Selection of Low Toxic Chemicals among 47 Pesticides Used in Cucumber for Aphid Predator, Aphidoletes aphidimyza Rondani (Diptera: Cecidomyiidae) (진디혹파리에 대한 오이에 등록된 47종 살충제의 저독성 농약 선발)

  • From the results achieved by the evaluation of toxicities to an aphid predator, Aphidoletes aphidimyza Rondani eggs with 47 pesticides used for cucumber farming, 22 pesticides including emamectin benzoate showed low toxicity. 12 pesticides with flonicamid among 22 pesticides tested on eggs showed low toxicity (mortality, 0~2.7%) to A. aphidimyza lavae and indoxacarb + teflubenzuron and chlorantraniliprole showed toxicity of 35.4% and 64.6%, respectively. 14 pesticides including indoxacarb + teflubenzuron and chlorantraniliprole demontrating low toxicity against A. aphidimyza lavae showed low toxicity to A. aphidimyza pupae and adults as well.

Elephant Hawk-Moth (Deilephila elpenor L.) as a Herbivore of the Bog-bean (Menyanthes trifoliata L.), an Endangered Plant Species (멸종위기식물인 조름나물의 섭식자로서의 주홍박각시)

  • Even though many researches are conducted for the conservation and restoration of endangered species Menyanthes trifoliata, recently, there is no study on the threatening factors to this plant. This is the first time in Korea to study growth and feeding characteristics of Deilephila elpenor as a threatening factor to Menyanthes trifoliata through an experiment. Experiment was done with 6 Eephant hawk-moth larvae and change of body weight, food preference, and ingestion amount of Bog-bean were investigated. It took 27 days from larva to pupa and maximum body weight of lavae was in the range of 4-7.5g. The food preference sequence of the lavae was Menyanthes trifoliata, Impatiens balsamina, Ampelopsis brevipedunculata var. heterophylla, Parthenocissus tricuspidata. Ingestion model shows the total amount of ingestion by a larva is 11-30g and this amount can be acquired at $0.03-0.08m^2$ of Menyanthes trifoliata pure stand. This study showed Deilephila elpenor as a potential threatening factor and suggests that the conservation and restoration plan of endangered species Menyanthes trifoliata include the control plan of Deilephila elpenor, also.

Expression of Human Stem Cell Factor with Recombinant Baculovirus in BmN Cell Line and Silkworm

  • A recombinant transfer vector pBacSCF was constructed by inserting huamn stem cell factor (hSCF) cDNA into plasmid pBacPAK8. BmN cells were co-transfected with modified Bombyx mori, nuclear polyhedrosis virus (BmBacPAK) DNA and the recmbinant transfer vector to construct a recombinant baculovirus containing hSCE gene. DNA dot blotting and RNA dot blotting demonstrated that the hSCE gene was contained in the recombinant virus and transcribed. The recombinant baculovirus was infectious to BmN cells and to silkworm. SDS-PAGE analysis showed a specific band of expressed product in the extract of infected cells and in the heamolymph of infected larvae. Bioactivity of the recombinant hSCE was determined with W-1 cell line and MTT colorimetric method in synergy with interlukin-3 (IL-3). These results revealed that the hSCF gene was over-expressed in cultured cells and lavae of silkworm.

The Expression of Corazonin Neurons in Larvae Stage of Scuttle Fly

  • Scuttle fly which moves abruptly after standing for a while and stop suddenly to rush off again, is a fly species in the Phoridae family. This species like rotten organic materials and it is known to proliferate even in the industrial materials including organic solvents. These characteristic behaviors of the scuttle fly seem to be related to muscular and nervous system or neurotransmitters. Thus, we focused at the neurotransmitter, corazonin (Crz) that is known to be related to resistance to stress and investigated the developmental process of the neurons in the scuttle fly. Corazonin is a neuropeptide being expressed in the central nervous system (CNS) and is known to control mainly physiological functions and behaviors. Its many functions that have been proposed are still in controversy. In this studies, we found that there are three groups of corazoninergic neurons in the larval CNS of the scuttle fly and these neurons undergo distinguishable changes through metamorphic process compared to different fly species. Larva has 3 pairs of Crz neurons at the dorsolateral area of the brain, 1 pair at the dorsomedial brain and 8 pairs at the ventral nerve cord.

Seasonal Occurrence of the Tobacco Moth, Ephestia elutella Hubner (Lepidoptera : Pyralididae). (다색알락명나방의 발생소장에 관한 연구)

  • The tobacco moth, Ephestia elutella Hubner (Lepidoptera: Pyralididae), was first reported one of the serious of stored tobacco in Korea in 1939 (8). In recent years, tobacco moth lavae were observed infesting in all of the tobacco storage warehouses in this Country. Adults of the tobacco moth for the seasonal occurrence were studied in the three and four regions in 1981 and 1982, respectively. This species was caught from early May to late October in all regions for both years. In 1981, the first emergence peak was in late May to late at Andong, early June at Suweon and the second one in middle August in both regions. In 1982, on the other hand, there were three peak times, late May and July, and middle September. But the relate size of the peaks was different in two years; the second peak showed about 66% of the total number of tobacco moths captured in the whole year in 1981 at Suweon and 59% of those at Andong, while the first peak had 32% at Suweon and 57% at Shintanjin in 1982. This difference in relative peak sizes was discussed.

