Honey bees (Apis mellifera) forage in agricultural areas, and are exposed to diverse pesticide poisoning. Toxic effects on Apis mellifera of different groups of pesticides were tested in the laboratory; fungicide (Metconazole), herbicide (Glyphosate), acaricide (Amitraz), organophosphate insecticide(Fenitrothion) and neonicotinoid insecticides(Thiacloprid, Thiamethoxam, Imidacloprid, Acetamiprid, Dinotefuran and Clothianidin). Commercial formulations were serially diluted from the recommended concentration (RC) to 10-6 times to carry out feeding and contact tests. Toxicity was transformed into lethal dose (LD50) and hazard question (HQ). The acute toxicity of pesticides showed similar patterns between feeding and contact tests. But feeding tests showed greater toxic to honey bee than contact test. The organophosphate and nitro-neonicotinoid insecticides were highly toxic with HQ values ranging greater than 1. However, cyano-neonicotinoids of Thiacloprid and Acetamiprid showed low toxicity. Even at the RC, 24 hr mortalities were 18 and 30%. The acaricide (Amitraz) showed intermediate level of toxicity at RC but negligible at the concentration lower than 10-1 times. A fungicide(Metconazole) and herbicide(Glyphosate) showed minimal impacts. The results imply that the selective use of pesticides could help conservation of pollinators in agricultural production systems.

The present study was carried out to investigate the potential toxicity of highly-developed rice varieties, Oryza sativa cvs. Jukjinjubyeo, Heuginjubyeo and Heughyangbyeo by a single oral dose in ICR mice. The test article was orally administered once by gavage at dose levels of 0, 2.5, 5.0, and 10.0 g/kg body weight (n=10 for male and female mice for each dose). We examined the number of deaths, general clinical signs, body weights and biochemical analysis for sexes and doses of mice between control and experimental groups. As a result all of the groups were alive during experimental periods and none of the significant clinical symptom and body weight changes were shown in all of the groups. Plasma glucose levels were valued for both control and experimental groups and there were no significant differences between the groups. The activities of aminotransferase were not increased in all experimental groups compared to control groups. The results suggest that the toxicity of the aleurone layer of Oryza sativa cvs. Jukjinjubyeo, Heugjinjubyeo and Heughyangbyeo are low and its $LD_{50}$ is over 10.0 g/kg body weight in both male and female mice.
This paper dealt with plasmid DNA profile in 98 Salmonella(S) isolated from pigs and cattle sources in Taegu, Gyeongbook and Gyeongnam during the period from 1984 to 1987. Also we were studied for restriction enzyme analysis of the plasmid DNA, and mouse infection, Sereny test and normal setum resistance test in guinea pig for S typhimurium and S enteritidis harbored or cured 60 megadalton(Md) plasmid and 36 Md plasmid, respectively. Of the 13 Salmonella isolated from cattle, 7 Salmonella harbored one or more plasmids and molecular sizes of the large plasmids were 60 Md for S typhimurium and 36 Md for S enteritidis. Of the 85 Salmonella isolated from pigs, 47 Salmonella were confirmed as being one or more plasmids, and all the S typimurium stains harbored 60 Md plasmid. In enzyme digestion with 8 types of restriction endonuclease for 60 Md plasmid DNA of S typhimurium, cleavage patterns were varied to enzymes, and the DNA was segmented into 4 to 15 fragments. In restriction enzyme analysis of 36 Md plasmid DNA obtained from four strains of S. enteritidis, the DNA showed the same cleavage patterns obtained with Eco RI, Hind III and Bam H I, and was segmented into 3 to 5 fragments. In virulence for mice by measuring the 50% lethal dose ($LD_{50}$), the $LD_{50}$ values obtained for 60 Md virulence-associated plasmid harbored strains of S typhimurium and 36 Md virulence-associated plasmid of S enteritidis were up to $10^4$-fold lower than the values obtained for the plasmid-cured strains of the same serotype. Only the plasmid harbored strains were resistant to the bactericidal activity of 90% guinea pig serum, and only they gave positive responses in sereny test. We suggested that their plasmid DNA might be associated with virulence for mice.
