We isolated 1,916 strains of bacteria from gut and feces of abalone. The most active agarolytic bacterium, DAG-130, was identified from the gut of the abalone Haliotis gigantea. Of the bacteria harbored by both H. discus hannai and H. gigantea, 59% were agarolytic. There was no significant difference in the number of agarolytic bacteria isolated from abalone fed on the seaweeds Gelidium amansii, Laminaria japonica, or Undaria pinnatifida. Of the agarolytic bacteria, 72% were isolated from the guts of all sources tested while 43% came from the feces. The strain DAG-130 showed 100% identity with the bacterium Vibrio cyclotrophicus based on phylogenetic analysis of l6S rDNA. The bacterium produced monomers and oligomers from the agar substrate.
This study was conducted to evaluate the potential of a microbial fermentation procedure to improve the functional qualities of seaweeds. Aspergillus oryzae, which has been used in traditional Korean fermented foods, was inoculated and cultivated in an aqueous extract of sea tangle (Laminaria japonica). Fermentation of the sea tangle extract by A. oryzae for 4 days resulted in a 3-fold increase in $\gamma$-aminobutyric acid (GABA) content. GABA is known to be a bioactive compound. Fungal fermentation of the extract also enhanced its antioxidant activity and increased its total content of phenolic compounds. It was assumed that these changes stemmed from the biodegradation of active compounds of the sea tangle packaged within its rigid structural matrix or occurred as result of fungal fermentation. These results suggested that the application of microbial fermentation to the processing of seaweeds will help in the development of processed foods to meet consumer demands.
In 2006, we studied herbivore consumption rate and preference for algal species using laboratory experiments. In the no-choice feeding experiment, three herbivores (two sea urchins, Stronglyocentrotus nudus and Stronglyocentrotus intermedius and an abalone, Haliotis discus hannai) showed significantly (P<0.05) distinct preferences for four algae (Laminaria japonica, Undaria pinnatifida, Costaria costata and Ecklonia stolonifera). The alga preferred by S. nudus was L. japonica followed by U. pinnatifida, C. costata and E. stolonifera with respective daily feeding rates of $5.7{\pm}:0.85\;g,\;4.4{\pm}0.89\;g,\;3.1{\pm}0.39\;g\;and\;2.1{\pm}0.32\;g(mean{\pm}SE)$. A similar trend was found for S. intermedius. Interestingly, the herbivore consumption rates for the algae differed with or without competition. The competition stimulated the feeding activity for S. nudus, but lowered the activity for S. intermedius. The multi-choice feeding experiment confirmed that L. japonica was significantly preferred by two herbivores: S. nudus and H. discus hannai (Duncan's multiple range test, ANOVA, p<0.05).
Seaweed biomass was estimated in the rocky subtidal zone of Daejin on the mid-east coast of Korea using a destructive sampling method from February to November 2007. Monthly sampling was conducted at depths of 5, 10 and 15 m using 50 $\times$ 50 cm quadrats. A total of 73 species, including 5 chlorophytes, 25 phaeophytes, 43 rhodophytes and one seagrass were identified. Average seaweed biomass was 2,852.3 g wet wt. $m^{-2}$ and biomass values varied monthly from 1,189.8 g to 7,523.7 g. The species dominant in biomass were Laminaria japonica (481.7 g), Sargassum confusum (470.85 g), Undaria pinnatifida (422.57 g), Costaria costata (282.16 g), Odonthalia corymbifera (174.46 g), S. sagamianum (163.22 g) and S. horneri (122.18 g). The vertical distribution of algae was characterized by S. confusum and U. pinnatifida at 5 m, L. japonica and U. pinnatifida at 10 m, and O. corymbifera and L. japonica at 15 m depth. The C/P, R/P and (R+C)/P values were 0.20, 1.72 and 1.92, respectively.
