It was recognized that lead intoxication reduces thiamine content in the brain of rat and this change produces the alterations of thiamine-related biochemical reactions. In the present study, it was tested whether the changes of myelin composition as well as seizure threshold induced by lead intoxication in rats may be related to these changes of thiamine status and thiamine related biochemical factors. Wistar rats were divided into five groups: Control group, Lead-treated group, Lead plus Thiamine-treated group, Thiamine-deficient group, Pyrithiamine-treated group. Each group was divided into three subgroups: 3, 7 and 16 week old group. Myelin protein and phospholipid, one of the compositions of myelin lipid, were measured in the myelin isolated from rat brain. Threshold of electric shock seizure was tested in each group. The amount of each myelin composition in lead-treated group and thiamine-deficient group was significantly lower than those of all the brains in control group, but recovery by supplementation with thiamine during lead intoxication was occurred only in the cerebellum of 3 week old animal. Thresholds of the electric shock seizure of lead treated group and thiamine deficient group in 3 and 7 week old rats were significantly lower than those of control group, while those of lead plus thiamine treated group were similar to those of control group.
In the present study, we tested whether lead intoxication induces change of the thiamine content and the seizure threshold in rats and the changes of seizure threshold are related to the changes of thiamine status. It was also tested whether administration of excessive thiamine could reverse the toxic manifestation of lead in rats. Four groups of Wistar rats were prepared: 1) control group, 2) lead treated group, 3) lead plusthiamine treated group, and 4) thiamine deficient group. Each group of animals was divided into three subgroups based on age: 3, 7 and 16 weeks. In each group, thresholds of electroshock seizure and thiamine contents in brain regions including telencephalon, brain stem, cerebellum were measured. Thiamine contents in brain regions of the lead treated group were significantly lower than those of the control group and thiamine treatment reversed the decrease back to the control level. Thresholds of the electroshock seizure of the lead treated group in 3, 7 week old rats and those of thiamine deficient group in 3 week old rats were significantly lower than those of the control group. These observations were reversed by the supplementation with thiamine. These results from the present study suggest that increased seizure sensitivity induced by lead intoxication in rats may be mediated at least in part through the changes of thiamine status.
This study was carried out to investigate the levels of copper, zinc, manganese, nickel, cadmium and mercury content in breast milk among urban, rural and industrial lactating women in Korea. A total of 59 samples, which were collected from 17 in urban, 20 in rural and 22 in industrial area, and from 21-38 years-old healthy lactating women, were analyzed by Rigaku Mercury Analyzer for mercury, and by atomic absorption apectrophotometry for the other metals. The results are summarized as follows : The mean trace metal contents in breast milk were determined to be 0.34$\pm$0.14 ppm for copper, 2.01$\pm$1.43 ppm for zinc, 8.49$\pm$5.11 ppb for manganese, 7.75$\pm$5.73 ppb for nickel, 1.65$\pm$2.42 ppm for cadmium, 34.45$\pm$26.71 ppb for lead and 0.90$\pm$0.68 ppb for mercury. For the trace metal content in breast milk by area, the highest of copper, zinc, cadmium and mercury content were in urban, the highest of manganese content was in industrial, and the highest of nickel and lesd content were in rural. For copper, zinc, manganese and lead content in breast milk by lactation period, the highest levels were found in under 4 weeks after lactating, and subsequently the levels declined as lactation progressed, but the levels of zinc and manganese content increased from over 25 weeks after lactating. For cadmium and mercury content in breast milk by lactation period, the lowest levels were found in under 4 weeks after lactating, the highest levels were found in 5-12 weeks after lactating, and subsequently the levels declined as lactation progressed. For nickel content in breast milk by lactation period, the highest level was in 13-24 weeks after lactating, the lowest level was in 5-12 weeks after lactating.
In order to investigate the content of heavy metals in soil according to vertical profiles, 72 soil samples were collected from 4 sampling sites : Ulchiro-2 ga, Dugdo, Sungnaedong, and Amsa- dong. The content of mercury was measured by a mercury analyzer and those of lead, copper and zinc were measured by an atomic absorption spectrophotometer. From the results of this study, it was shown that the content of heavy metals decreased gradually from the surface soil to the 2 m- depth soil. But, there was no difference in the contents of the heavy metals in the 3m, the 4m and the 5m-depth soil.
