Leuconostoc mesenteroides CJNU 0147 and Lactobacillus casei CJNU 0588 Improve Growth of a Bifidobacterium lactis Strain in Co-cultures

  • Previous studies have confirmed that fermented whey produced by Leuconostoc mesenteroides CJNU 0147 or Lactobacillus casei CJNU 0588 display bifidogenic growth stimulator (BGS) activity. The present study sought to determine if the strain itself can improve the growth of bifidobacteria in co-cultures. In reinforced clostridial medium (RCM), both strains stimulated the growth of a Bifidobacterium lactis strain during the exponential phase and also stimulated the growth during almost all growth phases in whey broth. Fermented whey containing viable Leu. mesenteroides CJNU 0147 and L. casei CJNU 0588 cells maintained viability of the B. lactis strain stored at $10^{\circ}C$ in MRS broth. Viable cell count of the B. lactis strain without the fermented whey was decreased to 5.6 log cfu/mL after 15 days, whereas that of the strain with the fermented whey was slightly increased to 7.1 log cfu/mL as compared with initial viable cell count of 6.9 log cfu/mL.

전분분해효소와 유산균에 의한 보리의 유산발효 (Lactic Fermentation of Steamed Barley with an Enzyme and a Lactobacillus)

  • Fermented barley food was produced by the combining action of an enzyme and a lactobacillus. When Lactobacillus sp. L-5 and commercial liquefying amylase from Tae Pyeong Yang Chemical Co. were selected, inoculated on steamed barley and cultivated at 37$^{\circ}C$ for 48hrs, the fermented product of good quality was obtained. In batch cultivation using rotary drum fermentor, viable cell count reached 1.1$\times$10CFU/g after 12hrs' cultivation, and specific growth rate in logarithmic phase was 0.6hr-1. Viable cell count, acidity, pH, concentration of reducing sugar and viscosity of the 48hrs' fermentation product from rotary drum fermentor was 4.3$\times$108CFU/g, 1.17%, 3.1, 10.7% and 1430cp.

Improvement of the Stability of Lactobacillus casei YIT 9018 by Microencapsulation Using Alginate and Chitosan

  • Lactobacillus casei YIT 9018 was microencapsulated within alginate or alginate/chitosan double membrane using an air atomizer. Microbiological analysis revealed that the viability of encapsulated L. casei in gastric juice, hydrogen peroxide, and pepsin was 2-3 log cycle higher than that of the nonencapsulated cell. However, the encapsulated cells did not show a signifciant increase in survival when subjected to in virto high acid and 0.6% bile salt condition. Alginate-encapsulated, alginate/chitosan-encapsulated, and nonencapsulated cells were stored at different temperatures eencapsulated cells showed similar stability at $4^{\circ}C$. However, at $22^{\circ}C$, the alginate/chitosan-encapsulted cell was the most stable.

매실(Prunes mime) 착즙액이 Yoghurt Starter의 산생성 및 증식에 미치는 영향 (The Effect of Maesil(Prunus mime) Extract on the Acid Production and Growth of Yoghurt Starter)

  • Skim milk에 매실 착즙액을 수준별로 첨가하고 4종(Streptococcus thermophilus, Lactobacillus acidophilus, Lactobacillus bulgaricus, Lactobacillus casei)의 젖산균을 단독균주 또는 혼합균주로 접종하여 매실 착즙액의 첨가가 젖산균의 산생성 및 생육에 미치는 영향과 저장성을 조사하였고, 매실 착즙액으로 사용한 매실의 일반 성분을 분석하였다. 매실의 일반 성분은 조회분 0.4%, 식이섬유 4.1%, 구연산 4.66%, 총당 0.264%, vitamin C 405.34mg%이었고, 매실 착즙액의 첨가는 젖산균의 증식을 촉진시켰으며 산생성도 증가하였다. 실험구 중 Str. thermophilus, Lac. acidophilus와 Lac. casei의 혼합균주에 3% 매실 착즙액을 첨가한 실험구가 가장 많은 양의 젖산(1023%)을 생성하였고, 가장 높은 생성균수(3.6$\times$$10^{11}$ cfu/mL)를 나타내었다. 매실 착즙액 3% 첨가 호상 요구르트를 대조구와 함께 4$^{\circ}C$와 2$0^{\circ}C$에서 30일 동안 저장한 결과, 세 개의 혼합균주(Str. thermophilus, Lac. acidophilus)를 사용한 호상 요구르트가 4$^{\circ}C$에서 pH는 4.25, 생균수는 4.1$\times$$10^{9}$ cfu/mL 이었고, 2$0^{\circ}C$에서 pH는 3.76, 생균수는 8.7$\times$$10^{8}$ cfu/mL으로 대조구인 매실 무첨가 호상 요구르트에 비하여 높은 저장성을 나타내었다. 이상의 결과로 보아 매실 착즙액을 첨가한 호상 요구르트의 제조가 가능할 것으로 사료된다.