Various bacterial strains were isolated from rice field ditch water, and their seaweed degrading activities were investigated. They were incubated in a liquid medium of sea tangle (Laminaria iaponica) and sea mustard (Undaria pinnatifida) powder for 3 weeks. Ratios of reduced sugar to total sugar of the liquid medium were measured once a week. Ratios of reduced sugar to total sugar of 27A311, 27C221 27A111 and 27B121 strains were highest. Accordingly, these four strains were incubated in 3 different liquid media of sodium alginate, sea tangle powder, and sea mustard powder for 3 or 4 weeks. The ratios of reduced sugar to total sugar and cell growth were measured once a week. Cell growth was higher in 27A111 and 27C221. Ratios of reduced sugar to total sugar was higher for 27C221 in the liquid mediums with sodium alginate and sea mustard powder, and for 27A111 in the liquid medium with sea tangle powder.
본 연구는 다시마의 활성성분을 열수추출하고, 그 추출물을 제 2형 당뇨병 환자에게 섭취시킴으로서 그들의 혈당, 혈중지질 및 항산화 체계 개선에 있어서 다시마추출물의 효능을 과학적으로 규명하고자 하였다. 다시마를 열수추출하고 동결건조한 후 캡슐화하여 1.4 g을 350 mg씩 4캡슐로 나누어, 아침과 저녁 식후에 2캡슐씩 제 2형 당뇨환자에게 12주간 섭취 시켰다. 위약군은 동량의 cellulose를 실험군과 동일하게 제조한 후 같은 방법으로 섭취하게 하였고 이중맹검법을 사용하였다. 섭취 전후 일반사항 및 식이섭취조사, 신체계측을 실시하였고, 혈액을 채취하여 혈당, 당화혈색소, 혈청지질, 지질과산화물 및 항산화 효소의 변화와 독성검사인 간 및 신장 기능의 변화를 측정하였다. 최종 연구는 총 37명으로 실험군 17명, 대조군 20명으로 양군간의 baseline 특성은 유의적 차이가 없었다. 다시마추출물 섭취군에서 공복혈당은 12주간 섭취 후 $156.0{\pm}15.2mg/dL$에서 $155.8{\pm}14.2mg/dL$로 나타났고, 당화혈색소 역시 $6.74{\pm}0.66%$에서 $6.65{\pm}0.64%$으로 유의적으로 감소하지 않았다. 다시마추출물의 섭취가 12주간 이루어지면서 총콜레스테롤은 $177.0{\pm}16.5mg/dL$에서 $168.6{\pm}15.0mg/dL$로 유의적 (p<0.05)으로 감소한 반면에, 위약군의 경우 $175.8{\pm}17.4mg/dL$에서 $179.8{\pm}19.9$로 변화되었다. 또한 다시마추출물 섭취군의 LDL-콜레스테롤, 중성지방의 변화에서도 섭취 전에 비해 섭취 후 유의적 (p<0.05) 감소를 나타내었으며, 특히 LDL-콜레스테롤의 농도를 고농도군, 정상군, 저농도군으로 분류시 LDL-콜레스테롤 농도가 높은 환자일수록 감소의 폭이 큼을 알 수 있었다. 지질과산물 농도의 변화를 보면 다시마추출물 섭취군은 $1.58{\pm}0.88nmol$에서 $1.13{\pm}0.03nmol$로 유의적(p<0.05)으로 감소되었고, GSH-px활성도 위약군에 비해 다시마추출물 섭취군은 $13.98{\pm}0.66I.U/mg$ protein에서 $14.58{\pm}0.22I.U/mg$ protein로 활성도가 약간 상승하였다. SOD 활성은 실험군($1.18{\pm}0.22$ vs. $1.28{\pm}0.18unit/mg$ protein)에서 유의적(p<0.05)으로 증가하였으며, 간 및 신장 기능 검사에서도 실험 전후 모든 군이 정상범위를 나타내었다. 따라서 본 연구는 당뇨병 환자들의 혈청지질 및 혈액 항산화 효소계에 도움을 줄 수 있는 안전한 보조제로서 다시마추출물의 사용 가능성을 제시하였다.
