Altered expression of norepinephrine transporter and norepinephrine in human placenta cause pre-eclampsia through regulated trophoblast invasion

  • Objective: We investigated the norepinephrine transporter (NET) expression in normal and pre-eclamptic placentas and analyzed the invasion activity of trophoblastic cells based on norepinephrine (NE)-NET regulation. Methods: NET and NE expression levels were examined by western blot and enzyme-linked immunosorbent assay, respectively. Trophoblast invasion activity, depending on NE-NET regulation, was determined by NET-small interfering RNA (siRNA) and NET transfection into the human extravillous trophoblast cells with or without NE treatment and invasion rates were analyzed by zymography and an invasion assay. Results: NET mRNA was expressed at a low level in pre-eclamptic placentas compared with normal placentas and NE concentration in maternal plasma increased significantly in pre-eclamptic women compared to normal pregnant women (p<0.05). NET gene upregulation and NE treatment stimulated trophoblast cell invasion up to 2.5-fold (p<0.05) by stimulating matrix metalloproteinase-9 activity via the phosphoinositol-3-kinase/AKT signaling pathway, whereas NET-siRNA with NE treatment reduced invasion rates. Conclusion: NET expression is reduced by inadequate regulation of NE levels during placental development. This suggests that a complementary balance between NET and NE regulates trophoblast cell invasion activities during placental development.

  • Road pavements in Korea generally show shorter service life than the predicted one. There are many reasons for this phenomenon including increased traffic load and other attacks from exposure conditions. In order to extend a service life and upgrade the pavement, a new multi-functional composite pavement system is being developed in Korea. This study is to investigate the performances of fiber-reinforced lean concrete for pavement base. This study considered mineral admixtures of fly ash and reject ash. The reject ash is defined as ash that does not meet the specifications for fly ash so that it cannot be used as a supplemental material for cement replacement. Due to the inherent property of lean concrete, compaction during the fabrication of specimens is a key factor. Therefore, this study suggests an appropriate compaction method. From the test results, the compressive strengths of the concrete satisfied the required limit of 5 MPa at 7 days. When a compaction roller was used to mimic actual field conditions, the strength development seemed to be influenced by the compaction energy rather than hydration of cement itself.

  • The station area development planed to improve the old and narrowed station and to enhance the train passenger's convenience through private capital attraction in the past. But it has been faced with difficulty for the creative of transfer demand. Because of lean upon building facilities were caused by shortage of business profitability as to development of the upside a railway line, and lack of public utilities. The station area development requires that it's trend to large scale development of station areas, korail's direct development, deputation of specialty company to development, collaboration between local government and korail in development, diversification of process & installed facility, building multi-transfer center and one-stop transfer system. This trend caused by low profitability in development of the upside a railway line. The purposes of study are to make the system and law reflected to the property of station area development, investment in kind of needed land, and change railroad areas to high-rise and complex development, and make a forum between government and korail, etc.

Distinguishing Aroma Profile of Highly-Marbled Beef according to Quality Grade using Electronic Nose Sensors Data and Chemometrics Approach

  • Fat deposition in animal muscles differs according to the genetics and muscle anatomical locations. Moreover, different fat to lean muscle ratios (quality grade, QG) might contribute to aroma development in highly marbled beef. Scientific evidence is required to determine whether the abundance of aroma volatiles is positively correlated with the amount of fat in highly marbled beef. Therefore, this study aims to investigate the effect of QG on beef aroma profile using electronic nose data and a chemometric approach. An electronic nose with metal oxide semiconductors was used, and discrimination was performed using multivariate analysis, including principal component analysis and hierarchical clustering. The M. longissimus lumborum (striploin) of QG 1++, 1+, 1, and 2 of Hanwoo steers (n=6), finished under identical feeding systems on similar farms, were used. In contrast to the proportion of monounsaturated fatty acids (MUFAs), the abundance of volatile compounds and the proportion of polyunsaturated fatty acids (PUFAs) decreased as the QG increased. The aroma profile of striploin from carcasses of different QGs was well-discriminated. QG1++ was close to QG1+, while QG1 and QG2 were within a cluster. In conclusion, aroma development in beef is strongly influenced by fat deposition, particularly the fat-to-lean muscle ratio with regard to the proportion of PUFA. As MUFA slows down the oxidation and release of volatile compounds, leaner beef containing a higher proportion of PUFA produces more volatile compounds than beef with a higher amount of intramuscular fat.

  • The amount of dredging clay will be greatly increased by the eco-rive project and port development in Korea. Geotechnical engineers have thrown their efforts into the new ways for effective re-uses of the dredging clay such as the material for reclamation, and so on. However, very high initial water content and low strength causes unexpected difficulties in the aspect of trafficablility or time for consolidation. Therefore, the injection of cement stabilizer is used as one of ways to improve reclaimed ground. However, it also makes an argument by heavy metal from cement stabilizer. In this paper constant rate of strain consolidation test and normal consolidation test were performed to investigate behavior characteristics of the consolidation about soil-cement include lean mixed cement to reduce the environmental loads by the cement. The experimental results of consolidation characteristics about soil-cement include lean mixed cement influenced by mixing ratio. Especially it was observed that mixing ratio of 4%~6% leads not only the reduction of consolidation settlement, but time for consolidation.