Continuous Cultivation of Lactobacillus rhamnosus with Cell Recy-cling Using an Acoustic Cell Settler

  • Continuous production of lactic acid from glucose by Lactobacillus rhamnosus with cell recycling using an acoustic cell settler was carried out. The performance of the system, such as the concentration of cell and product were compared with the control experiment without recycling. The acoustic settler showed cell separation efficiency of 67% during the continuous operation and the cell concentration in the fermentor with recycle exceeded that of the control by 29%. Com-pared with the control, tactic acid production was increased by 40%, while glucose consumption was only increased by 8%. The higher value of lactic acid production to substrate consumption (Yp/s, product yield coefficient) achieved by cell recycling is interpreted to indicate that the recycled cell mass consumes less substrate to produce the same amount of product than the control Within system environmental changes due to the longer mean cell residence time induced the cells maintaining the metabolic pathways to produce Less by-Product but more product, lactic acid.

김치유래 Lactobacillus sakei OPK2-59의 ${\gamma}$-Aminobutyric Acid 생성 및 Glutamate Decarboxylase 활성 (${\gamma}$-Aminobutyric Acid Production and Glutamate Decarboxylase Activity of Lactobacillus sakei OPK2-59 Isolated from Kimchi)

  • 김치로부터 분리한 유산균 Lactobacillus sakei OPK2-59는 ${\gamma}$-aminobutyric acid (GABA) 생성능력과 glutamate decarboxylase(GAD) 활성을 보유하고 있음이 확인되었다. Lactobacillus sakei OPK2-59를 59.13 mM과 177.40 mM monosodium glutamate (MSG)가 함유된 MRS 배지에서 배양하면 균주의 성장을 위한 최적 온도범위와 pH는 각각 $25-37^{\circ}C$와 6.5였다. 59.13 mM과 177.40 mM MSG 함유 MRS 배지에서 배양온도 $25^{\circ}C$ 조건에서, 48시간 배양하였을 경우 MSG의 GABA 전환율은 각각 99.58%와 31.00%였다. 또한 Lactobacillus sakei OPK2-59 세포추출액을 이용하여 MSG를 GABA로 전환할 수 있었으며, 추출물에 의한 GABA 전환율은 $30^{\circ}C$, pH 5 조건에서 78.51%로 가장 높았다. 세포추출액에 의한 MSG의 GABA 전환에 미치는 무기염의 영향을 조사한 결과 $CaCl_2$, $FeCl_3$, $MgCl_2$를 첨가한 반응액에서 염을 넣지 않고 반응한 control보다 GABA 전환율이 2-3배 증진되는 것으로 조사되었다. 이러한 결과들은 김치 유산균 Lactobacillus sakei OPK2-59의 GABA 생성능은 유산균 세포 내에 존재하는 GAD에 의한 것이며, GAD에 의한 GABA 전환율은 무기염에 의하여 증진될 수 있음을 제안해 주는 것이다.

Lactobacillus sp.균주를 이용한 산삼 배양근 발효물의 기능성 평가 (Evaluation of Functional Properties of the Tissue Cultured Wild Ginseng Fermented by Lactobacillus sp.)

  • A tissue cultured wild ginseng (TCWG) suspension was inoculated with lactic acid bacteria and fermented to improve the functionality of TCWG. The utilization of TCWG was increased directly using the freeze-dried powder. The optimal ratio of TCWG powder and water for fermentation was 1:19 (5%), which was selected by measuring the fluidity and viable cell count according to concentration. The effects on ADH activation and immune cell activation by each ferments with 10 kinds of Lactobacillus sp. strains were examined. The ferments with the Lactobacillus casei KFRI 692 strain showed 5.4 times higher ADH activity and 1.3 times higher ALDH activity than the non-fermented TCWG powder (control). The level of NO production and cytotoxicity was also measured by Raw 264.7 cells. The ferment with the Lac. casei KFRI 692 strain showed the highest level of NO production and lower cytotoxicity than the others. Therefore, the Lac. casei KFRI 692 strain was selected as a strain for fermentation of a TCWG suspension to maximize its functionality. To identify the optimal fermentation time of the selected Lac. casei KFRI 692 strain on the 5% TCWG suspension, the viable cell count of lactic acid bacterial and the changes in pH were observed for 72 hours. 24-hrs was found to be the optimal fermentation time. In this way, fermented TCWG with lactic acid bacteria showed higher ADH activation efficacy and immune cell activation than non-fermented TCWG.

