach, and liver. So, the roles of nebulin in non-muscle tissue have been studied. However, lack of information or studies on nebulin binding proteins and nebulin function in brain are available so far. Therefore, the current study have investigated a novel binding partner of Nebulin C-terminus by using yeast two-hybrid screening with human brain cDNA library. Nebulin C-terminus, containing simple repeats, serine rich and SH3 domain, interacts with osteonectin C-terminal region. The specific interaction of nebulin and osteonectin were confirmed in vitro by using GST pull-down assay and reconfirmed in vivo by using transfected COS-7 cells with EGFP-tagged nebulin and DsRed-tagged osteonectin. Consequently, this study identified SH3 domain in nebulin C-terminus specifically binds to extracellular Ca-binding (EeC domain in osteonectin. Also, nebulin C-terminus fusion protein colocalized with osteonectin EC domain fusion protein in transfected COS-7 cells. The current study found the interaction between nebulin and osteonectin in human brain for the first time and suggested the nebulin in brain may be associated with osteonectin, as a regulator of cell cycle progression and mitosis.

실행능력 평가도구에 대한 체계적 고찰 : 정상아동에서 국내.외 평가도구별 수행범위 제시 (Systematic Review of Assessment Tools for Praxis Ability of Sensory Integrative Function : Provide Range of Assessment Tools in Foreign and Domestic Normal Children)

  • 목적 : 본 연구의 목적은 국내 외 감각통합기능의 실행능력을 측정하는 평가도구에 대한 체계적 고찰을 통해 정상아동의 수행 평균이나 범위를 제시하고자 하였다. 연구방법 : Medline, PubMed, Ovid, Eric, 국회도서관, 국가과학기술정보센터, Kiss(한국학술정보), RISS(학술연구정보서비스), 구글 검색 엔진을 이용하여 감각통합이론의 틀을 바탕으로 한 실행능력 평가도구명을 검색하였다. 대상논문은 2000년 1월부터 2011년 4월까지 출판된 학위논문, 학회지, 저널이었다. 결과 : 총 24개의 논문이 검색되었고, 사용된 평가도구는 Bruinink-Oseretsky Test of Motor Proficiency (BOTMP), Bruininks-Oseretsky Test of Motor Proficiency, Short Form (BOT-SF), Bruininks-Oseretsky Test of Motor Proficiency, Second Edition (BOT-2), Bruininks-Oseretsky Test of Motor Proficiency, Second Edition, Short Form (BOT-2-SF), Clinical Observations of Motor and Postural Skills (COMPS), Motor Assessment Battery for Children (MABC), Clinical Test of Sensory Interaction with Balance (CTSIB), Sensory Integration and Praxis Test (SIPT) 소항목이었다. BOTMP-SF에서 그리스와 미국아동의 수행을 비교한 결과, '달리기 속도 및 민첩성', '카드분류하기', '점찍기' 항목에서 큰 수행의 차이를 보였다. BOT-2의 한국아동의 수행결과는 8개 소항목에서 모두 평균과 평균이상의 결과를 보였으며 BOT-2-SF의 인도아동의 수행결과는 연령이 증가할수록 모든 항목의 수행도가 높아짐을 보였다. 영국, 캐나다, 한국 아동이 수행한 COMPS에서는 세 나라 평균이 모두 정상범주에 있었고 특히, 한국아동의 평균 수행점수가 캐나다 아동에 비해 높았다. 결론 : 본 연구에서 제시하는 국내 외 정상 아동의 수행 결과는 임상의 평가 대상이 되는 장애아동의 실행능력 수행을 비교하는데 기준을 제시할 것이다.

