Response of Muscle Protein Synthesis to the Infusion of Insulin-like Growth Factor-I and Fasting in Young Chickens

  • In order to elucidate the physiological function of circulating IGF-I on muscle protein synthesis in the chicken under malnutritional conditions, we administrated recombinant chicken IGF-I using a osmotic mini pump to fasted young chickens and measured the rate of muscle protein synthesis and plasma metabolite. The pumps delivered IGF-I at the rate of $22{\mu}g/d\{300{\mu}g{\cdot}(kg\;body\;weight{\cdot}d)^{-1}\}$. Fractional rate of protein synthesis in the muscle was measured using a large dose injection of L-[$2,6-^3H$]phenylalanine. Constant infusion of chicken IGF-I did not affect plasma glucose level. Significant interaction between dietary treatment and IGF-I infusion was observed in plasma NEFA and total cholesterol concentrations. When chicks were fasted, IGF-I infusion decreased plasma NEFA and total cholesterol concentrations. On the other hand, IGF-I administration did not affect plasma levels of both metabolites. Fasting reduced plasma triglyceride concentration significantly. IGF-I infusion also decreased the level of plasma triglyceride. Plasma IGF-I concentration of young chickens was halved by fasting for 1 d. IGF-I infusion using an osmotic minipump for 1 d increased plasma IGF-I concentration in fasted chicks to the level of fed chicks. Fasting decreased body weight and the loss of body weight was significantly ameliorated by IGF-I infusion. There was a significant interaction between dietary treatment and IGF-I infusion in the fractional rate of breast muscle protein synthesis. There was no effect of IGF-I infusion on muscle protein synthesis in fed chicks. Muscle protein synthesis reduced by fasting was ameliorated by IGF-I infusion, but did not reach to the level of fed control. Muscle weight of fasted chicks infused with IGF-I was similar to fasted birds without IGF-I infusion, which suggests that muscle protein degradation would be increased by IGF-I infusion as well as protein synthesis in fasted chicks.

Capecitabine Pattern of Usage, Rate of Febrile Neutropaenia and Treatment Related Death in Asian Cancer Patients in Clinical Practice

  • Background: Oral capecitabine is increasingly replacing intravenous 5-fluorouracil in many chemotherapy regimens. However, data on the risk of febrile neutropaenia (FN) and treatment related death (TRD) with the drug remain sparse outside of clinical trial settings despite its widespread usage. This study aimed to determine these rates in a large cohort of patients treated in the University of Malaya Medical Centre (UMMC). Materials and Methods: We reviewed the clinical notes of all patients prescribed with oral capecitabine chemotherapy for any tumour sites in University Malaya Medical Centre (UMMC) from $1^{st}$ January 2009 till $31^{st}$ June 2010. Information collected included patient demographics, histopathological features, treatment received including the different chemotherapy regimens and intent of treatment whether the chemotherapy was given for neoadjuvant, concurrent with radiation, adjuvant or palliative intent. The aim of this study is to establish the pattern of usage, FN and TRD rates with capecitabine in clinical practice outside of clinical trial setting. FN is defined as an oral temperature > $38.5^{\circ}C$ or two consecutive readings of > $38.0^{\circ}C$ for 2 hours and an absolute neutrophil count < $0.5{\times}10^9/L$, or expected to fall below $0.5{\times}10^9/L$ (de Naurois et al., 2010). Treatment related death was defined as death occurring during or within 30 days of last chemotherapy treatment. Results: Between $1^{st}$ January 2009 and $30^{th}$ June 2010, 274 patients were treated with capecitabine chemotherapy in UMMC. The mean age was 58 years (range 22 to 82 years). Capecitabine was used in 14 different tumour sites with the colorectal site predominating with a total of 128 cases (46.7%), followed by breast cancer (35.8%). Capecitabine was most commonly used in the palliative setting accounting for 63.9% of the cases, followed by the adjuvant setting (19.7%). The most common regimen was single agent capecitabine with 129 cases (47.1%). The other common regimens were XELOX (21.5%) and ECX (10.2%). The main result of this study showed an overall FN rate of 2.2% (6/274). The overall TRD rate was 5.1% (14/274). The FN rate for the single agent capecitabine regimen was 1.6% (2/129) and the TRD rate was 5.4% (7/129). All the TRDs were with single agent capecitabine regimen were used for palliative intent. Conclusions: Oral capecitabine is used widely in clinical practice in a myriad of tumour sites and bears a low risk of febrile neutropaenia. However, capecitabine like any other intravenous chemotherapeutic agent carries a significant risk of treatment related death.

