ULTRASTRUCTURAL STUDY ON THE EFFECT OF RADIATION IN THE RAT FETUS TONGUE. (방사선조사가 태내백서의 설조직에 미치는 영향에 관한 전자현미경적 연구)

  • The author observed the effects of /sup 60/Co irradiation on the development and subcellular structure of tongue tissue of the fetal rats. The lower left abdomen of mothers were exposed to radiation on 15½th day of gestation with 300R. The fetuses were removed on the 6hr, 14hr, 24hr, 48hr and 72hr after irradiation and the light microscopic and electron microscopic observations of the lingual epithelium, lamina propria and muscle layer were carried out. The results were as follows: 1. The irradiated fetuses showed the retardation of filiform papillae formation. 2. Epithelial cells revealed fusion and myelination of mitochondria, large autolysosomes, increased lipid droplets, retardation of tonofilaments and desmosome formation. 3. In the lamina propria, undifferentiated cells showed bleb formation of nuclear membrane, pyknosis and fragmentation of nucleus, edema of cytoplasm I and nucleus, increased auto-lysosomes, dilatation of cell membrane and cell necrosis. Also, collagenous fibril formation was inhibited by irradiation. 4. In the muscle layer, growth of myotubes was inhibited. Myotubes showed swelling of mitochondria, loss of mitochondrial cristae, autolysosomes, retardation of myofibril formation, and large vacuoles. Undifferentiated cells adjacent myotube contained pyknotic nucleus and autolysosomes. 5. Among the various tissues of tongue, it seems that mesenchymal cells were most radiosensitive.

Fibrillar Collagens in Vocal Polyp and Nodule (성대폴립 및 결절에서의 원섬유성 교원질 (Fibrillar Collagen)의 발현양상)

  • Benign vocal fold lesions such as polyps, nodules and edema are known to be caused by vocal trauma such as voice misuse and/or abuse. Even though these lesions are known to be caused by the same etiology, phonotrauma, they show widely different clinical features and different responses to voice therapy. Previous studies suggested that benign vocal fold lesions represent disturbance in the balance of the extracellular matrix(ECM) constituents of the vocal folds. Collagen is one of the major constituents of ECM. Among collagens, fibrillar collagens are most important ones for maintaining the structural integrity. On the basis of gross morphology, vocal polyps wert divided into angiomatous one and edematous one, and nodules were divided into conical one and sessile one. In these four groups, the pattern of distribution of various fibrillar collagens(type 1, 2, 3, 5) was studied by immunohistochemical staining using paraffin embedded tissues. Within each group, differences among collagen subtypes were insignificant. In edematous polyp, collagens were sparsely dispersed in lamina propria by diffuse edema. In angiomatous polyp, collagens were displaced into submucosal layer by hemorrhagic space. In nodules, collagens were stained compactly in lamina propria. Quite different distributions of fibrillar collagens between polyps and nodules are thought to suggest that vocal polyps and nodules are totally different disorders in their pathophysiology.

Pathological characteristics on porcine enteritis associated with porcine circovirus type 2 in Jeju (제주지역의 돼지 써코바이러스 2형 관련 장염에 대한 병리학적 특징)

  • From 2006 to 2009, 50 pigs suspected of enteritis associated with porcine circovirus type 2 (PCV-2) (EAPC) were collected. Gross and histopathologic examinations and immunohistochemistry (IHC) were performed on the small intestine of 50 pigs. The pigs with EAPC were concentrated in grower pigs (68%), and diarrhea (84%) was the most common clinical sign. Grossly, the walls of the small intestine were thickened, and mesenteric lymph nodes were enlarged. The histopathologic features in the small intestine with EAPC were characterized by lymphoid depletion, histiocytic infiltration, and formation of basophilic grape-like inclusion bodies and multinucleated giant cells in gut-associated lymphatic tissue (GALT) and lamina propria. According to IHC, PCV-2 antigens were more intense and distributed widely in GALT (98%) than lamina propria (82%) of the small intestine. Co-infection with other enteric pathogens was detected in 26 pigs (52%). Twenty-four pigs (48%) were confirmed to be a PCV-2 single infection. Collectively, infected PCV-2 in the small intestine may induce an immunosuppressive status of individuals and then allow secondary co-infections in the digestive system. This study shows that PCV-2 can induce diarrhea and enteric lesions in pigs without a co-infection of other enteric pathogens.

