Biosorption of lead by Laminaria japonica

  • Characteristics of biosorption of lead by marine algae, Laminaria japonica, were examined. The biosorption capacity of lead by L. japonica was achieved up to 30% of its own weight and proportional to the initial lead concentration. However, the opposite result was shown in different initial weight of biomass. Ion exchange reaction between lead ions and calcium ions was observed on lead biosorption with Ca-Ioaded biomass. Stoichiometric coefficient, which can represent the exchange ratio between metal ions and protons during elution process, was determined as 1.39. Therefore, it was concluded that the reaction between lead ions already attached in biomass and protons in bulk solution was not fully stoichiometric ion exchange relation at elution process.

참다시마 다당체의 면역 증강 활성 (Immunomodulating Activity of Laminaria japonica Polysaccharides)

  • Laminaria japonica의 polysaccharides(LP)가 마우스 비장 세포에서 현저한 면역 증강 활성을 보였다. Balb/c 마우스 비장 세포를 mitogen인 Con A, T cell growth factor인 IL-2 및 alloantigen인 anti-C57로 자극한 뒤, LP를 농도별(0.25, 0.5, 1 mg/mL)로 첨가하여 마우스 비장 세포의 증식 활성을 측정한 결과, 비장 세포증식능이 유의적으로 증가함을 확인하였다. 이러한 결과를 근거로 항원 특이적인 세포 독성 T 림프구(CTLs)의 활성과 IL-2에 의해 야기되는 비특이적인 림포카인 활성 살해 세포(LAKs)의 세포독성능을 측정하였다. 작동 세포와 표적 세포의 비율을 10:1로 혼합한 뒤, LP를 농도별로 첨가하여 세포 용해 시 발생되는 LDH를 측정한 결과, 동종 항원 반응 비장 세포의 살해 세포 작용이 유의적으로 증가하였으며, LAKs의 활성 또한 유의적으로 증가함을 확인하였다. 그리고 T 림프구의 활성에 직접적으로 관여하는 IL-2의 생산능을 측정한 결과, LP를 첨가한 경우 농도 의존적으로 IL-2의 생산량이 증가됨을 확인하였다. 이상의 결과로, 참다시마에서 획득한 LP가 마우스 비장 세포의 T 림프구 및 NK 세포의 증식에 영향을 미치는 면역 증강 활성을 가지고 있음을 입증하였다.

Protective Effects of the BuOH Fraction from Laminaria japonica Extract on High Glucose-induced Oxidative Stress in Human Umbilical Vein Endothelial Cells

  • This study investigated the protective effect of the butanol (BuOH) fraction from Laminaria japonica (BFLJ) extract on high glucose-induced oxidative stress in human umbilical vein endothelial cells (HUVECs). Freeze-dried L japonica was extracted with distilled water, and the extracted solution was mixed with ethanol then centrifuged. The supernatant was subjected to sequential fractionation with various solvents. The BuOH fraction was used in this study because it possessed the strongest antioxidant activity among the various solvent fractions. To determine the protective effect of the BFLJ, oxidative stress was induced by exposing of HUVECs to the high glucose (30 mM) or normal glucose (5.5 mM) for 48 hr. Cell viability, lipid peroxidation, glutathione (GSH) concentration, and antioxidant enzyme activities such as catalase, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), and glutathion reductase (GSH-re) were measured. Exposure of HUVECs to high glucose for 48 hr resulted in a significant (p<0.05) decrease in cell viability, SOD, GSH-px and GSH-re and a significant (p<0.05) increase in thiobarbituric acid reactive substances (TBARS) formation in comparison to the cells treated with 5.5 mM glucose or untreated with glucose. BFLJ treatment decreased TBARS formation and increased cell viability, GSH concentration, and activities of antioxidant enzymes including catalase, SOD, GSH-px, and GSH-re in high glucose pretreated HUVECs. These results suggest that BFLJ may be able to protect HUVECs from high glucose-induced oxidative stress, partially through the antioxidative defence systems.

갯녹음 해역에 해중림 조성을 한 다시마의 생장과 성숙 (Growth and Maturation of Laminaria japonica Transplanted for Seaforest Construction on Barren Ground)

  • Barren ground is an abnormal phenomenon in coastal ecosystems where seaweeds are destroyed and largely replaced by coralline algae containing calcium carbonate components. To restore the seaweed forest, we attempted reconstruction in an area of Samcheok, Korea, where barrenness is extensive. We developed two methods of seaforest construction underwater longline and seed transplantation for the brown seaweed Laminaria japonica, a fast-growing edible seaweed. The blade length of L. japonica attached to an underwater long line was $93.9{\pm}38.2cm$ and blade weight $7.6{\pm}2.1g$ in February 2004. Seaweed size declined after August 2004. A Similar pattern was observed using the transplantation method with maxima of $179.3{\pm}40.3cm\;and\;14.9{\pm}3.2g$ for blade length and weight, respectively in July 2004. The transplanted seaweeds matured in October as did a wild population. These results indicate that transplanted seaweed acclimate to new environments.

알긴산이 Nannochloropsis oculata NIES-2145의 생장에 미치는 영향 (Effect of Alginate on the Growth of Nannochloropsis oculata NIES-2145)

  • The growth effect of Nannochloropsis oculata (N.oculata), unicellular microalgae, by alginate was investigated. Alginate was depolymerized with sulfuric acid ($H_2SO_4$) and heat ($121^{\circ}C$), simultaneously. Addition of 0.75% alginate oligomer depolymerized with 0.2 N $H_2SO_4$ showed the maximum yield and the growth rate of N. oculata. Chlorophyll content and reducing sugar was increased by alginate oligomer in a dose-dependent manner. Alginate oligomer promoted the growth of N. oculata, whereas the original alginate polysaccharides had no significant effect. Laminaria japonica (L. japonica) extract containing high level of alginate was also increased growth rate and chlorophyll content. $CO_2$ supply addition to L. japonica extract showed no change the growth rate, although addition to alginate oligomer showed prominently increased. N. oculata could use more saccharides in presence of $CO_2$ according to reducing sugar determination. From these results, it is useful to establish optimal condition for high cell density cultivation of N. oculata.

The Herbal Composition GGEx18 from Laminaria japonica, Rheum palmatum, and Ephedra sinica Inhibits High Fat Diet-Induced Obesity by Regulating Appetite Genes

  • The herbal composition Gyeongshingangjeehwan 18 (GGEx18), which is composed of three herbs, Laminaria japonica Aresch (Laminariaceae), Rheum palmatum L. (Polygonaceae), and Ephedra sinica Stapf (Ephedraceae), has been used as an anti-obesity drug in Korean local clinics. Thus, we investigated whether GGEx18 regulates obesity by suppressing appetite in high fat diet-induced obese C57BL/6J mice. Administration of GGEx18 to obese mice for 9 weeks significantly decreased body weight gain, epididymal adipose tissue weight, and food efficiency ratio. GGEx18 also caused a significant decrease in the circulating levels of leptin, which were increased by about 450% in obese control mice compared with normal lean mice. Concomitantly, GGEx18 decreased mRNA levels of a potent appetite-stimulating hormone neuropeptide Y, but increased an appetite-suppressing hormone pro-opiomelanocortin mRNA levels. These results suggest that GGEx18 may prevent obesity through regulating appetite in nutritionally obese mice.
