Mulberry (Morus sp.) of the family Moraceae is very economically important in Asian countries including Korea, because its leaf and fruit have been commercially used in sericulture and horticultural industries. Therefore it is necessary to develop the optimal production system for rapid and cost-effective propagation of mulberry. Our studies focused on establishing an acclimatization method for the successful plantlet production of new cultivar 'Cheongsu' which was transferred ex vitro after in vitro culture. In particular, effect of abscisic acid (ABA) addition into the last subculture medium on plantlet response to subsequent ex vitro transfer and its growth was investigated. During acclimatization, stomatal conductance and transpiration rate of ABA-pretreated plantlets were significantly lower than those of non-treated plantlets. Net photosynthetic rate of ABA-pretreated plantlets decreased after ex vitro transfer but increased after 14 days, and it was mostly higher than that of non-treated plantlets. Moreover, relative water content as well as chlorophyll contents and its ratio were also higher in ABA-pretreated plantlets. On the other hand, proline was considerably higher than in control plantlets. After 1 month of ex vitro transfer, survival rate of ABA-pretreated plantlets was 85.6%, which increased by 29.1% in comparison with control (56.5%). More vigorous growth was also observed in ABA-pretreated plantlets. From these results, it was found that application of ABA to the last subculture medium could improve acclimatization and promote survival of mulberry plantlets after ex vitro transfer, inducing water stress tolerance and alleviating abiotic stresses.
The spent mushroom substrate (SMS) of Lentinula edodes that was derived from sawdust bag cultivation was used as materials for controlling Phytophthora blight disease of pepper. Water extract from SMS (WESMS) of L. edodes inhibited mycelial growth of Phytophthora capsici, suppressed Phytophthora blight disease of pepper seedlings by 65% and promoted growth of the plant over 30%. In high performance liquid chromatography (HPLC) analysis, oxalic acid was detected as the main organic acid compound in WESMS and inhibited the fungal mycelium at a minimum concentration of 200 mg/l. In quantitative real-time PCR, the transcriptional expression of CaBPR1 (PR protein 1), CaBGLU (${\beta}$-1,3-glucanase), CaPR-4 (PR protein 4), and CaPR-10 (PR protein 10) were significantly enhanced on WESMS and DL-${\beta}$-aminobutyric acid (BABA) treated pepper leaves. In addition, the salicylic acid content was also increased 4 to 6 folds in the WESMS and BABA treated pepper leaves compared to water treated leaf sample. These findings suggest that WESMS of L. edodes suppress Phytophthora blight disease of pepper through multiple effects including antifungal activity, plant growth promotion, and defense gene induction.

This study evaluated the drought tolerance of Liriope platyphylla F.T.Wang & T.Tang, Dendranthema zawadskii var. lucidum (Nakai) J.H.Park, Hosta longipes (Franch. & Sav.) Matsum., Sedum sarmentosum Bunge and Zoysia japonica Steud. for an extensive green roof. In order to assess drought tolerance of green roof plants, several criteria were measured such as volumetric water content, leaf and soil moisture potential, chlorophyll a and b, chlorophyll fluorescence, photosynthesis, stomatal conductance, transpiration rate and antioxidants. The results of the drought tolerance measurement of green roof plants focused on the gradually withering of plants from lack of volumetric water content. D. zawadskii was the first to show an initial wilting point, followed by Z. japonica, H. longipes and L. platyphylla in order while S. sarmentosum showed no withering. It was concluded that H. longipes, L. platyphylla and S. sarmentosum were highly drought tolerant plants able to survive over three weeks. Furthermore, chlorophyll a and b were divided into two types: Type I, which kept regular content from the beginning to the middle of the period and suddenly declined, like H. longipes and Z. japonica; and Type II, which showed low content at the beginning, sharply increased at the middle stage and decreased, like D. zawadskii, L. platyphylla and S. sarmentosum. Volumetric water content and the amount of evapotranspiration consistently declined in all plant species. The analysis of chlorophyll fluorescence results that S. sarmentosum, which had relatively high drought tolerance, was the last to decline, while Z. japonica and S. sarmentosum withered after rapid reduction. At first, photosynthesis, stomatal conductance and transpiration rate showed high activity, but they lowered as the plant body closed stomata owing to the decrease in volumetric water content. Measuring antioxidants showed that when drought stress increased, the amount of antioxidants grew as well. However, when high moisture stress was maintained, this compound was continuously consumed. Therefore, the variation of antioxidants was considered possible for use as one of the indicators of drought tolerance evaluation.