Gastric mucosal immune response of Helicobacter pylori-infected children (Helicobacter pylori 감염 소아에서 위점막 면역반응)

  • Purpose : Helicobacter pylori infection is one of the most common gastrointestinal infections worldwide; it almost invariably causes chronic gastritis. Pediatric studies may provide important insights into the mucosal immune response of H. pylori-infection, as children are not submitted to environmental factors such as alcohol, tobacco and anti-inflammatory medication. The aim of the present study was to investigate the mucosal immune response against H. pylori in clinically well-defined groups: H. pylori-positive (divided into peptic ulcer disease and gastritis) and H. pylori-negative control. Methods : Antral biopsies were obtained from 45 children undergoing an upper GI endoscopy for dyspeptic symptoms. T cells (CD3+, CD4+, CD8+) and B cells (CD20+) were analyzed by quantitative immunohistochemistry. The correlation of lymphocyte subsets of gastric mucosa with histology was evaluated. Results : T cells (CD3+, CD4+, CD8+) and B cells (CD20+) were significantly increased in the lamina propria of H. pylori-positive group (P<0.01). CD8+ T cells were significantly increased in the lamina propria of the H. pylori-positive peptic ulcer disease (P<0.01). Within the epithelium, only CD4+ T cells were significantly increased in the H. pylori-positive group (P<0.01). Gastric histological parameters had a closer correlation with lymphocytes in the lamina propria than intraepithelial lymphocytes. Conclusion : This study suggests that both T cells and B cells in the lamina propria play important roles in the local immune response of H. pylori-infected children. Furthermore, it remains to be elucidated whether CD8+ T cells in the lamina propria may contribute to peptic ulcer formation in H. pylori-infected children.

  • The author observed the distribution of mast cells in the hard palate wound healing of healthy male albino rats weighing about 100gm. The mild wounds were made antero-posterior linealy by sugical knife. The results were as follow : 1. At the from three days to five days after wound, the number of mast cells were markedly increased. 2. In the wound healing process, mast cells were more distributed in the lamina propria than in the submucosa.

  • This study was carried out to investigate the distribution and morphological properties of mast cells in the ileum of Korean pheasant. In the ileum mast cells were located in the lamina propria of mucoase. The cells were mainly oval with irregular cell surface occasionally spindle-like or triangular in shape. Some mast cells had one or more tail-like long cytoplasmic processes. All mast cells had many blue granules stained by toluidine blue in the cytoplasm.

  • The present study was designed to investigate the morphological developments of the stomach in the prenatal and postnatal rats. The gastric fundus of 16- to 22-day-old fetuses, neonates, 7-day-old, 14-day-old, 21-day-old, and adult rats were observed by transmission electron microscopy. 1. In the 19-day-old fetuses, the gastric pits and the parietal cells were the primitive state, but the parietal cells contained numerous microvilli and mitochondria. 2. In the 20-day-old fetuses, the intracellular canaliculus appeared in the parietal cells, and glycogen granules were localized mainly in the basal part of the epithelial cells and parietal cells. 3. In the 20-day-old fetuses, the mucous granules and zonular occludens appeared in the gastric glandular cells adjacent to the parietal cells. 4. In the 21-day-old fetuses, the chief cells were identified in the lamina propria and these cells contained granular endoplasmic reticulum and zymogen granules. 5. In the 20- to 22-day-old fetuses, the definitive parietal cells were firstly appeared in pairs or groups within the lamina propria. These cells tended to be placed near the basement membrane and blood capillary, and frequently tended to be extruded into lumen of gland. 6. The endocrine cells were appeared at the basal areas of the gastric gland after the 14-day-old.

