이온도입의 전류밀도에 따른 메틸렌 블루의 경피전달 효과 비교 (Comparison of Current Density on Iontophoretic Transdermal Delivery of Methylene Blue in Rat Skin)

  • PURPOSE: The purpose of this study was to compare the effect of current density on penetration depth, tissue concentration and transdermal transport of methylene blue(MB) by iontophoretic transdermal delivery. METHODS: Twenty-four male Sprague-Dawley rats were randomly divided into 1 mA($0.11mA/cm^2$), 2 mA($0.22mA/cm^2$), 4 mA($0.44mA/cm^2$), and 8 mA($0.89mA/cm^2$) groups. These rats were exposed to anodic iontophoresis of 1% MB using a direct current for 15 minutes. The penetration depth were measured using light microscopy from cryosections of skin tissue. The tissue concentration and transdermal transport were measured using biochemical analysis from target skin tissues. The data were analyzed with one-way analysis of variance. RESULTS: The significant differences in the penetration depth, tissue concentration and transdermal transport were detected among the groups(p<.001). Post hoc comparisons of the penetration depth, tissue concentration and transdermal transport of he 2 mA, 4 mA, and 8 mA iontophoresis groups were greater than in the 1 mA iontophoresis group(p<.05). There was no significant difference, however, among 2 mA, 4 mA, and 8 mA iontophoresis group. CONCLUSION: There was no difference in the efficiency of iontophoresis from 2 mA($0.22mA/cm^2$) to 8 mA($0.89mA/cm^2$). Higher current density can cause skin injury and discomfort sensation. In general, $0.5mA/cm^2$ is proposed to be the maximum iontophoretic current which should be used on human. The appropriate current amplitude should be selected by considering the safety current density and the depth of the target tissue.

  • Moringa oleifera Lam, family Moringaceae, is a perennial plant which is called various names, but is locally known in Malaysia as ''murungai'' or ''kelor''. Glucomoringin, a glucosinolate with from M. oleifera is a major secondary metabolite compound. The seeds and leaves of the plant are reported to have the highest amount of glucosinolates. M. oleifera is well known for its many uses health and benefits. It is claimed to have nutritional, medicinal and chemopreventive potentials. Chemopreventive effects of M. oleifera are expected due to the existence of glucosinolate which it is reported to have the ability to induce apoptosis in anticancer studies. Furthermore, chemopreventive value of M. oleifera has been demonstrated in studies utilizing its leaf extract to inhibit the growth of human cancer cell lines. This review highlights the advantages of M. oleifera targeting chemoprevention where glucosinolates could help to slow the process of carcinogenesis through several molecular targets. It is also includes inhibition of carcinogen activation and induction of carcinogen detoxification, anti-inflammatory, anti-tumor cell proliferation, induction of apoptosis and inhibition of tumor angiogenesis. Finally, for synergistic effects of M. oleifera with other drugs and safety, essential for chemoprevention, it is important that it safe to be consumed by human body and works well. Although there is promising evidence about M. oleifera in chemoprevention, extensive research need to be done due to the expected rise of cancer in coming years and to gain more information about the mechanisms involved in M. oleifera influence, which could be a good source to inhibit several major mechanisms involved in cancer development.

시험환경 내 입자수에 대한 OPC, CNC 장비 간 마스크 밀착 성능비교 (Fit Performance Comparison Between OPC and CNC Based on Number of Ambient Aerosol Particles)

  • Objectives: Fit performance may vary depending on the ambient aerosol number and ratio in respective test environment. Although several instrument can measure it, they differ with respect to the measurement principle and the range of ambient aerosols collected to calculate the fit factor. Methods: In this study, the fit performance between a condensation nuclei counter(CNC) and an optical particle counter(OPC) was compared according to ambient aerosol number concentration, and evaluated consistency. One type respirators(N95 masks) was worn by 50 participants PortaCount®(Pro+ 8038) and MT®(05U) were connected with one probe to one mask, and Fit Factors(FFs) were measured simultaneously. Results: The interclass correlation coefficient of the fit factor and ambient aerosol number, as measured by the two instrument, was 0.82 and 0.79, respectively, indicating a high consistency level. On the other hand there was a significant correlation between the successful test performance of the OPC instrument and the ambient aerosol number. Conclusions: The test was passed with the CNC and OPC instruments when the ambient aerosol number was 635-3,332 particles/cm3 and 368-1,976 particles/cm3, respectively. Thus, the ideal ambient aerosol number of particles differed between the two instrument.