This experiment was carried out to study the effect of the autumnal cutting times on the regrowth, the accumulated carbohydrate and dry matter yield of Italian ryegrass The results were summarized as follows: 1. In dry matter yield, the plot of earlier cutting was shown the highest yield (p<0.05), and that of the last-cutting was shown lower yield of dry matter than that the none-cutting plot. 2. TSC (Total Water Soluble Carbohydrate) content slightly decreased after the first cutting and gradually increased according to the regrowth, and then decreased again to the second cutting time. And also the TSC content levels of stubble, stem and leaf at one week before falling to sub-zero temperature were all the highest in the eariler cutting plot (p<0.01), and there was significant correlation between the TSC content level and the second harvested dry matter yield (p<0.05). 3. CGR (Crop Growth Rate) was decreased below $8^{\circ}C$. RLGR (Relative Leaf area Growth Rate) and NAR (Net Assimilation Rate) were both high during 30 days after regrowth, and low after regrowth in all plots. LAI (Leaf Area Index) rapidly increased during 50 days after cutting, and then slowly increased in all the plots, and maximum LAI was 3.4-5.8. Also dry matter yield increased in the plots having a high LAI to 70 days after cutting. 4. It was recognized that there were significant correlation between TSC, LAI, CGR, NAR, LWR (Leaf Weight Ratio) and the second harvested dry matter yield during the low temperature periods, and the degree of contribution to dry matter yield was in order of LWR>LAI>TSC>NAR>CGR.
The morphological, anatomical and physiological traits were eximined for Populus alba ${\times}$ glandulosa which is an important planting species in Korea. The results obtained are as follows: 1. External characters in the leaf shape and chaff shape in the catkin were inherited as incomplete dominance but nectar gland was inherited as dominance. 2. Among the 15 selected clones, 9 clones were male, 2 clones female and 2 clones monoecious. 3. There were well-developed cork layers and bast fiber bundles in the bark. 4. Primordial leaves composed of 3 layers of cells and those undifferentiated into palisade and spongy parenchymas differed in its origin. 5. Leaf scare consisted of two kinds of tissues; one is connected to vascular bundle and the other not to vascular bundle. Tissues which had been connected to vascular bundle were isolated with only 2 or 3 layers of cork cells from the outside. 6. There was complicated arrangement in the vascular bundle of petioles. 7. Growth of the hybrid was sensitively influenced by external temperature, day-length and amount of light. In particular, it was apparent in height growth. 8. Flatness, loam soils and a $60{\times}60cm$ spacing might be best factors for the growth of P. alba ${\times}$ glandulosa. 9. The rooting of 15 clones was dependant upon external factors. 10. The growth of P. alba ${\times}$ glandulosa was best at around 80% of soil moisture content on the basis of plot water capacity. 11. Temperature difference between inside and outside stems below 100cm during the winter was the greatest at the south among seasons and among directions. 12. The sap movement was markedly influenced by air temperature, relative humidity in forest stand and moisture content in stem. 13. Total sugars in the cortex changed with season but did not differ in the dircetion of the stem. 14. Isoperoxidase variations in the leaf were different among 15 clones. Thus, it may be useful as a criterium for clonal identification. 15. The rate of soil moisture content decreased at a rapid slope was faster than that at a slow slope. Poor growth of P. alba ${\times}$ glandulosa at the slope was probably due to depletion of soil moisture.
To investigate the germanium content in paddy soil in Gyeongnam province, 310 paddy soil samples were collected at 19 areas in Gyeongnam, Ulsan and Busan. Contents of germanium in paddy soils were analyzed in different topographies, soil types and soil textures. Average content of germanium in Gyeongnam was 0.24 mr/kg, those of Masan, Jinju and Hadong were above 0.30 mg/kg. Germanium content with different topographies were no difference. Germanium contents in different soil types were 0.27 mg/kg in well adapted soil and 0.23 mg/kg in poorly drained soil. Germanium contents in different soil textures were 0.27 mg/kg in silt loam and 0.23 mg/kg in sandy loam. To determine germanium content on agricultural product in the field, content of germanium in cereals, vegetables and fruits were analyzed. Germanium content of agricultural product was high in the order of cereals>vegetables>fruits. In case of vegetables, germanium contents were high in the order of leaf vegetables>root vegetables>fruit vegetables. Germanium contents were high with 62 and $65{\mu}g/kg$ in lettuce and young radish, respectively. To analyze the germanium content in medicinal plant, samples were collected from 19 medicinal plants at Hamyang areas. Germanium contents in Angelica keiskei, Ligusticum chuanxiong, Panax ginseng and Atractylodes macrocephala were relatively high with $100{\mu}g/kg$ above.

Severe droughts in spring have occurred frequently in Korea in recent years, exerting a critical impact on corn yield. Therefore, it is necessary to find physiological and/or molecular indicators of the response to drought stress in maize plants. In this study, we investigated the effects of water-deficit stress on two Korean elite F1 maize hybrids, Ilmichal and Gwangpyeongok, by withholding water for 10 days at tassel initiation. The water deficit drastically reduced the relative leaf water content, leaf number, leaf area, and stem length, leading to dry matter reduction. Moreover, it reduced the SPAD values and stomatal conductance of leaves in drought-stressed plants of both hybrids. Importantly, the number of leaves and SPAD value were non-destructive and easy to investigate in response to water-deficit stress, suggesting that they may be useful indicators for screening drought-tolerant genetic resources. We detected more than 100 spots that were differentially accumulated under drought stress. Of these spots, a total of 21 protein spots (≥1.5-fold) from drought-exposed maize leaves were successfully analyzed by MALDI-TOF-TOF mass spectrometry. Functional annotation using Gene Ontology analysis revealed that most of the identified proteins were involved in carbohydrate metabolism, stress response fatty acid catabolism, photosynthesis, energy metabolism, and transport. The protein expression levels were increased in both Ilmichal and Gwangpyeongok, except for triosephosphate isomerase, fructose-bisphosphate aldolase, and an uncharacterized protein. The lactoylglutathione lyase delta (3,5)-delta (2,4)-dienoyl-CoA isomerase was overexpressed in Gwangpyeongok only. The results obtained from this study suggest that the drought-specific genes may be useful as molecular markers for screening drought-tolerant maize genotypes.