In vivo Antagonistic Effect of Lactobacillus helveticus CU 631 against Salmonella enteritidis KU101 infection

  • In vivo antagonistic effect of Lactobacillus helveticus CU 631 and Lactobacillus spp. against typical enteritis causing pathogen Salmonella enteritidis KU 101 have been determined, which showed an increase in survival rate and the decline in viable cell numbers of pathogen in liver and spleen at sacrifice. A signifcant difference in the antagonistic effect against KU 101 were observed, which was species and/or strain dependent of Lactobacillus (p<0.01), the survival rate of the mice in the Salmonella infection by feeding L. helveticus CU 631 has been shown to be 157%, whereas those of L. rhamnosus GG ATCC 53103, L. acidophilus ATCC 4356, L. johnsonii C-4 were 137%, 132%, 119% respectively on the basis of lactobacilli non-associated control KU101 fed mice to be 100%. Viable cells of S. enteritidis KU101 in the liver and in the spleen at sacrifice were decreased in Lactobacillus spp. fed group with no significant difference. The higher level of total secretory IgA concentration in the intestinal fluid of lactobacilli fed mice than control mice have been observed. In vitro antagonistic activity of Lactobacillus spp. against KU101 have been determined, a prominent antagonistic activity of CU 631 against KU 101 were demonstrated.

  • Lactobacillus helveticus YM-1 과 Streptococcus thermophilus CH-1을 환원탈지유 배지에 접종하여 생육특성 및 배양조건을 검토한 결과 두 균은 전형적인 공생을 나타냈으며 혼합배양시 최적 pH와 온도는 각각 6.5와 4$0^{\circ}C$였다. 또한 두 균 사이에는 단백질 분해력의 차이가 현저하였고 L.helveticus의 배양여액에는 S. thermophilus의 생육을 촉진시키는 물질이 함유되어 있음을 관찰하였다.

Probiotic균주의 Pathogenic Organism에 대한 억제 활성과 송아지분변 분리균주의 억제활성 특성 (Inhibition Activity Against Pathogenic Organism of Probiotic Bacteria and Characterization of Inhibition Activity of Isolated Bacteria from Calf Dejecta)

  • 본 실험은 Lactobacillus spp.와 Bacillus spp. 의 Salmonella typhimurium, E. coli 및 Listeria monocytogenes에 대한 억제활성과 송아지 분변 분리균주의 생장 및 억제활성의 특성을 알아보기 위해 실시되었다.Lactobacillus spp.와 Bacillus spp.의 병원성 세균 Salmonella typhimurium에 대한 억제활성은 Lactobacillus helveticus CU631이 가장 높았고, Bacillus spp.는 활성이 약하였다. 송아지 분변 분리균주를 동정한 결과 Lactobacillus pentosus CU13과 CU05, Pediococcus pentosaceus CUR02, Lactococcus lactis ssp lactis CUM14로 확인되었다. Lactobacillus rhamnosus CU02와 Lactobacillus pentosus CU13의 Listeria monocytogenes에 대한 억제활성에 영향을 미치는 배지성분과 첨가수준은 Tween 80 1.0%, peptone 3.0%, yeast extract 3.0%, glucose 3.0% beef extract 3.0%, NaCl 1.0~3.0%이며, whole cell과 세포벽 물질은 Listeria monocytogenes에 대하여 억제활성을 나타내었다. 억제 성향을 강하게 보인 균체 배양액을 80℃로 열처리한 결과 억제력은 나타나지 않았으나 catalase 및 Proteinase-K 처리는 억제활성에 영향을 미치지 않았다. 이 결과 억제활성 물질은 유기산에 의한 것으로 사료된다. Lactobacillus pentosus CU13과 Lactobacillus rhamnosus CU02의 21균주에 대한 억제 능력을 측정한 결과 병원성균주를 포함한 16균주에서 억제활성을 보였으나 5균주에서는 억제성향을 나타내지 않았다. Escherichia coli O157:H7을 감염시킨 mouse 중 Lactobacillus pentosus CU13을 접종한 경우 다소 체중 회복 현상이 나타났으나, Lactobacillus rhamnosus CU02을 접종한 경우 체중 회복이 빠르게 나타났다.