Diagnosis of fibroadenoma using radiolabeled bladder cancer specific peptide

  • Most tumors are believed to overexpress several receptors, and small peptides targeting these receptors were developed for diagnosis and tumor therapy during past decade. Here we report that fibroadenoma can be visualized by bladder cancer specific peptide. A 9-mer bladder cancer specific peptide, which was discovered from the phage display method, was synthesized by peptide synthesizer, and additional tyrosine was conjugated at the N-terminal for radioiodination (Y-BP). Y-BP was radiolabeled with $^{131/124}I$ using Iodogen tube. The rat treated with N-butyl-N-(4-hydroxybutyl)nitrosamine for 8 weeks was allowed to grow until large size tumor was developed under axilla. The tumor model was microPET imaged sequentially using [$^{18}F$]FDG and radioiodinated $^{124}I-Y-BP$. The tumor was excised and examined by immunostaining studies. Radioiodinated $^{124}I-Y-BP$ was purified using fast protein liquid chromatography (FPLC) in > 90% radiochemical purity. The whole tumor was well visualized by [$^{18}F$]FDG with several intense focal uptake within tumor. The tumor was also clearly seen with $^{124}I-Y-BP$ at 4 h post-injection, and to our surprise the tumor uptake of $^{124}I-Y-BP$ lasted up to three days. The tumor was diagnosed histologically as a fibroadenoma derived from mammary gland. In conclusion, the bladder cancer specific peptide showed the good potential as a new radiotracer for the detection of breast fibroadenoma.

레토르트식품, 분유, 비스킷 및 피자 내에 함유되어 있는 트랜스지방산 함량 분석 (Analysis of Trans Fatty Acid Content in Retort Food, Powdered Milk, Biscuit and Pizza Products)

  • 본 연구에서는 식품 내에 존재하는 트랜스지방산 함량에 대한 기초 자료를 수집하기 위하여 서울 지역에서 수거한 레토르트식품류 2종, 분유류 6종, 비스킷류 7종 피자류 3종을 선택하여 총 4종류의 18종의 시료를 분석하였다. 레토르트식품류 2종, 분유류 6종, 비스킷류 1종은 chloroform-methanol(CM) 추출법에 의하여 조지방을 추출하였으며 비스킷류 6종, 피자류 3종은 chloroform-methanol(CM) 추출법과 acid digestion 추출법을 각각 이용하여 조지방을 추출한 후 gas chromatography(GC)에 의해 트랜스지방산 함량을 분석하였다. 레토르트식품류의 총 지방 중 트랜스지방산 함량은 1-2.8%이며 분유류의 트랜스지방산 함량은 0.4-2.4%로서 제품의 브랜드와 제조회사에 따라 차이가 많이 났다. 비스킷류의 트랜스지방산 함량은 0-2.95%로서 제품 간 가장 많은 차이를 나타내었으며 이는 첨가물의 종류에 따라 달라지는 것으로 사료된다. 피자류에서는 2.8-3.45%의 높은 트랜스지방산 함유율을 나타내었다.

칠면조와 육계에서 청색증 발생기전에 관한 고찰 (Degenerative myopathy of the supracoracoideus (DMS) in turkeys and broiler chickens, Review)

  • Deep pectoral myopathy (DPM), also known as Oregon muscle disease or green muscle disease, was first described in 1968 by Dickinson et al as "degenerative myopathy" in turkeys. Even though this condition was first recognized in adult meat-type turkey and chicken breeders, it is becoming more and more common in meat-type growing birds. DPM occurs exclusively in birds that have been specially selected for breast muscle development. It is generally recognized that DPM is an ischemic necrosis that develops in the deep pectoral muscle (supracoracoideus or pectoralis minor muscle) mainly because this muscle is surrounded by inelastic fascia and the sternum, which do not allow the muscle mass to swell in response to the physiological changes occurring when muscle are exercised, as in wing flapping. The lesion does not impair the general health of birds and is generally found during cut-up and deboning, moreover, it can be both unilateral or bilateral, affecting just one or both pectoralis minor muscle, respectively. No public health significance is associated to DPM, but it is aesthetically undesirable. The fillet should be removed, whereas the rest of the carcass is still fit for human consumption. However, the required trimming operations determine the downgrading of the products and produce an economic loss for the industry, especially because it affects the more valuable part of the carcass. The incidence of DPM increases with market weight in broilers, with more cases reported in higher-yielding strains and in males. Increased bird activity (flock nervousness, flightiness, struggle, and wing flapping) induced by factors such as feed or water outages, lighting programs and intensity, human activity, and excessive noises in and around chicken houses should be looked at as a trigger for the development of DPM in broiler. However, most of the studies conducted to evaluate the incidence of DPM in poultry are concerned with parental commercial breeding stocks under experimental conditions (Bianchi et al. 2006. Poult Sci 85 : 1843-1846). There is a possible genetic relationship between the selection for large-breasted birds and this condition. Management procedures that discourage excessive wing flapping would reduce the incidence (Jordan and Pattison. 1998. Poultry diseases. 398-399).