To cope with increasing importance of water stress in food crop production, some physiological characteristics, their cultivar-differences and grain yield of winter barley cultivars in response to water stress during reproductive stages were studied employing three covered-barley cultivars, Milyang 12, Durubori, and Olbori, one naked-barley cultivar, Baegdong, and one two-row malting-barley cultivar, Hyangmaeg. The barley grown in pot-soil was conditioned for 10 days under water stress, varying the time of water stress : 20 days before heading, 10 days before heading and the time of heading. The decrease in growth due to water stress varied greatly with the cultivars and time of water stress. The greatest injury occurred when water stress was imposed for 10 days from 10 days before heading : the culm length of water-stressed plants have shown reduced by 85∼98% of the non-stressed; the number of spikes per plant by 52∼83%; the number of grains per spike by 71∼86%; 1,000-grain weight by 80∼84%; yield per pot by 60∼94%. The number of spikes per plant as one of yield components was most sensitively affected. As a whole, the drought resistance of cultivars was high in the order of Olbori> Milyang 12 and Durubori> Hyangmaeg>Baegdong. On rewatering the plants after termination of the water stress treatment the recovery rate of free proline content and relative turgidity of flag leaf were higher in 3 covered-barley cultivars, and lower in cultivars Baegdong and Hyangmaeg.
The study was investigated to compare growth characteristics, the antioxidant activity, and the triterpenoid content of two Centella asiatica cultivars ('Giant Tiger Care' and 'Good Tiger Care'). At 41 days after transplanting, lengths of leaf and petiole were significantly longer in 'Good Tiger Care' than in 'Giant Tiger Care'. However, the growth characteristics (leaf area, petiole thickness, petiole length, and weight) were greater in 'Giant Tiger Care' than in 'Good Tiger Care' at 104 days after transplanting (harvest time). Antioxidant activity and total phenol content in four extracts (WE, water extract; HWE, hot water extract; 50E, 50% EtOH extract; 70E, 70% EtOH extract) of the two cultivars were high in 70E of 'Giant Tiger Care'. As a result of the triterpenoids analysis, the major triterpenoids of the two cultivars were identified as madecassoside and asiaticoside. The total triterpenoid content was high in 50E and 70E of 'Giant Tiger Care' and 'Good Tiger Care', respectively but the total triterpenoid content was highest in Good Tiger Care'. However, at the 104 days after planting, the yield of 'Giant Tiger Care' was three times higher than that of 'Good Tiger Care'. In addition, the total triterpenoid content that can be produced in the same cultivation area (3.3m2) was 2.459mg in 50E of 'Giant Tiger Care', which was ~2.2times higher than that of 70E of 'Good Tiger Care' (1.103 mg). Thus, it is considered economical to cultivate 'Giant Tiger Care' which has the highest antioxidant activity and high total triterpenoid production per cultivation area.

Background: This research aims to study the effect of climate change on the phenology, growth, and physiological traits of Silene capitata Kom., a Korean endangered species II. This study increased $CO_2$ concentration in a closed glass greenhouse, with the daily mean temperature and $CO_2$ concentration respectively being $4.61^{\circ}C$ and 93.63 ppm higher than the outside temperature (ambient conditions, control). The seeds of S. capitata were sown in control and treatment environments in March 2013 while seedlings were transplanted into individual pots in May 2013. To research phenological changes, the first day of the flowering and ripening of the plants transplanted in 2013 and first day of leafing in 2014 were observed. The growth and physiological responses of mature leaves were also studied in 2013. Results: There was no difference in the first day of flowering, but the first day of ripening was earlier in the treatment group than the control group. There was no difference in the number of rosette leaves between the two groups, but leaf area was wider in the treatment group than the control group. Transpiration rate and stomatal conductance were higher in the treatment group than the control group, chlorophyll content decreased, and photosynthetic rate and water use efficiency were the same for both groups. As a result of simple regression analysis among the transpiration rate, stomatal conductance, photosynthetic rate, and water use efficiency, stomatal conductance increased when transpiration rate increased. Stomatal conductance increased with photosynthetic rate in the control unlike in the treatment group. The photosynthetic rate and water use efficiency increased with transpiration rate in the control group unlike in the treatment group. Furthermore, water use efficiency increased as photosynthetic rate increased in both groups. Conclusion: Due to high $CO_2$ concentration, the photosynthetic rate was no longer controlled by the stomata, which appeared to suppress the excessive production of photosynthetic products by reducing chlorophyll content. It is believed that the phenological responses of S. capitata under climate change conditions will advance and that stable growth will be difficult in regions lacking moisture due to the high transpiration rate.
