On-line Monitoring of IPTG Induction for Recombinant Protein Production Using an Automatic pH Control Signal

  • The response of IPTG induction was investigated through the monitoring of the alkali consumption rate and buffer capacity during the cultivation of recombinant E. coli BL21 (DE3) harboring the plasmid pRSET-LacZ under the control of lac promoter. The rate of alkali consumption increased along with cell growth, but declined suddenly after approximately 0.2 h of IPTG induction. The buffer capacity also declined after 0.9 h of IPTG induction. The profile of buffer capacity seems to correlate with the level of acetate production. The IPTG response was monitored only when introduced into the mid-exponential phase of bacterial cell growth. The minimum concentration of IPTG for induction, which was found out to be 0.1 mM, can also be monitored on-line and in-situ. Therefore, the on-line monitoring of alkali consumption rate and buffer capacity can be an indicator of the metabolic shift initiated by IPTG supplement, as well as for the physiological state of cell growth.

  • The effects of adsorption and desorption of alkali-metals on Si(111) surface were investigated by using AES and RHEED-system. The adsorption system is a fundamental interest because of its unique electronic properties such as measurement of work function change, adatom-core level shift. It was found that the growth node of K on Si(111) surface was layer by layer growth and the saturation coverage was 2.0ML at room temperature. Superstructure changes on Si(111) surface according to the alkali-metal thickness and substrate temperatures were accurately defined. By applying the isothermal desorption method, the desorption energies of Li/Si(111) and K/Si(111) surfaces was measured. On Li/Si(111) and K/Si(111) surfaces, the desorption energies were 3.07 eV, 2.19 eV respectively.

PMA Activates Stat3 in the Jak/Stat Pathway and Induces SOCS5 in Rat Brain Astrocytes

  • Suppressors of cytokine signaling (SOCS) family members are negative feedback regulators of the Jak/Stat pathway, which is an essential inflammatory signaling pathway. We investigated expression of eight members of the SOCS family in rat astrocytes, using two inflammatory stimulants, PMA and IFN-${\gamma}$. Only a few SOCS genes were induced by both stimulants, and we detected an increase in SOCS5 protein with PMA. PMA activated the Jnk, Erk, p38, and Jak/Stat signal pathways. In addition, it increased the level of activated-Stat3 resulting from tyrosine phosphorylation. A gel-shift assay showed that a protein in nuclear extracts from PMA-treated cells was able to bind to Stat binding elements. These results suggest that activated Stat3 binds to SOCS promoters and leads to their transcriptional induction.

  • Control of shock may be important in the hydraulic system and necessary to avoid failure and to improve the efficiency of operation. This study addresses the design and use of an orifice to provide the desired control of the hydraulic actuator system. The experimental apparatus is an idealization of an automobile shift system. Control is accomplished by installing three different types of orifices at appropriate locations in the system. Experimental results show that the orifice can be used to obtain the control of shock and control level depends on the orifice size, orifice type, operating pressure and flow rate.

  • One of the ever-increasing demands on the performances of heterodyne interferometers is to improve the measurement resolution, of which current state-of-the-art reaches the region of sub-nanometers. We propose a new scheme of phase-measuring electronics that reduces the measurement resolution without further increase in clock speed. Our scheme adopts a super-heterodyne technique that lowers the original beat frequency to a level of 1 MHz by mixing it with electrically generated reference signal. The technique enables us to measure the phase of Doppler shift with a resolution of 1.58 nanometer at a sampling rate of 1 MHz. To avoid the undesirable decrease in the maximum measurable speed caused by the lowered beat frequency, a special from of frequency up-down counting technique is combined with the super-heterodyning. This alloys performing required phase unwrapping simply by using programmable digital gates without 2$\pi$ ambiguities up to the maximum velocity of 2.35 m/s.

  • In this paper, We present demodulation and synchronization method of phase shift keying signal using Double Frequency Vector Technique for Reference Vector. 2nd Harmonic Vector for Reference Vector is utilized in discriminating between noise and carrier signal, and in producting correlation value for data bit logical level. And we applied this demodulator to Automatic Meter Reading System being communicated with electric distribution power lines.

  • Annexin I is a member of the in family of calcium dependent phospholipid banding proteins and is an in vitro phospholipase $A_2$ (PLA$_2$) inhibitor. The mechanism of PLA$_2$ inhibition by annexin I is still ambiguous. The structure of annexin I was studied at the atomic level by using nuclear magnetic resonance (NMR), circular dichrotsm (CD) and fluorescence spectroscopy. Recombinant human annexin I and N-terminally truncated annexin I (1-31 deleted: d-annexin I) were purified and their NMR spectra were compared. The NMR spectra of the two were similar. When $Ca^{2+}$ ion added to annexin I ad d-annexin I, peak broadening occurred, but no significant spectroscopic change was observed. When porcine pancreatic PLA$_2$ was added to deuterium labeled annexin I, an interaction of annexin I with PLA$_2$ was observed as indicated by the disappearance and shift of several peaks in the NMR spectrum. This result supports a protein-protein interaction mechanism for PLA$_2$ inhibition by annexin I.I.

  • Crosstalk is the primary cause of image distortion in active matrix liquid crystal displays (AMLCD). Crosstalk produces voltage errors that limit gray scale fidelity and consequently, degrades display resolution, contrast ratio, color fidelity, and image quality. In this study, crosstalk phenomena of some methods to compensate level shift voltages has been simulated. This will be contributed to find the way to design the excellent image quality of the TFT-LCDs.

Acute Cerebral Infarction after Head Injury

  • Cerebral infarction rarely occur following head injury. The authors present the case of a 39-year-old man with complete infarction in the middle cerebral artery[MCA] and anterior cerebral artery[ACA] territories ccurred immediately after head injury. He had compound depressed fracture in right frontal bone with no neurological deficit. After the depressed bone elevation, postoperative computed tomography scan showed the right MCA and ACA territory infarction with midline shift. Cerebral angiography obtained on the day after emergent decompressive craneictomy showed the complete occlusion of the internal carotid artery[ICA] at the level of lacerum ICA segment. There was no evidence of neck vessel dissection and basal skull fracture. Cerebral infarction can occur in an ultraearly period after head injury without neck vessel dissection or basal skull fracture. We stress the need for attention to the cerebral infarction as the cause of a rare neurological deterioration of the head trauma.