대전지역 미취학 어린이의 비수유성 빨기 습관에 대한 조사 연구 (Non-nutritive Sucking Habits of Preschool Children in Daejeon)

  • 이 연구의 목적은 대전지역 미취학 어린이의 비수유성 빨기 습관의 유병률을 조사하고 이 유병률에 영향을 미치는 기여요인과의 관련성을 평가하며 비수유성 빨기 습관이 유치열의 교합에 어떤 영향을 주는지 알아보고자 하는 것이다. 구강검진을 위해 개인치과에 내원한 만 18개월에서 65개월 사이의 어린이 841명을 검사하고 그 보호자를 대상으로 설문조사를 시행하였다. 전체 설문에 대답한 어린이 중 32.8%에서 비수유성 빨기 습관이 있었으며 여아에서 37.7%로 남아보다 유의하게 더 높았다. 모유 수유의 기간이 길어짐에 따라 비수유성 빨기 습관 유병률은 유의한 감소를 보였으며 출생순서와 엄마의 직업유무는 관련성이 없었다. 손가락빨기가 전체 비수유성 빨기 습관 어린이 중 62.6%로 인공젖꼭지빨기(37.4%)에 비해 더 우세하였고 습관의 지속기간 또한 유의하게 길었다. 비수유성 빨기 습관의 지속기간이 길어질수록 유치열 교합이상이 더 많이 관찰되었다.

기능성 브래지어 패턴디자인 연구 -돌출.하수 유방유형을 중심으로- (A Study on the functional pattern design for brassiere-focusing on large-breasted women-)

  • The purpose of this study is to develop designs and patterns of a functional brassiere. For this purpose, women who have protruding and drooping breasts with 75B for their brassiere size were sampled. The fitting test for the fitness and function of test brassiere and control brassiere were carried out twice and compared the results from the both in terms of front, rear and side looks. The results of this study can be summarized as follows; 1. The results from measuring the level of sag and spread, and volume and protrusion of breasts shows that there are some distinctive differences in the measurements before and after wearing the test brassiere and the test brassiere is proved to be very effective in correcting breasts' shapes. 2. Front look: Since the two front cutting lines of the test brassiere served to have the curved main part pressing against the breasts, it could support breasts to more than satisfactory level. In addition, upperline panel and side panels of the test brassiere were found to have the upperline of the upper cup pressed against the breasts and at the same time it pressed and gathered the flab around upper part of the breasts and armpits that helped to rearrange the shape of the breasts. It is also proved that the two-pieced test brassier was found to be more functional in supporting the lower cup of the breasts. 3. Rear look: The test brassier was made up of two wide U-shaped wings with 2.5cm tape on its lower sides. Each wing has 4 lines and 3 hems (5.5cm). For this reason, the test brassiere was tighter and better fitted on its back. 4. Side look and entire look: The wings of the test brassiere might look wider than that of the control brassier which has conventional straight-shaped wings, but it was analyzed that the test brassiere held the upper and lower sides of the wings more effectively to be more pressed against the breast. Therefore, the test brassiere scored higher in terms of adjusting to body movements, while the control brassiere looked better in overall terms. 5. The functional brassier fur protruding and drooping breasts developed from the experiments of this study is a full side stretch brassiere which covers the entire breasts satisfactorily. Hadoop기반의 공개의료정보 빅 데이터 분석을 통한 한국여성암 검진 요인분석 서비스 (Analysis of Factors for Korean Women's Cancer Screening through Hadoop-Based Public Medical Information Big Data Analysis)