  • Gross and histological findings were obtained on cysts formed in the reproductive organs, particularly in the mesosalpinx, mesovarium, and fimbria, of 144 gilts and 37 sows. Of the 181 gilts and sows, 63 (34.8%) were found to have cysts which totaled 89. The number of cysts totaled 38 for the gilts and 51 for the sows, with relative frequencies of 18.1% and 48.6%, respectively. The total number of cysts and the incidence were greater in the sows than in the gilts. With regard to the location of cysts, the highest incidence was found in the mesosalpinx (59.6%). The incidence of cysts in the mesovarium and fimbria was relatively low, with 16.9% and 23.6%, respectively. Most cysts were spherical or oval in shape, and below 5.9mm in diameter. A pedunculated cyst was also seen. The bilateral, incidence of the cysts was lower than unilateral, although the incidence was approxmately the same between the gilts and the sows. With regard to the sexual cycle, the high incidence of cysts was found in the follicular phase than in the luteal phase. The cysts putting pressure upon the oviduct were classified into 3 types according to the shape of epithelium and lamina propria. In type I the epithelium had ciliated cells and the lamina propria formed folds. I type 2 ciliated cells existed, but no folds were formed. In type 3, neither ciliated cells nor folds were seen, the histological change, including partial atrophy and lack of folds in the tubal mucosa were seen.

Morphological study on the development of the tongue in fetuses and neonates of Korean native goat (Capra hircus) (한국재래산양(Capra hircus)의 태아 및 신생아의 혀 발달에 관한 형태학적 연구)

  • This study was carried out to investigate the morphological development of the tongue in the 60-, 90- and 120-day-old fetuses and neonates of Korean native goats by light microscopy. In 60-day-old fetuses, the tongue tissues were differentiated into epithelium, lamina propria, and muscle layer. The primordia of filiform, conical, lentiform, fungiform, and vallate papillae appeared by this time, and rudiments of taste buds were observed in the epithelia of the primordia gustatory papillae. The dorsal lingual epithelia were PAS positive. Collagenous fibers and blood vessels were present in the lamina propria. In 90-day-old fetuses, the gustatory glands were moderately positive for PAS and muscle fibers, and connective tissues were developed. In 120-day-old fetuses, many taste buds were observed in the gustatory papillae. The muscle bundles, collagenous fibers, blood vessels and gustatory glands were well developed. In neonates, over 40 taste buds were found in a transverse section of the vallate papillae. Muscle layers, blood vessels, collagenous fibers and gustatory glands were very well developed.

A morphological study of vomeronasal organ of Korean black goat (Capra aegagrus hircus) (한국흑염소 보습코기관의 형태학적 관찰)

  • The vomeronasal organ (VNO) plays an important role in reproduction and social activities in ruminants including goats. A morphological study on the structure of VNO and its epithelial cells was carried out in Korean black goats. Grossly, the VNO of Korean goats opens into mouth through incisive ducts. Microscopically, the epithelium of VNO consisted of medial sensory epithelium and lateral non-sensory epithelium. Several blood vessels and nerve bundles were observed in the lamina propria encased by vomeronasal cartilage. Immunohistochemical staining showed that protein gene product (PGP) 9.5 was immunostained in the receptor cells of the sensory epithelium and in some cells of the non-sensory epithelium. Galectin-3 was mainly observed in the supporting cells of sensory and non-sensory epithelium. Lectins including wheat germ agglutinin, Ulex europaeus agglutinin, Bandeiraea simplicifolia lectin Isolectin B4, Dolichos biflorus agglutinin and soybean agglutinin used in this study were bound in VNO sensory, non-sensory epithelia as well as in the lamina propria with varying intensity. Collectively, this is a first descriptive morphological study of VNO of Korean black goat with special reference to lectin histochemistry.