광중합용 DLP 프린터로 제작한 전악 인공치아의 UV-C 초음파 세척에 따른 체적 안정성 평가 (Evaluation of dimension stability according to UV-C ultrasonic cleaning of full arch artificial teeth made with DLP printer for photopolymerization)

  • Purpose: The purpose of this study was to compare the dimension safety evaluation between a general ultrasonic cleaner and an ultrasonic cleaner equipped with UV-C (ultraviolet-C). Methods: An edentulous model was prepared. A denture base and an occlusal rim were fabricated, and scanning was performed. After scanning, a denture base and full arch artificial teeth were designed. The full arch artificial teeth were printed using a three-dimensional printer (n=10). The residual resin was washed with alcohol and then scanned (reference data). The printed specimens were classified and cleaned using a general ultrasonic cleaner (GU group) and an ultrasonic cleaner equipped with UV-C (UC group). After each washing, a rescan was performed (scan data). Reference data and scan data were superimposed using overlapping software. Data were statistically analyzed using the Mann-Whitney test (α=0.05). Results: In the deviation values of full arch artificial teeth, the GU group showed a high deviation of 18.02 ㎛ and the UC group showed a low deviation of 15.02 ㎛. The two groups demonstrated a statistically significant difference (p<0.05). Conclusion: Full arch artificial teeth prepared using photopolymerized resin were deformed according to the temperature of water generated in the ultrasonic cleaner. It is judged that there is no deformation according to the UV-C ultrasonic cleaner.

Identification of Long Non-Coding RNAs and Their Target Genes from Mycelium and Primordium in Model Mushroom Schizophyllum commune

  • Schizophyllum commune has emerged as the most promising model mushroom to study developmental stages (mycelium, primordium), which are two primary processes of fruit body development. Long non-coding RNA (lncRNA) has been proved to participate in fruit development and sex differentiation in fungi. However, potential lncRNAs have not been identified in S. commune from mycelium to primordium developmental stages. In this study, lncRNA-seq was performed in S. commune and 61.56 Gb clean data were generated from mycelium and primordium developmental stages. Furthermore, 191 lncRNAs had been obtained and a total of 49 lncRNAs were classified as differently expressed lncRNAs. Additionally, 26 up-regulated differently expressed lncRNAs and 23 down-regulated between mycelium and primordia libraries were detected. Further, Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that differentially expressed lncRNAs target genes from the MAPK pathway, phosphatidylinositol signal, ubiquitin-mediated proteolysis, autophagy, and cell cycle. This study provides a new resource for further research on the relationship between lncRNA and two developmental stages (mycelium, primordium) in S. commune.

A wireless sensor with data-fusion algorithm for structural tilt measurement

  • Tilt is a key indicator of structural safety. Real-time monitoring of tilt responses helps to evaluate structural condition, enable cost-effective maintenance, and enhance lifetime resilience. This paper presents a prototype wireless sensing system for structural tilt measurement. Long range (LoRa) technology is adopted by the sensing system to offer long-range wireless communication with low power consumption. The sensor integrates a gyroscope and an accelerometer as the sensing module. Although tilt can be estimated from the gyroscope or the accelerometer measurements, these estimates suffer from either drift issue or high noise. To address this challenging issue and obtain more reliable tilt results, two sensor fusion algorithms, the complementary filter and the Kalman filter, are investigated to fully exploit the advantages of both gyroscope and accelerometer measurements. Numerical simulation is carried out to validate and compare the sensor fusion algorithms. Laboratory experiment is conducted on a simply supported beam under moving vehicle load to further investigate the performance of the proposed wireless tilt sensing system.

Robust wireless sensor network configuration design for structural health monitoring with optimal information-energy tradeoff

  • In this paper, a robust wireless sensor network configuration design method is proposed to develop the optimal configuration under the consideration of sensor failure and energy consumption. A malfunctioned sensor in a wireless sensor network may lead to data transmission failure of the entire sensing cluster, inducing severe deterioration in system identification performance. The proposed method determines a wireless sensor network configuration that is robust against sensor failure. By utilizing Bayesian inference, we introduce a robust indicator to evaluate the impact on estimation accuracy of sensor configurations with various malfunctioned sensors. Moreover, a network formation strategy is proposed to optimize the energy efficiency of the wireless sensor network configuration. Therefore, the resultant robust wireless sensor network configuration can operate with the minimum energy consumption while the measurement information of the sensor network with malfunctioned sensors can be guaranteed. The proposed method is illustrated by designing the robust wireless sensor network configurations of a truss model and a bridge model.