  • 본 논문에서는 공개의료정보 빅데이터 분석을 위해 클라우드 환경에서 아파치 하둡 기반의 클라우드 환경을 도입하여 컴퓨팅 자원의 유연한 확장성을 제공하고 실제로, 로그데이터가 장기간 축적되거나 급격하게 증가하는 상황에서 스토리지, 메모리 등의 자원을 신속성 있고 유연하게 확장을 할 수 있는 기능을 포함했다. 또한, 축적된 비정형 로그데이터의 실시간 분석이 요구되어질 때 기존의 분석도구의 처리한계를 극복하기 위해 본 시스템은 하둡 (Hadoop) 기반의 분석모듈을 도입함으로써 대용량의 로그데이터를 빠르고 신뢰성 있게 병렬 분산 처리할 수 있는 기능을 제공한다. 빅데이터 분석을 위해 빈도분석과 카이제곱검정을 수행하고 유의 수준 0.05를 기준으로 단변량 로지스틱 회귀분석과 모델별 의미 있는 변수들의 다변량 로지스틱 회귀분석을 시행 하였다. (p<0.05) 의미 있는 변수들을 모델별로 나누어 다변량 로지스틱 회귀 분석한 결과 Model 3으로 갈수록 적합도가 높아졌다.

활엽수림(闊葉樹林)에서 간벌(間伐)에 의(依)한 번식기(繁殖期) 조류(鳥類) 군집(群集)의 변화(變化) (Changes in Breeding Bird Community Caused by Thinning in Deciduous Forest)

  • 본 연구는 간벌로 인한 산림환경구조와 번식기 조류 군집의 변화를 파악함으로써 조류 군집을 보호 및 유지할 수 있는 합리적인 산림 시업 방안을 모색하고자 1997년 4월에서 1998년 8월까지 평창군 국유림내 활엽수 천연림 지역에서 벌채 전과 후 세력권 도식법을 이용하여 실시되었다. 간벌 전과 후, 수종구성에 있어서는 큰 차이가 없었으나 헥타당 임목의 밀도, 종 풍부도, 종다양도지수, 흉고단면적에 있어서는 차이가 있었다. 산림환경의 수직적 구조는 간벌 후 하층과 상층의 피도량이 증가하였으며 흉고직경의 분포 역시 변화하였다. 간벌 전에는 14종 23쌍의 조류가 번식을 한 반면 간벌작업 후에는 8종 12쌍의 조류가 번식을 하여 번식 종 수와 쌍수가 감소하였으며, 조류의 종다양도 지수 역시 2.57에서 1.98로 감소하였다. 또한 조류 군집의 영소길도와 채이길드에 있어서도 변화가 발생하였는데 이는 간벌로 인해서 대경목의 수목과 수관층이 제거됨에 따라 이들이 이용할 수 있는 둥지자원과 먹이자원의 감소와 서식지 구조가 변화했기 때문인 것으로 판단된다. 그러므로 산림 내에서 조류 군집의 보호 및 유지를 위해서는 상층임관과 대경목의 존치를 고려한 산림작업이 실시되도록 해야 할 것이다. 이러한 산림작업을 통해서 산림생태계 내에서 생물다양성을 확보할 수 있으며 또한 지속 가능한 산림 경영이 가능할 것으로 판단된다.

잣나무 클론 채종원에서 간벌 강도에 따른 개량효과와 유전다양성 (Genetic Gain and Diversity in a Clonal Seed Orchard of Pinus Koraiensis Under Various Thinning Intensities)

  • 잣나무 채종원의 유전간벌을 위하여 재적생장에 대한 개량 효과와 유효클론수를 이용한 유전다양성을 평가하였다. 유전 간벌은 클론의 일반조합능력으로 나타나는 육종가(breeding value), 암꽃과 수꽃 개화량을 이용한 종자생산 기여도, 클론별 ramet수 등를 이용한 클론별 변이를 기초로 하였다. 차대 검정 결과에 의하여 산출된 재적생장에 대한 클론별 일반조합능력(genetic value) 및 1991년부터 2003년까지 조사된 개화량 성적을 이용하여 잣나무 채종원(1981년 조성) 179클론에 대하여 간벌 강도에 따른 유전다양성 및 개량효과를 분석하였다. 이때 채종원 외부로부터의 화분오염율은 30%(gene migration = 15%)로 설정하였다. 그 결과 재적생장에 대한 개량효과는 간벌율이 10%에서 60%까지 높아짐에 따라서 점차 증가하였다. 하지만 이보다 더 높은 간벌 강도에서 현저한 개량효과 증가는 나타나지 않았다. 유효클론수는 간벌강도 10%에서 60% 사이에서는 40클론으로 나타났으며, 70% 간벌율 이상에서는 증가하거나 감소하는 변동을 나타냈다. 따라서 본 잣나무 채종원에 대해서는 50%에서 60% 수준의 간벌 강도가 적당한 것으로 판단된다.