This study was performed to investigate the effect of dietary calcium level on cadmium and lead toxicity in rats. Fifty-four male rats of Sprague-Dawely strain weighing 152$\pm$12g were blocked into 9 groups according to body weight, and were raised for 30 days. Nine experimental diets different with cadmium(0%, 0.04%), lead (0%, 0.071%) and calcijm(0.5%, 1.0%, 1.5%) levels were prepared. The results are summarized as follow. Weight gain, F.E.R.(food efficiency ratio), and weights of liver, kidney and femur were lower in cadmium exposed groups than those of heavy metal free groups. Weight gain F.E.R. and ash weight of lead groups were lower than those of heavy metal free groups. But, these were increased with increasing dietary calcium level. Cadmium and lead concentrations in blood, liver, kidney and femur were lower in rats fed 1.5% calcium than 0.5% calcium diet. Fecal cadmium and lead excretions were remarkably increased in 1.5% calcium groups, and cadmium and lead retention rates were decreased in 1.5% calcium groups. Metallothionein concentrations in liver, kidney and small intestine were higher in rats exposed to cadmium and lead. Calcium content in blood, femur and daily urinary and fecal calcium excretion were decreased by cadmium and lead additions, and increased in 1.5% calcium groups. Creatinine clearance were decreased with cadmium administratino and calcium addition. In conclusion, weight gain and organ weights were decreased with cadmium or lead administration. But, cadmium administration was more toxic than lead adminstration. Cadmium or lead toxicity was alleviated by increasing dietary calcium level. Especially, lead toxicity was alleviated in proportion to dietary calcium level.
The effect of various supplies of lead singly and in combination with aluminium on growth and chlorophyll biosynthesis was investigated in 7-day-old Vigna anguluris seedlings. Expose to 50 $\mu$N Pb or more drastically reduced root elongation rate. Significant depressions in root growth was observed within 1 day and no recovery of growth was seen over the duration of treatment period. Root elongation decreased depending on the Pb concentrations. Root growth inhibition was stronger than shoot growth inhibition. The initiation of lateral roots appeared to be more sensitive to Pb than the growth of main roots. Inhibition of root and shoot elongation by Pb was lessened by combined exposure of Pb and Al, suggesting that the presence of AA reverse the inhibitory effect of Pb alone. With the histochemical sodium rhodizonate method the rate of Pb uptake was dependent on the Pb concentration and exposure time of the roots to Pb salts. Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. 5-Aminolevulinic acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.

A highly sensitive, selective and rapid method for the determination of lead based on the reaction of lead (II) with 5-(4'-chlorophenylazo)-6-hydroxypyrimidine-2,4-dione (CPAHPD) and the solid phase extraction of the Pb(II)-CPAHPD complex with Amberlite XAD-2000 was developed, in the presence of pH 5.6 buffer solution and Triton X-114 medium. CPAHPD reacts with lead to form a violet complex with a molar ratio of 2:1 (CPAHPD to lead). This complex was enriched by the solid phase extraction with Amberlite XAD-2000. An enrichment factor of 500 was obtained by elution of the complex from the resin with a minimal amount of isopentyl alcohol(0.2 mL). In isopentyl alcohol medium, the molar absorptivity of the complex is $1.13{\times}10^6L\;mol^{-1}cm^{-1}$ at 647 nm. Beer's law is obeyed in the range of $5.0-160ng\;mL^{-1}$ in the measured solution. The relative standard deviation for 10 replicate samples of $50ng\;mL^{-1}$ level is 1.26%. The detection and quantification limits reaches 1.5 and $4.7ng\;mL^{-1}$ in the original samples. The presented procedure was successfully applied for determination of lead content in real samples such as vegetables, waters, biological and soil samples with satisfactory results.
The Food and Agriculture Organization (FAO) has been estimating the potential of insects as human food and animal feed for convincing food security since 2010. On account of this reason, Tenebrio molitor larvae have been gaining considerable attention as an alternative protein source for various foods. However, consumers do not prefer them because of their disgusting appearance. Therefore, it is necessary to develop a method that improve their appearance as to lead to T. molitor larvae consumption. In this study, T. molitor larvae which is prepared for optimally sampling conditions (shade drying, 30 min boiling after hot air drying, 30 min steaming after hot air drying), were cultured with Cordyceps militaris mycelia, and measured growth and density of mycelial. Also analyzed the cordycepin and adenosine content compared with commercial C. militaris. As a result, T. molitor larvae cultivated with C. militaris mycelia showed the highest cordycepin content (13.75 mg/g) was observed in optimal sampling conditions (shade drying). Therefore, we report that the methods which T. molitor larvae cultivated with C. militaris mycelia not only improve their appearance but also increase cordycepin content, which can be contributed to lead T. molitor larvae consumption.
