Optimization of Hydrogen Production Process using 50 Nm3/h Biogas (50 Nm3/h급 바이오가스 직접 이용 수소 생산 공정 최적화)

  • This study presents a novel approach to hydrogen production by biogas from organic waste without CO2 removal. A process model was developed to reduce the costs associated with biogas pretreatment and purification processes. Through optimization of heat exchange networks, the simulation aimed to minimize process costs, maximizing hydrogen production and flue gas temperature. The results reveal that the most efficient process model maximizes the flue gas temperature while following the constraint of the number of heat exchangers. These findings hold promise for contributing to the expansion of "Biogas-to-clean hydrogen" energy conversion technology.

Proteome-wide Characterization and Pathophysiology Correlation in Non-ischemic Cardiomyopathies

  • Background and Objectives: Although the clinical consequences of advanced heart failure (HF) may be similar across different etiologies of cardiomyopathies, their proteomic expression may show substantial differences in relation to underlying pathophysiology. We aimed to identify myocardial tissue-based proteomic characteristics and the underlying molecular pathophysiology in non-ischemic cardiomyopathy with different etiologies. Methods: Comparative extensive proteomic analysis of the myocardium was performed in nine patients with biopsy-proven non-ischemic cardiomyopathies (3 dilated cardiomyopathy [DCM], 2 hypertrophic cardiomyopathy [HCM], and 4 myocarditis) as well as five controls using tandem mass tags combined with liquid chromatography-mass spectrometry. Differential protein expression analysis, Gene Ontology (GO) analysis, and Ingenuity Pathway Analysis (IPA) were performed to identify proteomic differences and molecular mechanisms in each cardiomyopathy type compared to the control. Proteomic characteristics were further evaluated in accordance with clinical and pathological findings. Results: The principal component analysis score plot showed that the controls, DCM, and HCM clustered well. However, myocarditis samples exhibited scattered distribution. IPA revealed the downregulation of oxidative phosphorylation and upregulation of the sirtuin signaling pathway in both DCM and HCM. Various inflammatory pathways were upregulated in myocarditis with the downregulation of Rho GDP dissociation inhibitors. The molecular pathophysiology identified by extensive proteomic analysis represented the clinical and pathological properties of each cardiomyopathy with abundant proteomes. Conclusions: Different etiologies of non-ischemic cardiomyopathies in advanced HF exhibit distinct proteomic expression despite shared pathologic findings. The benefit of tailored management strategies considering the different proteomic expressions in non-ischemic advanced HF requires further investigation.

  • As considering a comparison study of the structure of mind and psychologic type of C.G Jung to Ji-In-Lon and Sa-Sang constituition of Lee Je Ma, I've got the following conclusion. 1. The study of the structure of mind of Jung and Ji-In-Lon of Lee Je ma have a important meaning to each theory of type of men. They developed their own peculiar theory of type men by the analysis of the mind of men. 2. Jung explained the organization of recognition by four phase theorical structure; consciousness, personal unconsciousness, collective unconsciousness, self. Lee Je Ma explained the organization of recognition by four phase theorical structure;Sa, Sim, Sin, Mul. And theory of type of men, Jung established four type; Tae-Yang-In, Tae-Um-In, So-Yang-In, So-Um-In, They all established the four phase structure, which have things in common. 3. Jung studied the consciousness and unconsciousness and Lee Je Ma studied the Sung-In, Eui, Ye, Ji, and Sim-Yock-Bi, Bak, Tam, Na. I think they have a close relationship. And I think there is relationship between consciousness, unconsciousness to Gun-Ja-Ji-Sim, So-In-Ji-Sim. 4. I think there is thought of relationship between Lee Je Ma's Dae-Dong, Gak-lip, Hang-gi-ji, Hang-gi-hang and Ho-sun, Oh-ak, Sa-sim, Tae-hang and relationship of internal organ's size to Jung's the first function, second function, third function, fourth function. 5. I think the relationship between the type of Jung;thinking type, feeling, type, sensation type, intuition type to the type of Lee Je Ma; Tae-Yang-In, Tae-Um-In, So-Yang-In, So-Yang-In, So-Um-In needs more study.