  • In general, DI gasoline engine has the advantages of higher power output, higher thermal efficiency, higher EGR tolerance and lower emissions due to the operation characteristics of increased volumetric efficiency, compression ratio and ultra-lean combustion scheme. In order to apply the concept of stratified charge into direct injection gasoline engine, some kinds of methodologies have been adapted in various papers. In this study, a reflector was adapted around the injector nozzle to apply the concept of stratified charge combustion which leads the air-fuel mixture to be rich near spark plug. Therefore, the mixture near the spark plug is locally rich to ignite while the lean mixture is wholly introduced into the combustion chamber. The characteristics of combustion is analyzed with the variations of fuel injection pressure and load in a stratified -charge direct injection single cylinder gasoline engine. The obtained results are summarized as follows ; 1. The MBT spark timing approached to TDC with the increase of load on account of the increase of evaporation energy, but has little relation with fuel injection pressure. 2. The stratification effects are apparent with the increase of injection pressure. It is considered by the development of secondary diffusive combustion and the increase of heat release of same region, but proceed rapidly than diesel engine. Especially, in the case of high pressure injection (l70bar) and high load (3.0kgf m), the diffusive combustion parts are developed excessively and results in the decrease of peak pressure than in the case of middle load. 3. The index of engine stability, COVimep value, is drastically decreased with the increase of load. 4. To get better performance of DI gasoline engine development, staged optimizaion must be needed such as injection pressure, reflector, intake swirl, injection timing, chamber shape, ignition system and so on. In this study, the I50bar injection pressure is appeared as the optimum.

  • Fablab is a global network of digital fabrication facilities. Fablab is a digital workshop for individual manufacturing. In addition, Fablab is a next-generation digital infrastructure that connects education, training, R&D and production. The Fablab is a facility lab that makes the products you want, and it is a space where users-led products or services are discovered for the community. Furthermore, through various citizen-led projects, it is playing the role of innovation that changes the region and society. In this study, we examine the operating conditions of Fablabs in Korea and Japan (Fablab Seoul, Fablab Busan, Fablab Kamakura and Fablab Kitakagaya). It also explores the business model and sustainable development potential of each fablab. To this end, first, we compare and analyze the use of fablabs in both countries. Second, the purpose of the fablabs of both countries is analyzed. Third, we analyze the business models that the fablabs of both countries are taking for sustainable development through Lean Canvas. Based on the results obtained through case analysis of both countries, we make suggestions for the development of fablabs in Korea.

Combustion Enhancemen of Premixed Mixtures Using Laser-Induced Cavity Ignition (레이저 유도 공동 점화방식을 이용한 예혼합기 연소 특성 향상)

  • In this study, a new type of laser-induced ignition using a conical cavity has been developed to utilize all the available incident laser energy. In the method, it is possibile to ignite combustible methane/air mixtures by directing a laser beam of a constant small diameter into a small conical cavity, without focusing the laser beam. Shadow graphs for the early stage of combustion process show that a hot gas jet is ejected from the cavity, especially with lean mixture. After a very show time, the hot gas jet finishes issuing and the flame behavior is quite similar to flame propagation initiated by a conventional spark ignition. The combustion process using the new method exhibits more rapid pressure increase and a higher maximum pressure rise than that of the center ignition using laser-induced spark, with significant decrease in the combustion time. Also, the new ignition method is numerically modeled to simulate the flame kernel development and subsequent combustion process using the KIVA-IIcode. The calculated results show satisfactory agreement with experimental results.

  • Body fat distribution in patients with Prader-Willi syndrome (PWS) is characterized by reduce lean body mass (LBM), increased total body fat mass (FM), and lower percentage of visceral adipose tissue (VAT). Individuals with PWS seem to have a lower risk for insulin resistance with high levels of adiponectin, an anti-atherogenic adipocytokine that is decreased in visceral fat hypertrophy subjects compared to simple obese subjects, both in children and in adults. The mechanism of the reduction in visceral adiposity in PWS is still unclear. It might be related to qualitative intrinsic characteristics of adipocyte or novel genetic influences on the control of fat distribution. However, obesity remains a critical problem, and obesity status plays a crucial role in individual metabolic risk clustering and development of metabolic syndrome (Mets) in PWS children and adults. Long-term growth hormone (GH) treatment after cessation of skeletal growth improved body composition, with an increase in lean body mass and a reduction in total body fat and subcutaneous and visceral fat in PWS adults. Thus, the role of GH is important after childhood because it might attenuate obesity and Mets in PWS adult by adipocyte modification.

  • Vitamin and mineral deletion from swine diets can result in reduced growth if done during the period wher muscle and bone development is occurring. Several of the vitamins and minerals decline in the serum during the starter period, suggesting a higher dietary inclusion may be necessary postweaning. Vitamin research with grower-finisher pigs is limited, but results suggest that rapidly growing lean pigs may have a higher dietary requirement for the B vitamins. Several studies have suggested that early weaning and pigs of a lean genotype may have a dietary requirement for vitamin C, CI and Cr. High dietary vitamin E levels are fortified in the diet and seems to be effective in preventing mulberry heart problems in weanling and grower pigs. Organic Se is more effectively retained in muscle tissue than inorganic Se, approximately 20% less is excreted, but the bioavailability of organic Se for glutathione peroxidase activity is only 80 to 90% to that of sodium selenite. The active form of thyroxine (T4) is dependent upon a Se containing enzyme. Withdrawal of vitamins and minerals during the latter part of the finisher period has not affected pig performance responses, but studies with poultry suggest that the vitamin content of the meat may be reduced if the vitamins are withdrawn prior to marketing. High levels of vitamin E have been shown to improve pork quality, by reducing drip loss. Studies with vitamin C and Se have suggested that they may also be involved in pork quality.