본 연구는 생리활성이 높은 다시마와 소비를 촉진해야 할 쌀을 이용하여 햄버거 패티가 지닌 고에너지, 고지방 및 고콜레스테롤의 문제점을 해결하면서 건강기능성이 향상된 패티를 개발하고자 수행되었다. 표준패티는 지방을 첨가하지 않는 재료 조성으로 제조하였고, 세 종류의 실험패티는 표준패티 재료 중 육류의 2.5%를 다시마 분말로 대체한 패티 L과 육류의 25% 또는 50%를 동 분말을 함유한 밥으로 대체한 패티 LI과 LII이었다. 이들 세 종류의 실험패티에 대해 일반성분과 나트륨 및 콜레스테롤 함량을 분석했고, 품질 특성을 측정했으며, 관능성과 기호도를 평가했다. 실험패티의 일반성분은 표준패티와 크게 달라졌다. 즉 탄수화물 함량은 L은 낮았으나 LI과 LII는 높았고, 조지방과 콜레스테롤 함량은 세 실험패티 모두 낮았는데 LII와 LI은 더욱 낮았으며, 조단백 함량은 LI과 LII만 낮았다. 회분과 나트륨 함량은 세 실험패티 모두 높았는데 이중에서도 L이 가장 높았다. 실험패티의 품질특성도 표준패티와 상당히 변화되었다. 즉 가열 후 중량 감소율은 세 실험패티 모두 낮았는데 LII가 가장 낮았으며, 직경 감소율은 세 실험패티 모두 적었고, 반면에 두께 감소율은 L과 LII가 높았다. 색도는 세 실험패티 모두 명도와 적색도가 낮은 반면에 황색도는 높았다. 수분 함량은 세 실험패티 모두 높았는데 특히 LI과 LII가 크게 높았다. pH 값은 세 실험패티 모두 높았다. 경도는 L은 높았으나 반대로 LI과 LII는 낮았다. MDA 함량은 세 실험패티 모두에서 낮았으며, LII, LI 및 L 순으로 낮았다. 패티의 관능평가 결과, LI과 LII는 표준패티에 비해 대칭성, 색상, 윤기, 경도 및 다즙성이 우수하였고, 향미, 이취, 고소함, 느끼함, 맛, 이미 등 6개 항목을 같았다. 패티의 기호도 평가에서는 세 실험패티 모두 표준패티에 비해 윤기가 높았고, 이외에 전체 풍미를 포함한 11개 항목은 동일하였다. 이들 패티를 이용하여 제작한 버거의 기호도 역시 전체 풍미를 비롯한 10개 항목은 모두 차이를 보이지 않았다. 이러한 본 연구결과는 패티 재료 중 육류의 2.5%를 다시마 분말로 대체하거나 25% 또는 50%를 동 분말을 함유한 밥으로 대체하면 패티의 수분 함량은 높아지고, 에너지와 지방 및 콜레스테롤 함량은 낮아지며, 품질 특성에 긍정적인 효과가 있고 또한 관능성이나 기호도에 바람직한 영향을 끼친다는 점을 확인해주었다. 이러한 결과는 다시마 분말이나 동 분말을 함유한 밥을 이용한 건강기능성 패티의 개발이 매우 긍정적이며 가능성이 높음을 알려준다.
갈조류에서 에스트로겐 활성소재를 평가하기 위하여 in vitro 검출시스템을 사용하여 에스트로겐 효과를 가지는 분획을 검증하였다. 각각의 갈조류의 분획물들은 에탄올 추출로부터 물, 헥산, 부탄올 그리고 메탄올의 용매를 이용한 분획과정을 통해 제조하였다. 갈조류들의 수층 분획물에서 가장 높은 에스트로겐 활성을 보였다. 미역과 다시마의 수층 분획물들은 $500\;{\mu}g/ml$의 농도에서 $10^{-7}\;M$의 표준물질($17{\beta}$-estradiol)과 비슷한 활성을 보였으며, 곰피와 돌자반의 수층 분획물들은 $500\;{\mu}g/ml$의 농도에서 $10^{-8}\;M$의 표준물질보다 높은 활성을 보였다. 이 연구의 결과로 갈조류인 미역, 다시마, 곰피 및 돌자반의 수층 분획물에 에스트로겐 대체 작용을 할 수 있는 성분이 포함되어 있는 것을 확인하였으며, 이를 이용하여 폐경기 장애를 예방할 수 있는 에스트로겐 활성소재의 개발이 가능할 것으로 기대된다.