  • Here we report an antifungal peptide isolation from G. mellonella larvae. The peptide shows a high degree of sequence homology to an insect defensin, named heliomicin, first reported in Lepidoptera. The peptide was purified by a three-step procedure consisting of acid extraction, gel permeation chromatography and reversed-phase HPLC. First the N-terminal amino acid sequence of the purified peptide was determined by automated Edman degradation. (omitted)

  • For the investigation of seasonal fluctuation of immature (lavae and pupae) beanfly, infestation rates, and habit, several soybean-cultivars were planted at Kumgok Experiment Farm of KAERI by three different planting times at 25-day intervals. Infestation rates of beanflies were ranged from $85\%\;to\;100\%$ in accordance. with the planting dates, where an increase in infestation rate was found with delay in planting dates. Immature beanflies were already observed from lune 20 by plant dissection counts. Three peaks of the seasonal fluctuation of lava were shown by occurring on July 10, August 10 and 50 during the soybean cultivation, while two peaks of pupal appearances were found. The most high peaks of lavae and pupae occurred on August 10 and 30, respectively. The lava were habitable in the pith or cortex tissue of soybean stem. They prepared tiny hole on the axilla as well as the internode of stem, and then they pupated in the holes from which the adults are able to escape. Immature beanfly seemed to prefer invading to the underground part of the stem when the soybean plants were in young stage.

Morphological and Skeletal Development of Larvae and Juveniles of Entomacrodus stellifer lighti (Herre) (저울베도라치, Entomacrodus stellifer lighti 자치어(仔稚魚)의 형태(形態)와 골격발달)

  • The blenniid fish, Entomacrodus stellifer light(Herre) is distributed in the coastal waters of southern Japan and south-eastern Korea. Matured adults of blenniid fish were collected from the rocky shore of Namchun-dong, Nam-gu, Pusan, Korea in July, 1991. The observations were made on the morphology and skeletal development of the reared larvae and juveniles. The lavae reached 5.80mm in mean total length(MTL) in 10 days after hatching. A this time the larvae have absorbed the yolk completely and became postlarvae. Melanophores are distributed on the fin membrane of the lower part of pectoral fin and ventral margins of tail. The larvae averaged 6.20mm in TL and the caudal notochord flex at $45^{\circ}$ in 13 days after hatching. The part of the fin-fold of the future dorsal and anal fins became high. The lavae reached juvenile stage in 31 days after hatching and attained 15.80mm in MTL, and all fin-rays was formed. Ossification of the cranium took place at 6.20mm of MTL(11 days after hatching) in parasphenoid. Vertebrae began to develop from the anterior end to ossify posteriorly. Ossification of all bones nearly completed when the larvae reached to 15.80mm in MTL (31 days after hatching).

LIFE CYCLE AND CHEMOTHERAPEUTIC CONTROL OF A FILARIAN WORM, PHILOMETROIDES CARASSII PARASITIC IN CARASSIUS AURATUS (붕어에 기생하는 철사충 Philometroides carassii의 생활사와 치료에 관하여)

  • Philometroides carassii (ISHII, 1931) is a parasitic nematode of Carassius auratus and gold fish. Recently a large number of parasitized fishes by this nematode have been reported by fish farmers in Korea. The present investigation was aimed to find out the life cycles of the parasites and effects of chemotherapeutic medicines on the emerged larvae from the adult parasites. Results of the study are summarized as follows : 1. Intermediate host of p. cararsii is a freshwater copepod, Cyclops sp. and the invaded lavae in the body cavity of copepod are capable for parasitizing the fish host after 3 hours of invasion. 2. The lavae eaten directly by the fish host are digested or excreted. 3. The larvae invaded in the fish intestine through the intermediate host pass into the body cavity in 1 to 10 days, and become subadult stage. After 18 to 20 months the subadults parasitic in the body cavity of the Carassius auratus, penetrate through muscle fissile into tile caudal fin where they become adults. Whereas those parasitic in the body cavity of the gold fish, 7 to 8 months the sutadults penetrate through muscle tissue into the caudal fin where they become adults. 1. Male subadults found in the body cavity of the Carassius auratus and gold fish, were larger than females, This is the first record of male subadults occurring in the fish host. 5. Among four different :drugs tested, most of all larvae died in 4 ppm of dipterex in 5 days. But no adults were harmed even in 6 ppm dipterex in 11 days.

Cytotoxicity of Acetoxyscirpendiol from Paecilomyces tenuipes (Paecilomyces tenuipes로부터 분리한 Acetoxyscirpendiol의 세포사멸작용)

  • Paecilomyces tenuipes is one of the famous Chinese medicinal entomopathogenic fungi that parasite in the lavae of silkworm. A cytotoxic compound, 4$\beta$-acetoxyscirpendiol (ASD) was isolated from a methanolic extracts of Paecilomyces tenuipes. The ASD compound belongs to scirpenol subfamily of trichothecene mycotoxin. In a continuation of the elucidation of the mechanism of ASD, we report here the evidences of induction of apoptosis by ASD in human Jurkat T cell line. In MTT reduction assay for monitoring cell viability, ASD showed strong toxicity. The 50 percent inhibitory concentrations of ASD against human T lymphoid Jurkat cell was 59.5 ng/$m\ell$. Phosphatidylserine externalization was increased by ASD at 3 and 6 hrs when compared with that of 6 hrs in the cell line showing in a time-dependent manner. When whole lysates of cells treated with ASD were subjected to western blot assay, 113 kDa poly(ADP-ribose) polymerase (PARP) was significantly cleaved to 89 kDa fragment. Time-dependent DNA fragmentation was also observed when Jurkat T cells were treated with ASD at 100 ng/$m\ell$ for 6 hrs and 18 hrs at the ratios of 8.5% and 15.0%, respectively. From these data, Jurkat T lymphocytes treated with ASD from Paecilomyces tenuipes underwent typical cascades of apoptotic cell death.