Automated assessment of cracks on concrete surfaces using adaptive digital image processing

  • Monitoring surface cracks is important to ensure the health of concrete structures. However, traditional visual inspection to monitor the concrete cracks has disadvantages such as subjective inspection nature, associated time and cost, and possible danger to inspectors. To alter the visual inspection, a complete procedure for automated crack assessment based on adaptive digital image processing has been proposed in this study. Crack objects are extracted from the images using the subtraction with median filter and the local binarization using the Niblack's method. To adaptively. determine the optimal window sizes for the median filter and the Niblack's method without distortion of crack object an optimal filter size index (OFSI) is proposed. From the extracted crack objects using the optimal size of window, the crack objects are decomposed to the crack skeletons and edges, and the crack width is calculated using 4-connected normal line according to the orientation of the local skeleton line. For an image, a crack width nephogram is obtained to have an intuitive view of the crack distribution. The proposed procedure is verified from a test on a concrete reaction wall with various types of cracks. From the crack images with different crack widths and patterns, the widths of cracks in the order of submillimeters are calculated with high accuracy.

Molecular Characterization of Plasmids Encoding CTX-M β-Lactamases and their Associated Addiction Systems Circulating Among Escherichia coli from Retail Chickens, Chicken Farms, and Slaughterhouses in Korea

  • Extended-spectrum β-lactamases (ESBLs), particularly those of the CTX-M types, are the predominant resistance determinants of Escherichia coli that are rapidly spreading worldwide. To determine CTX-M types, E. coli isolates were collected from retail chickens (n = 390) and environmental samples from chicken farms (n = 32) and slaughterhouses (n = 67) in Korea. Fifteen strains harboring blaCTX-M genes were isolated from 358 E. coli isolates. The most common CTX-M type was eight of CTX-M-15, followed by six of CTX-M-1 and one of CTX-M-14. The blaCTX-M genes were identified in the isolates from retail chickens (n = 9), followed by feces, water pipes, floors, and walls. Conjugations confirmed the transferability of the plasmids carrying blaCTX-M genes to the recipient E. coli J53 strain. Furthermore, eight addiction systems carried by the replicons in CTX-M types were confirmed. The dominant system was identified as ccdAB, vagCD, and pndAC in donor strains and transconjugants. The clonal relationship between the two strains carrying blaCTX-M genes indicates that E. coli may transmit from the farm to retail chickens, suggesting a possible public health risk. Our findings demonstrate that the detection of CTX-M types in E. coli isolates is important for tracking ESBL production in animals, and suggest linkage of multiple addiction systems in plasmids bearing blaCTX-M genes.

Enhanced Expression and Functional Characterization of the Recombinant Putative Lysozyme-PMAP36 Fusion Protein

  • The porcine myeloid antimicrobial peptide (PMAP), one of the cathelicidin family members, contains small cationic peptides with amphipathic properties. We used a putative lysozyme originated from the bacteriophage P22 (P22 lysozyme) as a fusion partner, which was connected to the N-terminus of the PMAP36 peptide, to markedly increase the expression levels of recombinant PMAP36. The PMAP36-P22 lysozyme fusion protein with high solubility was produced in Escherichia coli. The final purified yield was approximately 1.8 mg/L. The purified PMAP36-P22 lysozyme fusion protein exhibited antimicrobial activity against both Gram-negative and Grampositive bacteria (Staphylococcus aureus, Salmonella enterica serovar Typhimurium, Pseudomonas aeruginosa, and Bacillus subtilis). Furthermore, we estimated its hemolytic activity against pig erythrocytes as 6% at the high concentration ($128{\mu}M$) of the PMAP36-P22 lysozyme fusion protein. Compared with the PMAP36 peptide (12%), our fusion protein exhibited half of the hemolytic activity. Overall, our recombinant PMAP36-P22 lysozyme fusion protein sustained the antimicrobial activity with the lower hemolytic activity associated with the synthetic PMAP36 peptide. This study suggests that the PMAP36-P22 lysozyme fusion system could be a crucial addition to the plethora of novel antimicrobials.