Objectives: Bee venom (BV) is a complex mixture of proteins and contains proteins such as phospholipase and melittin, which have an effect on blood clotting and blood clots. The mechanism of action of honey bee venom (HBV, Apis mellifera) on human plasma proteins and its anti-thrombotic effect were studied. The purpose of this study was to investigate the anti-coagulation effect of BV and its effects on blood coagulation and purification. Methods: Crude venom obtained from Apis mellifera was selected. The anti-coagulation factor of the crude venom from this species was purified by using gel filtration chromatography (sephadex G-50), and the molecular weights of the anti-coagulants in this venom estimated by using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Blood samples were obtained from 10 rabbits, and the prothrombin time (PT) and the partial thromboplastin time (PTT) tests were conducted. The approximate lethal dose (LD) values of BV were determined. Results: Crude BV increased the blood clotting time. For BV concentrations from 1 to 4 mg/mL, clotting was not observed even at more than 300 seconds, standard deviations $(SDs)={\pm}0.71$; however, clotting was observed in the control group 13.8 s, $SDs={\pm}0.52$. Thus, BV can be considered as containing anti-coagulation factors. Crude BV is composed 4 protein bands with molecular weights of 3, 15, 20 and 41 kilodalton (kDa), respectively. The $LD_{50}$ of the crude BV was found to be $177.8{\mu}g/mouse$. Conclusion: BV contains anti-coagulation factors. The fraction extracted from the Iranian bees contains proteins that are similar to anti-coagulation proteins, such as phospholipase $A_2(PLA_2)$ and melittin, and that can increase the blood clotting times in vitro.

The acute toxicity and sublethal effects of nitrite on the dark-banded rockfish, Sebastes inermis (mean body weight: $83.3{\pm}7.2$ g), were studied under static conditions for a period of 96 h. The acute toxicity of nitrite was at the 50% lethal concentration ($LC_{50}$) of 700 mg/L. The sublethal effects on selected hematological parameters of the dark-banded rockfish, such as its osmolality, hematocrit, cortisol, alanine aminotransferase (ALT), and aspartate aminotransferase (AST), were measured after 0, 6, 12, 24, 48, 72, and 96 h of exposure to 0, 50, 100, 200, 400, or 700 mg/L nitrite. Sublethal nitrite caused a progressive reduction in the hematocrit of the fish, depending on the nitrite concentration and the exposure period. Exposure to 100-700 mg/L nitrite for 96 h caused a reduction in the hematocrit and an increase in cortisol, ALT, and AST compared with the control levels. Abnormal ultrastructural changes in the gills and liver tissues were observed in fish exposed to 700 mg/L nitrite for up to 96 h compared with the control tissues. Ultrastructural changes included atrophic gill mitochondria and hepatocytes that developed smooth endoplasmic reticulum and atrophic mitochondria. Although no rockfish mortality occurred at 500 mg/L nitrite, all the hematological parameters examined responded adversely to a nitrite dose of 200 mg/L for 96 h. These results show that although the acute toxic concentration of nitrite for the dark-banded rockfish is > 700 mg/L, sublethal concentrations of nitrite also negatively affect its hematological parameters.
1. Objectives: The aim of this study is data analysis for acute toxicity and safety of Cheongsimyeonja-tang. 2. Methods: We investigated the acute toxicity for water-extracted Cheongsimyeonja-tang. Fifty five male and female mice were observed for 14 days after one day oral administration of Cheongsimyeonja-tang at the respective doses of 0 (control group), 2560, 3200, 4000 and 5000 mg/kg. 3. Results: We observed survival rates, general toxicity, change of body weight and autopsy. In animals administered with Cheongsimyeonja-tang, there were nither dead animals nor significant changes of body weights. In addition, no differences were found between control and treated groups in clinical sign and autopsy. 4. Conclusion: The data confirmed that Cheongsimyeonja-tang is free from the toxicity and safety problems in treated groups. Compared with the control group, we could not find any toxic alteration in all treated groups(2560, 3200, 4000 and 5000 mg/kg). Lethal Dose 50 (LD50) value for mice was more than 5000 mg/kg per oral for both male and females. It suggest that Cheongsimyeonja-tang in mice is considered to be safe.