  • Pharmacopuncture, or herbal acupuncture, is a new form of therapy derived from combinations of two traditional therapeutic methods, herbal medicine and acupuncture therapy. To compare the efficacy between loratadine-pharmacopuncture (LP) and loratadine-oral administration (LO), the effect of loratadine was investigated in murine models. Anti-anaphylactic effects of loratadine treatments were investigated in compound 48/80-induced systemic anaphylactic reaction and passive cutaneous anaphylaxis (PCA). LP treatment significantly inhibited the compound 48/80-induced systemic anaphylactic reaction and PCA. The effect between LP and LO were on a similar level. These results indicate that LP can be used as an alternative method for LO in case of emergency.

An Analytical Solution of Flow and Progressive Wave-Induced Residual Pore Water Pressure in Seabed (흐름과 진행파에 의한 해저지반 내 잔류간극수압의 해석해)

  • This study extended the Lee et al.'s (2015a) solution which improved the existing analytical solution for prediction of the residual pore water pressure into progressive wave and flow coexisting field. At this time, the variation of incident wave period and wave length should be incorporated to Lee et al.'s (2015a) analytical solution, which does not consider flow. For the case of infinite thickness, the new analytical solution using Fourier series was compared to the analytical solution using Laplace transformation proposed by Jeng and Seymour (2007). It was verified that the new solution was identical to the Jeng and Seymour's solution. After verification of the new analytical solution, the residual pore water pressure head was examined closely under various given values of flow velocity's magnitude, direction, incident wave's period and seabed thickness. In each proposed analytical solution, asymptotic approach to shallow depth with the changes in the soil thickness within finite soil thickness was found possible, but not to infinite depth. It is also identified that there exists a discrepancy case between the results obtained from the finite and the infinite seabed thicknesses even on the same soil depth.

Effect of Callus Type and Antioxidants on Plant Regeneration and Transformation of Tall Fescue (캘러스의 형태와 항산화물질 첨가가 톨 페스큐의 식물체 재분화와 형질전환효율에 미치는 영향)

  • An efficient transformation system for the production of transgenic plants has been developed for tall fescue (Festuca arundinacea Schreb.) via Agrobacterium-mediated transformation of seed-derived callus. From the point of morphogenetic capacity, three types of callus were selected. High frequency of plant regeneration was obtained by selection of type II callus, and the plant regeneration frequency was 52.6% when embryogenic callus were cultured on the regeneration medium. Supplementation of the media with 10 mg/L $AgNO_3$ and 40 mg/L cysteine enhanced frequencies of plant regeneration up to 65.3%. The highest transformation efficiency was also obtained when type II callus were inoculated with Agrobacterium. Southern blot analysis of PCR products of transgenic plants demonstrated that transgenes were successfully integrated into the genome of tall fescue. Efficient regeneration system and transformation established in this study will be useful for molecular breeding of tall fescue through genetic transformation.

Expression of Heat Shock Protein and Antioxidant Genes in Rice Leaf Under Heat Stress