본 연구는 햄버거 패티 재료 중 육류의 일부를 다시마 분말 또는 다시마 분말을 함유한 밥으로 대체하여 식후 혈당과 혈장 지질 농도에 미치는 영향이 개선되는지 알아보고자 수행되었다. 표준패티 C와 시험패티 L, LI 및 LII를 제조하였다. L은 육류의 2.5%를 다시마 분말로 대체하였고, LI과 LII는 각각 육류의 25%와 50%를 다시마 분말을 함유한 밥으로 대체하여 제조했다. 인체시험 대상자는 외견상 건강하고 혈당과 혈장 지질 농도가 정상이며 본 실험에 참여하고자 자발적으로 동의서를 작성한 성인 여성 10명이었다. 이들 대상자에게 매 시험 시작 전 사흘 동안 영양적으로 균형 잡힌 식사를 제공했다. 각각의 패티를 200 g씩 섭취하게 하였으며, 섭취 전에 공복 혈액을 채취하였고, 섭취 후 30분, 60분, 120분, 180분 및 240분에 채혈하였다. C는 섭취 후 240분까지 혈당을 크게 올렸고 중성지방 농도는 미미하게 올렸으며, LDL-콜레스테롤 농도를 상당히 높인 반면에 HDL-콜레스테롤 농도를 현저하게 낮추어 총 콜레스테롤 농도를 약간 낮추는 결과를 나타냈다. 이에 비해 L은 혈당을 전혀 높이지 않았으며, 중성지방 농도의 저하 효과는 거의 없었으나 총 콜레스테롤 농도를 크게 낮추었는데, 이는 HDL-콜레스테롤 농도도 떨어졌지만 LDL-콜레스테롤 농도의 저하가 크게 기여한 결과였다. 한편 LI과 LII는 혈당은 C와 비슷하게 올렸지만, 중성지방 농도를 현저하게 낮추었고, 총 콜레스테롤 농도를 L과 비슷한 양상으로 낮추었다. LI과 LII는 L에 비해 비록 통계적 유의성은 없었으나, LDL-콜레스테롤 농도의 저하 효과가 약한 편이었다. 반면에 HDL-콜레스테롤 농도를 낮춘 효과는 비슷했다. 이러한 연구결과는 햄버거 패티의 재료 중 육류 일부를 다시마 분말로 대체하거나 또는 다시마 분말을 함유한 밥으로 대체함으로써 식후 혈당이나 혈장 지질 양상을 개선시키는 건강기능성 패티의 개발 가능성을 증명해준다. 혈당 상승을 경계해야 할 사람에게는 혈당저하 효과가 강력한 다시마 분말로 육류 2.5%를 대체한 L이 적합하며, 중성지방이나 콜레스테롤 농도의 상승을 억제해야 할 사람에게는 다시마 분말을 함유한 밥으로 육류의 25% 또는 50%을 대체한 LI과 LII가 유용할 것이다. 즉, 혈당이나 혈장 지질 농도가 정상인 사람에서 확인되었는바 고혈당이나 고지혈증을 지닌 사람에서는 그 효과가 더 크게 나타날 것이라 추측되며 향후 확인해야 할 과제라고 생각한다.
This study was conducted to optimize the processing conditions, including the ingredient ratio and extraction time, for a water-soluble fraction of Alaska pollock head and non-forming sea tangle by response surface methodology. Our results indicated that the optimal independent variables for obtaining extracts with a high yield and desirable sensory characteristics were 1.32 for $X_1$ (extraction time), 1.36 for $X_2$ (sea tangle concentration) and 0.93 for $X_3$ (water volume) in coded values, and 5.48 h for $X_1$, 18.18% for $X_2$ and 6.86 times for $X_3$ in uncoded values. The predicted values of $Y_1$ (yield), $Y_2$ (TCA soluble-N) and $Y_3$ (overall acceptance) for extracts produced under these optimized conditions were 22.10%, 1.83 g/100 mL and 5.9, respectively, their experimental values were 21.4%, 1.7 g/100 mL and 5.7, respectively. No significant differences between the actual and predicted values were found.