Sterigmatocystin bears a close structural relationship to aflatoxin $B_1$ and is a carcinogenic compound that has been shown to affect various species of experimental animals. Reaction and toxicity of sterigmatocystin in the artificial gastric juice were investigated. Sterigmatocystin was degraded in artificial gastric juice and extracted by the method of A.O.A.C. After cleaned up by silica gel column chromatography, this substance was detected and characterized by thin layer chromatography, UV, IR and mass spectra. It showed $R{\mathcal{f}}$ 0.4 and brick-red color by TLC. Especially, in the mass spectrum of it, fragment peak at m/e 327 was due to the loss of the $-CH_3$ and $-H_2O$, fragment peak at m/e 341 was due to the loss of the $H_2O$ and $-H^+$, and fragment peak at m/e 239 was due to the loss of the 2-chloro-tetrahydrofuran and methyl group from the parent molecule. Therefore, a degraded substance of sterigmatocystin reacted in artificial gastric juice (Sub. K) was estimated with additional formation of hydrochloric acid. In four-day-old chicken embryos, the mean lethal dose $(LD_{50})$ was $140\;{\mu}g/egg$, and 90 to 100% of the embryos were killed with 1 mg/egg. This $LD_{50}$$140\;{\mu}g/egg$ compared with an $LD_{50}$$14.69\;{\mu}g/egg$ for sterigmatocystin (acute toxicity) showed the substance to be much less toxic than sterigmatocystin.
The sterilization effects of methylene blue (MB), formalin, and iodine on the egg of marine medaka, Oryzias dancena, were investigated for disinfecting naididae worm, Chaetogaster diastrophus through sterilization. To determine harmfulness of MB, formalin, and iodine, lethal concentrations 50 ($LC_{50}$) of three chemicals were analyzed in the eggs of marine medaka. The sterilized periods of each chemical were set at 1 hr. Sterilized rates of naididae worm in each chemical were significantly affected and increased drastically as the concentration of each chemical increased (p<0.05). Sterilization abilities of naididae worm were most effective for formalin, but survival rates of egg and hatched rates for formalin were lowest among each chemical. The $LC_{50}$ of MB over 96 hrs were 185.26, 103.84, and 127.15 ppm for adults, juveniles, and eggs respectively. The toxic effects of MB were clearly dose dependent for each life stage (p<0.05). The toxicity sensitivity of juveniles to MB was dramatically higher than that of other groups. In 48 hrs after sterilization, cortisol and glucose concentrations of the adult group with MB treatment were significantly higher than those of the adult group with no treatment (p<0.05). This research provides useful data on sterilization effect of MB, formalin, and iodine, acute toxicity in marine medaka egg and toxicity, sensitivity of life stage of MB in marine medaka.

Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.
Vespa velutina nigrithorax du Buysson, 1905 is the invaded species in Korea since 2003. Since its importance as the honey bee pest, beekeepers use insecticides to kill the adult and immature hornets. However, its legality and effectiveness has not been confirmed. This study investigated the susceptibility of insecticides commonly used to control hornets by beekeepers in Korea. Eight insecticides were tested on adult worker and larvae by topical or oral treatment. Adults showed more than 70% mortalities from Clothianidin, Dinotefuran, and Carbosulfan treatment within 30 minutes. Bifenthrin and Cartap hydrochloride showed relatively low toxicity. The median lethal dose (LD50) for Clothianidin, Dinotefuran, and Carbosulfan was 0.29, 0.65, and 2.21 ㎍/bee, respectively. In larval feeding test where 5th instar larvae were fed 3 times every 24 hours, the mortality began after second treatments. After 3rd treatments (72 h), all insecticides showed mortality more than 70%. The LD50 values of Clothianidin, Dinotefuran, and Carbosulfan to V. velutina were approximately 10 to 100 times higher than those to honey bee, Apis mellifera. This study provides the basic information of those chemical toxicities to Vespa hornet and honey bees.
