  • This study investigated the effects of mulberry leaf extract on lipid metabolism in rats fed a high cholesterol diet. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly assigned either to one of two normal diet groups, with (NE group) or without (N group) mulberry extract, or one of four high cholesterol groups containing 1% cholesterol and various levels of dietary mulberry leaf extract. The rats fed high cholesterol diets were subdivided into 4 groups according to level of mulberry extract; Mulberry extract free group (HC group), 0.8% mulberry leaf extract group (HCL group), 1.6% mulberry leaf extract (HCM group) and 3.2% mulberry leaf extract (HCH group). The rats were fed their respective diets ad libitum for 4 weeks. The levels of serum triglyceride, total cholesterol and LDL-cholesterol of the HC group were higher than mulberry leaf extract supplemented groups. In contrast, the levels of serum HDL-cholesterol in groups supplemented with mulberry leaf extract were significantly lower than that of HC group. Hepatic total lipids, triglycerides, and cholesterol were significantly higher in the high cholesterol groups compared to those of the normal group, but were lower in the HCL, HCM and HCH groups than in the HC group. HMG-CoA reductase activity was significantly decreased in the HC and HCL groups compared to the normal and NE groups. However, the activities in the HCM and HCH group were similar to that of the normal group. The activity of acyl-CoA-cholesterol acyl transferase (ACAT) was increased in high cholesterol groups compared to the normal group. However, the activity was lower for all of the high cholesterol groups fed mulberry leaf extracts, and was lowest for the highest supplemented group (HCH), with no significantly difference from the normal group. In conclusion, the reduction in serum and hepatic lipid composition by mulberry leaf extract may be due to its modulation of HMG-CoA reductase and ACAT activities.

  • The antimicrobial and antioxidant activities of different cultivars and parts of broccoli were investigated. The screening of antimicrobial activities for the floret and leaf extracts of 11 cultivars against selected gram positive (Staphylococcus aureus and Listeria monocytogenes) and gram negative bacteria (Escherichia coli O157:H7 and Salmonella enteritidis) were conducted. The AMG cultivar showed the highest antimicrobial activity against S. aureus with the floret and leaf extracts (12.83 and 13.00 mm). Antibacterial activity against L. monocytogenes was strongest with floret extract of NJ cultivar (13.58 mm) and leaf extract of YDR cultivar (13.92 mm). Moreover, the size of inhibition zone against L. monocytogenes was bigger than those of 4 kinds of pathogenic bacteria. Both floret and leaf extracts of Grd cultivar showed the highest antimicrobial activity against E. coli O157: H7, but there was no difference between floret and leaf extracts. Floret extract o f NJ cultivar and leaf extract of NY and 0c cultivars were effective against S. enteritidis whereas leaf extract exhibited better antimicrobial effect than floret extract. These results showed that floret extract of NJ and leaf extract of 0c had the highest antioxidant activity which was 39.90 and 43.64%, respectively. The antioxidant activity of leaf extract was 1.5 times higher than that of floret extract. All cultivars except NJ showed that electron donating ability of leaf extract was higher than that of floret extract.

  • The natural dyeing of silk fabrics with guava leaf extract was investigated. Mixed components of flavonoid and tannin seem to be the components of guava leaf extract. The temperature and time for dyeing of silk fabrics with guava leaf extract were $40^{\circ}C$ and $90^{\circ}C$ for eighty minutes, respectively. The dyeing equilibrium was shown at the fourth time of repeated dyeing. The highest K/S value was recorded at pH 3. Fe-mordanted fabrics showed the highest K/S value. High K/S values were shown in the post-mordanting of dyed fabrics at $40^{\circ}C$ and the pre-mordanting of dyed fabrics at $90^{\circ}C$. After repeated dyeing and mordanting, various color change occurred with mordant treatment. Surface colors were changed to YR color in alkaline water extract and to Y color in acidic water and ethanol extract, respectively. The washing fastness was level 3-4 for Sn-mordanted fabrics and level 4 for Fe-mordanted fabrics, and the dry cleaning fastness also showed very excellent result with level 4-5. The rubbing fastness was satisfactory with level 3-4. The light fastness was above level 4 only when Fe-mordanting was conducted, and the rest of dyed fabrics was not fast enough. In the antibacterial activity, the powder of guva leaf extract showed 99.9% of high antibacterial activity and Staphylococcus aureus showed 99.6% in dyed fabric. All dyed fabrics showed higher deodorization and UV protection rate than control fabric.

  • This study was carried out to evaluate the physicochemical characteristics of fermented milks containing mulberry leaf extract. The mulberry leaf extracts were added to fermented milk at $0{\sim}2.0%$. The pH, acidity, numbers of viable cells, chemical composition and color values of fermented milk preparation were analyzed, and these samples were subjected to a taste test panel. As the ratio of mulberry leaf extract increased, the pH value of the fermented milk decreased proportionally and acidities increased significantly. The numbers of viable cells was highest in the fermented milk sample containing 0.5% mulberry leaf extract. In the chemical composition analyses, increases in the concentration of mulberry leaf extract led to a significant increase in crude protein, crude fat, and crude ash contents and a significant decrease in lactose content. Ca, Mg and K in the fermented milk were also significantly increased with the addition of mulberry leaf extract. As mulberry leaf extract amount increased, the lightness and redness values decreased, while the yellowness value increased. From the sensory evaluation of the fermented milk containing mulberry leaf extract, color, taste, texture and overall acceptability of the fermented milk sample containing 1% mulberry leaf extract was found to be much better than those of the other groups.

