Evidence shows that probiotic bacteria can undergo substantial structural and morphological changes in response to environmental stresses, including antibiotics. Therefore, this study investigated the effects of penicillin G (0.015, 0.03, and 0.06 mg/l) on the morphology and adhesion of Lactobacillus acidophilus ATCC 4356, including the colony morphotype, biofilm production, hydrophobicity, $H_2_O2$ formation, S-layer structure, and slpA gene expression. Whereas only smooth colonies grew in the presence of penicillin, rough and smooth colony types were observed in the control group. L. acidophilus ATCC 4356 was found to be hydrophobic under normal conditions, yet its hydrophobicity decreased in the presence of the antibiotic. No biofilm was produced by the bacterium, despite testing a variety of different culture conditions; however, treatment with penicillin G (0.015-0.06 mg/l) significantly decreased its production of $H_2_O_2$ formation and altered the S-layer protein structure and slpA gene expression. The S-protein expression decreased with 0.015 mg/l penicillin G, yet increased with 0.03 and 0.06 mg/l penicillin G. In addition, the slpA gene expression decreased in the presence of 0.015 mg/l of the antibiotic. In conclusion, penicillin G was able to alter the S-layer protein production, slpA gene expression, and certain physicochemical properties of Lactobacillus acidophilus ATCC 4356.
Statins are a class of lipid-lowering drugs commonly used in the prevention of cardiovascular diseases. However, statin therapy presents many limitations, which have led to an increased interest in non-drug therapies, such as probiotics, to improve blood cholesterol levels. Indeed, probiotic strains such as Lactobacillus acidophilus have been found to improve blood lipid profiles, especially in reducing total cholesterol and LDL-C levels. In this study, we established a high-cholesterol rat model and studied the effect of Lactobacillus acidophilus administration alone or in combination with rosuvastatin. We were able to show that Lactobacillus exerts a cholesterol-lowering effect. Additionally, we observed that when administered together, rosuvastin and Lactobacillus exert a combined cholesterol-lowering effect. Altogether, our data advocate for the possibility of establishing probiotics as non-drug supplements for the treatment of hypercholesterolemia.
건강에 유익한 작용을 하는 프로바이오틱스는 까다로운 미생물로 다양한 환경조건에 매우 민감하여, 이들의 생존율은 항생제, 노화, 스트레스와 식이 등과 같은 요인들에 크게 영향을 받는다. 이에 본 연구에서는 한국 식단에서 많이 이용되고 있는 항균활성이 있는 각종 향신료(마늘, 생강, 파, 양파, 청양고추, 홍고추)와 프로폴리스가 한국인의 장에서 분리한 유산균주의 생존율에 미치는 영향을 조사하였다. 그 결과 대부분의 한국형 유산균주들은 모든 향신료와 프로폴리스에 저항성을 나타냈으며, 심지어 일부 한국형 유산균주(Bifidobacterium lactis, Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus rhamnosus, Streptococcus thermophiles)는 특정 향신료 또는 프로폴리스에 의해 증식률이 증가하였다. 하지만 이와 다르게 대부분의 외래종 유산균주는 다양한 향신료와 프로폴리스에 의해 증식률이 감소 하였으며, 일부 외래종 유산균주(L. helveticus, S. thermophiles)는 특정 향신료에 의해 증식률이 크게 감소 하였다. 마찬가지로 각 향신료와 프로폴리스가 한국형 유산균주만 사용하여 제조된 프로바이오틱스 제품과 외래종 유산균주가 사용된 해외 프로바이오틱스 제품에 미치는 영향을 조사한 결과 한국형 프로바이오틱스 제품은 모두 큰 영향을 받지 않았다. 하지만 해외 프로바이오틱스 제품은 마늘, 양파, 파, 프로폴리스에 의해 크게 감소하는 경향을 나타냈다. 따라서 본 연구결과, 한국인의 장에서 분리한 유산균은 대체적으로 항균활성이 있는 각종 향신료에 저항성을 가지는 생존력이 강한 균주로 판단되며, 한국인의 장에 적합한 유산균으로 사료된다.