  • We have previously investigated the proteome changes of rice leaves under heat stress (Lee et al. in Proteomics 2007a, 7:3369-3383), wherein a group of antioxidant proteins and heat shock proteins (HSPs) were found to be regulated differently. The present study focuses on the biochemical changes and gene expression profiles of heat shock protein and antioxidant genes in rice leaves in response to heat stress ($42^{\circ}C$) during a wide range of exposure times. The results show that hydrogen peroxide and proline contents increased significantly, suggesting an oxidative burst and osmotic imbalance under heat stress. The mRNA levels of chaperone 60, HSP70, HSP100, chloroplastic HSP26, and mitochondrial small HSP responded rapidly and showed maximum expression after 0.5 or 2 h under heat stress. Transcript levels of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) and Cu-Zn superoxide dismutase (Cu-Zn SOD) showed a rapid and marked accumulation upon heat stress. While prolonged exposure to heat stress resulted in increased transcript levels of monodehydroascorbate reductase, peroxidase, glyoxalase 1, glutathione reductase, thioredoxin peroxidase, 2-Cysteine peroxiredoxin, and nucleoside diphosphate kinase 1, while the transcription of catalase was suppressed. Consistent with their changes in gene expression, the enzyme activities of APX and DHAR also increased significantly following exposure to heat stress. These results suggest that oxidative stress is usually caused by heat stress, and plants apply complex HSP- and antioxidant-mediated defense mechanisms to cope with heat stress.

  • In order to optimize tissue culture conditions for genetic transformation of orchardgrass, the effects of culture medium supplements on tissue culture responses were investigated with mature seeds of 3 cultivars, 'Frode'. 'Roughrider' and 'Frontier' as explant tissues. Callus induction medium containing 3mg/L 2,4-D or 3mg/L dicamba each with 0.1mg/L BA was optimal fer embryogenic callus formation from mature seed and had a strong effect on successive plant regeneration. The regeneration frequency from embryogenic callus among cultivars were descending order of Roughrider > Frode > Frontier. Supplementation of the regeneration media with 10mg/L $AgNO_3$ and 40mg/L cysteine enhanced frequency of plant regeneration. Efficient regeneration system established in this study will be useful fur molecular breeding of orchardgrass through genetic transformation.

A New Rice Cultivar "Jogwang" with RSV Resistance and Short Growth Duration (벼줄무늬잎마름병 저항성 단기성 벼 신품종 "조광")

  • Jogwang is a new early maturing japonica rice developed in 2007 from a cross between Milyang187 and YR21113-B-B at the Department of Functional Crop Science, NICS, RDA. This cultivar is very suitable to the rice-cash crop double cropping system. Heading date of Jogwang is 2 days earlier than Keumobyeo under the late transplanting cultivation on July 10 at the Yeongnam plain. The tolerance level of this variety to leaf discoloration at seedling stage is very similar to Keumobyeo. It showed slightly lower viviparous germination and premature heading. This cultivar showed resistant reactions to leaf blast and rice stripe virus disease but susceptible to bacterial blight disease and major insect pests. The ratio of milling and head rice recovery of Jogwang is 76.5% and 64.5%, respectively. The milled kernels are translucent with non glutinous endosperm. This cultivar has 7.3% protein and 18.5% amylose content. In local adaptability test, showed that the milled rice yield of Jogwang is $4.90\;MT\;ha^{-1}$. This cultivar is suitable for planting in the plain paddy fields of Honam and Yeonnam regions in Korea.

Effect of Acetosyringone and Variety on Transformation of Orchardgrass (오차드그래스의 형질전환에 있어서 Acetosyringone과 품종이 미치는 영향)

  • Effects of acetosyringone and on Agrobacterium-mediated transformation of orchardgrass were investigated. Embryogenic calli induced from 3 varieties, Frontier, Potomac and Roughrider, were infected and co-cultured with Agrobacterium EHA101 carrying standard binary vector pIG121Hm encoding the hygromycin phosphotransferase(HPT), neomycin phosphotransferase II(NPTII) and intron-containing ${\beta}-glucuronidase$ (intron-GUS) genes in the T-DNA region. The effects of varieties and acetosyringone(AS) concentrations on transformation and the expression of the GUS gene were investigated. Inclusion of $200{\mu}M$ AS in inoculation and co-cultivation media lead to a significant increase in stable transformation efficiency. Hygromycin resistant calli were developed into complete plants via somatic embryogenesis. GUS histochemical assay and PCR analysis of transgenic plants demonstrated that transgenes were integrated into the genome of orchardgrass.