In vitro anticancer effects of persimmon leaf extracts (water extract at 8$0^{\circ}C$ for 3 hours, water extract at room temperature for 48 hours, 50% ethanol extract at 8$0^{\circ}C$ for 3 hours, 50% ethanol extract at room temperature for 48 hours, 75% ethanol extract at 8$0^{\circ}C$ for 3 hours and 75% ethanol extract at room temperature for 48 hours) on SNU16 (Korean gastric cancer cell) were investigated by MTT assay. Persimmon leaf extracts exhibited strong in vitro anticancer effects. We found that the higher the ethanol content of the solvent, the stronger the in vitro anticancer effects. Extraction yields, contents of flavonoids, vitamin A, vitamin C and vitamin E were measured. We found that the higher the ethanol content of the solvent, the higher the extraction yields and the contents of flavonoids, vitamin A and vitamin E. Among persimmon leaf extracts, 75% ethanol 8$0^{\circ}C$ extract showed the highest extraction yield, the highest contents of flavonoids, vitamin A and vitamin E and exhibitied the strongest in vitro anticancer effect on SNU16. Therefore, 75% ethanol 8$0^{\circ}C$ extract was chosen as the material to investigate in vivo anticancer effects. In vivo anticancer effect of persimmon leaf 75% ethanol 8$0^{\circ}C$ extract was investigated in SNU16 transplanted nude mice. Twenty five female nude mice (BALB/c) were blocked into five groups according to body weight and raised for 4 weeks with diets containing 4% (w/w), 8% (w/w) persimmon leaf 75% ethanol 8$0^{\circ}C$ extract, with IT (intratumoral) injection treatment with 1.65 mg/100 $\mu$1, 3.3 mg/100 $\mu$1 concentration every other day 3 weeks after SNU16 was transplanted. Persimmon 75% ethanol 8$0^{\circ}C$ extract significantly lowered tumor weight and tumor volume in SNU16 transplanted nude mice. Tumor weight and tumor volume in all experimental groups were significantly lower than those in the control group. Helper T cell (CD4) levels of mice injected with 3.3 mg/100 $\mu$1 extract significantly increased. Cytotoxic T cell (CD8) levels in all experimental groups significantly increased and helper/cytotoxic T cell ratios in all experimental groups significantly decreased. Natural killer cell and MHC class II molecule in all experimental groups significantly increased. In conclusion, persimmon leaf 75% ethanol 8$0^{\circ}C$ extract exhibited strong in vitro and in vivo anticancer effects against SNU16 cells and it increased cytotoxic T cell, natural killer cell and MHC classII molecule in experimental groups in SNU16 transplanted nude mice.

  • To examine the possibility of using Musa sapientum L. (Banana) leaf extract as a cosmetic raw material, banana leaves grown in Jeju Island were extracted with 70% ethanol. Polysaccharides present in banana leaf extract were discarded by precipitation with cold ethanol. Polysaccharide-discarded banana leaf extract promoted procollagen and COL1A1 gene expression, but inhibited matrix metalloproteinase (MMP)-1 and MMP-2 gene expression in human skin fibroblasts when examined by real-time reverse-transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA). The active compound in banana leaves was identified by fractionation with various solvents. The chloroform fraction showed the highest anti-wrinkle efficacy and the active compound of chloroform fraction was identified as corosolic acid by NMR, FT-IR, EA, and HPLC-MS. In addition, banana leaf extract showed anti-oxidative efficacy with an IC50 value of 67.91 ppm, as determined by DPPH free radical scavenging assay. Finally, the anti-wrinkle efficacy of banana leaf extract-containing cream was confirmed by clinical tests. Based on these results, banana leaves could have an application as a cosmetic raw material with anti-wrinkle efficacy.

  • Objectives: This study assessed the cytotoxicity of the metallic mordant cupric acetate (CA) and the protective effect of Albizzia julibrissin (AJ) leaf extract on CA-induced cytotoxicity in NIH3T3 fibroblasts. Methods: For this study, cell viability and antioxidative effects such as the inhibitory activity of lipid peroxidation (LP) and superoxide anion-radical (SAR) scavenging activity were assessed. Results: CA significantly decreased cell viability in a dose-dependent manner, and the $XTT_{50}$ value was measured as $55.0{\mu}M$ of CA. The cytotoxicity of CA was determined as highly toxic by Borenfreund and Puerner's toxic criteria. The catalase antioxidant significantly increased cell viability diminished by CA-induced cytotoxicity in these cultures. Regarding the protective effect of AJ leaf extract on CA-induced cytotoxicity, AJ leaf extract remarkably increased the SAR scavenging ability and the inhibitory ability of LP. From these findings, it is suggested that oxidative stress is involved in the cytotoxicity of CA, and AJ leaf extract effectively protected CA-induced cytotoxicity via antioxidative effects. Conclusions: Natural resources like AJ leaf extract may be a putative therapeutic agent for treatment or alleviation of the toxicity induced by CA metallic mordant.