Probiotics로서의 사용을 위한 우수 젖산균주 선발을 위해 김치 및 발효유 제품으로부터 분리한 젖산균과 공시 젖산균주를 대상으로 내산성, 내담즙성, 장내 점착능, 병원성균 억제능 등의 probiotics 특성과 장관면역활성, mitogenic activity 및 대식세포 활성화 등의 면역활성을 검토하였다. 내산성 실험의 결과, Lactobacillus acidophilus DDS-1, Lb. acidophilus B-3208, Lb. plantarum과 Leuconostoc mesenteroides ssp. mesenteroides ATCC 8293의 경우 생존률이 50% 이상으로 나타났으며, 특히 Lb. acidophilus DDS-1과 Lb. acidophilus B-3208의 경우에는 70% 정도의 높은 생존률을 나타내었다. 또한 젖산균주의 담즙산 내성에서는 Lactococcus lactis ssp. lactis ATCC 7962를 제외한 모든 균주가 3% oxgall이 함유된 MRS배지에서도 성장이 가능한 것으로 관찰되었다. 젖산균주의 장내 점착능 면에서 볼 때, Lb. acidophilus DDS-1, Lb. acidophilus B-3208과 Bifidobacterium bifidum KCTC 3357의 장내 점착능이 실험에 사용된 다른 젖산균주와 비교하여 우수하다고 관찰되었으며, 특히 Lb. acidophilus DDS-1과 B-3208의 장내점착능이 높게 나타났다. 젖산균주의 병원성균 억제능 실험의 결과, Lb. acidophilus DDS-1, Lb. acidophilus B-3208 및 B. bifidum KCTC 3357은 Staphylococcus aureus를 제외한 Escherichia coli O157:H7 ATCC 43895, Listeria monocytogenes ATCC 59414, 그리고 Salmonella enteritidis ATCC 49313에 대해 높은 증식억제능을 나타내었다. 젖산균의 세포질 획분과 세포벽 획분을 대상으로 면역활성 실험을 실시한 결과, 장관면역 활성은 젖산균의 세포질 획분에서 양성 대조군으로 사용한 LPS와 같은 수준의 면역활성을 나타내었고, 특히 Lb. acidophilus속 균주들의 세포벽 획분의 장관 면역활성이 다른 균주보다 높게 나타났다. 한편 젖산균주의 세포질과 세포벽 성분에 대한 비장 림프구의 증식능은 대조군과 비슷한 수준으로 낮게 관찰되었으나, 이들의 대식 세포증식능은 세포벽 및 세포질 모두의 획분에서 대조군보다 높았으며 특히 세포벽 획분의 경우에는 양성대조군인 LPS 보다 높거나 유사한 정도의 높은 활성을 확인할 수 있었다. 이상의 결과로부터 시험 균주 중에서 Lb. acidophilus 균주인 DDS-1과 B-3208이 프로바이오틱스로서 요구되는 조건을 충족시킨다는 것을 알 수 있었으며, 이들 균주의 상업적 이용 가능성을 재차 확인할 수 있었다.
Lactic acid bacteria were isolated from piglets and chicken and characterized. Lactic acid bacteria showing resistance to low pH and bile, adhesion to intestinal epithelium cells, and the inhibition of Escherichia coli and Salmonella spp. were identified as Lactobacillus acidophilus. L. acidophilus PF01 survived for 2 h in MRS broth adjusted to pH 2. L. acidophilus CF07 was less resistant than L. acidophilus PF01 to pH 2, but survived at pH 2.5 for 2 h. Both of isolates were able to grow in MRS broth containing 0.3% (w/v) bile, with L. acidophilus CF07 being more tolerant to bile than L. acidophilus PF01. L. acidophilus PF01 and CF07 adhered specifically to the duodenal and jejunal epithelium cells of piglet, and the cecal and duodenal epithelium cells of chicken, respectively. Both of isolates did not adhere to the epithelium cells of the various animal intestines from which they were isolated. When L. acidophilus was cultured with E. coli and Salmonella spp. in MRS broth, MRS broth containing 2% skim milk powder or modified tryptic soy broth at $37^{\circ}C$, L. acidophilus PF01 and CF07 inhibited the growths of E. coli K88 and K99, and S. enteritidis and S. typhimurium, respectively. Both of isolates were found to possess the essential characteristics of probiotic lactic acid bacteria for piglet and chicken.
Lactobacillus acidophilus is a gram-positive, microaerophilic, and acidophilic bacterial species. L. acidophilus strains in the gastrointestinal tracts of humans and other animals have been profiled, but strains found in the canine gut have not been studied yet. Our study helps in understanding the genetic features of the L. acidophilus C5 strain found in the canine gut, determining its adaptive features evolved to survive in the canine gut environment, and in elucidating its probiotic functions. To examine the canine L. acidophilus C5 genome, we isolated the C5 strain from a Korean dog and sequenced it using PacBio SMRT sequencing technology. A comparative genomic approach was used to assess genetic relationships between C5 and six other strains and study the distinguishing features related to different hosts. We found that most genes in the C5 strain were related to carbohydrate transport and metabolism. The pan-genome of seven L. acidophilus strains contained 2,254 gene families, and the core genome contained 1,726 gene families. The phylogenetic tree of the core genes in the canine L. acidophilus C5 strain was very close to that of two strains (DSM20079 and NCFM) from humans. We identified 30 evolutionarily accelerated genes in the L. acidophilus C5 strain in the ratio of non-synonymous to synonymous substitutions (dN/dS) analysis. Five of these thirty genes were associated with carbohydrate transport and metabolism. This study provides insights into genetic features and adaptations of the L. acidophilus C5 strain to survive the canine intestinal environment. It also suggests that the evolution of the L. acidophilus genome is closely related to the host's evolutionary adaptation process.