HtrA2 Interacts with Aβ Peptide but Does Not Directly Alter Its Production or Degradation

  • HtrA2/Omi is a mammalian mitochondrial serine protease homologous to the E. coli HtrA/DegP gene products. Recently, HtrA2/Omi was found to have a dual role in mammalian cells, acting as an apoptosis-inducing protein and being involved in maintenance of mitochondrial homeostasis. By screening a human brain cDNA library with $A{\beta}$ peptide as bait in a yeast two-hybrid system, we identified HtrA2/Omi as a binding partner of $A{\beta}$ peptide. The interaction between $A{\beta}$ peptide and HtrA2/Omi was confirmed by an immunoblot binding assay. The possible involvement of HtrA2/Omi in $A{\beta}$ peptide metabolism was investigated. In vitro peptide cleavage assays showed that HtrA2/Omi did not directly promote the production of $A{\beta}$ peptide at the ${\beta}/{\gamma}$-secretase level, or the degradation of $A{\beta}$ peptide. However, overexpression of HtrA2/Omi in K269 cells decreased the production of $A{\beta}40$ and $A{\beta}42$ by up to 30%. These results rule out the involvement of HtrA2/Omi in the etiology of Alzheimer's disease. However, the fact that overexpression of HtrA2/Omi reduces the generation of $A{\beta}40$ and $A{\beta}42$ suggests that it may play some positive role in mammalian cells.

Circular RNA expression profiles in the porcine liver of two distinct phenotype pig breeds

  • Objective: An experiment was conducted to identify and characterize the circular RNA expression and metabolic characteristics in the liver of Jinhua pigs and Landrace pigs. Methods: Three Jinhua pigs and three Landrace pigs respectively at 70-day were slaughtered to collect the liver tissue samples. Immediately after slaughter, blood samples were taken to detect serum biochemical indicators. Total RNA extracted from liver tissue samples were used to prepare the library and then sequence on HiSeq 2500. Bioinformatic methods were employed to analyze sequence data to identify the circRNAs and predict the potential roles of differentially expressed circRNAs between the two breeds. Results: Significant differences in physiological and biochemical traits were observed between growing Jinhua and Landrace pigs. We identified 84,864 circRNA candidates in two breeds and 366 circRNAs were detected as significantly differentially expressed. Their host genes are involved in lipid biosynthetic and metabolic processes according to the gene ontology analysis and associated with metabolic pathways. Conclusion: Our research represents the first description of circRNA profiles in the porcine liver from two divergent phenotype pigs. The predicted miRNA-circRNA interaction provides important basis for miRNA-circRNA relationships in the porcine liver. These data expand the repertories of porcine circRNA and are conducive to understanding the possible molecular mechanisms involved in miRNA and circRNA. Our study provides basic data for further research of the biological functions of circRNAs in the porcine liver.

Selection and Analysis of Genomic Sequence-Derived RNA Motifs Binding to C5 Protein

  • Escherichia coli RNase P is a ribonucleoprotein composed of M1 RNA and C5 protein. Previously, analysis of RNA aptamers selected for C5 protein from a synthetic RNA library showed that C5 protein could bind various RNA molecules as an RNA binding protein. In this study, we searched cellular RNA motifs that could be recognized by C5 protein by a genomic SELEX approach. We found various C5 protein-binding RNA motifs derived from E. coli genomic sequences. Our results suggest that C5 protein interacts with various cellular RNA species in addition to M1 RNA.

Electrochemical Detection of $17{\beta}-estradiol$ by using DNA Aptamer Immobilized Nanowell Gold Electrodes

  • Aptamer is the single-stranded oligonucleotide which binds to various target molecules such as proteins, peptides, lipids and small organic molecules with high affinity and specificity. DNA aptamers specific for the $17{\beta}-estradiol$ were selected by SELEX (Systematic Evolution of Ligands by EXponential enrichment) process from a random DNA library. These DNA aptamers have a high affinity to $17{\beta}-estradiol$ as an endocrine disrupting chemical. Nanowell and $200{\mu}m$ gold electrode were used as substrate for DNA aptamer immobilization and electrochemical analysis. Especially, nanowell gold electrode was fabricated by e-beam lithography. The size of single nanowell is 130nm and 40,000 nanowells were deposited on one gold electrode. The immobilization method was based on the interaction between the biotinylated aptamer and streptavidin deposited on gold electrode previously. Immobilization procedure was optimized by surface plasma resonance (SPR) and electrochemical analysis. After the immobilization of DNA aptamer on streptavidin modified gold electrode, $17{\beta}-estradiol$ solution was treated on aptamer immobilized gold electrode. The current of gold electrode was decreased by the binding of $17{\beta}-estradiol$ to DNA aptamer immobilized on gold electrode. However, in negative control experiments of 1-aminoanthraquinone and 2-methoxynaphthalene, the current was rarely decreased. And more sensitive data was obtained from nanowell gold electrode comparing with $200{\mu}m$ gold electrode.

