  • A bioflocculant producing bacterium for wastewater treatment was isolated and identified as Lactobacillus jensenii. The bioflocculant was supposed as a polysaccharide. Lactobacillus jensenii YW-33 produced the flocculant with highest activity in the medium composed of 2% sucrose, 0.05% tryptone, 0.5% yeast extract, 0.01% K$_{2}$HPO$_{4}$, 0.01% KH$_{2}$PO$_{4}$, 0.01% NaCl, 0.005% MnSO$_{4}$, 5H$_{2}$O and 0.2% Tween 80. The optimal culture pH and temperature were 7.0 and 25$\circ$C, respectively. In the time course of production with jar fermentor, the productivity of the flocculant was increased in proportion to the cell growth and the cultivation time for maximum production was 24 hrs, showing flocculating activity of 1,130 units per ml.

Rapid Detection of Lactobacillus Genus Inhabiting in Vagina of 20's Healthy Women Using Multiplex PCR (Multiplex PCR 분석을 통한 건강한 20대 여성의 질 내에 서식하는 Lactobacillus 속 유산균의 신속한 검출)

  • The purpose of this study is to simultaneously detect and identify specific genus of Lactobacillus distributed in vagina of healthy women in their twenties by using multiplex primer. Vaginal fluid samples were taken from 166 women who were healthy and did not have vaginosis symptoms caused by bacterial infection. Six species of lactic acid bacteria belong to Lactobacillus genus that are known to inhabit in healthy vagina were selected to make a species-specific multiplex primer. Multiplex primer I was specified to selectively detect L. iners, L. crispatus, L. gasseri and multiplex primer II was specified to selectively detect L. acidophilus, L. jensenii, L. fermentum. L. crispatus (77%) was most frequently detected, and L. acidophilus (57%) and L. jensenii (57%) were relatively higher than others. Although the proportion of L. iners (59%) was a little higher than those of L. acidophilus and L. jensenii, this should be further validated in healthy women's vagina to be sure since they often appear in women having bacterial vaginosis and/or during the antibiotic therapy. Conclusively, the multiplex PCR technique using the species-specific primer could a useful tool to predict variation of vaginal health condition and process of recovery from vaginal disease.

Purification and Characterization of Bioflocculant Producing from $Lactobacillus$ $jensenii$ YW-33 ($Lactobacillus$ $jensenii$ YW-33이 생산하는 생물응집제의 정제 및 특성)

  • The distribution of flocculating activity of culture broth was examined and the major constituent with flocculating activity was identified. Most of flocculating activity was found in culture broth without cells. As the activity was maintained by the digestion with pronase, it suggests that the activity is due to the polysaccharide. The bioflocculant obtained from $Lactobacillus$ $jensenii$ YW-33 was precipitated by 60~80% EtOH fractionation (LJ-80). LJ-80 was separated by ion-exchange chromatography using DEAE-Toyopearl 650C and LJ-80-II showed more potent flocculating activity than those of other fractions. The major activity fraction LJ-80-II was further purified on the gel permeation using Sepharose CL-6B to LJ-80-II-1. GPC (Sepharose CL-6B) and HPLC were used to determine whether LJ-80-II-1 has a homogenecity. The molecular weight of purified LJ-80-II-1 was estimated over 800,000 dalton by gel permeation chromatography. Purified LJ-80-II-1 contained 98.4% total sugar, 0.6% protein. Main sugar of purified LJ-80-II-1 was composed of mannose : galactose : glucose with a molar ratio of 1.61 : 0.25 : 1.00.

Genetic Analysis of Natural Microflora in the Stored Joraengyi Rice Cake and Their Capability of Propionic Acid Production (조랭이 떡에 존재하는 자연균총 유전자 군집분석 및 천연유래 프로피온산 생성능 분석)

  • This study was conducted to analyze the microbial community and propionic acid production ability of natural microflora in the rice cakes. Genetic analysis of natural microflora in Jorangyi rice cake was performed to select propionic acid - producing bacteria. Selected propionic acid-producing bacteria were cultivated in TSB (tryptic soy broth) supplemented with glucose, and growth characteristics were analyzed by temperature and production of propionic acid was analyzed by gas chromatography (GC-FID). Linearity, detection limit, quantitative limit, and recovery rate were measured to verify propionic acid assay. A total of 98 microbial strains were detected from microflora of Joraengyi rice cake that grew after expiration of shelf life. Lactobacillus casei group accounted for 50.48% and Lactobacillus buchneri was 29.60%. Propionic acid - producing bacteria were Propionibacterium thoenii, P. cyclohexanicum, Propionibacterium_uc, P. jensenii, and P. freudenreichii. Natural bacteria and Lactobacillus spp. did not produce propionic acid during 14 days but P. cyclohexanicum, P. freudenreichii subsp. Shermanii, P. thoenii and P. jesenii produced $263.47{\mu}g/mL$, $338.90{\mu}g/mL$, $325.43{\mu}g/mL$ and $222.17{\mu}g/mL$ during 4 days and 2,462.02 and 2,904.78, 2,220.64, $3,519.17{\mu}g/mL$ during 14 days. As a result of this study, it was affirmed that the natural microflora of Joraengyi rice cake during storage can produce propionic acid from natural sources even if a high concentration of propionic acid is not intentionally added. Because of characteristics of rice cake composed of starch and glucose. This study will be used as a recognition criterion to detect natural preservatives such as propionic acid in starchy foods such as rice cakes and as reference standard safety management data.