The effects of yogurt supplementation with enzyme-bioconverted ginseng (EBG), ascorbic acid, and yeast extract on the bacterial counts of Streptococcus thermophilus, Lactobacillus acidophilus LA-5, and Bifidobacterium BB-12 were investigated to develop healthy yogurts with high probiotic counts during storage. In addition, the colors and viscosities of the yogurts were determined. EBG, ascorbic acid, and yeast extract did not affect S. thermophilus counts. EBG and ascorbic acid enhanced the viabilities of L. acidophilus LA-5 and Bifidobacterium BB-12 during storage. Yeast extract improved growth of L. acidophilus LA-5 and Bifidobacterium BB-12 during fermentation. EBG turned the yogurt into brown color. We conclude that supplementation of yogurt with EBG, ascorbic acid, and yeast extract may enhance its health-promoting functions by increasing the viability of probiotics, which can thus promote consumption of the yogurt.
Probiotics로서의 활성이 높은 Lactobacillus acidophilus 30SC의 생존성을 증진시키기 위한 기초 자료를 얻고자, heat shock stress를 가한 후 생균수를 측정하고, 생존율의 변화를 통해 고온 처리에 의한 고온 및 냉동 내성의 증진 효과를 평가하였다. 또한 열처리 동안 새로이 발현되는 단백질을 1차원 및 2차원 전기영동을 이용하여 확인하였으며, 주사전자현미경을 사용하여 세포 모양을 관찰하였다. L. acidophilus 30SC는 $55^{\circ}C$의 heat shock stress를 받았을 때 생존 균수가 감소하는 것으로 나타났다. 나머지 처리구는 $37^{\circ}C$에서 계속 배양한 것과 별다른 차이를 나타내지 않았다. 특히 $45^{\circ}C$로 heat shock stress를 준 경우 $37^{\circ}C$에서 배양한 것과 거의 동일하였다. L. acidophilus 30SC에 $45^{\circ}C$로 heat shock stress를 가한 뒤 추가로 55 및 $60^{\circ}C$에 노출시켰을 때 가장 높은 생존율을 나타냈고, 치사 수준인 $55^{\circ}C$의 heat shock stress를 받은 후 $55^{\circ}C$ 및 $60^{\circ}C$에 노출되었을 때 생존율이 급격히 감소하는 경향을 보였다. L. acidophilus 30SC에 $55^{\circ}C$로 15분 Heat shock stress를 준 경우 약 22와 25 kDa의 단백질들이 새로이 발현된 것으로 나타났으나, 24와 27 kDa로 추정되는 단백질의 발현 정도는 낮았음을 확인하였다. 2차원 전기영동을 실시한 결과, $37^{\circ}C$에서 배양한 대조구와 비교할 때 $55^{\circ}C$로 heat shock stress를 준 경우 새로이 5개의 protein spot을 발견할 수 있었다. 주사전자현미경으로 세포의 형태를 관찰한 결과 heat shock stress를 준 경우에는 세포의 길이가 신장되는 경향을 나타내었다.
An experiment was conducted with three hundred and twenty broiler chickens to evaluate the influence of supplementation of probiotic on growth, microbiological status and carcass quality of chickens. The probiotic contained similar proportions of six strains of variable organisms namely Lactobacillus acidophilus, Lactobacillus casei, Bifidobacterium bifidum, Aspergillus oryzae, Streptococcus faecium and Torulopsis sps and was fed at 100 mg/kg diet. The body weight and feed conversion of probiotic fed groups were superior (p<0.05) compared to the control group in the 4th, 5th and 6th weeks. The chickens fed the diet with probiotic had lower (p<0.05) numbers of coliforms and Campylobacter than chickens fed the control diet. All chickens' carcasses on the control diet were positive for Salmonella while only 16 of the 40 carcasses were positive from chickens fed diets containing probiotic. The leg and breast meat of probiotic fed chickens were higher (p<0.05) in moisture, protein and ash, and lower in fat as compared to the leg and breast meat of control chickens.
본 연구는 남은 음식물의 사료화를 목적으로 Lactobacillus acidophilus와 Saccharomyces cerevisiae를 이용하여 남은 음식물을 발효시켜 고품질의 생균사료를 생산하기 위하여 실시하였다. 액상으로 충분히 마쇄시킨 남은 음식물 시료에 공기주입량을 0v.v.m 0rpm, 0.25v.v.m 100rpm, 0.5v.v.m 200rpm과 1v.v.m 500rpm으로 다르게 조절하여 $30^{\circ}C$에서 발효시킨 결과 0.5v.v.m 200rpm의 조건에서 생균수가 $4.5{\times}10^9CFU/m{\ell}$로 가장 높은 것으로 나타났다. 또한 공기주입량을 조절함으로써 최종 발효물의 pH를 조절할 수 있었고 사료로서의 기호성에 알맞고 보존성도 좋은 4-5의 pH범위를 얻을 수 있었다.