Differential expression and in situ localization of a pepper defensin (CADEFl) gene in response to pathogen infection, abiotic elicitors and environmental stresses in Capsium annuum

  • Pepper defensin ( CADEFl) clone was isolated from cDNA library constructed from pepper leaves infected with avirulent strain Bv5-4a of Xanthomonu campestris pv. vesicatoria. The deduced amino acid sequence of CADEFl is 82-64% identical to that of other plant defensins. Putative protein encoded by CADEFl gene consists of 78 amino acids and 8 conserved cysteine residues to form four structure-stabilizing disulfide bridges. Transcription of the CADEF1 gene was earlier and stronger induced by X campestris pv. vesicatoria infection in the incompatible than in the compatible interaction. CADEF1 mRNA was constitutively expressed in stem, root and green fruit of pepper. Transcripts of CADEFl gene drastically accumulated in pepper leaf tissues treated With Salicylic acid (SA), methyl jasmonate (MeJA), abscisic acid (ABA), hydrogen Peroxide (H$_2$O$_2$), benzothiadiazole (BTH) and DL-${\beta}$-amino-n-butyric acid (BABA). In situ hybridization results revealed that CADEF1 mRNA was localized in the phloem areas of vascular bundles in leaf tissues treated with exogenous SA, MeJA and ABA. Strong accumulation of CADEF1 mRNA occurred in pepper leaves in response to wounding, high salinity and drought stress. These results suggest that bacterial pathogen infection, abiotic elicitors and some environmental stresses may play a significant role in signal transduction pathway for CADEF1 gene expression.

  • Interactions between GRA proteins of dense granules in Toxoplasma gondii and host cell proteins were analyzed by yeast two-hybrid technique. The cMyc-GRA fusion proteins expressed from pGBKT7 plasmid in Y187 yeast were bound to host cell proteins from pGADT7-Rec-HeLa cDNA library transformed to AH109 yeast by mating method. By the selection procedures, a total of 939 colonies of the SD/-AHLT culture, 348 colonies of the $X-\alpha-gal$ positive and PCR, 157 colonies of the $X-\beta-gal$ assay were chosen for sequencing the cDNA and finally 90 colonies containing ORF were selected to analyze the interactions. GRA proteins interacted with a variety of host cell proteins such as enzymes, structural and functional proteins of organellar proteins of broad spectrum. Several specific bindings of each GRA protein to host proteins were discussed presumptively the role of GRA proteins after secreting into the parasitophorous vacuoles (PV) and the PV membrane in the parasitism of this parasite.

Simultaneous Comparison of Efficacy and Adverse Events of Interventions for Patients with Esophageal Cancer: Protocol for a Systematic Review and Bayesian Network Meta-analysis

  • Background: Esophageal cancer is one of the most serious malignancies. Due to the aggressive nature of this cancer, the prognosis is poor. A network meta-analysis with simultaneous comparison of multiple treatments can help determine better treatment options that have higher effects on overall survival of patients with lower adverse events. The aim of this review is to simultaneously compare efficacy and adverse events of treatment interventions for esophageal cancer. Materials and Methods: In this review, only randomized control trials (RCT) will be considered for network meta-analysis. All international electronic databases including Medline, Web of Sciences, Scopus, Cochran's library, EMBASE and Cancerlit will be searched to find randomized control trials which compared two or more treatment interventions for esophageal cancer. A network plot will be drawn for visual representation of all available treatment interventions. Bayesian approach will be used to combine the direct and indirect evidence. Treatment effects (e.g. hazard ratio for time to event outcomes, risk ratio for binary outcomes, and rate ratio for count outcomes with 95% credible interval) will be reported. Moreover, cumulative probability of the treatment ranks will be reported using the surface under the cumulative ranking (SUCRA) graphs. Consistency assumption will be assessed by the loop-specific and design-by-treatment interaction approaches. Conclusions: The results of this study may be helpful for the patients, clinicians and health policy makers in selecting treatments that have the best effect on survival and lowest adverse events.
